Abstract

본 발명은 하이드로젤을 이용한 교차 전극 바이오센서에 관한 것으로서, 기판상에 빗(Comb) 모양으로 복수의 제 1 돌출 전극이 배열되도록 구성된 제 1 교차 전극, 제 1 교차 전극과는 마주하여 기판상에 빗 모양으로 복수의 제 2 돌출 전극이 배열되도록 구성되되 복수의 제 2 돌출 전극들이 제 1 교차 전극에 형성된 복수의 제 1 돌출 전극들과 각각 교차 배치되도록 구성된 제 2 교차 전극, 및 서로 교차하여 배치된 제 1 교차 전극과 제 2 교차 전극의 사이 공간에 채워진 하이드로젤을 포함하는바, 단백질 등과 같은 생체물질의 유무 및 그 농도를 각 교차 전극 사이의 임피던스 측정을 통해 검출함에 있어 각각의 교차 전극 간에 하이드로젤이 구비되도록 하여 임피던스 검출 폭과 검출 한계를 수십 내지 수백배 이상 늘이고 검출 정확성을 향상시킬 수 있는 효과가 있다.

Description

하이드로젤을 이용한 교차 전극 바이오센서
본 발명은 교차 전극 바이오센서에 관한 것으로서, 특히 단백질 등과 같은 생체물질의 유무 및 그 농도를 각 교차 전극 사이의 임피던스 측정을 통해 검출함에 있어 각각의 교차 전극 간에 하이드로젤이 구비되도록 하여 임피던스 검출 폭과 검출 한계를 수십 내지 수백배 이상 늘이고 검출 정확성을 향상시킬 수 있도록 한 하이드로젤을 이용한 교차 전극 바이오센서에 관한 것이다.
최근 들어 유전자, 단백질 등의 다양한 생체물질의 유무 및 그 농도를 전기적 방법으로 검출하는 바이오센서가 많이 개발되고 있다. 그 하나의 예가 교차전극(interdigitated microelectrode)을 이용하는 것이다. 이는 생체물질과 특이적 결합을 하는 수용체가 고정되는 영역이 지그재그 형태로서 실질적으로 매우 넓기 때문에 생체물질의 농도가 낮더라도 측정이 제대로 이루어진다는 평가를 받고 있다.
대한민국 등록특허 제777973호(2007.11.29.공고)에 개시된 내용이 이와 같은 교차전극을 이용하는 것이다. 그러나 상기 등록특허의 경우에는 전극 사이에 흐르는 전류를 통하여 농도 측정이 이루어지기 때문에, 전극 사이에 전류가 통하도록 도전성입자를 별도로 이용해야 하는 번거로움이 있다.
또한, 상기 등록특허의 경우에는 임피던스에 영향을 주는 전기장(Electric filed)이 각각의 전극 사이에서 발생하는 양보다 전극의 위쪽으로 빠져나가는 양이 더 많은 문제가 있었다. 다시 말해, 임피던스의 변화가 각각의 전극 사이에서 발생하는 반응보다는 각 전극들의 위쪽에서 발생하는 변화에 대한 영향을 더 많이 받게 되어, 임피던스 검출 정확도가 저하될 뿐만 아니라, 임피던스 검출 폭과 검출 한계가 좁고 낮아서 그 신뢰성과 활용성이 저하되는 문제가 있었다.

상기와 같은 문제점을 해결하기 위한 본 발명의 목적은 단백질 등과 같은 생체물질의 유무 및 그 농도를 각 교차 전극 사이의 임피던스 측정을 통해 검출함에 있어 각각의 교차 전극 간에 하이드로젤이 구비되도록 하여 임피던스 검출 폭과 검출 한계를 수십 내지 수백배 이상 늘이고 검출 정확성을 향상시킬 수 있도록 한 하이드로젤을 이용한 교차전극 바이오센서를 제공하는 것이다.
상기와 같은 목적을 달성하기 위한 본 발명의 실시 예에 따른 하이드로젤을 이용한 교차 전극 바이오센서는 기판상에 빗(Comb) 모양으로 복수의 제 1 돌출 전극이 배열되도록 구성된 제 1 교차 전극, 제 1 교차 전극과는 마주하여 기판상에 빗 모양으로 복수의 제 2 돌출 전극이 배열되도록 구성되되 복수의 제 2 돌출 전극들이 제 1 교차 전극에 형성된 복수의 제 1 돌출 전극들과 각각 교차 배치되도록 구성된 제 2 교차 전극, 및 서로 교차하여 배치된 제 1 교차 전극과 제 2 교차 전극의 사이 공간에 채워진 하이드로젤을 포함한다.
상기에서 설명한 본 발명의 하이드로젤을 이용한 교차 전극 바이오센서에 의하면 단백질 등과 같은 생체물질의 유무 및 그 농도를 각 교차 전극 사이의 임피던스 측정을 통해 검출함에 있어 각각의 교차 전극 간에 하이드로젤이 구비되도록 하여 임피던스 검출 폭을 수십 내지 수백배 이상 늘이고 검출 정확성을 향상시킬 수 있는 효과가 있다.
또한, 하이드로젤의 포어 크기 조절 특성을 이용하여 생체 물질들의 검출 효율과 검출량을 늘여서 임피던스의 검출 한계를 향상시켜 하이드로젤을 이용한 교차 전극 바이오센서의 신뢰성과 활용성을 더욱 높일 수 있는 효과가 있다.

이하, 본 발명의 실시 예를 첨부한 도면들을 참조하여 상세히 설명하기로 한다.
도 1은 본 발명의 실시 예에 따른 하이드로젤을 이용한 교차 전극 바이오센서의 구성을 나타낸 구성도이다. 그리고, 도 2는 도 1에 도시된 I-I' 단면을 나타낸 단면도이다.
도 1 내지 도 3에 도시된 하이드로젤을 이용한 교차 전극 바이오센서는 기판(PL) 상에 빗(Comb) 모양으로 복수의 제 1 돌출 전극이 배열되도록 구성된 제 1 교차 전극(100), 제 1 교차 전극(100)과는 마주하여 기판(PL) 상에 빗(Comb) 모양으로 복수의 제 2 돌출 전극이 배열되도록 구성되되 복수의 제 2 돌출 전극들이 제 1 교차 전극(100)에 형성된 복수의 제 1 돌출 전극들과 각각 교차 배치되도록 구성된 제 2 교차 전극(200), 및 서로 교차하여 배치된 제 1 교차 전극(100)과 제 2 교차 전극(200)의 사이 공간에 채워진 하이드로젤(300)을 포함하여 구성된다.
우선, 이와 같이 구성된 하이드로젤을 이용한 교차 전극 바이오센서를 이용한 임피던스 검출 특성을 먼저 설명하면, 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이의 임피던스는 다음과 같이 정리된다.
Z = R + jX
= R + j(XL-XC)
= R - jXC
= R - j(1/wC)
여기서, Z는 임피던스(impedance), R은 저항(resistance), X는 리액턴스(reactance), C는 정전용량(capacitance), w는 각주파수(angular frequency)이다. 리액턴스 X는 인덕터 성분인 XL과 커패시터 성분인 XC 나뉘는데, 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이는 전기적으로 직접 연결되어 있지 않기 때문에 인덕터 성분(XL)은 무시되고 커패시터 성분(XC)만 존재한다고 볼 수 있다.
이에, 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이 공간에 채워진 하이드로젤(300)에 표적(target) 생체물질에 특이적으로 반응하는 수용체(주로 항체, 압타머 등)을 고정하고 표적 생체물질이 수용체에 반응했을 때의 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이 공간, 즉 하이드로젤(300) 형성 공간의 임피던스 변화를 확인하면 표적 생체물질의 정량분석이 가능하게 된다.
도 2 및 도 3에 각각 도시된 바와 같이, 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이 공간들에 모두 하이드로젤(300)이 형성되도록 하면, 제 1 교차 전극(100)과 하이드로젤(300) 및 제 2 교차 전극(200)이 배치된 수평 방향으로 대부분의 전기장과 임피던스 변화가 일어난다. 즉, 제 1 교차 전극(100)과 하이드로젤(300) 및 제 2 교차 전극(200)의 상부나 수직 방향으로 전기장과 임피던스가 이탈되는 현상을 방지하여 수평 방향으로 전기장과 임피던스를 발생시키므로 임피던스 검출 폭을 수십 내지 수백배 이상 높일 수 있게 된다.
도 3은 도 1에 도시된 I-I' 단면을 나타낸 다른 단면도이다. 그리고, 도 4는 도 1 및 도 2에 도시된 교차 전극들을 실제로 구현하여 각각의 세부 구성과 단면을 보여주는 도면이다,
도 3은 제 1 교차 전극(100)과 제 2 교차 전극(200)을 형성하기 위한 패턴 형성 방법을 도 2와는 달리하여 제 1 교차 전극(100)과 제 2 교차 전극(200)을 형성하고, 제 1 교차 전극(100)과 제 2 교차 전극(200)의 양 측면을 모두 감싸는 형태로 금속 패턴(210)이 구성되도록 한 예를 도시하였다.
구체적으로, 도 2의 경우는 기판(PL) 상에 포토 레지스트나 폴리머 및 실리콘 구조체를 이용하여 제 1 교차 전극(100)과 제 2 교차 전극(200)의 패턴을 포토리소그래피 공법으로 패터닝하여 빗살 모양으로 구성한 후, 포토리소그래피 공법으로 다시 패터닝하여 제 1 및 제 2 교차 전극 패턴들의 양 측면을 모두 싸는 형태로 금속 패턴(210)을 구성한 예를 도시하였다.
반면, 도 3의 경우는 실리콘 재질의 기판(PL)을 패터닝하여 포토리소그래피 공법으로 패터닝하여 빗살 모양으로 제 1 및 제 2 교차 전극 패턴들을 형성한 후, 다시금 포토리소그래피 공법으로 다시 패터닝하여 제 1 및 제 2 교차 전극 패턴들의 양 측면을 모두 싸는 형태로 금속 패턴(210)을 구성한 예를 도시하였다.
이 후, 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이 공간에 하이드로젤(300)을 채워 하이드로젤(300) 형성 공간의 임피던스 변화를 확인함으로써, 표적 생체물질의 정량분석을 수행하게 된다.
도 5는 도 1에 도시된 하이드로젤의 포어 변화 구조를 나타낸 단면 모식도이다.
도 5를 참조하면, 하이드로젤(300)은 3차원 고분자 네트워크 구조를 갖는 물질로 PEGDA(Polyethylene glycol diacylate) crosslinker, PEG(Polyehylene glycol) porogen 및 다양한 기능성 그룹으로 구성되어있다. PEGDA crosslinker는 네트워크 구조체를 형성하며, PEG porogen은 포어(Pore)의 크기를 결정한다.
액체 상태인 하이드로젤(300)을 자외선 (UV)에 노출시키면 PEGDA crosslinker가 활성화되어 3차원 네트워크 구조를 형성하며, PEG porogen을 씻어내는 과정을 통해 포어가 형성된다.
도 5는 자외선 노출 (UV curing) 및 PEG porogen을 씻어내는 과정(Rinsing) 전/후 하이드로젤의 단면 모습을 나타낸 것으로, 서로 다른 PEG porogen을 사용하여 하이드로젤(300)을 형성했을 경우 포어의 크기 변화를 보여준다. 이렇게, 하이드로젤(300)은 네트워크 구조에 존재하는 기능성 그룹에 항원 및 다양한 수용체들을 3차원적으로 고정화할 수 있으며, PEG porogen의 종류에 따라 포어의 크기를 조절할 수 있다.
도 6은 도 5에 도시된 하이드로젤의 포어 크기 변화에 따른 내부 변화를 나타낸 다른 단면 모식도이다.
도 6에 도시된 바와 같이, 하이드로젤(300)의 포어의 크기가 생체 물질보다 작으면 생체 물질이 하이드로젤(300) 내부를 통과할 수 없게 된다. 그리고, 하이드로젤(300)의 포어의 크기가 생체물질보다 커지게 되면 생체 물질이 하이드로젤(300)의 내부를 통과할 수 있게 된다.
이에, 하이드로젤(300)을 이용한 교차 전극 센서를 이용하여 생체물질을 검출하기 위하여 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이의 공간에 표적 (Target) 생체물질에 특이적으로 반응하는 수용체가 존재하는 하이드로젤(300)을 채워 활성화 한 후, 표적이 생체물질에 반응했을 때의 임피던스 변화를 확인하여 타겟 물질의 정량분석을 가능하게 한다.
도 7은 항체-표적 생체분자 반응에 의한 교차 전극 바이오센서의 임피던스 변화를 나타낸 그래프이다.
도 7에 도시된 바와 같이, 항체와 표적 생체물질이 특이결합을 하게 되면 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이에 타겟 분자가 위치하게 되므로 저항이 변화하게 된다. 또한, 리액턴스도 표적분자의 성질에 의해서 캐패시턴스(C)의 값이 감소하게 되어 XC 값은 커지고 같이 XC 값은 감소하게 된다. 이러한 저항과 리액턴스의 변화량을 확인하여 타겟 분자의 양을 정확하게 검출할 수 있다.
종래에는 교차 전극 센서를 이용한 생체 물질 검출시 각각의 전극의 사이에 항체를 고정하고 표적분자와 결합 시 임피던스의 변화를 관찰하였다. 이 경우에는 항체가 전극 사이 표면에만 2차원적으로 고정된다. 하지만, 본 발명의 실시 예에 따라 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이에 하이드로젤(300)을 형성하여 이용할 경우 항체를 하이드로젤(300) 내의 네크워크 구조에 항체를 3차원적으로 고정할 수 있어 센서의 다이내믹레인지(Dynamic range)를 넓힐 수 있다.
도 8은 도 1 및 도 2에 도시된 하이드로젤을 이용하는 교차 전극 바이오센서의 또 다른 특성을 나타낸 구성도 및 단면 모식도이다.
도 8을 참조하면, 본 발명에서는 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이에 형성된 하이드로젤(300)의 포어 크기를 표적 물질의 크기와 비슷하게 조절함으로써, 표적 물질보다 크기가 큰 표적 외 물질을 분류할 수 있게 된다. 즉, 표적 물질의 크기에 따라 하이드로젤(300)의 포어 크기를 조절함으로써 표적 물질의 분류가 가능해질 수 있다.
하이드로젤(300)의 포어 크기는 하이드로젤(300) 형성 후 자외선을 이용하여 PEGDA crosslinker를 활성화시켜 3차원 네트워크 구조를 형성하는 공정 시간과 강도를 설정하여 조절할 수 있다.
도 9는 도 1 및 도 2에 도시된 교차 전극 바이오센서의 제조 방법을 설명하기 위한 단면도이다.
교차 전극 바이오센서는 MEMS(Micro Electro Mechanical Systems) 미세 공정 기술을 이용하여 2가지 방법으로 제작할 수 있다.
우선, 도 9의 경우는 실리콘 재질의 기판(PL)을 포토리소그래피(photolithography) 공법으로 패터닝하여 빗살 모양으로 제 1 및 제 2 교차 전극 패턴들을 형성한 후, 다시금 포토리소그래피 공법으로 패터닝하여 제 1 및 제 2 교차 전극 패턴들의 양 측면을 모두 싸는 형태로 금속 패턴(210)을 구성한 예를 도시하였다.
구체적으로, (a) 실리콘 재질의 기판(PL) 상에 포토 레지스터(PR; Photoresist)을 패터닝 한 후, (b) DRIE(Deep Reactive Ion Etching)를 이용하여 포토리소그래피 공법으로 실리콘 재질의 기판(PL)을 식각하여 빗살 모양으로 제 1 및 제 2 교차 전극 패턴을 형성한다.
다음으로, (c) PECVD(Plasma Enhanced Chemical Vapor Deposition)을 이용하여 300nm의 실리콘 산화막(SiO2)을 제 1 및 제 2 교차 전극 패턴을 포함한 기판(PL)의 전면에 증착함으로써 절연층(201; Insulating layer)을 형성한다.
이어, (d) 금속 패턴(210)으로 미세전극을 형성하기 위해, 절연층(201) 상에 스퍼터링(Sputtering) 방식으로 Ti(Titanium) 50nm, Pt(Platinum) 200nm로 순차 증착한다. 여기서, Ti 층과 Pt 층은 Silicon Dioxide layer의 결합력을 증가시키기 위한 Adhesion layer로 사용될 수 있다. 이렇게, SiO2/Ti/Pt의 순으로 증착된 실리콘 구조체는 ICP-RIE(Inductively Coupled Plasma Reactive Ion Etcher)를 이용하여 Pt 층과 Ti 층을 식각함으로써, 하여 금속 패턴(210)으로 미세전극을 형성할 수 있다.
도 10은 도 1 및 도 3에 도시된 교차 전극 바이오센서의 제조 방법을 설명하기 위한 다른 단면도이다.
도 10은 기판(PL) 상에 포토 레지스트나 폴리머 및 실리콘 구조체를 이용하여 제 1 교차 전극(100)과 제 2 교차 전극(200)의 패턴을 포토리소그래피 공법으로 패터닝하여 빗살 모양으로 구성한 후, 포토리소그래피 공법으로 다시 패터닝하여 제 1 및 제 2 교차 전극 패턴들의 양 측면을 모두 싸는 형태로 금속 패턴(210)을 구성한 예를 도시하였다.
구체적으로, (a) 포토 레지스트나 폴리머 및 실리콘 구조체의 패터닝 방법은 절연층(201)을 형성하기 위해, PECVD을 이용하여 300nm의 실리콘 산화막을 실리콘 재질의 기판(PL) 상에 증착한다.
그리고, (b) 제 1 및 제 2 교차 전극 패턴 형성을 위해 포토레지스트(PR)를 마이크로 패터닝한다.
(c) 금속 패턴(210)으로 미세전극을 형성하기 위해서는 Ti 50nm, Pt 200nm를 스퍼터링 방식으로 증착 한 후, (d) ICP-RIE를 이용하여 Pt 층과 Ti 층을 순차적으로 식각함으로써, 하여 금속 패턴(210)으로 미세전극을 형성할 수 있다.
도 11은 도 1에 도시된 교차 전극 바이오센서의 하이드로젤 형성 방법을 설명하기 위한 단면도이다.
도 11에 도시된 바와 같이, 제 1 교차 전극(100)과 제 2 교차 전극(200) 사이 공간에 하이드로젤(300)을 형성하기 위해서는 먼저 제 1 교차 전극(100)과 제 2 교차 전극(200)을 포함한 교차 전극 바이오센서 영역을 모두 덮도록 하이드로젤(300)을 도포한다.
이 후, 하이드로젤의 평탄화를 위해 PDMS(Polydimethylsiloxance)를 제 1 교차 전극(100)과 제 2 교차 전극(200) 상에 덮고, 그 후에 자외선 노출(UV curing) 방식을 이용하여 PEGDA crosslinker를 활성화시켜 3차원 네트워크 구조를 형성한다. 그리고, PPEG porogen을 씻어내는 과정(Rinsing)을 통해 포어를 형성함과 아울러 포어의 크기를 결정한다. PEG porogen의 종류와 첨가 비율에 따라 포어의 크기를 조절할 수 있다.
도 12는 도 1에 도시된 하이드로젤을 이용한 교차 전극 바이오센서의 임피던스 검출 효율 변화를 나타낸 그래프이다.
도 12에 도시된 바와 같이, 하이드로젤(300)이 교차 전극 바이오센서 즉, 제 1 교차 전극(100)과 제 2 교차 전극(200) 간에 형성된 경우, 하이드로젤(300)이 형성되지 않은 종래의 바이오센서보다 검출 한계가 100배 향상되었고 다이나믹 레인지(Dynamic range)도 확장됨을 확인할 수 있다. 이는 하이드로젤(100) 내에 형성되어 있는 항체의 개체수가 증가하여 반응 확률이 증가 되었고, 이를 민감하게 검출할 수 있는 것에서 기인한다고 할 수 있다.
이상에서 상술한 바와 같이, 본 발명의 하이드로젤(300)을 이용한 교차 전극 바이오센서에 의하면 단백질 등과 같은 생체물질의 유무 및 그 농도를 각 교차 전극 사이의 임피던스 측정을 통해 검출함에 있어 각각의 교차 전극 간에 하이드로젤(300)이 구비되도록 하여 임피던스 검출 폭을 수십 내지 수백배 이상 늘이고 검출 정확성을 향상시킬 수 있는 효과가 있다.
또한, 하이드로젤(300)의 포어 크기 조절 특성을 이용하여 생체 물질들의 검출 효율과 검출량을 늘여서 임피던스의 검출 한계를 향상시켜 하이드로젤(300)을 이용한 교차전극 바이오센서의 신뢰성과 활용성을 더욱 높일 수 있는 효과가 있다.
상기에서는 본 발명의 실시 예를 참조하여 설명하였지만, 해당 기술 분야에서 통상의 지식을 가진 자라면 하기의 특허 청구의 범위에 기재된 본 발명의 사상 및 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.

Claims (5)

  1. 기판상에 빗(Comb) 모양으로 복수의 제 1 돌출 전극이 배열되도록 구성된 제 1 교차 전극;
    상기 제 1 교차 전극과는 마주하여 상기 기판상에 빗 모양으로 복수의 제 2 돌출 전극이 배열되도록 구성되되 상기 복수의 제 2 돌출 전극들이 상기 제 1 교차 전극에 형성된 복수의 제 1 돌출 전극들과 각각 교차 배치되도록 구성된 제 2 교차 전극; 및
    상기 서로 교차하여 배치된 제 1 교차 전극과 제 2 교차 전극의 사이 공간에 채워진 하이드로젤을 포함하는 하이드로젤을 이용한 교차 전극 바이오센서.
  2. 제 1 항에 있어서,
    상기 제 1 교차 전극과 상기 제 2 교차 전극은
    상기 기판과 일체로 형성된 상기 기판 재질의 제 1 및 제 2 교차 전극 패턴; 및
    상기 제 1 및 제 2 교차 전극 패턴들의 양 측면을 모두 감싸는 형태로 패터닝되어 형성된 금속 패턴을 포함하는 것을 특징으로 하는 하이드로젤을 이용한 교차 전극 바이오센서.
  3. 제 1 항에 있어서,
    상기 제 1 교차 전극과 상기 제 2 교차 전극은
    상기 기판상에 포토레지스터나 폴리머 및 실리콘 구조체가 패터닝되어 형성된 제 1 및 제 2 교차 전극 패턴; 및
    상기 상기 제 1 및 제 2 교차 전극 패턴들의 양 측면을 모두 감싸는 형태로 패터닝되어 형성된 금속 패턴을 포함하는 것을 특징으로 하는 하이드로젤을 이용한 교차 전극 바이오센서.
  4. 제 1 항에 있어서,
    상기 하이드로젤은
    3차원 고분자 네트워크 구조를 갖는 PEGDA(Polyethylene glycol diacylate) crosslinker 및 PEG(Polyehylene glycol) porogen 를 포함하여 구성되며,
    상기 하이드로젤을 자외선 에 노출시켜 상기 PEGDA crosslinker를 활성화시킴과 아울러, 상기 PEG porogen을 씻어내어 포어가 형성되도록 하는 하이드로젤을 이용한 교차 전극 바이오센서.
  5. 제 1 항에 있어서,
    3차원 고분자 네트워크 구조를 갖는 PEGDA(Polyethylene glycol diacylate) crosslinker 및 PEG(Polyehylene glycol) porogen 를 포함하여 구성되며,
    PDMS(Polydimethylsiloxance)를 상기 제 1 및 제 2 교차 전극 상에 덮고 자외선을 노출함으로써 상기 PEGDA crosslinker를 활성화시켜 3차원 네트워크 구조를 형성하고,
    상기 PEG porogen의 종류와 첨가 비율을 조절함와 아울러 상기 PPEG porogen을 씻어내는 과정을 통해 상기 포어의 크기를 조절하는 하이드로젤을 이용한 교차 전극 바이오센서.
