Abstract

Systems, apparatus, devices and methods for automated processing of nucleic acids and other samples using a container assembly with a plunger support member comprising at least one plunger extending a distance below the plunger support member, a vessel member comprising at least one vessel residing under the plunger support member, and a flexible impermeable barrier comprising an outer wall with an upper perimeter portion sealably attached to the plunger support member. The outer wall of the flexible barrier merges into a closed bottom surface defining a segment of a bottom of the container assembly. The bottom surface extends laterally a distance to define an inner flexible barrier segment that is sealably attached to the vessel member to define an enclosed interior chamber that resides under the plunger support member inside the flexible barrier with the at least one vessel with the liquid cavity residing inside the interior chamber.
