Abstract

본 발명은 상기와 같은 문제점을 해결하기 위해 안출된 것으로서, 본 발명자들은 중간엽 기질세포-기반 세포 치료의 유효성를 향상시키기 위하여 예의 노력한 결과, 중간엽 줄기세포 (MSC)의 니치 활성 또는 조혈 줄기세포에 대한 유지 활성은 공여자, 즉 개개인의 차이가 아닌, 체외 배양 조건에 따라 가역적으로 변화하고, 상기 니치 활성은 배양된 중간엽 줄기세포의 CFU-F (colony-forming unit fibroblast)를 통해 예측 가능함을 확인하고, 이에 기초하여 본 발명을 완성하였다.
본 발명의 목적은 니치 활성이 증진된 중간엽 줄기세포의 선별방법을 제공하는데 있다.
본 발병의 다른 목적은 상기 선별된 중간엽 줄기세포를 포함하는 조혈 줄기세포의 자가생산 촉진용 조성물을 제공하는데 있다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기와 같은 본 발명의 목적을 달성하기 위하여, 본 발명은 체외로 분리된 중간엽 줄기세포를 배양하는 단계; 및 상기 배양된 전체 중간엽 줄기세포 중 10% 이상이 CFU-F (colony-forming unit fibroblast)를 형성하는, 중간엽 줄기세포를 선택하는 단계를 포함하는, 니치 활성이 증진된 중간엽 줄기세포의 선별방법을 제공한다.
또한, 본 발명은 체외에서 배양된 중간엽 줄기세포의 CFU-F (colony-forming unit fibroblast) 개수를 평가하는 단계; 및 상기 배양된 전체 중간엽 줄기세포 중 10% 이상이 CFU-F를 형성하는 경우의 배양 조건을 선정하는 단계를 포함하는, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법을 제공한다.
본 발명의 일 구현예로서, 상기 중간엽 줄기세포는 2 내지 5회 계대배양된 것일 수 있다.
본 발명의 다른 구현예로서, 상기 CFU-F는 배양된 줄기세포를 플레이팅한 지 10일 내지 17일 경과 후, 평가되는 것일 수 있다.
본 발명의 또 다른 구현예로서, 상기 중간엽 줄기세포는 인간 지방조직, 골수, 말초혈액 또는 제대혈에서 유래한 것일 수 있다.
본 발명의 또 다른 구현예로서, 상기 니치 활성은 조혈 줄기세포의 미분화능을 유지하고, 자가 재생능을 촉진시키는 것일 수 있다.
또한, 본 발명은 상기 선별된 중간엽 줄기세포를 포함하는 조혈 줄기세포의 자가생산 촉진용 조성물을 제공한다.
본 발명의 일 구현예로서, 상기 조성물은 급성 백혈병, 만성 골수성 백혈병, 골수이형성 증후군, 림프종, 다발성 골수종, 생식세포 종양, 유방암, 난소암, 소세포폐암, 신경아세포종양, 재생불량성빈혈, 적혈구병증, 고셔씨병, 헌터 증후군, ADA 효소결핍증, Wiskot-Aldrich 증후군, 류마티스 관절염, 전신성 홍반성 낭창, 또는 다발성 경화증을 앓고 있거나 항암제 투여 또는 방사선 조사로 인해 조혈 세포가 손상된 환자에게 이식하기 위한 것일 수 있다.
본 발명의 다른 구현예로서, 상기 조성물은 조혈 줄기세포와 함께 이식되며, 상기 조혈 줄기세포는 원시미분화상태의 표지자로서 Lin-Sca-1+c-kit+(LSK)를 갖는 것일 수 있다.
또한, 본 발명은 상기 선별된 중간엽 줄기세포 및 조혈 줄기세포를 공동-배양하는 단계를 포함하는, 조혈 줄기세포의 자가생산 촉진방법을 제공한다.
본 발명에서는 MSC의 니치 활성 차이는 ex-vivo 팽창 단계에서 이루어지며, 조혈 회복에서의 다양한 결과로 이어질 수 있음을 보여주었다. 특히, 이러한 차이는 클론의 이질성으로부터 기인하기 보다는, 고유의 상류 신호 경로에 의해 유도되는 MSC의 기능적 상태로부터 기인하며, 이러한 기능적 상태는 MSC의 CFU-F를 통해 유추할 수 있었다. 따라서, 본 발명은 종래의 중간엽 기질세포-기반 세포 치료의 문제점으로 지적되고 있는 치료 효과의 가변성을 해결함에 기여할 것으로 기대된다.

이하, 본 발명을 상세히 설명하기로 한다.
본 발명은, 체외로 분리된 중간엽 줄기세포를 배양하는 단계; 및 상기 배양된 전체 중간엽 줄기세포 중 10% 이상이 CFU-F (fibroblast colony-forming unit)를 형성하는, 중간엽 줄기세포를 선택하는 단계를 포함하는, 니치 활성이 증진된 중간엽 줄기세포의 선별방법을 제공한다.
또한, 본 발명은 체외에서 배양된 중간엽 줄기세포의 CFU-F (colony-forming unit fibroblast) 개수를 평가하는 단계; 및 상기 배양된 전체 중간엽 줄기세포 중 10% 이상이 CFU-F를 형성하는 경우의 배양 조건을 선정하는 단계를 포함하는, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법을 제공한다.
본 발명에서 사용되는 용어, 중간엽 줄기세포 (mesenchymal stem cell; MSC)는 연골, 뼈, 지방, 골수간질, 근육, 신경 등을 만드는데 원조가 되는 세포로서, 성인에서는 일반적으로 골수에 머물러 있지만 제대혈, 말초혈액, 기타 조직 등에도 존재하며, 이들로부터 수득할 수 있는 세포를 의미한다. 본 명세서에서는, 중간엽 줄기세포는 중간엽 기질세포 또는 기질세포와 같은 의미로 사용된다. 상기 중간엽 줄기세포는 인간(human), 원숭이(monkey), 돼지(pigs), 말(horses), 소(cows), 양(sheep), 개(dogs), 고양이(cats), 생쥐(mice), 토끼(rats) 등의 모든 동물 유래의 세포를 포함하나, 바람직하게는 인간 유래의 세포일 수 있다.
본 발명에서 사용되는 용어, 니치(Niche)는 줄기세포 및 그 외 체세포와 같은 조직세포의 발달 및 증식을 지원하는 조직 또는 기관으로 구성된 구성요소(세포 및/또는 물질)를 의미하며, 세포간 상호작용을 유발하고 전능성을 갖기 위하여 필요한 인자들을 분비하는 것으로 알려져 있다. 니치는 소위 미세환경(microenvironment)이라고도 말하며, 줄기세포의 모든 특징을 표현하는 줄기세포성(stemness)을 유지하는데 있어 핵심적인 역할을 한다. 줄기세포는 점착성 분자 성장 요소(adhesion molecules growth factors)로 이루어진 일종의 미세환경에 자리 잡고 있는바, 학계에서는 이를 니치라고 명명하며, 줄기세포의 이러한 영역은 입지(location), 점착성(adhesiveness), 귀소성(homing), 휴면성(quiescence), 활동성 (activation)을 유지하고 조정하는 역할을 하고 있다. 즉 니치는 줄기세포의 분화를 조정하며, 다른 곳으로의 이동 혹은 세포 자살을 막고 보호해주는 역할을 하는 줄기세포를 둘러싼 주요한 미세환경이라고 볼 수 있다. 특히, 줄기세포 니치 중 활발히 연구가 된 분야는 조혈 모세포(조혈 줄기세포) 니치로, 본원의 조혈모세포 니치는 조혈모세포가 거주하는 곳으로 골수세포의 일종인 조혈모세포는 모든 유형의 혈액 세포로 분화할 수 있는 대신, 자신의 니치 즉, 조혈모세포 니치를 벗어나면 이같은 능력을 제대로 발휘하지 못하는 것으로 알려져 있다. 여타 다른 니치와 마찬가지로 상기 조혈모세포 니치는 단순히 조혈모세포를 위한 안식처일 뿐만 아니라 해당 줄기세포인 조혈모세포의 수까지도 조절하는 것으로 알려져 있다. 본 발명의 목적상, 상기 니치 활성은 조혈 줄기세포의 미분화능을 유지하고, 자가 재생능을 촉진시키는 것을 의미할 수 있다.
본 발명에서 사용되는 용어, "자가 재생 (self-renewal)"은 동일한 성상 및 특징을 갖는 세포를 생산해내는 능력으로 자가복제, 자기복제, 자가재생이라고도 하며, 줄기세포가 가지는 중요한 특징의 하나이다. 특히, 본원 발명에서 자가 재생이란 분화되지 않은 상태를 유지하면서 증식을 계속하는 능력을 의미한다.
한편, 체외 팽창된 중간엽 줄기세포 (MSCs)는 조혈 기능 회복의 파라크라인 (Paracrine) 보조 물질로 널리 이용되어 왔으나, 일관되지 않은 효능을 보여 임상적 적용에 어려움을 겪고 있는 실정이다. 따라서, 중간엽 줄기세포-기반 세포 치료의 유효성을 향상시키기 위하여, 니치 활성이 증진된 중간엽 줄기세포를 선별하는 것은 중요한 기술적 과제이다.
본 발명자들은 체외 팽창된 중간엽 줄기세포의 CFU-F를 니치 활성을 파라미터로 채택하여 (CFU-F 10% 이상), 자극성 또는 비자극성 배지 조건을 분류하고, 이에 따른 효과의 차이를 관찰하였다. 그 결과, 자극성 조건에서 계대배양된 중간엽 줄기세포는 상호 간섭 분자 (Jagged-1 및 CXCL-12)의 발현이 증진되었으며, 중간엽 줄기세포의 조혈 생착 또는 회복 대한 향상 효과는 자극성 조건에서 배양된 중간엽 줄기세포와 조혈 줄기세포를 공동-배양할 때만이 관찰되었다. 특히, 이러한 효과는 배지 조건을 역전시킴에 따라 가역적으로 전환되었는바, 니치 활성의 차이는 클론의 이질성에 의한 것이라기 보다는, 중간엽 줄기세포의 기능적 상태, 즉 배양 조건의 변화 따라 발생하는 것임을 최초로 규명하였다. 실제로, 자극성 조건에서 배양된 중간엽 줄기세포는, 비자극성 조건에서 배양된 경우와 달리, P53의 저해 및 ATF의 활성화와 같은 고유 신호 전달 경로를 지니고 있음을 확인함으로써, 중간엽 줄기세포의 니치활성는 체외 배양 기간 동안의 외재적 요소에 의해 결정되는 것임을 재차 검증하였다. 본 발명은 이러한 실험 결과들을 토대로, 니치 활성이 증진된 중간엽 줄기세포를 선별함으로써, 세포 치료제의 품질을 표준화 및 향상시킬 수 있고, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 새로운 배양 조건을 스크리닝할 수 있다는 점에 기술적 특징이 있다.
본 발명에서, 니치 활성의 정량적인 평가를 위하여, 상기 중간엽 줄기세포는 바람직하게 2 내지 5회, 가장 바람직하게 2회 계대배양시킬 수 있고, 상기 CFU-F의 산출 또는 평가는 배양된 중간엽 줄기세포를 배지에 플레이팅한 지 10일 내지 17일 경과 후, 가장 바람직하게 14일째 이루어질 수 있다.
본 발명에서, 배양 조건은 바람직하게, 배지 첨가되는 보조 물질일 수 있으나, 이 밖에도 물리적 자극, 생리적 변화 (저산소 상태, 특정 인자의 발현 등), 배양 방법의 변화 (3차원 배양) 등을 포함할 수 있다.
본 발명의 다른 양태로서, 상기 선별된 중간엽 줄기세포를 포함하는, 조혈 줄기세포의 자가생산 촉진용 조성물; 상기 선별된 중간엽 줄기세포 및 조혈 줄기세포를 공동-배양하는 단계를 포함하는, 조혈 줄기세포의 자가생산 촉진방법; 및 상기 선별된 중간엽 줄기세포 및 조혈 줄기세포를 개체에 투여하는 단계를 포함하는, 조혈 줄기세포의 자가생산 촉진방법을 제공한다.
본 발명의 조혈 줄기세포의 자가생산 촉진용 조성물 등은 전술한 니치 활성이 증진된 중간엽 줄기세포의 선별방법에서 기술한 바 있는 중간엽 줄기세포 등을 이용하기 때문에, 이들 사이에 공통된 내용은 본 명세서의 과도한 복잡성을 피하기 위하여, 그 기재를 생략한다.
본 발명에서 사용되는 용어, "조혈 줄기세포 (hematopoietic stem cell: HSC)"는 적혈구, 백혈구, 혈소판 등의 혈액 세포를 생산하는 미분화된 골수 조혈세포의 조상세포로서, 골수가 파괴된 숙주에 이식하였을 때, 자가 재생능 (self-renewing capacity)을 가지고 장기간 재증식 (repopulation)하는 능력을 보여준다. 상기 조혈 줄기세포는 인간(human), 원숭이(monkey), 돼지(pigs), 말(horses), 소(cows), 양(sheep), 개(dogs), 고양이(cats), 생쥐(mice), 토끼(rats) 등의 모든 동물 유래의 세포를 포함하나, 바람직하게는 인간 유래의 세포일 수 있다. 일례로서, 상기 조혈 줄기세포는 원시미분화상태의 표지자로서 Lin-Sca-1+c-kit+(LSK)를 갖는 것일 수 있다.
본 발명의 조성물은 조혈 세포가 손상된 생리적 상태에 있는 환자를 대상으로 치료적 학적 유효량의 조혈 줄기세포와 함께 이식된다. 상기 조혈 세포가 손상된 생리적 상태는 급성 백혈병, 만성 골수성 백혈병, 골수이형성 증후군, 림프종, 다발성 골수종, 생식세포 종양, 유방암, 난소암, 소세포폐암, 신경아세포종양, 재생불량성빈혈, 적혈구병증, 고셔씨병, 헌터 증후군, ADA 효소결핍증, Wiskot-Aldrich 증후군, 류마티스 관절염, 전신성 홍반성 낭창, 또는 다발성 경화증을 앓고 있거나, 항암제 투여 또는 방사선 조사로부터 야기될 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예
1. 실험재료 및 실험준비
1-1. 제대혈,
중간엽
줄기세포, 및 체외 배양
서면 동의 하에서 건강한 임산부 공여자로부터 제대혈 (CB)을 수득하였다. 본 연구에서, 서면 동의서 및 모든 실험은 한국 카톨릭대 임상시험심사위원회 (institutional review board)로부터 승인을 받았다 (CUMC11U077). 또한, 한국 카톨릭대 임상시험심사위원회의 승인을 받고 (KC13MDMS0839), 건강한 공여자의 서면 동의 하에서 골수 흡인을 통해 MSC를 수득하였다. 18세 미만의 공여자에 대해서는 공여자 대신, 공여자의 부모로부터 서면 동의서를 받았다 (MC12TNSI0120).
MSC 배양물은 BM 단핵구 세포로부터 구축되었고 (established), 이를 10% 우태아 혈청 (FBS)를 함유하는 Dulbecco's modified Eagle's medium (DMEM)에서 계대배양하였다. 배양 기간 동안 각각 다른 FBS 배치를 구입 및 적용함으로써, MSC에 미치는 영향을 시험하였다. 저 산소 조건 하에서 MSC의 배양은 1%의 O2로 조절된 CO2 수분-자켓형 저산소 배양기 (Thermo Fisher, Heracell 150i, Waltham, MA)에서 이루어졌다.
FBS 배치를 스크리닝하기 위하여, 무작위로 선택된 FBS 배치 7개 및 5개를 각기 다른 공급업체 (Gibco 또는 Hyclone)로부터 수득하였고, 두 세트의 독립적인 스크리닝을 통해 MSC에 미치는 영향을 시험하였다. 모든 FBS 배치는 세포 배양 등급으로, 여과되어(3X, 100nm filter) 내독소, 바이러스 또는 마이코플스마가 없는 상태를 유지하였다. 각각의 다른 배양 조건에서의 팽창 (expand)을 비교하기 위해, 분석 전 MSC를 적어도 2번 계대배양하였다. 배가 시간 (doubling times)은 t/n으로 산출되었고, 여기서 t는 배양기간, n은 n=log (NH-NI)/log2 식으로 산출된 배가된 개체의 수를 나타낸다 (상기 NI는 본래 플레이팅된 세포의 수: NH는 계수할 떄 수득된 세포의 수).

동물 실험은 한국 카톨릭대학교 동물 실험위원회 및 검토 위원회의 승인을 받아 수행되었다. 유사유전자형 이식 (congenic transplantation)에서, C57BL/6J-Ly 5.2 (BL6) 또는 C57BL/6J-Pep3b-Ly5.1 (Pep3b) 마우스를 각각 수용자 또는 공여자로 사용하였다. 5-플루오로우라실 처리 (5-FU BMC)에 의한 마우스 골수 세포의 농축 (enrichment)은 종래의 방법에 의해 수행되었다. 마우스 MSC는, 10% FBS가 첨가된 배지에 부착된 세포를 CD45 음성에 이를 때까지 계대배양시켜 마우스의 골수로부터 수득하였다. 치사적으로 조사된 (900rad) 유사유전자형 수용자 마우스로의 BMC 이식은 종래의 방법에 의해 수행되었다. 공동-이식에 대한 실험은, 마우스에 HSC 및 MSC를 동시-주사하거나 (직접적), 주사 2시간 전 동일한 시험 튜브에서 혼합한 후 (프라이밍), 이의 혼합물을 주사하는 방식으로 수행되었다. 골수의 재증식는 말초 혈액 샘플에서 공여자-유래 CD45.1+ 백혈구 세포 (WBC)의 비율을 측정하여 평가하였다. 재증식된 조혈 세포의 계통은 면역 염색을 통해 분석되었다; 골수 세포 또는 B-림프구 세포 각각을 확인하기 위하여 각각 anti-Mac-1/Gr-1 항체 (BD Pharmingen, San Diego, CA) 및 anti-B220 항체 (BD Pharmingen)를 사용하였다.
이식 9 내지 12주 후, 마우스를 희생시켰고 공여자-유래 세포는 재증식의 수준을 평가하기 위해 사용되었으며, CD45에 대한 항체 (BD Pharmingen), 계통 마커 (StemCell Technologies, Vancouver, BC, Canada), Sca-1-PEcy7(BD Pharmingen), c-kit-APC (eBioscience, San Diego, CA, USA)를 이용한 유세포 분석 (flow cytometry analysis)을 통해 계층화하였다.

분석
MSC 표면 마커에 대한 유세포 분석을 실시하였다. 세포들을 단일 클론 항체, anti-human CD73-PE, CD-34-APC, CD146-PEcy7, CD271-APC, Streptavidin-PEcy7, CD140a-PE (BD Pharmingen), SSEA4-Biotin (R&D Systems, Minneapolis, MN), CD166-FITC (Serotec, Oxford, UK)로 염색하였고, FACSCalibur (Becton Dickinson) 및 CellQuest 소프트웨어를 이용하여 분석하였다. 상호간섭 분자 (cross-talk molecule)의 발현을 조사하기 위하여, MSC를 삼투화시키고, Jagged-1 특이적 항체 (28H8, Cell signaling, Danvers, MA) 또는 CXCL-12 특이적 항체 (79018, R&D Systems)로 세포 내 염색을 실시하였다. 상대적인 발현 수준은 △MFI, 평균 형광 강도의 차이에 의해 확인되었다. MSC의 골 형성 분화 및 지방 세포 분화는 각각의 특이적 분화 배지에서 유도되었고, 알리자린 레드 염색법 및 지방 드럽렛에 의해 정량화하였다. 콜로니 형성 (CFU-F)을 위하여, MSC는 100mm 디쉬 당 500 세포의 밀도로 분주하고, 14일 동안 인큐베이션시켰으며, 메탄올 용액에서 크리스탈 바이올렛으로 염색한 후, 콜로니의 수를 산출하였다.

PCR
RT-PCR 분석을 위하여, RNA는 MSC로부터 정제되었고, 무작위 Hexamer와 SuperScriptTM II (Invitrogen, Carlsbad, CA, USA)를 사용하여 상기 RNA로부터 cDNA를 제조하였으며, Jagged-1 (5'-GTG TCT CAA CGG GGG AAC TT-3' 및 5'-ACA CAA GGT TTG GCC TCA CA-3') 또는 CXCL12 (5'-TCA GCC TGA GCT ACA GAT GC-3' 및 5'-TCA GCC TGA GCT ACA GAT GC-3')의 특이적 프라이머를 사용하여 증폭시켰다. 실시간 정량 PCR (PQ-PCR)은 oter-gene 6000 system (Corbett life science, Australia) 및 SYBR premix Ex taq (Takara, Japan)로 수행되었다. 내인성 GAPDH 대조군으로 정규화시킨 후, PCR 산물의 상대적인 발현 수준을 확인하였다. 각 유전자에 대한 임계 주기 (Ct) 값은 글라이세르 알데하이드-3-포스페이트 디하이드로제나제 (GAPDH)의 임계 주기 값으로 정규화하였다. 상대적인 mRNA 발현은 식 2-△△
Ct 을 이용하여 산출되었으며, 여기서, △Ct = Ct샘플 - Ct△

1-5. 조혈 세포의 정제, 체외 배양, 및 장기 배양
CD34+ 세포는 면역자기 세포 분리 (Dynabeads; Invitrogen, https://www.thermofisher.com)를 사용하여 UCB의 단핵구세포로부터 정제되었다. 상기 세포들을 100ng/ml의 인간 Flt-3 리간드 (Prospec Tany, Rehovot, Israel), 100ng/ml의 인간 SCF (Prospec Tany), 40ng/ml의 인간 IL-6 (R&D Systems), 40ng/ml의 인간 IL-3 (R&D Systems), 및 10-6M의 하이드로코티손 소듐 헤미석신네이트 (Sigma)가 보충된 40ng/ml의 인간 G-CSF (Prospec Tany)를 함유하는 DMEM에 각가 다른 우태아 혈청 배치를 첨가하여 배양하였다. 공동-배양에 사용하기 전, MSC에 조사를 실시한 후(1500 cGy), 유사한 배지 조건에서 CD34+ 세포와 5일 동안 공동-배양하였다. 조혈 전구세포의 콜로니 형성을 분석하기 위하여, 조혈 세포를 사이토카인을 함유하는 반-고체 메틸셀룰로오스 배지 (MethoCult; StemCell Technologies)에서 14일 동안 배양하였으며, 전술한 바와 같이, 콜로니 수 및 계통을 분석하였다. 장기 배양-개시 세포 (LTC-IC) 분석을 위하여, CD34+ 세포를 정상 MSC와 5일간 공동 배양하였고, 6주간 장기 배양 배지로 옮긴 후 반-고체 배지에서 콜로니-형성 어세이를 실시하였다.
1-6. 마이크로 어레이 분석
RNA 추출물은 연속적으로 증폭되고, 올리고뉴클레오타이드 DNA 마이크로어레이에 혼성화되었다. 이중 가닥 DNA 주형은 T7 MEGAscript kit (Ambion, Austin, TX)를 사용하여 T7 RNA 폴리머라제-기반 선형 증폭 프로토콜의 변형인, Eberwine 방법에 의해 증폭되었다. 바이오틴-표지된 cRNA 샘플은 Illumina Human HT-12_V4-BeadChip (48 K) (Illumina, Inc., San Diego, CA)에 혼성화되었다. 어레이를 스캐닝하고, 어레이 실험결과의 처리 및 분석은 Illumina BeadStudio 소프트웨어를 사용하여 수행되었다. 마이크로어레이 연구는 한국 기초 과학 지원 연구원 (Korea Basic Science Institute, MEST)의 Shared Research Equipment Assistance Program에 의해 수행되었다.
계층적 클러스터링은, 평균 관계를 거리 척도로 나타내는 피어슨 상관 계수를 사용하여 수행되었다. 유전자 온토로지 (GO) 프로그램 (http://david.abcc.ncifcrf.gov/)을 사용하여 기능적 하위 그룹의 유전자를 분류하고, 유전자 세트 농축 분석 (gene set enrichment analysis; GSEA)을 수행하였다. 상기 GSEA에서, Kolmogorov-Smirnov 통계는 비-자극 조건에서의 MSC보다 자극 조건에서의 MSC에서 상향-발현되는 유전자에 대한 농축의 유의적 수준을 계산하는데 사용되었다. MSC에서 유전자 전사적 변화를 일으킬 수 있는 상향 조절 유전자 후보를 확인하기 위하여, Ingenuity Pathway Analysis (IPA, Ingenuity Systems, www.ingenuity.com)를 수행하였다.
1-7. 통계 분석
전사체 수준의 유의성을 확인하기 위하여, MSC간 유의성 차이를 t-검정을 이용하여 확인하였다 (p<0.01). 상향 조절된 유전자에서 중첩된 P-값은 Fish's exact test에 의해 산출되었고, 이들은 실험 결과 내 유전자와 조절 유전자에 의해 조절된 유전자 사이의 중첩 유의성을 통계학적으로 측정하는데 사용되었다. 활성화 Z-점수는, 경로 분석 과정에서 상향 조절 유전자가 활성화 되거나 (양의 점수), 억제되는 (음의 점수) 가능성을 조사하고자, 각 조절 유전자의 타겟 신호 강도를 이용하여 측정하였다.

2. 이질적인
CFU
-F에 의한 다른 혈청 조건에서 배양된
MSC의
선별
본 발명자들은, MSC의 줄기세포-유지 활성 (stem cell-supporting activity)에 대한 이질성은 MSC의 ex-vivo 팽창 과정에서 배양 조건의 차이에 따라 발생하는 것으로 전제하였다. 특히, 배양 배지의 보충제로 종래 널리 이용되고 있는 우태아 혈청 (FBS)을 사용하여 MSC의 기능적 이질성에 미치는 영향을 조사하였다.
우선, 본 발명자들은 농축된 CFU-F를 지닌 MSC 하위 집단은 골수에서 니치 세포의 역할을 수행한다는 사실에 기반하여, 배양된 MSC의 이질성에 대한 파라미터로, CFU-F의 빈도를 선정하였다. 여러 우태아 혈청(FBS) 배치에 대한 두 가지의 코호트 검사에서, 많은 (자극성) 또는 적은 (비자극성) CFU-F가 생성될 수 있는 두 쌍의 혈청 배치 (자극성: SS-1 및 SS-2 [총 500개의 중간엽 줄기세포를 플레이팅 한지 14일째 CFU-F가 >50인 경우], 비자극성: NSS-1 및 NSS-2 [총 500개의 중간엽 줄기세포를 플레이팅 한지 14일째 CFU-F가 <50])를 분류하였다 (도 1a).
이러한 자극성 (SS) 또는 비-자극성 (NSS) 혈청 배치는, 7명의 정상 공여자로부터 독립적으로 유래한 MSC에 대한 실험에서, 유의적인 CFU-F의 차이를 발생시켰다 (도 1b). 이러한 결과들은 공여자 개개인 편차보다는 혈청 배치의 차이가 CFU-F에 더 큰 영향을 미침을 나타낸다. 또한, 높은 증식성 콜로니 (large colony) 또는 낮은 증식성 콜로니 (small colony)에 대한 각각 효과를 확인한 결과, 혈청 배치의 차이는 두 유형의 콜로니 모두 상기와 유사한 영향을 미쳤고 (도 1c), 자극성 혈청 조건 하에서 MSC의 배가 시간은 비자극성 조건에 비해 더 짧게 관찰되었는바, 자극성 혈청 조건에서의 MSC가 높은 증식 활성을 보였다 (도 1d). 한편, 표면 표현형을 비교하였을 때, 자극성 조건 하에서 MSC는 비자극성 조건에 비해 CD146+ 세포의 비율이 높게 관찰되었으며, 그 밖에 CD34, CD271, CD140a, SSEA4, 또는 CD73은 유사하게 관찰되었다. (도 1e). 또한, 광학 현미경 상에서는, 자극성 조건 하에서 MSC는 방추형 형태로 나타내었던 반면, 비자극성 조건 하에서 MSC는 보다 평평한 형태를 나타내었고 (도 1f), 유세포 분석에서 전방 측면 산란 (forward/side scatter)으로 평가된 바와 같이, 물리적 특성의 독특한 프로파일을 나타내었다 (도 1g). 그러나, 자극성 또는 비자극성 조건의 차이에 의한, MSC의 골 형성 또는 지방 형성 분화와 관련되 유의적인 차이는 관찰되지 않았다 (도 1h).

3.
MSC의
니치
활성에 따른 혈액학적 회복의 변화 확인
본 실시예에서는, 상기 실시예 2의 자극성 또는 비자극성 조건 하에서 배양된 MSC에 대한, 조혈 줄기세포 (HSCs)의 유지와 관련된 니치 활성 및 이에 따른 조혈 회복 효과를 비교하였다.
3-1. CD34+ 세포와의 공동-배양을 통한 조혈 줄기세포의 유지 활성 확인
우선, 본 발명자들은 in-vivo 및 in-vitro 조건에서 HSC를 유지하는데 중요한 역할을 하는 것으로 알려진 두 개의 상호 간섭 분자, Jagged-1 및 CXCL-12의 발현 수준을 비교하였다. 자극성 조건에서 배양된 MSC는 비자극성 조건에서 배양된 경우보다 높은 Jagged-1 및 CXCL-12의 발현을 나타내었다 (도 2a 및 2b). 상기 MSC에 대한 결과들은, HSC에 대한 니치 상호 간섭 (niche cross-talk) 차이의 가능성을 암시하는 것이다.
이러한 가능성을 조사하기 위하여, 본 발명자들은 각각의 MSC 군 (SS-1,2 또는 NSS-1,2)과 UCB-유래 인간 CD34+ 세포를 5일간 공동-배양한 후, 이들의 HSC 유지 활성을 비교하였다 (도 3a). 자극 조건에서 팽창된 각각의 개별 공여자 유래-MSC (hMSC #1, #2, #7)는 조혈 줄기세포에 대한 높은 유지 활성을 나타내었고, 이는 높은 CFU 수를 통해 확인되었다 (도 3b). 더욱이, 자극성 조건에서 배양된 MSC는, 높은 CD34+90+의 팽창으로 나타낸 바와 같이, 조혈 전구세포의 원시 구획에 대한 높은 유지 활성을 보였고, 장기 SCID-재증식 활성 및 장기 배양 개시 세포 (LTC-IC)의 높은 팽창을 갖는 조혈 세포 하위집단은 6 주간의 장기 배양 후에도 검출되었다 (도 3d). 이와 대조적으로, MSC와의 공동-배양없이 CD34+ 세포만을 단독으로 배양하였을 때, 이러한 배양 조건과 관련된 효과의 차이는 관찰되지 않았다 (도 3b 내지 3d). 즉, 상기 배양 조건의 차이에 따른 효과는, HSC에 직접적으로 영향을 미치기 보다는, MSC 기능의 변화로 유도된 배양물의 차이로부터 유래하는 것임을 나타낸다. 이러한 결과들은, MSC는 배양 조건에 따라 의존적으로 뛰어난 HSC 유지 활성을 발휘할 수 있으며 (자극성 혈청 또는 배지), HSC 자가-재생에 대한 높은 유지 활성은 배양 기간 동안 높은 빈도의 CFU-F (>50)를 나타내는 MSC 배양 조건과 높은 관계가 있음을 보여준다.
3-2. 동물 모델을 이용한 조혈 회복 효과 확인
이러한 결과에 기초하여, 본 발명자들은, 이러한 MSC의 유지 활성의 차이가 MSC 및 HSC의 공동-이식 후에 조혈 회복의 다양한 수준을 평가할 수 있는 인자가 될 수 있는지에 주목하여, 임상적으로 MSC를 공동주입 (이식)하는 상황과 유사하도록 임의로 설계하였다. 이식 후 9 내지 12주에 걸쳐 말초 혈액에서의 생착에 대한 동역학을 평가하고자 유사유전자형 마우스 재증식 모델 (congenic murine repopulation model)을 이용하였다. 상기 마우스는 말초 혈액 내 인간 조혈 세포의 생착을 평가함에 있어서, 마우스 BM 내 사이토카인의 종 특이성으로 인한 이종 동물모델의 한계와 초기에 정체기에 도달하는 공여자-유래 세포의 조혈모 생착에 대한 동역학을 고려하여 제조되었다.
인간 MSC와 마찬가지로, 마우스 BM-MSC에서도 자극성 또는 비자극성 조건에 따라 CFU-F의 수의 유의적인 차이를 관찰하였으며 (도 4a), 자극성 또는 비자극성 혈청 배치에 대한 반응으로서, 자극성 조건에서 배양된 마우스 BM-MSC는 비자극성 조건에서 배양된 경우보다 높은 Jagged-1 및 SDF-1의 발현을 나타내었다 (도 4b 내지 4d). 이어서, 치사적으로 조사된 수용자 마우스에 공여자의 HSC와 함께 자극성 및 비자극성 배양 조건 하에서 배양된 MSC를 공동-이식하였다. 특히, HSC 및 MSCs 혼합물의 세포간 접촉에 의한 영항을 배제하기 위하여, 본 발명자들은, 2 시간 혼합 후 이식하는 방식 (프라이밍) 또는 이러한 전 처리없이 수용자에게 상기 혼합물을 동시에 주사하는 방식 (직접 방식)으로 공동-이식하였으며, 이에 따른 효과를 개별적으로 조사하였다 (도 5a). 비-자극성 조건의 MSC 및 HSCs의 공동-이식은 두 실험동물 (프라이밍 및 직접 방식) 모두에서, HSC 단독 이식에 비해 초기 조혈 생착을 향상시키지 못하였던 반면, 자극성 조건 하에서 팽창된 MSC와 HSC를 공동-이식한 경우, 이식 후 9주 내지 12주 동안의 초기 조혈 회복 단계에서, 두 실험동물 모두 생착 수준이 현저하게 증가되었다 (도 5b). 또한, 이식 전 48시간 동안 저 산소 상태 (1% O2)에 노출된 MSC를 공동-이식에 이용한 경우, 자극성 또는 비자극성 배양 조건에 의한 생착 수준의 변화와 유사한 경향을 보여주었다 (도 5b). 따라서 상기의 생착 향상 효과는 정상 산소 상태 또는 저산소 상태 모두에서 일관성 있게 관찰됨을 알 수 있었다.
주목할 점은, 상기 생착의 향상에 있어서, 공여자-유래 세포의 림프-골수 계통 분포의 현격한 이동이 관찰되지 않았으며, 이는 다계통 재증식 세포의 수준에서 재증식의 증가가 일어남을 나타내는 것이다 (도 5c). 이러한 결과를 뒷받침하는 것으로, 자극성 조건에서 배양된 MSC를 이식한 수용자의 BM에서는, 재증식된 BM에서 다량의 원시 (Lin-Sca-1+c-kit+) 세포에 의해 나타나는 공여자-유래 줄기세포의 수가 더욱 높게 관찰되었다 (도 5D). 즉, 이러한 결과들은 MSC의 니치 활성의 차이를 유도하는 배양 조건은 조혈 회복 및 HSC의 재생의 이질성과 높은 관련이 있음을 보여주는 것이다.

4. 가역적으로 전환되는
MSC의
니치
활성 확인
본 실시예에서는, MSC의 HSC 유지 활성 차이를 배양 조건에 대한 함수로 나타내기 위하여, 도 6a와 같이 실험 조건을 설정하였다. 우선, 본 발명자들은 이러한 MSC의 차이가 두 조건간 클론의 이질성을 갖는 MSC 아집단에 대한 선택적인 팽창에 기인하는 것인지를 조사하였다. 이를 위하여, 본 발명자들은, 다른 배양 조건 사이의 MSC 배양물을 교체하였고, 이들이 MSC의 기능에 미치는 영향을 조사하였다.
초기에 자극성 조건 하에서 성장시킨 MSC는 비 자극성 조건으로 전환됨에 따라 CFU-F는 급격하게 감소되었던 반면, 비자극성에서 자극성 조건으로 교체한 MSC에서는, 높은 증식성 (large) 콜로니를 포함하는 이들의 CFU-F는 다시 증진되었다 (도 6b).
또한, 배양 조건을 전환시킴에 따른 MSC의 HSC에 대한 유지 활성에 미치는 효과를 확인한 결과, 원시적 조혈 세포 집단(CD34+90+)의 팽창에 대한 자극성 또는 비자극성 조건에 의한 차이는 제3의 배지로 교체하였을 때, 완전히 사라졌다 (도 6c). 이와 유사하게, CD34+90+ 팽창에 대한 자극성 또는 비자극성 조건에 의한 효과는, 배양물이 또 다른 조건에 놓였을 때, 가역적으로 전환되었다 (도 6d). 이를 통해, MSC의 니치 활성 및 조혈 회복 효과는 배양 조건의 변화에 따라 가역적으로 전환됨을 알 수 있었다. 아울러, 팽창 기간 내 이러한 MSC의 니치 활성 차이는, 특정 아집단에 대한 선택적 성장에 따른 클론의 이질성에 기인하기 보다는, 배양 조건으로부터 유래한 외재적 인자에 의해 유도된, MSC의 기능적 상태의 차이로부터 유래하는 것임을 보여주었다.

특성을 조절하는 신호

본 실시예에서는, 니치 활성의 차이가 MSC의 기능적 상태에 의해 야기될 수 있는지 추가적으로 확인하고자, 자극성 또는 비자극성 조건에서 배양된 3개의 독립적인 MSC를 대상으로, 마이크로 어레이를 실시하여 유전자 발현 프로파일을 비교하였다.
총 47,323개의 유전자 중 785개의 유전자가 2개의 MSC 그룹 사이에서 유의적인 발현 차이를 보였다 (도 7a). 이러한 유전자의 발현 차이를 유전자 집합 농축 분석 (GSEA)에 의해 기능적 주석으로 분석한 결과, 유전자 온톨로지 (GO) 카테고리 중 15개가 상향 조절되었고, 4개의 GO 카테고리는 비자극성 MSC에 비해 자극성 MSC에서 하향 조절되었다 (도 7b 내지 7g, FDR<25%). 이러한 결과는 두 개의 MSC 그룹은 실제로 별개의 생물학적 기능을 위한 고유의 신호 전달 경로 하에 있음을 나타내는 것이다.
전사체 차이를 일으킬 수 있는 상향 조절 인자를 확인하기 위하여, 본 발명자들은, 차별적으로 발현된 유전자 세트를 대상으로 경로 분석 (IPA, Ingenuity Systems, www.ingenuity.com)을 실시하였다. 그 결과, 하류 표적에 매우 중요한 변화를 나타내는 5개의 상류 신호 전달 경로를 확인하였으며, 그 결과는 표 1에 나타내었다.

상기 표 1에 나타낸 바와 같이, tumor-growth factor- signals (TGF-1)의 다중화와 함께, p53 및 tribbles pseudokinase 3 (TRIB3)의 저해, RAS oncogene family-like 6 (RABL6) 및 activating transcription factor 4 (ATF4)의 활성화를 확인하였다. 이러한 결과는, 자극성 또는 비자극성 조건에서 배양된 MSC는 다른 니치 활성을 부여할 수 있는 고유의 신호 전달 경로 하에 있음을 나타내는 것이다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.

- 제5항에 있어서, 상기 조성물은 급성 백혈병, 만성 골수성 백혈병, 골수이형성 증후군, 림프종, 다발성 골수종, 생식세포 종양, 유방암, 난소암, 소세포폐암, 신경아세포종양, 재생불량성빈혈, 적혈구병증, 고셔씨병, 헌터 증후군, ADA 효소결핍증, Wiskot-Aldrich 증후군, 류마티스 관절염, 전신성 홍반성 낭창, 또는 다발성 경화증을 앓고 있거나 항암제 투여 또는 방사선 조사로 인해 조혈 세포가 손상된 환자에게 이식하기 위한 것인, 조혈 줄기세포의 자가생산 촉진용 조성물.
- 제5항에 있어서, 상기 조성물은 조혈 줄기세포와 함께 이식되는 것인, 조혈 줄기세포의 자가생산 촉진용 조성물.
- 제7항에 있어서, 상기 조혈 줄기세포는 원시미분화상태의 표지자로서 Lin-Sca-1+c-kit+(LSK)를 갖는 것인, 조혈 줄기세포의 자가생산 촉진용 조성물.
- 체외에서 배양된 중간엽 줄기세포의 CFU-F (colony-forming unit fibroblast) 개수를 평가하는 단계; 및체외에서 배양된 전체 중간엽 줄기세포 중 10% 이상이 CFU-F를 형성하는 경우의 배양 조건을 선정하는 단계를 포함하는, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법.
- 제9항에 있어서, 상기 중간엽 줄기세포는 2 내지 5회 계대배양된 것인, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법.
- 제9항에 있어서, 상기 CFU-F 개수를 평가하는 단계는 상기 배양된 줄기세포를 배지에 플레이팅한 지 10일 내지 17일 경과 후, 평가하는 것인, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법.
- 제9항에 있어서, 상기 중간엽 줄기세포는 인간 지방조직, 골수, 말초혈액 또는 제대혈에서 유래한 것인, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법.
- 제9항에 있어서, 상기 니치 활성은 조혈 줄기세포의 미분화능을 유지하고, 자가 재생능을 촉진시키는 것인, 중간엽 줄기세포의 니치 활성을 증진시키기 위한 배양 조건의 스크리닝 방법.
