본 발명은 물질 비파괴 분리농축 장치 및 물질 비파괴 분리농축 방법에 관한 것이다. 보다 상세하게는, 물질 내에 존재하는 타겟 물질에 작용하는 전단응력을 최소화하여 비파괴적으로 타겟 물질인 입자, 미립자 등의 분리/농축을 수행할 수 있는 물질 비파괴 분리농축 장치 및 물질 비파괴 분리농축 방법에 관한 것이다.
시료 내에서 생체 물질, 바이오 디젤, 중금속 등의 타겟 물질을 검출하기 위해서는 고가의 검출기를 사용하거나 시료 준비 단계에서 타겟 물질의 농도를 증폭하는 작업이 반드시 필요하다.
상대적으로 분자량이 작은 단분자 물질의 경우 다양한 방식의 농축 방법이 존재하고 있으나, 세포 수준의 물질의 경우, 원심 분리 방식이 가장 많이 사용되고 있다. 하지만, 원심 분리 방식의 경우, 강한 회전력에 의해 분리 과정에서 세포가 일정량 파괴되는 문제점이 발생한다. 시료의 양이 증가할수록 낭비되는 세포의 양이 비례해서 증가되기 때문에, 타겟 물질을 비파괴적으로 농축하는 장치 또는 방법이 요구되고 있다. 하나의 예로써, 적혈구를 파괴되는 양을 줄이고 농축되는 양을 늘릴 수 있다면, 환자로부터의 혈액 채취량을 줄일 수 있으며, 환자의 고통을 최소화하면서 정밀한 검사가 가능해질 수 있다.
이온농도분극(Ion Concentration Polarization, ICP) 현상을 이용하여 물질을 농축하는 현상이 학계에 보고되고 있다. 그러나, 이온농도분극 현상을 이용하는 경우 모든 물질들을 농축할 수 있는 것은 아니다. 이온농도분극층 내부에서 전기장이 증폭됨에 따라 발생한 강한 전기대류성 와류(electroconvective vortex)가 물질에 강한 전단응력을 유발하여 파괴시키는 경우가 존재하기 때문이다. 예를 들어, 지질이중층(lipid bilayer) 등의 약한 막에 쌓인 세포들의 경우 강한 전기대류성 와류에 의해 이온농도분극층의 경계면에서 손상 및 파괴가 일어난다. 이는 이온농도분극 현상을 활용한 비파괴 분리/농축에 있어서 한계점으로 작용한다.
본 발명은 상기와 같은 문제점을 포함하여 여러 문제점들을 해결하기 위한 것으로서, 비파괴적으로 타겟 물질인 입자, 미립자 등의 분리/농축을 수행할 수 있는 물질 분리농축 장치 및 물질 분리농축 방법을 제공하는 것을 목적으로 한다.
그리고, 본 발명은 간단한 구조로 구현할 수 있고, 간단한 공정으로, 경제적이며, 대량 생산이 가능한 물질 분리농축 장치 및 물질 분리농축 방법을 제공하는 것을 목적으로 한다.
그러나 이러한 과제는 예시적인 것으로, 이에 의해 본 발명의 범위가 한정되는 것은 아니다.
상기 과제를 해결하기 위한 본 발명의 일 관점에 따르면, 분리/농축을 위한 물질을 주입하는 입구를 일단에 구비한 메인 마이크로 채널; 상기 메인 마이크로 채널의 길이 방향에 평행하도록, 상기 메인 마이크로 채널의 일면에 배치되는 이온 선택성 멤브레인; 상기 메인 마이크로 채널의 타단에서 분기되는 복수의 사이드 마이크로 채널; 상기 메인 마이크로 채널의 타단에 이격되도록 배치되며, 상기 이온 선택성 멤브레인과 적어도 일부가 접하게 배치되는 버퍼 마이크로 채널을 포함하며, 상기 이온 선택성 멤브레인은 상기 메인 마이크로 채널과 전기적으로 병렬 배치되는, 물질 분리농축 장치가 제공된다.
또한, 본 발명의 일 실시예에 따르면, 상기 물질 분리농축 장치에 전기장을 인가하면, 상기 메인 마이크로 채널 내에서, 상기 이온 선택성 멤브레인과 인접한 부위에 이온농도분극(ICP; Ion Concentration Polarization) 현상이 발생함으로써 이온공핍영역(ion depletion zone)이 형성될 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 이온 선택성 멤브레인은 상기 메인 마이크로 채널의 전반에 걸쳐 배치될 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 메인 마이크로 채널의 일면에 접하는 상기 이온 선택성 멤브레인의 부분의 길이는, 상기 메인 마이크로 채널의 길이와 동일하거나 짧고, 상기 이온공핍영역이 형성되는 길이보다 길 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 물질 분리농축 장치에 전기장을 인가하면, 상기 이온공핍영역은 상기 메인 마이크로 채널의 타단에서 일단 방향으로 점점 영역이 커질 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 메인 마이크로 채널 내와 상기 이온공핍영역의 경계면에서 전기장의 크기가 일정하게 유지될 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 이온 선택성 멤브레인은 나피온(Nafion) 재질일 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 물질은 마이크로-나노 크기를 갖는 극성의 미립자를 포함할 수 있다.
그리고, 상기 과제를 해결하기 위한 본 발명의 일 관점에 따르면, (a) 이온 선택성 멤브레인이 길이 방향에 평행하도록 일면에 배치되고, 전기적으로 병렬 배치되는 메인 마이크로 채널을 구비한, 물질 분리농축 장치에 분리/농축을 위한 물질을 공급하는 단계; (b) 상기 물질 분리농축 장치에 전기장을 인가하여, 상기 이온 선택성 멤브레인과 상기 메인 마이크로 채널이 인접하는 소정 부위에 이온농도분극(ICP;Ion Concentration Polarization) 현상이 발생함으로써 이온공핍영역(ion depletion zone)을 형성하는 단계; 및 (c) 상기 이온공핍영역을 기준으로 상기 물질로부터 미립자를 분리농축하는 단계를 포함하는, 물질 분리농축 방법이 제공된다.
또한, 본 발명의 일 실시예에 따르면, 상기 물질 분리농축 장치에 전기장을 인가하면, 상기 이온공핍영역은 상기 메인 마이크로 채널의 타단부에서 일단부 방향으로 점점 영역이 커질 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 물질 내에 포함된 미립자는 상기 이온공핍영역의 경계면에서 전기적 반발력에 의해 밀려나 상기 물질로부터 분리농축 될 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 메인 마이크로 채널 내와 상기 이온공핍영역의 경계면에서 전기장의 크기가 일정하게 유지될 수 있다.
또한, 본 발명의 일 실시예에 따르면, 상기 이온공핍영역의 부근에서 상기 물질에 포함된 미립자에 가해지는 전단응력은, 상기 미립자가 분쇄되는 전단응력보다 작은 세기일 수 있다.
상기한 바와 같이 이루어진 본 발명의 일 실시예에 따르면, 비파괴적으로 타겟 물질인 입자, 미립자 등의 분리/농축을 수행할 수 있는 효과가 있다.
그리고, 본 발명의 일 실시예에 따르면, 간단한 구조로 구현할 수 있고, 간단한 공정으로, 경제적이며, 대량 생산이 가능한 효과가 있다.
물론 이러한 효과에 의해 본 발명의 범위가 한정되는 것은 아니다.

이하에서는, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 본 발명을 용이하게 실시할 수 있도록 하기 위하여, 본 발명의 바람직한 실시예들에 관하여 첨부된 도면을 참조하여 상세히 설명하기로 한다.
본 발명에 있어서, 물질(시료)이라 함은 마이크로-나노 미터 수준의 크기를 가지는 미립자를 포함하는 물질을 의미한다. 물질은 혈액, 미세조류, 기타 유체 등이 이에 해당될 수 있으며, 이때 물질에 포함된 미립자는 적혈구(red blood cell), 조류세포 등이 이에 해당될 수 있지만, 이러한 예로서 제한되는 것은 아니다. 다만, 이하에서는 혈액(물질) 안에 포함된 적혈구[미립자(15)]를 분리농축하는 실시예를 주로 상정하여 설명한다.
도 1은 제1 비교예에 따른 물질 분리농축 장치를 나타내는 개략도, 도 2는 제2 비교예에 따른 물질 분리농축 장치를 나타내는 개략도이다. 그리고, 도 3은 본 발명의 일 실시예에 따른 물질 분리농축 장치를 나타내는 개략도, 도 4는 본 발명의 일 실시예에 따른 물질 분리농축 장치의 사진이다. 제1 비교예와 제2 비교에는 본 발명의 일 실시예와 이온 선택성 멤브레인(40, 50, 60)의 형태, 배치, 크기 면에서 차이가 있고, 그에 의한 물질 분리농축 과정에서 차이를 나타낸다. 이 외의 구성에서, 동일한 참조 부호를 가지는 구성은 동일한 구성이므로, 그 설명이 생략될 수도 있음을 밝혀둔다.
도 3 및 도 4를 참조하면, 본 발명의 물질 분리 농축 장치는 메인 마이크로 채널(10), 복수의 사이드 마이크로 채널(20), 버퍼 마이크로 채널(30) 및 이온 선택성 멤브레인(60)을 포함할 수 있다.
메인 마이크로 채널(10)은 분리/농축을 위한 물질을 주입하는 입구(11)를 일단에 구비할 수 있다. 물질이 경로를 따라 이동하기 쉬운 구조를 가지도록, 메인 마이크로 채널(10)은 한 방향으로 길게 형성된 형태를 가질 수 있다. 메인 마이크로 채널(10)은 약 6,000㎛ 정도의 길이, 약 200㎛ 정도의 너비, 약 15㎛ 정도의 두께를 가지도록 설계될 수 있지만, 이 크기에 제한되는 것은 아니다.
메인 마이크로 채널(10)은 타단에서 복수의 사이드 마이크로 채널(20)로 분기될 수 있다. 본 발명에서는 메인 마이크로 채널(10)과 수직하도록 2개의 사이드 마이크로 채널(20)로 분기되지만 이에 제한되지는 않는다.
버퍼 마이크로 채널(30)은 메인 마이크로 채널(10)의 타단에 소정 간격 이격되도록 배치될 수 있다. 버퍼 마이크로 채널(30)과 메인 마이크로 채널(10)은 직접 연결된 구성은 아니지만, 이온 선택성 멤브레인(40-60)으로부터 간접적으로 연결된 구성일 수 있다. 버퍼 마이크로 채널(30)에는 메인 마이크로 채널(10)에 주입하는 물질과 상응하는 농도를 가진 전해질 수용액을 주입할 수 있다. 버퍼 마이크로 채널(30)은 "ㄷ" 형태를 가지고, 약 6,000㎛ 정도의 총 길이, 약 200㎛ 정도의 채널 너비를 가지도록 설계될 수 있지만, 이 크기에 제한되는 것은 아니다.
이온 선택성 멤브레인(40-60)은 메인 마이크로 채널(10)의 길이 방향과 평행하게, 메인 마이크로 채널(10)의 일면(하면)에 배치될 수 있다. 이온 선택성 멤브레인(40-60)은 다공성 나노 물질인 나피온(Nafion)을 포함하는 재질일 수 있다.
물질 분리농축 장치의 일단과 타단(11, 31)에 전기장을 인가하면, 메인 마이크로 채널(10) 내에서 이온 선택성 멤브레인(40-60)과 인접한 부위에 이온농도분극(Ion Concentration Polarization, ICP) 현상이 발생함으로써 이온공핍영역(ion depletion zone; P, P1, P2, P3, ...)이 형성될 수 있다.
이온농도분극 현상은 나노막을 갖는 구조 주변에서 관찰되는 전기화학 전달 현상 중의 하나이다. 전기 이중층의 두께가 나노막의 크기와 비슷할 때 나노막 내부에서 전기 이중층이 겹침으로써 단일 이온 투과성을 보인다는 것이 이론적으로 알려져 있다. 벽면 전하와 같은 전하를 갖는 이온들은 확산과 표류력에 의해 나노막을 통과하지 못하고 벽면 전하와 반대 전하를 갖는 이온들만이 통과하게 되면서, 나노막 경계면에서는 이온들의 공핍과 과다 현상이 나타난다. 나노막을 통과하지 못한 이온들 사이에서는 강한 전기적인 반발력이 작용하여 양이온과 음이온 모두 영향을 받게 되고, 이에 따라 이온농도구배 현상이 나타난다. 이 때, 이온공핍영역(P) 경계면 주위에서 소용돌이가 형성되고, 전하를 띄고 있는 입자나 세포, 액적들도 이온공핍영역(P)의 경계면에서 이온들의 전기적 반발력에 영향을 받아 밀려나게 된다.
물질(혈액) 안에 포함된 미립자(15)(적혈구)는 마이크로-나노 크기를 가질 수 있다. 물질을 메인 마이크로 채널(10)의 입구에 주입하고, 전기장을 가하면 적혈구(15)들은 압력과 전기삼투 흐름에 의해 이동할 수 있다.
도 1에 도시된 비교예 1에서 이온 선택성 멤브레인(40)은 메인 마이크로 채널(10)의 타단부에서 사이드 마이크로 채널(20)에 겹치지 않을 정도로 매우 짧은 길이(약 100㎛)만큼만 중첩되도록 배치되어 있다. 전기장 인가시 이온공핍영역(P)이 형성될 수 있으나 물질 내의 미립자(15)들을 밀어내어 메인 마이크로 채널(10) 내에 농축할 정도의 전기적 반발력이 생기지는 않으며, 미립자(15)들은 사이드 마이크로 채널(20)을 통해 이동할 수 있다.
도 2에 도시된 비교예 2에서 이온 선택성 멤브레인(50)은 메인 마이크로 채널(10)의 타단부에서 소정 길이(약 1,000㎛)만큼 중첩되도록 배치되어 있다. 전기장 인가시 이온공핍영역(P)이 형성될 수 있고, 도 1과 비교하여 이온공핍영역(P)은 메인 마이크로 채널(10)의 타단부에서 상당히 좌측으로 이동한 정도까지 형성될 수 있다. 이 때, 전기삼투 흐름에 의한 힘과 이온농도분극 현상에 의한 힘에 의해 미립자(15)들이 이온공핍영역(P)의 경계면에서 밀려날 수 있다.
미립자(15)는 이온 공핍층 경계 면에서 발생한 소용돌이로부터 전단응력 τ=Q/A (Q는 전단력, A는 면적)을 받아 분쇄될 수 있고, 미립자(15)가 분쇄되면서 나온 유체, 오일 등이 수용액으로 나올 수 있다. 미립자 분쇄 조각들과 유체는 이온농도분극 현상으로 생긴 이온 공핍층 가장자리에서 정전기적 힘을 받아 밀려나는 힘을 받게 된다. 유체 흐름에 의해 받는 힘 Fdrag=-6πλUα(U는 유속, λ는 유체의 점도 및 α는 입자의 대표반경)에 의해 밀려 들어오고 이온농도분극 현상에 의한 힘 FICP과 전기삼투 흐름에 의한 힘 FEOF의 합에 의해 반대 방향으로 밀려나게 된다.
도 2를 참조하여 다시 설명하면, 전단응력을 받아 적혈구(15)가 분쇄(15')되면서 용혈(h)이 일어날 수 있다. 용혈(h)된 헤모글로빈은 이온 공핍층을 통과할 수도 있다. 그리고, 분쇄된 적혈구(15')들과 용혈(h)된 헤모글로빈들이 이온공핍영역(P)의 경계면에서 밀려나 농축되어 있을 수 있다. 혈액으로부터 적혈구를 분리농축을 할 수 있으나, 큰 전단응력을 받아 분쇄되는 문제점이 나타나는 것이다.
다시 말하면, 물질 분리농축 장치에 전기장을 가하였을 때, 메인 마이크로 채널(10) 내의 유동량과 이온공핍영역(P)을 통과하는 유동량의 차이로 인하여 이온공핍영역(P)의 경계면에서 반대방향으로의 역흐름이 발생하고, 이에 따른 소용돌이가 발생한다. 이 소용돌이가 미립자(15)에 강한 전단응력을 가하여, 미립자(15)를 파괴시킬 수 있다.
한편, 도 3에 도시된 본 발명의 실시예는, 이온 선택성 멤브레인(60)이 메인 마이크로 채널(10)의 전반에 걸친 길이(약 6,000㎛)만큼 중첩되도록 배치되어 있는 것을 특징으로 한다.
도 1 및 도 2와 비교하여 이온공핍영역(P1, P2, P3, ...)은 메인 마이크로 채널(10)의 더욱 좌측으로 이동한 정도까지 형성될 수 있다. 물질 분리농축 장치의 양단에 전기장이 인가되므로, 전기장의 세기에 따라 이온공핍영역(P1, P2, P3, ...)이 좌측으로[메인 마이크로 채널(10)의 타단에서 일반 방향으로] 확장(DP)하면서 점점 영역이 커질 수 있다. 그리고, 인가된 전기장의 세기에 대하여 이온공핍영역(P1, P2, P3, ...)이 확장(DP)되는 한계에 다다르면 영역이 더이상 커지지 않게 된다.
다른 관점으로, 이온 선택성 멤브레인(60)은 메인 마이크로 채널(10)의 길이와 적어도 동일하거나 짧게 형성되고, 이온공핍영역(P1, P2, P3, ...)이 최대화될 수 있는 길이보다는 길게 형성되어, 메인 마이크로 채널(10)의 일면에 접하도록 배치될 수 있다.
물질 분리농축 장치에 전기장을 가하였을 때, 초기에는 메인 마이크로 채널(10) 내의 유동량과 이온공핍영역(P)을 통과하는 유동량을 차이로 인하여 이온공핍영역(P)의 경계면에서 반대방향으로의 역흐름이 발생하고, 이에 따른 소용돌이가 발생한다. 여기까지는 도 2에서 설명한 제2 비교예와 동일하다. 이후에, 이온공핍영역(P)은 확장(DP)을 계속하게 되어 P1, P2, P3와 같이 커질 수 있고, 이온공핍영역(P)의 경계면에서 반대방향으로의 역흐름, 즉, 소용돌이의 세기가 약해지거나 없어질 수 있다. 소용돌이의 전단응력은 미립자(15)가 분쇄되는 전단응력보다 작은 세기를 가지므로, 미립자(15)가 분쇄되지 않고 농축될 수 있다.
도 3를 참조하여 다시 설명하면, 이온공핍영역이 확장(DP)됨에 따라 그 경계면에서 적혈구(15)가 밀려날 수 있다. 그러나, 소용돌이의 전단응력은 적혈구(15)를 분쇄할 정도의 크기가 아니므로, 이온공핍영역이 확장(DP)되어 이동하는 경계면을 따라 적혈구(15)들이 계속 분리농축될 수 있다.
한편, 본 발명의 물질 분리농축 장치는 이온 선택성 멤브레인(60)이 메인 마이크로 채널(10)과 전기적으로 병렬을 이루도록 배치되는 것을 특징으로 한다. 상기와 같이 미립자(15)들이 분쇄되지 않고, 최소한의 전단응력만을 받는 이유는, 이온 선택성 멤브레인(60)이 메인 마이크로 채널(10)과 전기적으로 병렬을 이루도록 배치됨에 따라, 이온공핍영역(P)의 경계면에서 전기장의 크기가 급격하게 일어나지 않고, 일정하게 크기가 유지되며, 전기장에 의해 유도되는 전기삼투 현상이 메인 마이크로 채널(10)에 걸쳐 안정적으로 분포하기 때문이다.
이온 선택성 멤브레인(60)과 메인 마이크로 채널(10)이 전기적으로 병렬을 이룬다는 의미는, 물질 분리농축 장치에 전기장을 인가하여 형성된 전류가 메인 마이크로 채널(10)의 일단에서 타단까지 전달되는 가장 작은 저항의 경로를 따라 이동하는 의미로 이해될 수 있다. 물질 분리농축 장치에서 전류가 전달되는 주된 저항을 구성하는 요소는 메인 마이크로 채널(10), 이온 선택성 멤브레인(40-60) 및 이온공핍영역(P)을 들 수 있다.
도 5는 비교예에 따른 물질 분리농축 장치를 등가회로로 도식화한 개략도이다. 도 5는 도 2의 제2 비교예의 등가회로에 대응할 수 있다.
이온 공핍층 내부(P 경계의 내부)의 이온 농도는 원래의 전해질 농도[메인 마이크로 채널(10)의 농도]에 비해 낮아지기 때문에 해당 영역에서 저항 값은 증가한다(Rm< Rd1
, Rd2). 이온 공핍층이 형성되면서 이온 선택성 멤브레인(50) 바깥 부분[이온 선택성 멤브레인(50)이 중첩되지 않는 부분]에 존재하는 이온 공핍층의 저항값 Rd1이 증가하면서 높은 전압이 Rd1에 인가될 수 있다. 이온 선택성 멤브레인(50)이 중첩되는 부분은 메인 마이크로 채널(10)과 전기적으로 병렬 배치(Rd2, Rn2)될 수 있다. P 경계와 이온 선택성 멤브레인(50) 사이의 매우 짧은 길이에 걸쳐 높은 전압이 인가되며, 이는 큰 크기의 전기장(V/m)이 생김을 의미할 수 있다.
반면, 메인 마이크로 채널(10)에 해당하는 영역인 Rm은 Rd1에 비해 저항의 크기가 낮으므로, 낮은 전압이 긴 길이에 걸쳐 인가되고, 낮은 크기의 전기장이 생길 수 있다. 따라서, 메인 마이크로 채널(10)의 Rm에 대한 전기장의 크기와 이온 공핍층의 Rd1에서의 전기장의 차이가 크게 되어, 전기장에 의해 유도되는 전기대류 현상에 의한 강하고 불안정한 와류가 Rd1 영역에서 형성된다. 그 결과, 물질들이 농축되는 과정에서 큰 전단응력을 받게 되어, 파괴와 손상이 일어날 수 있다.
도 6은 본 발명의 일 실시예에 따른 물질 분리농축 장치를 등가회로로 도식화한 개략도이다.
도 6을 참조하면, 도 5와는 다르게, 이온 선택성 멤브레인(60)이 메인 마이크로 채널(10)의 전반에 걸쳐 병렬 연결을 이룰 수 있다. 이온공핍영역이 확장되어 P1에서 포화된 경우를 상정하여 설명하면, 이온 공핍층이 형성되어 이온 공핍층 내부(P1 경계의 내부)의 이온 농도가 전해질 농도[메인 마이크로 채널(10)의 농도]에 비해 낮아지기 때문에 해당 영역에서 저항 값은 증가한다. Rm< Rd2).
하지만, 병렬 연결된 이온 선택성 멤브레인(60)이 존재하므로, 원래의 이온 공핍층 내부의 저항값 Rd2가 증가하더라도 이온 선택성 멤브레인(60)의 저항값 Rn2의 비에 따라 새로운 전기장의 경로가 형성될 수 있다. 즉, 도 5와 같이, P 경계와 이온 선택성 멤브레인(50) 사이의 매우 짧은 길이에 걸쳐 높은 전압이 인가되는 구간이 없어지게 되므로, P 경계의 바깥에서는 메인 마이크로 채널(10)의 저항 Rm과 이온 선택성 멤브레인(60)의 저항 Rn1에 의한 전기장의 경로가 형성되고, P 경계의 내부에서는 이온 공핍층의 저항 Rd2와 이온 선택성 멤브레인(60)의 저항 Rn2 에 의한 전기장의 경로가 형성될 수 있다.
그 결과, 전압이 메인 마이크로 채널(10)의 Rm에 대한 전기장과 이온 선택성 멤브레인(60)의 Rn2 에 대한 전기장이 긴 길이에 걸쳐 인가되며, 두 영역의 경계면에서 전기장의 크기 변화가 일정한 수준으로 유지될 수 있다. 전기장의 크기 변화가 급격하지 않으므로, 전기장에 의해 유도되는 전기삼투 현상이 메인 마이크로 채널(10)에 걸쳐 안정적으로 분포할 수 있다. 그 결과, 물질들이 농축되는 과정에서 최소한의 전단응력을 받게 되어, 물질(혈액)으로부터 미립자(적혈구; 15)를 비파괴적으로 분리농축 할 수 있다.
물질 분리농축 장치는 투명 재료를 제 1 기판으로 사용할 수 있다. 예를 들면, 제 1 기판으로 파이렉스, 실리콘 이산화물, 실리콘 질화물, 석영 또는 SU-8 중 하나를 사용할 수 있다. 또, 상기 제 1 기판 이외에도 제 2 기판을 포함할 수 있다. 제 2 기판은 물질 분리농축 장치를 커버하거나 실링(seal)하기 위해 이용될 수 있다. 제 2 기판은 제 1 기판과 동일한 재료로 이루어질 수 있다. 일부 실시예들에서 제 1 기판과제 2 기판은 상이한 재료들로 이루어질 수도 있다.
한편, 물질 분리농축 장치의 제조는 제 1 기판을 제 2 기판에 플라즈마 접합하는 것을 통해 완성할 수 있다. 또, 기판은 물질 분리농축 장치의 지지 구조이다. 기판의 적어도 일부분은 실리콘으로 만들어 질 수 있다. 본 발명의 일 실시예에서, 기판 또는 장치의 부분들은 중합체로 만들어질 수 있다. 상기 중합체는 PDMS(polydimethylsiloxane)를 사용할 수 있다. PDMS를 사용할 경우, 친수성(hydrophilic)을 갖도록 산소(O2) 플라즈마 처리될 수 있으나, 경우에 따라서 산소 플라즈마 처리를 생략할 수도 있다.
또한, 물질 분리농축 장치는 물질이 유입되는 입구(11)를 포함하며, 사이드 마이크로 채널(20)의 단부에는 출구(미도시)가 형성될 수 있다. 메인 마이크로 채널(10), 버퍼 마이크로 채널(30) 사이에는 이온 선택성 멤브레인(60)이 배치될 수 있다. 이온 선택성 멤브레인(60)에 유입된 물질 중에서 농축 대상이 아닌 나머지 물질은 사이드 마이크로 채널(20)을 통해 배출될 수 있다. 여기서, 이온 선택성 멤브레인(60)은 예를 들어, 나피온(Nafion)을 사용할 수 있다.
도 7은 제1 비교예에 따른 물질 분리농축 과정을 나타내는 사진이다.
도 7의 (a)를 참조하면, 약 6,000㎛ 의 길이, 약 200㎛ 의 너비를 가지는 메인 마이크로 채널(10)의 타단부에서 사이드 마이크로 채널(20)에 겹치지 않을 정도로 약 100㎛만큼 중첩되게 이온 선택성 멤브레인(40)을 배치하였다. 그리고 메인 마이크로 채널(10)의 입구(11)를 통해 혈액[적혈구(15) 포함]을 주입하였다.
도 7의 (b)를 참조하면, 전기장을 인가하면, 유체 흐름과 전기삼투 흐름에 의해 적혈구(15)들이 이동하는 것을 확인할 수 있다. 도 7의 (c)를 참조하면, 이온 선택성 멤브레인(40)이 메인 마이크로 채널(10)과 중첩된 길이가 짧아 이온공핍영역이 잘 형성되지 않고, 적혈구(15)들이 사이드 마이크로 채널(20)로 빠져나가는 것을 확인할 수 있다.
도 8은 제2 비교예에 따른 물질 분리농축 과정을 나타내는 사진이다.
도 8의 (a)를 참조하면, 약 6,000㎛ 의 길이, 약 200㎛ 의 너비를 가지는 메인 마이크로 채널(10)의 타단부에서 약 1,000㎛만큼 중첩되게 이온 선택성 멤브레인(50)을 배치하였다. 그리고 메인 마이크로 채널(10)의 입구(11)를 통해 혈액[적혈구(15) 포함]을 주입하였다.
도 8의 (b)를 참조하면, 전기장을 인가하면, 유체 흐름과 전기삼투 흐름에 의해 적혈구(15)들이 이동하고, 이온공핍영역이 메인 마이크로 채널(10)의 타단부에서 점차 생성되어 좌측 방향으로 영역이 커지는 것을 확인할 수 있다.
도 8의 (c)를 참조하면, 이온공핍영역이 좌측으로 커지면서, 이온 공핍층 경계면에서 적혈구(15)들이 반발력을 받아 반대 방향으로 밀려나면서 농축되는 것을 확인할 수 있다.
도 8의 (d)를 참조하면, 이온공핍영역이 상당히 좌측으로 커진 후에 더 이상 커지지 않게 되고, 이온 공핍층 경계면에서 적혈구(15)들이 상당히 농축된 상태를 확인할 수 있다.
도 8의 (e)를 참조하면, 이온 공핍층 경계면에서, 적혈구(15)들은 유체 흐름에 의해 받는 힘에 의해 밀려 들어오고, 이온농도분극에 의한 힘에 의해 반대 반향으로 밀려나면서 전단응력을 받는다. 이온 공핍층 경계면과 이온 선택성 멤브레인(50) 사이의 매우 짧은 길이에 걸쳐 큰 전기장이 형성되어 있으므로, 강한 전단응력에 의해 적혈구(15)들이 분쇄(15')되면서 용혈(h)(사진에서 붉은 색 표시)이 일어나는 것을 확인할 수 있다.
도 8의 (f)를 참조하면, 용혈(h)된 헤모글로빈 중 일부는 이온 공핍층을 통과하기도 하고, 분쇄된 적혈구(15')들과 용혈(h)된 헤모글로빈들(사진에서 붉은 색 표시가 두꺼워짐)이 이온공핍영역의 경계면에서 밀려나 농축된 것을 확인할 수 있다.
도 9는 본 발명의 일 실시예에 따른 물질 분리농축 과정을 나타내는 사진이다.
도 9의 (a)를 참조하면, 약 6,000㎛ 의 길이, 약 200㎛ 의 너비를 가지는 메인 마이크로 채널(10)의 타단부에서 약 6,000㎛만큼 중첩되게 이온 선택성 멤브레인(60)을 배치하였다. 그리고 메인 마이크로 채널(10)의 입구(11)를 통해 혈액[적혈구(15) 포함]을 주입하였다.
도 9의 (b)를 참조하면, 전기장을 인가하면, 유체 흐름과 전기삼투 흐름에 의해 적혈구(15)들이 이동하고, 이온공핍영역이 메인 마이크로 채널(10)의 타단부에서 점차 생성되어 좌측 방향으로 영역이 커지는 것을 확인할 수 있다.
도 9의 (c)를 참조하면, 이온공핍영역이 좌측으로 커지면서, 이온 공핍층 경계면에서 적혈구(15)들이 반발력을 받아 반대 방향으로 밀려나면서 농축되는 것을 확인할 수 있다.
도 9의 (d)를 참조하면, 이온공핍영역이 확장(DP)을 계속하게 되어 커지며, 전기장의 세기에 대하여 이온공핍영역이 확장(DP)되는 한계에 다다르면 영역이 더이상 커지지 않게 된다. 이온공핍영역이 한계에 다다르는 크기보다는 적어도 길게, 이온 선택성 멤브레인(60)의 길이를 메인 마이크로 채널(10)의 전반에 걸치도록 구성할 수 있다.
도 9의 (e)를 참조하면, 이온공핍영역이 확장(DP)을 멈추는 부분의 경계면에서 적혈구(15)들이 계속 농축되는 상태를 확인할 수 있다. 이온 공핍층 경계면에서, 적혈구(15)들은 유체 흐름에 의해 받는 힘에 의해 밀려 들어오고, 이온농도분극에 의한 힘에 의해 반대 반향으로 밀려나면서 전단응력을 받는다. 다만, 메인 마이크로 채널(10)과 이온 선택성 멤브레인(60)의 긴 길이에 걸쳐 전압이 낮게 인가되며, 메인 마이크로 채널(10)과 이온 공핍층 경계면 사이에서 전기장의 크기 변화가 일정하므로, 적혈구(15)들을 농축할 수 잇는 최소한의 전단응력만이 가해지게 된다. 그리하여 적혈구(15)들은 파괴되지 않고 혈액으로부터 분리농축 될 수 있다. 이후에 별도의 추출수단을 통해 농축된 적혈구(15)들을 추출할 수 있다.
이처럼 본 발명은, 비파괴적으로 타겟 물질인 입자, 미립자 등의 분리/농축을 수행할 수 있는 효과가 있다. 그리고, 간단한 구조로 분리농축 장치를 구현할 수 있으며, 경제적이고 간단한 공정으로, 대량의 분리농축 공정을 수행할 수 있는 효과가 있다.
본 발명은 상술한 바와 같이 바람직한 실시예를 들어 도시하고 설명하였으나, 상기 실시예에 한정되지 아니하며 본 발명의 정신을 벗어나지 않는 범위 내에서 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자에 의해 다양한 변형과 변경이 가능하다. 그러한 변형예 및 변경예는 본 발명과 첨부된 특허청구범위의 범위 내에 속하는 것으로 보아야 한다.
Claims (13)
- 분리/농축을 위한 물질을 주입하는 입구를 일단에 구비한 메인 마이크로 채널;상기 메인 마이크로 채널의 길이 방향에 평행하도록, 상기 메인 마이크로 채널의 일면에 배치되는 이온 선택성 멤브레인;상기 메인 마이크로 채널의 타단에서 분기되는 복수의 사이드 마이크로 채널;상기 메인 마이크로 채널의 타단에 이격되도록 배치되며, 상기 이온 선택성 멤브레인과 적어도 일부가 접하게 배치되는 버퍼 마이크로 채널을 포함하며,상기 이온 선택성 멤브레인은 상기 메인 마이크로 채널과 전기적으로 병렬 배치되는, 물질 분리농축 장치.
- 제1항에 있어서,상기 물질 분리농축 장치에 전기장을 인가하면, 상기 메인 마이크로 채널 내에서, 상기 이온 선택성 멤브레인과 인접한 부위에 이온농도분극(ICP; Ion Concentration Polarization) 현상이 발생함으로써 이온공핍영역(ion depletion zone)이 형성되는, 물질 분리농축 장치.
- 제1항에 있어서,상기 이온 선택성 멤브레인은 상기 메인 마이크로 채널의 전반에 걸쳐 배치되는, 물질 분리농축 장치.
- 제2항에 있어서,상기 메인 마이크로 채널의 일면에 접하는 상기 이온 선택성 멤브레인의 부분의 길이는, 상기 메인 마이크로 채널의 길이와 동일하거나 짧고, 상기 이온공핍영역이 형성되는 길이보다 긴, 물질 분리농축 장치.
- 제2항에 있어서,상기 물질 분리농축 장치에 전기장을 인가하면, 상기 이온공핍영역은 상기 메인 마이크로 채널의 타단에서 일단 방향으로 점점 영역이 커지는, 물질 분리농축 장치.
- 제2항에 있어서,상기 메인 마이크로 채널 내와 상기 이온공핍영역의 경계면에서 전기장의 크기가 일정하게 유지되는, 물질 분리농축 장치.
- 제1항에 있어서,상기 이온 선택성 멤브레인은 나피온(Nafion) 재질인, 물질 분리농축 장치.
- 제1항에 있어서,상기 물질은 마이크로-나노 크기를 갖는 극성의 미립자를 포함하는, 물질 분리농축 장치.
- (a) 이온 선택성 멤브레인이 길이 방향에 평행하도록 일면에 배치되고, 전기적으로 병렬 배치되는 메인 마이크로 채널을 구비한, 물질 분리농축 장치에 분리/농축을 위한 물질을 공급하는 단계;(b) 상기 물질 분리농축 장치에 전기장을 인가하여, 상기 이온 선택성 멤브레인과 상기 메인 마이크로 채널이 인접하는 소정 부위에 이온농도분극(ICP;Ion Concentration Polarization) 현상이 발생함으로써 이온공핍영역(ion depletion zone)을 형성하는 단계; 및(c) 상기 이온공핍영역을 기준으로 상기 물질로부터 미립자를 분리농축하는 단계를 포함하는, 물질 분리농축 방법.
- 제9항에 있어서,상기 물질 분리농축 장치에 전기장을 인가하면, 상기 이온공핍영역은 상기 메인 마이크로 채널의 타단부에서 일단부 방향으로 점점 영역이 커지는, 물질 분리농축 방법.
- 제10항에 있어서,상기 물질 내에 포함된 미립자는 상기 이온공핍영역의 경계면에서 전기적 반발력에 의해 밀려나 상기 물질로부터 분리농축되는, 물질 분리농축 방법.
- 제9항에 있어서,상기 메인 마이크로 채널 내와 상기 이온공핍영역의 경계면에서 전기장의 크기가 일정하게 유지되는, 물질 분리농축 방법.
- 제12항에 있어서,상기 이온공핍영역의 부근에서 상기 물질에 포함된 미립자에 가해지는 전단응력은, 상기 미립자가 분쇄되는 전단응력보다 작은 세기인, 물질 분리농축 방법.
