Abstract

본 발명은 분석 물질의 농축 형태 판단 방법 및 분석 물질의 농축 형태 전환 방법에 관한 것이다. 본 발명의 일 실시예에 따른 분석 물질의 농축 형태 판단 방법은, (a) 임계 이동도 모델을 설립하는 단계 (b) 임계 이동도 모델에 변수값을 적용하여 임계 이동도를 산출하는 단계, 및 (c) 산출된 임계 이동도와, 분석 물질의 전기영동 이동도의 절대값을 비교하여 분석 물질의 농축 형태를 판단하는 단계를 포함하는 것을 특징으로 한다.

분석 물질의 농축 형태 판단 방법 및 분석 물질의 농축 형태 전환 방법
본 발명은 분석 물질의 농축 형태 판단 방법 및 분석 물질의 농축 형태 전환 방법에 관한 것이다. 보다 상세하게는, 물질 내에 존재하는 입자, 미립자 등의 분석 물질을 농축하는 과정에서 농축되는 형태를 미리 판단할 수 있고, 물질 농축 장치의 설계 변수를 조정하여 분석 물질의 농축 형태를 전환할 수 있는 분석 물질의 농축 형태 판단 방법 및 분석 물질의 농축 형태 전환 방법에 관한 것이다.
시료 내에서 생체 물질, 바이오 디젤, 중금속 등의 타겟 물질을 검출하기 위해서는 고가의 검출기를 사용하거나, 시료 준비 단계에서 타겟 물질의 농도를 증폭하는 작업이 선행될 수 있다.
다양한 방식의 농축 방법이 존재하나, 세포 수준의 물질의 경우, 원심 분리 방식이 가장 많이 사용되고 있다. 하지만, 원심 분리 방식의 경우, 강한 회전력에 의해 분리 과정에서 세포가 일정량 파괴되는 문제점이 발생한다. 시료의 양이 증가할수록 낭비되는 세포의 양이 비례해서 증가되기 때문에, 타겟 물질을 비파괴적으로 농축하는 방법이 요구되고 있다. 하나의 예로써, 적혈구를 파괴되는 양을 줄이고 농축되는 양을 늘릴 수 있다면, 환자로부터의 혈액 채취량을 줄일 수 있으며, 환자의 고통을 최소화하면서 정밀한 검사가 가능해질 수 있다.
이온농도분극(Ion Concentration Polarization, ICP) 현상을 이용하여 물질을 농축하는 현상이 학계에 보고되고 있다. 이온농도분극에 의해 형성되는 이온공핍영역(ion depletion zone) 외부의 확산층(diffusion layer)에 존재하는 분석 물질은 유동에 따라 이송되는 이류(advection)와 전기영동 기작으로 인해 발생하는 전기적 이송(electro-migration)에 의해 농축 평형지점(equilibrium positon of preconcentration)이 결정될 수 있다.
확산층의 전반에서 이류가 우세한 경우 또는 전기적 이송이 우세한 경우에 따라 분석 물질이 농축되는 형태는 다르게 나타날 수 있다. 하지만, 전압, 유량, 분석 물질의 전기영동 이동도 등의 임의의 인가 조건과 이에 따른 분석 물질의 농축 형태에 대한 정량적 상관관계는 도출된 바가 없는 실정이다..
본 발명은 상기와 같은 문제점을 포함하여 여러 문제점들을 해결하기 위한 것으로서, 분석 물질의 농축 형태를 미리 판단할 수 있는 분석 물질의 농축 형태 판단 방법을 제공하는 것을 목적으로 한다.
그리고, 본 발명은 분석 물질의 농축 형태를 미리 판단하는 기준값을 마련하여 물질 농축 장치의 설계에 응용할 수 있는 분석 물질의 농축 형태 판단 방법을 제공하는 것을 목적으로 한다.
그리고, 본 발명은 물질 농축 장치의 설계 변수에 따라 농축 형태를 전환하는 분석 물질의 농축 형태 전환 방법을 제공하는 것을 목적으로 한다.

상기 과제를 해결하기 위한 본 발명의 일 관점에 따르면, 물질 농축 장치 내의 분석 물질의 농축 형태를 전환하는 방법으로서, (a) 임계 이동도 모델을 설립하는 단계; 및 (b) 분석 물질의 전기영동 이동도의 절대값을 기준으로 물질 농축 장치의 설계 변수를 변경하는 단계를 포함하고, 상기 설계 변수를 상기 임계 이동도 모델에 적용하여 산출된 임계 이동도가, 상기 분석 물질의 전기영동 이동도의 절대값보다 크면 이류 기작이 우세한 농축 형태가 되고, 상기 분석 물질의 전기영동 이동도의 절대값보다 작으면 전기적 이송 기작이 우세한 형태가 되는, 분석 물질의 농축 형태 전환 방법이 제공된다.

본 발명에 있어서, 물질(시료)이라 함은 마이크로-나노 미터 수준의 크기를 가지는 미립자를 포함하는 물질을 의미한다. 물질은 혈액, 미세조류, 기타 유체 등이 이에 해당될 수 있으며, 이때 물질에 포함된 미립자는 적혈구(red blood cell), 조류세포 등이 이에 해당될 수 있지만, 이러한 예로서 제한되는 것은 아님을 밝혀둔다.
도 1은 본 발명의 일 실시예에 따른 물질 농축 장치(1)를 나타내는 개략도이다.
도 1을 참조하면, 본 발명의 물질 농축 장치(1)는, 이온공핍층(ion depletion layer; 10)과 확산층(diffusion layer; 20)을 포함하는 메인 마이크로 채널(5), 이온 선택성 멤브레인(30) 및 그라운드 마이크로 채널(40)을 포함할 수 있다.
메인 마이크로 채널(5)은 분석 물질(P)이 유입/유출되는 입구/출구가 일단/타단에 구비될 수 있다. 물질이 경로를 따라 이동하기 쉬운 구조를 가지도록, 메인 마이크로 채널(5)은 한 방향으로 길게 형성된 형태를 가질 수 있다.
이온 선택성 멤브레인(30)은 메인 마이크로 채널(10)의 메인 마이크로 채널(10)의 적어도 일면(하면)에 배치될 수 있다. 이온 선택성 멤브레인(30)은 다공성 나노 물질인 나피온(Nafion)을 포함하는 재질일 수 있다. 이 외에도 이온 선택성 멤브레인(30)은 양이온 또는 음이온을 통과시킬 수 있는 재질일 수 있다.
메인 마이크로 채널(5)의 일단/타단에 전기장(VHIGH, VLOW)을 인가하면, 메인 마이크로 채널(5) 내에서 이온 선택성 멤브레인(30)과 인접한 부위에 이온농도분극(Ion Concentration Polarization, ICP) 현상이 발생함으로써 이온공핍층(10)이 형성될 수 있다. 반대로, 이온 선택성 멤브레인(30)과 그라운드 마이크로 채널(40) 사이에는 이온과다층(도면부호 미도시)이 형성될 수 있다.
이온농도분극 현상은 나노막을 갖는 구조 주변에서 관찰되는 전기화학 전달 현상 중의 하나이다. 전기 이중층의 두께가 나노막의 크기와 비슷할 때 나노막 내부에서 전기 이중층이 겹침으로써 단일 이온 투과성을 보인다는 것이 이론적으로 알려져 있다. 벽면 전하와 같은 전하를 갖는 이온들은 확산과 표류력에 의해 나노막을 통과하지 못하고 벽면 전하와 반대 전하를 갖는 이온들만이 통과하게 되면서, 나노막 경계면에서는 이온들의 공핍과 과다 현상이 나타난다.
나노막과 동일한 극성을 가지는 분석 물질(P)이 이온공핍층(10)을 통과하지 못하는 성질을 이용하여, 이온공핍층(10)을 분석 물질(P)의 농축 기작으로 활용할 수 있다. 분석 물질(P)은 이온공핍층(10)과 확산층(20)의 경계(11)를 기점으로, 확산층 내에서 농축될 수 있다.
분석 물질(P)은 유동에 따라 이송되는 이류(advection; A)와 전기영동 기작으로 인해 발생하는 전기적 이송(electro-migration; E)에 의해 농축 평형지점이 결정될 수 있고, 이류(A) 또는 전기적 이송(E)의 우세에 따라 분석 물질(P)의 농축 형태가 다르게 나타날 수 있다.
구체적으로, 분석 물질(P)의 전기영동 이동도의 절대값과 임계 이동도를 비교하여 농축 형태를 판단할 수 있다. 분석 물질(P)의 전기영동 이동도의 절대값이 임계 이동도보다 작은 경우는 이류(A) 기작이 우세하고, 분석 물질(P)의 전기영동 이동도의 절대값이 임계 이동도보다 큰 경우는 전기적 이송(E) 기작이 우세할 수 있다. 임계 이동도보다 작은 전기영동 이동도의 절대값을 갖는 분석 물질(P)은 확산층(20)의 특정 지점 상에서 농축(stacking)될 수 있고, 큰 전기영동 이동도의 절대값을 갖는 분석 물질(P)은 확산층(20) 내에서 농축 평형지점을 변경하면서 저수조 방향(입구/출구 방향)으로 이동(propagating)할 수 있다.
위 상관관계를 정량적으로 도출하기 위한, 본 발명의 물질 농축 장치(1) 내의 분석 물질(P)의 농축 형태 판단 방법은, (a) 임계 이동도 모델을 설립하는 단계(S10), (b) 상기 임계 이동도 모델에 변수값을 적용하여 임계 이동도를 산출하는 단계(S20), 및 산출된 임계 이동도와, 분석 물질의 전기영동 이동도의 절대값을 비교하여 상기 분석 물질의 농축 형태를 판단하는 단계(S30)를 포함하는 것을 특징으로 한다.

도 2는 본 발명의 일 실시예에 따른 확산층(20)에서 이류(A) 기작이 우세한 경우를 나타내는 개략도이다. 도 3는 본 발명의 일 실시예에 따른 확산층(20)에서 전기적 이송(E) 기작이 우세한 경우를 나타내는 개략도이다.

임계 이동도의 수치해석적 증명을 위한 일 실시예로서, 양이온 선택성 멤브레인(30)으로부터 저수조(입구/출구)까지의 길이가 1 X 10-5 m, 1㎛ 깊이, 1㎛ 너비의 메인 마이크로 채널(5)을 이용하였다. 채널 양단에 2.5V, 5V의 비대칭 전압을 인가하여 4.19 X 10-5 A의 이온전류가 흐르고, 2.11 X 10-10 m3/s의 유량이 유도되었다. 이 물질 농축 장치(1)로부터 도출된 임계 이동도(μcr)는 1.88 X 10-8 m2 V-1 s-1이었다.
도 4의 (a)는 임계 이동도(μcr ) 대비 90%의 전기영동 이동도 절대값(μA)을 가지는 입자(P)에 대한 결과이고, 도 4의 (b)는 임계 이동도(μcr ) 대비 110%의 전기영동 이동도 절대값(μA)을 가지는 입자(P)에 대한 결과이다. 상술한 바와 같이, 도 4의 (a)에서는 시간의 흐름에 따라 농축이 계속되면, 농축 평형지점이 확대(stacking; ST)되면서, 분석 물질(P)의 농축량이 증가(50->50')하는 것을 확인할 수 있다. 도 4의 (b)에서는 시간의 흐름에 따라 농축이 계속되면, 농축 평형지점이 저수조 방향(입구/출구 방향)으로 이동(propagating; PR)하면서, 분석 물질(P)의 농축량이 증가(60->60')하는 것을 확인할 수 있다.
도 5는 본 발명의 일 실험예에 따른 물질의 농축 형태를 나타내는 사진이다.
임계 이동도의 실험적 증명을 위한 일 실시예로서, 양이온 선택성 멤브레인(30)으로부터 저수조(입구/출구)까지의 길이가 7 X 10-3 m, 15㎛ 깊이, 200㎛ 너비의 메인 마이크로 채널(5)을 이용하였다. 채널 양단에 15V, 30V의 비대칭 전압을 인가하여 1.01 X 10-8 A의 이온전류가 흐르고, 2.29 X 10-14 m3/s의 유량이 유도되었다. 이 물질 농축 장치(1)로부터 도출된 임계 이동도(μcr)는 2.7 X 10-8 m2 V-1 s-1이었다.
도 5의 (a)는 1.3 X 10-8 m2 V-1 s-1의 전기영동 이동도 절대값을 가지는 sulforhodamine B 염료 분자의 농축 형태를 나타내고, 도 5의 (b)는 3.6 X 10-8 m2 V-1 s-1의 전기영동 이동도 절대값을 가지는 alexa488 염료 분자의 농축 형태를 나타내는 실험 결과이다. 임계 이동도(μcr) 2.7 X 10-8 m2 V-1 s-1보다 작은 전기영동 이동도 절대값을 가지는 도 5의 (a)에서 쌓이는 형태(stacking), 큰 전기영동 이동도 절대값을 가지는 도 5의 (b)에서 전파하는 형태(propagation)가 나타나므로, 도출된 임계 이동도가 농축 형태 판별에 대한 기준값으로 활용될 수 있음을 확인할 수 있다.
도 6은 본 발명의 다른 실험예에 따른 임계 이동도를 활용한 농축 형태를 나타내는 사진이다.
양이온 선택성 멤브레인(30)으로부터 저수조(입구/출구)까지의 길이가 7 X 10-3 m, 15㎛ 깊이, 200㎛ 너비의 메인 마이크로 채널(5)을 이용하였다. 채널 양단에 15V, 30V의 비대칭 전압을 인가하여 1.01 X 10-8 A의 이온전류가 흐르고, 2.29 X 10-14 m3/s의 유량이 유도되었다. 이 물질 농축 장치(1)로부터 도출된 임계 이동도(μcr)는 2.7 X 10-8 m2 V-1 s-1이었다.
도 6의 (a)는 는 1.3 X 10-8 m2 V-1 s-1의 전기영동 이동도 절대값을 가지는 sulforhodamine B 염료 분자의 농축 형태로서, 특정 지점에 농축되는 형태(stacking)를 나타낸다.
도 6의 (b)는 막다른(dead-end) 채널 구조로 설계된 물질 농축 장치(1)이다. 막다른 채널 구조는 입구/출구에 에어를 공급하거나 폐쇄하는 방법으로 구현될 수 있다. 저수조(입구/출구)로의 유로가 차단되어 있으므로, 유량 Q는 0이 된다. (식 20)에 Q=0을 대입하면, 임계 이동도(μcr)는 이온전류 i와 관계없이 0이다. 따라서, sulforhodamine B 염료 분자는 임계 이동도보다 큰 전기영동 이동도 절대값을 가지므로, 농축되며 이동되는 형태(propagation)를 나타낼 것으로 예측되며, 실험 결과 도 6의 (b)에서 이를 만족함을 확인할 수 있다. 또, 막다른 채널 구조는 전기영동 이동도 값에 관계없이 농축되는 형태가 이동되는 형태로 예측할 수 있으므로, 이 형태의 농축이 필요한 경우에는 곧바로 막다른 채널 구조를 적용할 수 있는 이점이 있다.
이를 통해, 동일한 분석 물질을 사용하더라도, 물질 농축 장치(1)의 구동 조건 및 설계를 변경함에 따라 농축 형태를 다르게 도출할 수 있음을 확인할 수 있다.
위와 같이, 본 발명은 임계 이동도 모델에 변수값을 적용하여 분석 물질의 농축 형태를 미리 판단할 수 있는 효과가 있다. 변수값은 전해질(μ+, μ-, D+, D-), 이온전류(I), 유량(Q), 전해질의 벌크농도(c0) 등으로, 물질 농축 장치(1)의 설계, 구동 조건으로서, 임계 이동도 모델에 이를 대입함에 따라 농축 형태간 전환 여부를 미리 판단할 수 있다. 따라서, 이온공핍층(10)을 활용한 물질 농축 장치(1)의 구동 조건을 변경함으로써, 구조적 설계 변경 없이도 분석 물질의 농축 형태를 변환할 수 있다. 예를 들어, 고농도 농축을 위해서는 특정 지점에 농축되는 형태(stacking)로 조건을 설정하고, 농축 물질의 추출을 위해서는 농축되며 이동되는 형태(propagation)로 조건을 설정할 수 있다.
한편, 유도된 임계 이동도에 근거하여, 물질 농축 장치(1)의 설계 변수값을 변경함에 따라, 특정 지점에 농축되는 형태(stacking)에서 농축되며 이동되는 형태(propagation)로 전환할 수 있다. 반대로, 농축되며 이동되는 형태(propagation)에서 특정 지점에 농축되는 형태(stacking)로 전환할 수도 있다.
본 발명은 상술한 바와 같이 바람직한 실시예를 들어 도시하고 설명하였으나, 상기 실시예에 한정되지 아니하며 본 발명의 정신을 벗어나지 않는 범위 내에서 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자에 의해 다양한 변형과 변경이 가능하다. 그러한 변형예 및 변경예는 본 발명과 첨부된 특허청구범위의 범위 내에 속하는 것으로 보아야 한다.

Claims (14)

  1. 물질 농축 장치 내의 분석 물질의 농축 형태를 판단하는 방법으로서,
    (a) 임계 이동도 모델을 설립하는 단계;
    (b) 상기 임계 이동도 모델에 변수값을 적용하여 임계 이동도를 산출하는 단계; 및
    (c) 상기 산출된 임계 이동도와, 분석 물질의 전기영동 이동도의 절대값을 비교하여 상기 분석 물질의 농축 형태를 판단하는 단계
    를 포함하는,
    분석 물질의 농축 형태 판단 방법.
  2. 제1항에 있어서,
    상기 (a) 단계에서, 상기 임계 이동도 모델은,

    로 설립하는, 분석 물질의 농축 형태 판단 방법.
  3. 제1항에 있어서,
    상기 (b) 단계에서 상기 변수값의 적용은, 설계한 상기 물질 농축 장치의 조건을 상기 임계 이동도 모델에 대입하는 것인, 분석 물질의 농축 형태 판단 방법.
  4. 제1항에 있어서,
    상기 물질 농축 장치 내의 상기 분석 물질은 유동에 따라 이송되는 이류(advection)과 전기영동 기작으로 발생하는 전기적 이송(electro-migration)의 합으로 농축 평형지점이 결정되는, 분석 물질의 농축 형태 판단 방법.
  5. 제4항에 있어서,
    상기 (c) 단계에서, 상기 분석 물질의 전기영동 이동도의 절대값이 상기 산출된 임계 이동도보다 작으면, 이류 기작이 우세한, 분석 물질의 농축 형태 판단 방법.
  6. 제5항에 있어서,
    상기 분석 물질의 농축 형태는 소정 지점 상에서 스택킹(stacking)되는 형태인, 분석 물질의 농축 형태 판단 방법.
  7. 제4항에 있어서,
    상기 (c) 단계에서, 상기 분석 물질의 전기영동 이동도의 절대값이 상기 산출된 임계 이동도보다 크면, 전기적 이송 기작이 우세한, 분석 물질의 농축 형태 판단 방법.
  8. 제7항에 있어서,
    상기 분석 물질의 농축 형태는 농축 평형지점이 이동(propagating)되는 형태인, 분석 물질의 농축 형태 판단 방법.
  9. 제1항에 있어서,
    상기 물질 농축 장치는, 상기 분석 물질을 주입하는 입구를 일단에 구비한 메인 마이크로 채널, 및 상기 메인 마이크로 채널의 적어도 일면에 배치되는 이온 선택성 멤브레인을 포함하고,
    상기 물질 농축 장치에 전기장을 인가하면, 상기 메인 마이크로 채널 내에서, 상기 이온 선택성 멤브레인과 인접한 부위에 이온농도분극(ICP; Ion Concentration Polarization) 현상이 발생함으로써 이온공핍영역(ion depletion zone)이 형성되는, 분석 물질의 농축 형태 판단 방법.
  10. 제9항에 있어서,
    상기 이온 선택성 멤브레인은 나피온(Nafion) 재질인, 분석 물질의 농축 형태 판단 방법.
  11. 제9항에 있어서,
    상기 메인 마이크로 채널은 막다른(dead-end) 채널 구조를 가지고, 상기 물질 농축 장치의 임계 이동도는 0인, 분석 물질의 농축 형태 판단 방법.
  12. 물질 농축 장치 내의 분석 물질의 농축 형태를 전환하는 방법으로서,
    (a) 임계 이동도 모델을 설립하는 단계; 및
    (b) 분석 물질의 전기영동 이동도의 절대값을 기준으로 물질 농축 장치의 설계 변수를 변경하는 단계
    를 포함하고,
    상기 설계 변수를 상기 임계 이동도 모델에 적용하여 산출된 임계 이동도가, 상기 분석 물질의 전기영동 이동도의 절대값보다 크면 이류 기작이 우세한 농축 형태가 되고, 상기 분석 물질의 전기영동 이동도의 절대값보다 작으면 전기적 이송 기작이 우세한 형태가 되는, 분석 물질의 농축 형태 전환 방법.
  13. 제12항에 있어서,
    상기 (a) 단계에서, 상기 임계 이동도 모델은,
