Abstract

본 발명은 백금계 항암제에 의해 유도된 세포 내 증가된 p53 수준을 유지시키는 방법 및 이의 활용에 관한 것으로, 보다 상세하게는 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiquitin ligase)에 대한 siRNA를 이를 필요로 하는 개체에 병용 또는 순차적으로 투여하여 세포에서 증가된 수준의 p53을 유지시키는 방법 및 이를 이용한 암 세포의 사멸을 촉진용 조성물에 관한 것이다. 본 발명의 방법에 따르면, 낮은 농도의 백금계 항암제를 처리하여도 세포 내 p53의 증가된 발현 수준을 오랜 시간동안 유지할 수 있어 암 세포의 사멸을 효과적으로 유도할 수 있을 뿐만 아니라, 백금계 항암제의 투여에 따른 약물 부작용을 최소화 할 수 있어 암의 예방 또는 치료제 개발에 유용하게 사용될 수 있다.

본 출원은 2015년 4월 16일에 출원된 미국 특허출원 제 62/148,403 호를 우 선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다. 본 발명은 백금계 항암제에 의해 유도된 세포 내 증가된 p53 수준을 유지시 키는 방법 및 이와 활용에 관한 것으로, 보다 상세하게는 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 이를 필요로 하는 개 체에 병용 또는 순차적으로 투여하여 세포에서 증가된 수준의 p53을 유지시키는 방 법 및 이를 이용한 암 세포의 사멸 촉진용 조성물에 관한 것이다.
【배경기술】
종양 억제 단백질 p53은 DNA 복구, 세포 주기 및 성장 정지, 및 아품토시스 를 책임지는 유전자의 다양한 어레이의 발현을 조절함으로써 세포에서 게놈의 완전 성을 유지하는 데 있어서 중추적 역할을 한다 [문헌 [May et al . , Oncogene 18 (53)' (1999) p. 7621-7636]; [Or en, Cel l Death Di f fer . 10 (4) (2003) p. 431- 442]; [Hal l and Peters , Adv. 【발명의 상세한 설명】
【기술적 과제】 이에, 본 발명자들은 백금계 항암제의 처리에 의해 암 세포에서 아품토시스
(apoptosis)가 유발되기 위해서는 p53의 발현이 임계값 (threshold) 이상으로 상승 하여야 하며, 일정한 시간 이상 P53의 발현증가가 유지 되도록 하는 방법 , 즉 백금 계 항암제에 의해 증가된 세포 내 p53의 발현을 보다 오랜시간 동안 유지시켜 암 세포를 효과적으로 사멸시킬 수 있는 치료전략을 개발하기 위해 예의 노력을 기울 인 결과, p53에 대한 내재적 저해제로 알려진 p53에 대한 유비퀴틴 리가아제를 저 해하는 물질을 시스플라틴과 병용투여 하면, 시스플라틴에 의해 증가된 세포 내 p53의 발현이 더 오랫동안 지속되어 암 세포에 대한 치료효과가 더 우수할 수 있다 는 것을 확인하고 본 발명을 완성하게 되었다. 따라서, 본 발명의 목적은 백금계 항암제 및 P53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 이를 필요로 하는 개체에 병용 또는 순차적으 로 투여하는 것을 특징으로 하는 세포에서 증가된 수준 (elevated level )의 p53을 유지시키는 방법을 제공하는 것이다. 본 발명의 다른 목적은 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제
(ubiqui t in l igase)에 대한 siRNA를 유효성분으로 포함하는, 세포에서 p53의 수준 올 증가된 수준 (elevated level )으로 유지시키는 것에 의한 세포 사멸 촉진용 조 성물을 제공하는 것이다. 본 발명의 다른 목적은 백금계 항암제 및 P53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 유효성분으로 포함하는, 암세포에서 p53의 수 준을 증가된 수준 (elevated level )으로 유지시키는 것에 의한 암 예방 또는 치료 용 약학적 조성물을 제공하는 것이다. 본 발명의 다른 목적은 (a) 인유두종바이러스 (Human Papi 1 lomar ius , HPV)의 E6 및 E7에 대한 siRNA를 자궁경부암 세포주에 형질도입하는 단계; (b) 상기 (a) 단계에서 형질도입된 세포주에 자궁경부암 치료제 후보물질을 처리하는 단계; (c) 상기 치료제 후보물질을 처리한 직후 내지 48시간 동안 일정한 시간 간격마다 세포 내 p53(tumor protein 53)의 발현수준을 측정하는 단계; 및 (d) 치료제 후보물질을 처리하지 않은 세포와 비교해 세포 내 p53(turaor protein 53)의 발현증가지속시간 이 연장된 물질을 선별하는 단계를 포함하는 자궁경부암 예방 또는 치료제 스크리 닝 방법을 제공하는 것이다.
【기술적 해결방법】
상기한 본 발명의 목적을 달성하기 위하여 본 발명은 백금계 항암제 및 p53 에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 이를 필요로 하는 개체에 병용 또는 순차적으로 투여하는 것을 특징으로 하는 세포에서 증가된 수준 (elevated level )의 p53을 유지시키는 방법을 제공한다. 본 발명의 다른 목적올 달성하기 위하여 본 발명은 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 유효성분으로 포함하 는, 세포에서 p53의 수준을 증가된 수준 (elevated level )으로 유지시키는 것에 의 한 세포 사멸 촉진용 조성물을 제공한다. 본 발명의 다른 목적올 달성하기 위하여 본 발명은 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 유효성분으로 포함하 는, 암세포에서 p53의 수준을 증가된 수준 (elevated level )으로 유지시키는 것에 의한 암 예방 또는 치료용 약학적 조성물을 제공한다. 본 발명의 다른 목적을 달성하기 위하여 본 발명은 (a) 인유두종바이러스 (Human Papi 1 lomarius , HPV)의 E6 및 E 대한 siRNA를 자궁경부암 세포주에 형질 도입하는 단계; (b) 상기 (a) 단계에서 형질도입된 세포주에 자궁경부암 치료제 후 보물질을 처리하는 단계; (c) 상기 치료제 후보물질을 처리한 직후 내지 48시간 동 안 일정한 시간 간격마다 세포 내 p53(tumor protein 53)의 발현수준을 측정하는 단계; 및 (d) 치료제 후보물질을 처리하지 않은 세포와 비교해 세포 내 p53(tumor protein 53)의 발현증가 지속시간이 연장된 물질을 선별하는 단계를 포함하는 자궁 경부암 예방 또는 치료제 스크리닝 방법을 제공한다. 이하 본 발명에 대하여 상세히 설명한다. 본 발명은 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 이를 필요로 하는 개체에 병용 또는 순차적으로 투여하는 것을 특징으로 하는 세포에서 증가된 수준 (elevated level )의 p53을 유지시키는 방법을 제공한다. 본 발명에서 상기 백금계 항암제는 암 치료에 널리 사용되는 항암제 가운데 하나로 약 50%의 암 환자에서 사용되고 있는 약물이다. 백금계 항암제는 백금과 배위결합하고 있는 착물로서 본 발명에서 상기 백금계 항암제는 시스플라틴 (ci s- di amminedi chloroplat inum [ I I ] ) , 카보플라틴 (carbopl at in) , 옥살리퓰라틴 (oxal iplat in) , 네다플라틴 (nedaplat in) , 피코플라틴 (pi copl at in), 트리플라틴 테 트라나이트레이트 (tr iplat in tetrani trate) , 사트라플라틴 (satraplat in) 및 이들 의 흔합물로 이루어진 군으로부터 선택될 수 있으며, 바람직하게는 시스플라틴 또 는 카보플라틴일 수 있고, 가장 바람직하게는 시스플라틴일 수 있다. 이 중 시스플라틴은 백금 원자에 2개의 염소와 암모니아가 배위된 하기 화학 식 1로 표시되는 구조를 갖는 화합물이다:

"약학적으로 허용되는 염"은, 화합물이 투여되는 유기체에 심각한 자극을 유 발하지 않고 화합물의 생물학적 활성과 물성들을 손상시키지 않는, 화합물의 제형 을 의미한다. 용어 "수화물", "용매화물'' 및 "이성질체" 역시 상기와 같은 의미를 가진다. 상기 약제학적 염은, 본 발명의 화합물을, 염산, 브름산, 황산, 질산, 인 산 등의 무기산, 메탄술폰산, 에탄술폰산, P-를루엔술폰산 등의 술폰산, 타타르산, 포름산, 시트르산, 아세트산, 트리클로로아세트산, 트리플루오로아세트산, 카프릭 산, 이소부탄산, 말론산, 석신산, 프탈산, 글루콘산, 벤조산, 락트산, 푸마르산, 말레인산, 살리실산 등과 같은 유기 카본산과 반응시켜 얻어질 수 있다. 또한, 본 발명의 화합물을 염기와 반응시켜, 암모니움 염, 나트륨 또는 칼륨 염 등의 알칼리 금속염, 칼슘 또는 마그네슘 염 등의 알칼리 토금속염 등의 염, 디시클로핵실아민, N-메틸 -D-글루카민, 트리스 (히드록시메틸) 메틸아민 등의 유기염기들의 염, 및 아 르기닌, 리신 등의 아미노산 염을 형성함으로써 얻어질 수도 있다.
"수화물 (hydrate) "은 비공유적 분자간력 (non— covalent intermolecular force)에 의해 결합된 화학양론적 (stoichiometric) 또는 비화학양론적 (non- stoichiometric) 량의 물을 포함하고 있는 본 발명의 화합물 또는 그것의 염을 의 미한다.
"용매화물 (solvate) "은 비공유적 분자간력에 의해 결합된 화학양론적 또는 비화학양론적 량의 용매를 포함하고 있는 본 발명의 화합물 또는 그것의 염을 의미 한다. 그에 관한 바람직한 용매들로는 휘발성, 비독성, 및 /또는 인간에게 투여되기 에 적합한 용매들이다.
"이성질체 ( isomer)"는, 동일한 화학식 또는 분자식을 가지지만 광학적 또는 입체적으로 다른 본 발명의 화합물 또는 그것의 염을 의미한다. 예를 들어, 본 발 명의 화학식 1에 따른 화합물은 치환기들의 종류에 따라서는 입체생성 중심 (asymmetric center , 비대칭 탄소 원자)을 가질 수 있는 바, 이 경우 화학식 1의 화합물은 거울상 이성질체 및 부분 입체 이성질체와 같은 광학 이성질체로서 존재 할수 있다.
"프로드럭 (prodrug) ' '은 생체 내에서 모 약제 (parent drug)로 변형되는 물질 을 의미한다. 프로드럭은, 몇몇 경우에 있어서, 모 약제보다 투여하기 쉽기 때문에 종종 사용된다. 예를 들어, 이들은 구강 투여에 의해 생활성을 얻을 수 있음에 반 하여, 모 약제는 그렇지 못할 수 있다. 프로드럭은 또한 모 약제보다 제약 조성물 에서 향상된 용해도를 가질 수도 있다. 예를 들어, 프로드럭은, 수용해도가 이동성 에 해가 되지만, 일단 수용해도가 이로운 세포에서는, 물질대사에 의해 활성체인 카르복실산으로 가수분해되는, 세포막의 통과를 용이하게 하는 에스테르 ( "프로드 럭 " )로서 투여되는 화합물일 것이다. 프로드럭의 또 다른 예는 펩티드가 활성 부위 를 드러내도록 물질대사에 의해 변환되는 산기에 결합되어 있는 짧은 펩티드 (폴리 아미노산)일 수 있다. 본 발명에서 상기 유비퀴틴 리가아제 (ubi qui t in l igase)는 E3 유비퀴틴 리가 아제로 알려진 단백질로, 특정 단백질을 인식해 유비퀴틴화를 유도하는 유비퀴틴 연결효소이다. 유비퀴틴 리가아제는 p53에 대한 유비퀴틴 결합효소로 작용하여 p53 을 유비퀴틴화 시킴으로써 26S 프로테아좀 경로를 통해 분해시키는 기능을 나타낸 다. 따라서, 본 발명에서 상기 유비퀴틴 리가아제는 p53에 대한 유비퀴틴 결합효소 로서 작용하여 p53을 유비퀴틴화 시킴으로써 세포 내에서 p53의 발현을 저하시키는 단백질 뿐만 아니라, 이의 단편, 이의 기능적 동등물 및 기능적 유도체를 모두 포 함하는 개념으로 이해될 수 있다. 본 발명의 유비퀴틴 리가아제는 둘 이상의 단백 질이 복합체를 형성하여 유비퀴틴 리가아제로 기능하는 것을 포함하며, 이에 한정 되지는 않으나, 예를 들어 인유두종바이러스 (Human Papi 1 lomar ius , HPV)의 E6/E6— AP 복합체와 같은 유비퀴틴 리가아제 복합체일 수 있다. 상기 "단편" 이란 천연형 단백질의 아미노산 일부가 결실되어 있으나 단백 질의 생리학적 활성은 천연형 단백질과 동일한 것 또는 단백질 복합체를 형성하는 구성 단백질 중 일부를 포함한다. 상기 "기능적 동등물 "에는 천연형 단백질 아미노 산 중 일부 또는 전부가 치환되거나, 아미노산의 일부가 결실 또는 부가된 아미노 산 서열 변형체로서 천연형 유비퀴틴 리가아제 단백질과 실질적으로 동등한 생리활 성을 갖는 것을 말한다. 또한 상기 "기능적 유도체 "는 상기 유비퀴틴 리가아제 단 백질의 물리 /화학적 성질을 증가 또는 감소시키기 위한 변형을 가한 단백질로서 천 연형 단백질과 실질적으로 동등한 생리 활성을 갖는 것을 의미한다 본 발명에서 상기 유비퀴틴 리가아제는 바람직하게는 HDM2, 인유두종바이러 스 (Human Papi 1 lomar ius , HPV)의 E6/E6-AP 복합체, E6 또는 E6-AP6 단백질 일 수 있으며, 가장 바람직하게는 인유두종바이러스의 E6/E6-AP 복합체, E6 또는 E6-AP 단백질 일 수 있으나 이에 제한되는 것은 아니다. 본 발명의 일실시예에 따르면, 자궁경부암 세포주에 시스플라틴을 처리한 결 과 세포 내 p53의 발현량이 박동 (pulse) 형태를 나타내며 증가와 감소를 반복하는 것으로 확인되었다. 시스플라틴에 의해 종양세포의 아픕토시스 (apotosis)가 유발되 기 위해서는 세포 내 p53의 발현량이 일정 수준 (threshold, 역치) 이상으로 증가되 어야 하는데, 종양세포에 시스플라틴만을 단독으로 처리했을 때에는 세포독성을 나 타내기에 충분한 정도로 P53의 발현증가가유지되지 않는 것으로 확인되었다. 본 발명의 다른 일실시예에서 본 발명자들은 자궁경부암 세포주에서 p53의 유비퀴틴 리가아제로 작용하는 인유두종바이러스 (Human Papi 1 lomar ius , HPV)의 E6 및 E7에 대한 siRNA를 자궁경부암 세포주에 처리한 결과, 세포 내 p53의 발현량이 증가하는 경향을 나타내었으나, 이러한 p53의 발현증가 또한 세포독성이 층분히 나 타날 정도로 오랜 시간동안 지속되지 않는 것으로 확인되었다. 이에, 본 발명자들은 인유두종바이러스 E6 및 E7에 대한 siRNA를 시스플라틴 과 병용하여 자궁경부암 세포주에 처리한 결과, 세포의 아품토시스가 (apoptosis)가 유발될 수 있는 수준 (threshold) 이상으로 p53의 발현증가가 유지되는 것을 확인할 수 있었다. 본 발명에서 용어, "siRNA" 란 특정 mRNA의 절단 (cleavage)을 통하여 RNAi (RNA interference) 현상을 유도할 수 있는 짧은 이중사슬 RNA를 의미한다. 표 적유전자의 mRNA와 상동인 서열을 가지는 센스 RNA 가닥과 이와 상보적인 서열을 가지는 안티센스 RNA 가닥으로 구성된다. siRNA는 표적 유전자의 발현을 억제할 수 있기 때문에 효율적인 유전자 넉다운 방법으로서 또는, 유전자치료 (gene therapy) 의 방법으로 제공된다. siRNA는 RNA끼리 짝을 이루는 이중사슬 RNA부분이 완전히 쌍을 이루는 것에 한정되지 않고 미스매치 (대응하는 염기가 상보적이지 않음), 벌지 (일방의 사슬에 대웅하는 염기가 없음) 등에 의하여 쌍을 이루지 않는 부분이 포함될 수 있다. 전 체 길이는 10 내지 100 염기, 바람직하게는 15 내지 80 염기, 더욱 바람직하게는 20 내지 70 염기이다. siRNA 말단 구조는 표적유전자의 발현을 RNAi 효과에 의하여 억제할 수 있는 것이면 평활 (blunt )말단 흑은 점착 (cohesive) 말단 모두 가능하다. 점착 말단 구조는 3 말단 돌출한 구조와 5 말단 쪽이 돌출한 구조 모두 가능하다. 돌출하는 염기 수는 한정되지 않는다. 예를 들어, 염기 수로는 1 내지 8 염기 , 바 람직하게는 2 내지 6 염기로 할 수 있다. 본 명세서에서 siRNA의 전장은, 중앙의 이중사슬 부분의 길이와 양 말단의 단일사슬 돌출을 구성하는 길이의 합으로 나타 내었다. 또한, siRNA는 표적유전자의 발현억제 효과를 유지할 수 있는 범위에서 예 를 들어, 한 쪽 말단의 돌출 부분에 저분자 RNA (예를 들어, tRNA, rRNA, 바이러스 RNA와 같은 천연의 RNA분자 또는 인공의 RNA분자)를 포함할 수 있다. siRNA 말단구 조는 양측 모두 절단 구조를 가질 필요는 없고, 이중사슬 RNA의 일방의 말단 부위 가 링커 RNA에 의하여 접속된 스템 루프형 구조일 수도 있다. 링커의 길이는 스템 부분의 쌍을 이루는 데 지장이 없는 길이면 특별히 한정되지 않는다. 본 발명의 siRNA는 p53에 대한 유비퀴틴 리가아제 (ubiquit in l igase) mRNA 를 특이적으로 감소시킬 수 있으면 서열과 길이는 특별히 제한되지 않으며, 본 발 명의 구체적 실시예에서는 HPV E6 또는 E7에 특이적인 5종의 siRNA를 제작하였으 며, 상기 siRNA가 세포 내 p53의 발현올 증가시키며, 시스플라틴과 병용처리 되었 을 때 세포 내 p53의 발현을 일정 수준 (threshold) 이상으로 오랜시간동안 유지시 키는 효과가 있음을 확인하였다. 따라서, 본 발명에서 상기 siRNA는 바람직하게는 서열번호 1 내지 10으로 이 루어진 군에서 선택될 수 있으나 이에 특별히 제한되는 것은 아니다. 본 발명의 발명자들은 하기 표에 나타난 바와 같이 HPV type 16 및 18의 E6/E7 종양성 단백질에 대한 siRNA를 제작하였다. 하기 표에서 'm' 으로 표시된 siRNA 서열은 염기 잔기에 메틸기를 결합시킨 2' -0-Me 변형 뉴클레오타이드로 치환 된 염기를 나타낸다. 즉, 2 ' -0-Me 변형된 U의 경우 'mU' 로 표시 또는 2 ' -0-Me 변 형된 G의 경우 'mG' 로 표시하였다.

본 발명은 백금계 항암제에 의해 증가된 세포 내 p53의 발현을 유지하기 위 해서 백금계 항암제와 p53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 병용투여 하는 것을 특징으로 한다. 상기 병용투여 한다는 것은 백금계 항암제와 siRNA를 동시에 (simutaneously) , 개별적으로 (separately) 또는 순차적 (sequent i al ly) 투여되는 것 을 의미한다. 보다 구체적으로는, 백금계 항암제와 siRNA를 단일 조성물의 형태로 제조하여 동시에 투여하거나, 또는 백금계 항암제와 siRNA 중 어느 하나를 다른 하 나가 투여되기 전에 미리 투여하는 것을 의미한다. 본 발명에서 백금계 항암제와 siRNA의 투여 순서, 즉, 어느 것을 어느 시점에서 동시에, 개별적으로 또는 순차적 으로 투여할 것인지의 여부는 전문가의 판단에 의해 용이하게 선택될 수 있을 것이 다. 상기 본 발명의 방법에 따라 백금계 항암제와 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 개체에 병용투여 하면, 개체의 세포, 특히 암 세포 내에서 p53의 발현증가가 오랜 시간 지속될 수 있으며, 결과적으로 암 세포의 사멸이 유도되는 효과가 나타난다. 바람직하게는, 상기 암 세포의 사멸은 증가된 p53에 의해 유도되는 아폽토시스 (apoptosis) 경로를 통해 이루어지는 것일 수 있 다. 본 발명은 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiquit in l igase)에 대한 siRNA를 유효성분으로 포함하는, 세포에서 p53의 수준을 증가된 수 준 (elevated level )으로 유지시키는 것에 의한 세포 사멸 촉진용 조성물을 제공한 다. 본 발명은 또한 백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 유효성분으로 포함하는, 암세포에서 p53의 수준올 증가된 수준 (elevated level )으로 유지시키는 것에 의한 암 예방 또는 치료용 약학적 조 성물을 제공한다. 본 발명에 따른 약학적 조성물은 백금계 항암제와 siRNA가 단일 조성물 (single composi t ion)의 형태로 제제화되거나 또는 개별적인 조성물 (separate composit ion)의 형태로 제제화될 수 있다. 바람직하게는, 개별적인 조성물의 형태 로 제제화될 수 있다. 이들을 제제화하는 방법은 당업계에서 범용되는 기술을 이용 할수 있다. 본 발명에 따른 약학적 조성물에 있어서, 상기 각 성분은 동시에 (simultaneous) , 개별적 (separate) 또는 순차적 (sequent ial )으로 투여될 수 있다. 예를 들면, 본 발명의 약학적 조성물에 포함된 각 성분이 단일 조성물 (single composit ion)로 되어 있는 경우에는 동시에 투여될 수 있으며, 단일 조성물로 되어 있지 않을 경우에는 한 성분을, 다른 성분이 투여되기 전, 후 및 /또는 다른 성분과 함께 동시에 투여될 수 있다. 상기 본 발명에 따른 약학적 조성물의 투여 순서 즉, 어떤 것을 어느 시점에서, 동시에, 개별적 또는 순차적으로 투여할 것인지의 여부 는 의사나 전문가에 의해 결정될 수 있다. 이러한 투여 순서는 많은 인자에 따라 달라질 수 있다. 본 발명의 일실시예에 따르면, 인간 자궁경부암 세포가 이식된 동물모델에 시스플라틴과 HPV E6 및 E7에 대한 siRNA를 병용투여한 결과, 시스플라틴 단독투여 에 의해서는 종양성장 억제효과가 나타나지 않는 저농도에서도 우수한 종양성장억 제효과를 나타내는 것을 확인하였다. 즉, 본 발명의 약학적 조성물을 이용하여 백금계 항암제와 siRNA를 병용 투 여하는 경우, 통상적으로 항암효과를 위해 투여되는 백금계 항암제의 농도보다 낮 은 농도로 약물을 투여해도 우수한 항암효과를 나타내기 때문에 백금계 항암제를 과량 투여하거나 또는 장기 투여할 경우 야기될 수 있는 심각한 부작용들을 감소시 킬 수 있다는 장점이 있다. 따라서, 본 발명의 약학적 조성물은 암 세포 내에서 p53의 발현증가를 오랜 시간 지속시켜 우수한 항암효과를 나타낼 수 있을 뿐만 아니라, 항암치료에 따른 부작용을 경감시킬 수 있어 임상에 유용하게 적용될 수 있다. 본 발명에 따른 약학적 조성물은 상기 백금계 항암제와 siRNA만을 함유하거 나 약학적으로 허용되는 담체와 함께 적합한 형태로 제형화 될 수 있으며, 부형제 또는 희석제를 추가로 함유할 수 있다. 상기 담체로는 모든 종류의 용매, 분산매 질, 수중유 또는 유중수 에멀젼, 수성 조성물, 리포좀, 마이크로비드 및 마이크로 좀이 포함된다. 약학적으로 허용되는 담체로는 예컨대, 경구 투여용 담체 또는 비경구 투여 용 담체를 추가로 포함할 수 있다. 경구 투여용 담체는 락토스, 전분, 샐를로스 유 도체, 마그네슘 스테아레이트, 스테아르산 등을 포함할 수 있다. 아울러, 경구투여 용으로 사용되는 다양한 약물전달물질을 포함할 수 있다. 또한, 비경구 투여용 담 체는 물, 적합한 오일, 식염수, 수성 글루코오스 및 글리콜 등을 포함할 수 있으 며, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수 소나트륨, 아황산나트륨 또는 아스코르 브산과 같은 항산화제가 있다. 적합한 보존 제로는 벤즈알코늄 클로라이드, 메틸 -또는 프로필-파라벤 및 클로로부탄올이 있다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현택제 등을 추가로 포함할 수 있다. 그 밖의 약학적으로 허용되는 담체 및 제제는 다음의 문헌에 기재되어 있는 것을 참고로 할 수 있다 (Remington' s Pharmaceut ical Sciences , 19th ed. , Mack Publ i shing Company , East on, PA, 1995) . 본 발명의 조성물은 인간을 비롯한 포유동물에 어떠한 방법으로도 투여할 수 있다. 예를 들면, 경구 또는 비경구적으로 투여할 수 있다. 비경구적인 투여방법으 로는 이에 한정되지는 않으나, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장내 투여일 수 있다. 본 발명의 약학적 조성물은 상술한 바와 같은 투여 경로에 따라 경구 투여용 또는 비경구 투여용 제제로 제형화 할 수 있다.
경구 투여용 제제의 경우에 본 발명의 조성물은 분말, 과립, 정제, 환제, 당 의정제, 캡슐제, 액제, 겔제, 시럽제, 슬러리제, 현탁액 등으로 당업계에 공지된 방법을 이용하여 제형화될 수 있다. 예를 들어, 경구용 제제는 활성성분을 고체 부 형제와 배합한 다음 이를 분쇄하고 적합한 보조제를 첨가한 후 과립 흔합물로 가공 함으로써 정제 또는 당의정제를 수득할 수 있다. 적합한 부형제의 예로는 락토즈, 텍스트로즈, 수크로즈, 솔비를, 만니를, 자일리를, 에리스리를 및 말티를 등을 포 함하는 당류와 옥수수 전분, 밀 전분, 쌀 전분 및 감자 전분 등을 포함하는 전분 류, 셀를로즈,메틸 셀를로즈, 나트륨 카르복시메틸셀를로오즈 및 하이드록시프로필 메틸-셀를로즈 등을 포함하는 셀를로즈류, 젤라틴, 폴리비닐피롤리돈 등과 같은 층 전제가 포함될 수 있다. 또한, 경우에 따라 가교결합 폴리비닐피롤리돈, 한천, 알 긴산 또는 나트륨 알기네이트 등을 붕해제로 첨가할 수 있다. 나아가, 본 발명의 약학적 조성물은 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다. 비경구 투여용 제제의 경우에는 주사제, 크림제, 로션제, 외용연고제, 오일 제, 보습제, 겔제, 에어로졸 및 비강 흡입제의 형태로 당업계에 공지된 방½으로 제형화할 수 있다. 이들 제형은 모든 제약 화학에 일반적으로 공지된 처방서인 문 헌 (Remington' s Pharmaceut ical Science , 19th ed. , Mack Publ i shing Company, East on, PA, 1995)에 기재되어 있다. 본 발명의 조성물의 총 유효량은 단일 투여량 (single dose)으로 환자에게 투여될 수 있으며, 다중 투여량 (mult iple dose)으로 장기간 투여되는 분할 치료 방 법 ( fract ionated treatment protocol )에 의해 투여될 수 있다. 투여량은 제제화 방 법, 투여 경로 및 치료 흿수 뿐만 아니라 환자의 연령, 체중, 건강 상태, 성별, 질 환의 중증도, 식이 및 배설율 등 다양한 요인들을 고려하여 환자에 대한 유효 투여 량이 결정되는 것이므로, 이러한 점을 고려할 때 당 분야의 통상적인 지식을 가진 자라면 본 발명의 조성물의 적절한 유효 투여량을 결정할 수 있을 것이다. 식품의 약품안정청 (KFDA)자료에 따르면 통상적으로, 백금계 항암제는 1희 0.1 ~ 250 y M 의 용량으로 투여할수 있다.
백금계 항암제의 유효량은 0.1 ~ 250 μ Μ이 바람직하며 0.5 ~ 200 μ Μ이 더욱 바람직하여 0.625 ~ 160 μ Μ이 가장 바람직하나 이에 한정되는 것은 아니며 환자의 나이, 신장, 중증도, 부위, 배설량에 따라 상기 유효량은 의사에 의해 조정될 수 있다. 한편, 백금계 항암제와 siRNA를 포함하는 약학적 조성물이 단일제제의 형태 로 투여가 될 경우에는 1회 투여량이 상기한 백금계 항암제의 양 또는 그 이하가 되도톡 배합 비율을 적절하게 선택한 제제를 조제하여, 그 배합 제제를 1일에 1회 또는 수회로 나누어 투여할 수 있다. 바람직하게는, 암을 예방 또는 치료하기 위하 여 백금계 항암제는 1회 투여량 0.5 ~ 200 μ Μ 용량으로, ρ53에 대한 유비퀴틴 리 가아제 (ubiquit in l igase)에 대한 siRNA는 1일 0.5 mg ~ 5 mg의 용량으로, 수회 로 나누어 병용 투여할 수 있다.
p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA의 유효량은 0.1 ~ 20 mg/kg 이 바람직하며 0.2 ~ 15 mg/kg 이 더욱 바람직하며 0.4 - 10 mg/kg 이 가장 바람직하나 이에 한정되는 것은 아니며 환자의 나이, 신장, 중증도, 부위, 배설량에 따라 상기 유효량은 의사에 의해 조정될 수 있다. 본 발명에 따른 약학적 조성물은 본 발명의 효과를 보이는 한 그 제형, 투여 경로 및 투여 방법에 특별히 제한되지 아니한다. 본 발명은 또한
(a) 인유두종바이러스 (Human Papi 1 lomarius , HPV)의 E6 및 E7에 대한 siRNA 를 자궁경부암 세포주에 형질도입하는 단계;
(b) 상기 (a) 단계에서 형질도입된 세포주에 자궁경부암 치료제 후보물질을 처리하는 단계 ;
(c) 상기 치료제 후보물질을 처리한 직후 내지 48시간 동안 일정한 시간 간 격마다 세포 내 p53의 발현수준을 측정하는 단계;
(d) 치료제 후보물질을 처리하지 않은 세포와 비교해 세포 내 p53의 발현증 가 지속시간이 연장된 물질을 선별하는 단계를 포함하는 자궁경부암 예방 또는 치 료제 스크리딩 방법을 제공한다. 본 발명의 상기 (a) 단계에서 자궁경부암 세포주에 HPV E6 및 E7에 대한 siRNA를 형질도입하는 방법은 인산칼슘를 이용한 형질감염 (Graham, F.L. 등, Virology, 52 :456(1973) ), DEAE 덱스트란에 의한 형질감염, 미세주사에 의한 형질 감염 (Capecchi , M.R., Cel l , 22:479(1980) ) , 양이온성 지질에 의한 형질감염 (Wong, T.K. 등, Gene, 10:87(1980) ), 전기천공법 (Neumann E. 등, EMBO J . , 1 :841(1982) ) , 형질도입 또는 트랜스펙션과 같이 문헌 (Basic methods in molecular biology, Davis 등, 1986 및 Molecular cloning: A laboratory manual , Davis 등, 1986)에 기재된 바와 같이 본 발명이 속한 기술 분야에서 통상의 지식을 가진 자에게 익히 공지되어 있는 방법들에 따라 제조될 수 있다. 본 발명의 상기 (b) 단계에서 치료제 후보물질이란 세포 내 p53의 발현량에 영향을 미칠 것으로 예상되는 미지의 물질을 의미하며, 항체, 앱타머, 천연추출물 또는 화학물질을 포함하나 이에 제한되는 것은 아니다. 치료제 후보물질을 세포에 처리하는 것은 시험물질을 세포 배양 배지에 추가한 후 세포를 일정 시간 배양하는 것을 의미한다. 상기 단계 (c)에서 p53의 발현은 p53 mRNA 또는 단백질의 발현을 의미하는 것으로, RT-PCR, 경쟁적 RT-PCR(compet i t ive RT-PCR) , 실시간 RT-PCR(Real-t ime RT-PCR) , RNase 보호 분석법 (RPA:RNase protect ion assay) , 노던 블랏팅 (northern blott ing) , DNA 마이크로어레이 칩 분석법, 웨스턴 블럿팅 (Western Blott ing) , 효 소면역분석법 (enzyme- l inked immunosorbent assay) , 방사능면역분석법 (RIA), 방사 면역확산법, 오우크테로니 면역 확산법, 로케트 면역 전기영동, 면역조직화학분석 법, 면역침전법 ( immunoprecipi tat ion) , 보체 고정 분석법, 유세포 분석법 (FACS) 및 단백질 칩 분석법으로 이루어진 군에서 선택된 방법으로 측정할 수 있다. 상기 (c) 단계에서 일정한 시간 간격이란 시간의 경과에 따른 세포 내 p53의 발현량 변화를 관찰하기에 적합한 시간 간격을 의미하는 것으로, 특별히 제한되는 것은 아니나 바람직하게는 30분 내지 1시간일 수 있다. 본 발명의 스크리닝 방법은 상기 (d) 단계 이후에 (e) 선별된 물질의 효과를 세포 또는 동물에서 확인하는 단계를 추가로 포함하는 것을 특징으로 할 수 있다. 그 효과를 세포에서 확인하는 단계란 자궁경부암 세포주에 대한 세포독성을 나타내 는지 여부를 확인하는 것을 의미하며, 그 효과를 동물에서 확인하는 단계란 자궁경 부암 세포주가 이식된 종양 동물모델에서 종양성장 억제효과를 나타내는지 여부를 확인하는 것을 의미한다.
[유리한 효과】
백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 이를 필요로하는 개체에 병용 또는 순차적으로 투여하는 것을 특징으로 하 는 본 발명의 방법에 따르면, 낮은 농도의 백금계 항암제을 처리하여도 세포 내 P53의 증가된 발현 수준을 오랜 시간동안 유지할 수 있어 암 세포의 사멸을 효과적 으로 유도할 수 있을 뿐만 아니라, 백금계 항암제 투여에 따른 약물 부작용을 최소 화 할 수 있어 암의 예방 또는 치료제 개발에 유용하게 사용될 수 있다.

도 1A는 Hela 세포 (왼쪽 패널)와 CaSki 세포 (오른쪽 패널)은 20nM의 HPV E6/E? siRNA로 형질전환 시킨 뒤 ΙΟ μ Μ 시스플라틴에 노출시켰다. 24시간 후, ΤΡ53 의 유도 및 Ε6, Ε7의 효율의 면역 블랏 분석을 위해 전체 세포 용해물을 수집하였 다. β -act in은 대조군으로 사용하였다. .

도 1C는 HPV E6/E7 siRNA를 처리한 종양세포에서 TP53과 과인산화된 RB의 유 도 정도를 TP53 및 E2F 루시퍼라제 리포터 활성을 이용하여 평가한 것이다.
도 1D는 Hela 세포 (위쪽 패널)와 CaSki 세포 (아래쪽 패널)에 E6/E7 특이적 siRNA를 단독 및 /또는 시스플라틴 (CDDP)과 함께 처리하였다. 내인성 TP53, RB와 18E7, 16E7, 16E6 및 18E6의 침묵성 복원은 지정된 시점에서 다른 하위 표적 유전 자와 함께 분석하였다. β -act in은 대조군으로 사용하였다.

도 1F는 He 세포 (왼쪽 패널)와 CaSki 세포 (오른쪽 패널)에 E6/E7 특이적 siRNA 단독 및 /또는 CDDP와 함께 처리한 뒤, 각 세포에서 CDKN1A 전사체 레벨을 Real-t ime quant it ive PCR(qPCR)로 분석하였다. 오차 막대는 독립실험의 mean±SD 를 의미한다. 도 2는 HPV E6/E7 siRNA단독 또는 CDDP와의 병용처리가 TP53의 타겟 유전자 로 잘 알려진 유전자의 발현에 미치는 영향을 실시간 qPCR 분석을 통해 확인한 결 과이다 (A: Hela 세포, B: Caski 세포) . 도 3은 E6/E7결손이 TP53 동태와 세포의 생존에 미치는 영향을 분석한 결과 이다.
도 3A는 GFP-TP53 리포터 구조의 도식 표현을 안정적인 세포 주를 형성하는 데 사용하였다. 특성화된 TP53-RE는 TP53 일치 결합 사이트 (consensus binding site, 녹색 상자)를 포함하고, 화살표는 GFP-리포터 유전자를 갖는 최소한의 TATA box를 나타낸다. IncuCyte를 30분마다 모든 웰의 9개'이미지로 사용하였다. 살아있 는 Hela 세포의 타임랩스 현미경 이미지는 20nM의 SP, ΙΟ μ Μ의 CDDP 단일 치료법 및 SP와 CDDP의 흔합 치료법을 실시하였다. 포스트 형질전환 후 대표적인 이미지의 선택은 대조군으로서 control siRNA처리된 세포와 형광 및 phase-대조 이미지의 병 합 (merge)으로 나타내었다 (살아있는 세포 영상기록의 대표적인 IncuCyte 이미지는 5일 동안 만들었다. 도 3D는 HPV E6/E7 siRNA 단독, CDDP 단독 또는 이들의 결합 반웅하여 TP53 활성 시뮬레이션올 나타낸 것이다. 도 4는 HPV E6/E7 siRNA 단독투여 및 /또는 CDDP와의 병용투여에 대한 GFP- TP53 및 RFP-E2F의 발현을 관찰한 결과이다.
도 4A는 안정적으로 카옌 RFP-E2F 및 GFP-TP53이 형질전환된 He la 세포를 배 양하였고, Hela 세포에서 HPV E6/E7 oncogene의 침묵 효과를 제사하는 GFP-TP53에 RFP-E2F의 시간 의존적 변화에 대해 공초점 현미경 (confocal microscopy)으로 촬영 하였다 (빨간색은 Ex/Em = 565nm/650nm, 녹색은 Ex/Em = 495nm/545nra를 나타낸다. 세포의 형광 이미지는 위상차 이미지에 겹쳐있다) . ^
도 4B는 평균 강도 데이터를 Dual 리포터 안정된 세포의 타임랩스 공초점 이 미지로 획득하였다. 세포는 12시간부터 24시간까지 20분마다 촬영하였다. 특히, 전 이 패턴은 HPV E6/E7 siRNA 풀 (pool ) 형질전환 세포에서 19시간 내지 21시간에서 관찰되었다. 도 5는 3중 병용 (시스플라틴, 과크리탁셀 및 siRNA) 투여의 HPV양성인 자궁 경부암 세포에 대한 효과를 in vitro또는 in vivo로 확인한 결과이다 (CDDP: 시스 플라틴, PTX: 파크리탁셀) .
도 5A는 Hela 세포에 E6/E7 특이적 siRNA, CDDP+파클리탁셀 (pacl itaxel ) 또 는 그들의 흔합물을 처리하였다. 내인성 TP53, 저인산화된 RB hypophosphorylated RB) 및 침묵성 18E7, 16E7, 16E6 및 18E6와 유도는 나타난 시점에서 다른 하위 표 적 유전자와 함께 분석하였다. β -act in은 대조군으로서 사용하였다.
도 5B는 CDKN1A 전사 수준을 Real-t ime quant it ive (q)PCR로 분석하였다. 오 차막대는 독립 실험의 mean士 SD를 나타낸다.
도 5C는 GFP-TP53의 활성에 3가지 흔합물의 효과를 분석하였다. 세포 증식 비율 (왼쪽 상단), GFP 개수 (오른쪽 상단) 및 GFP 강도 (오른쪽 하단)를 나타냈다. 도 5D는 Hela 세포에 HPV E6/E7 siRNA 풀 (pool ) , CDDP 및 CDDP+파클리탁셀 (pacl itaxel ) 또는 그들의 흔합물을 처리하였다. 세포 주기 분석 및 각 단계 (G0- Gl , S 및 G2/M)에서 세포의 비율 (% eel Is)을 측정하였다. 도 6은 HPV에 감염된 자궁경부암 세포에서 HPV E6/E7 oncogene 및 HPV E6/E7 siRNA흔합 치료의 역할을 도면으로 간략하게 나타낸 것이다. 도 7은 HeLa 세포에 시스플라틴 단독, siRNA pool 단독 및 /또는 시스플라틴 을 병용처리했을 때 GFP-TP53의 발현량을 시간경과에 따라 확인한 결과이다.
【발명의 실시를 위한 형태】
이하 본 발명을 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실 시예에 한정되는 것은 아니다.
<실시예 1>
자궁경부암 세포에서 시스폴라틴과 HPV E6/E7 siRNA가 TP53에 미치는 효과 <실시예 1-1>
자궁경부암 세포에서 시스플라틴과 병용처리한 HPV E6/E7 siRNA의 효과 자궁경부암세포와 HPV type 18의 바이러스에 감염된 HeLa 자궁경부암 세포주 (HeLa; ATCC CCL-2) , HPV type 16의 바이러스에 감염된 SiHa(SiHa; ATCC HTB-35) 자궁경부암 세포주를 6-웰 플레이트에 5X 104또는 1 X 105 의 세포수로 분주한 후 37 °C , 이산화탄소 5% 조건하에서 RPMI 1640 또는 DMEM 배지에서 각각 24시간 동안 배 양하였다. siRNA 세포내 도입 (transfect ion)은 DharmaFec Dharmacon, Lafayette , CO, USA)로 수행하였으며, 제조사의 설명에 따라수행하였다.
HPV type 18의 바이러스에 감염된 HeLa 자궁경부암 세포주의 경우에는 426, 450 서열의 siRNA의 각각 및 pool ing하여 형질전환시켰으며, HPV type 16의 바이러 스에 감염된 siHa 세포주의 경우에는 366 , 448 , 497 서열의 siRNA를 각각 및 pool ing하여 형질전환시켰다. 웨스턴 블롯의 경우 Whole-cel l lysate는 RIPA 버퍼 (10 mM TrisCpH .4), 0.15 M NaCl , 5 mM EDTA, 1% Triton X— 100, 0.5% deoxychol ic acid sodium salt 및 0.1% SDS)를 이용하여 추출되었다. 원심분리 후, 상층액의 단백질 농도는 BCA 단백 질 어세이 키트 (Thermo Fisher Scient i f ic Inc)를 이용하여 측정하였다. 단백질 샘 플은 완전한 변성 (denaturation)을 위해 샘플버퍼에 넣고 5분간 끓여졌다. 이 후 샘플들은 6), 10%, 또는 15% 폴리아크릴아마이드겔에 적용했고, 0.4 iim PVDF membraneCMilipore, Bilerica)에 트랜스퍼 하였다. 트랜스퍼 후, 5% 탈지유를 이용 하여 membrane을 블로킹 하였으며, 절절한 농도의 일차 항체 및 horseradish peroxidase— conjugated IgG를 넣고 incubation 하였다. 최종적으로 ECL 용액을 이 용하여 단백질의 발현정도를 검출하였다. 웨스턴 블로팅 실험에 사용한 항체로는 TP53 [D07], HPV 18-E6 [G7] , HPV16-E6 [C1P5] , HPV 18-E7 [F7], HPV16-E7 [ED17] , β -Act in [C4] 경우는 Santa Cruz Biotechnology (St. Louis, CA)사의 항체를 사용 하였다. 자궁경부암 세포에서 시스플라틴 (CDDP)과 결합한 HPV E6/E7 siRNA가 TP53에 미치는 영향을 확인하기 위하여 다음과 같이 실험을 실시하였다.
자궁경부암 세포인 Hela 세포와 CaSki 세포를 HPV E6/E7 siRNA로 형질전환시 킨 뒤 ΙΟμΜ 시스플라틴 (cisplatin)을 처리하여 24시간 동안 배양하였다. 세포 용 해물을 수집한 뒤 당업계에 공지된 방법으로 웨스턴 블랏을 실시하였다.
그 결과 도 1A에 표시된 바와 같이, 시스플라틴을 처리한 군에서 HPV E6/E7 의' 단백질 발현량이 나타나지 않았고, TP53의 발현량이 증가하는 것을 확인할 수 있었다.
<실시예 1-2>
시스플라틴과 HPV E6/E7 siRNA의 세포사멸 효과
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA가 세포 생존능력 에 미치는 효과를 확인하기 위하여 다음과 같이 실험을 실시하였다.
자궁경부암 세포인 Hela 세포와 CaSki 세포를 HPV E6/E7 siRNA로 형질전환시 킨-뒤 ΙΟμΜ 시스플라틴 (cisplatin)을 처리하여 24시간 동안 배양한 뒤 세포 생존 능력을 확인하기 위하여 WST분석방법을 실시하였다.
세포수의 측정은 water soluble tetrazol ium salts (WST) 방법 (EZ—Cytox kit; Daei 1 Lab Service, Seoul , Korea)을 이용하였다. EZ-Cytox solution (50 μ 1)을 12-웰 플레이트의 각 웰에 넣은 후, 2~3시간 동안 배양시킨다. 96-웰 플레이 트에서 GENios Pro microplate reader (Tecan Trading AG, Mannedorf , Switzerland)장비를 사용하여 살아있는 세포를 측정 (485nm) 하였다. 그 결과 도 IB에 나타난 바와 같이, 시스플라틴을 처리하지 않은 군과 비교 하여 처리한 군에서 세포 생존능이 감소하는 것으로 나타났다
<실시예 1-3>
시스플라틴과 HPV E6/E7 siRNA에 의한 TP53의 효소활성 측정
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA가 효소활성에 미 치는 영향을 확인하기 위하여 다음과 같이 실험을 실시하였다.
시스플라틴 (CDDP)과 HPV E6/E7 siRNA를 처리한 Hela 세포와 CaSki 세포에 pTA-TP53-Luc (TP53 reporter) and pTA-E2F-Luc (E2F reporter) 백터를 이용하여 이중 형질전환을 시켰다. 상기의 luci ferase reporter 백터시스템은 Clontech pathway prof i l ing system (Mountain View, CA, USA)사로부터 구매하였다. 이중 형 질전환 시킨 24시간 푸에, Dual -Luc i f erase Reporter assay system (Promega)을 이 용하여 luci ferase assay를 수행하였으며 GENios Pro microplate reader (Tecan Trading AG, Mannedorf , Swi tzerland)장비를 사용하여 luminescence 활성도를 측정 하였다. 그 결과 도 1C에 나타난 바와 같이, Hela 세포와 CaSki 세포에서 시스플라틴 을 처리한 경우에 TP53의 활성이 더 많이 증가하는 것으로 나타났으나 E2F의 활성 은 더 많이 감소하는 것으로 나타났다.
<실시예 1-3>
시스플라틴과 HPV E6/E7 siRNA가 TP53의 발현에 미치는 영향
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA가 효소활성에 미 치는 영향을 확인하기 위하여 다음과 같이 실험을 실시하였다.
상기 실시예 1-1에 기재된 방법과 동일하게 Hela 세포와 CaSki 세포를 배양 하여 형질전환 시킨 뒤 시스플라틴을 처리하였다. 그 다음 세포 용해물을 획득하여 웨스턴 블랏을 질시하였다. 웨스턴 블롯에는 TP53 [D07] , HPV 18-E6 [G7], HPV16- E6 [C1P5] , HPV 18-E7 [F7] , HPV16-E7 [ED17] , E2F-1 [KH195] , Cycl in-E [M20] , β -Act in [C4] 경우는 Santa Cruz Biotechnology (St . Louis , CA)사의 항체를 사용하 였으며 RB [554164]의 경우는 BD Pharmingen사의 항체, Phospho-TP53 (ser-15) [9284]의 경우는 Cel l signal ing사의 항체를 사용하였다. 그 결과 도 ID 및 IE에 나타낸 바와 같이 , 시스플라틴을 처리한 경우 TP53의 발현량이 증가하는 것으로 나타났으며, pRB의 발현량이 증가하는 것으로 나타났다. 또한, 18-E6 및 16-E6, 18-E7 및 16-E7의 발현량은 감소하는 것으로 나타났다 (도 ID, 도 IE) .
<실시예 1-4>
시스플라틴과 HPV E6/E7 siRNA가 CDKN1A와 발현에 미치는 영향
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA가 효소활성에 미 치는 영향을 확인하기 위하여 다음과 같이 실험을 실시하였다.
상기 실시예에 기재된 방법과 동일하게 He la 세포와 CaSki 세포를 배양하여 형질전환 시킨 뒤 시스플라틴을 처리하였다. 그 다음 세포에서 RNA를 추출하여 real-t ime qPCR을 실시하였다.
PCR에 사용된 프라이머와 프로브 서열은 다음과 같으며 모두 TIB M0LBI0L (Berl in, Germany)에서 제작되었다.

Light Cycler Real-t ime PCR Detect ion system (Roche Diagnost ics , Basel , Switzer land) 장비를 사용하여 각각 530 and 705 nm 파장에서 mRNA레벨을 확인하였 다. 그 결과 도 1F에 나타난 바와 같이, Hela 세포 (왼쪽)와 CaSki 세포 (오른쪽) 에 시스플라¾과 E6/E7 siRNA를 함께 처리한 경우에 CDKN1A의 mRNA 발현량이 증가 하는 것을 확인하였다.
<실시예 2>
자궁경부암 세포에서 시스폴라틴과 HPV E6/E7 siRNA가 TP53관련 유전자 발현 에 미치는 영향
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA가 TP53 표적 유전 자의 발현에 미치는 효과를 확인하기 위하여 다음과 같이 실험을 실시하였다.
잘 알려진 TP53 표적 유전자를 세포주기 정지 및 DNA 회복, TP53 발현 조절, 세포사멸 및 노화의 조절 등으로 기능에 따라 분류하였다. 상기 실시예에 기재된 방법과 동일하게 Hela 세포와 CaSki 세포를 배양하여 형질전환 시킨 뒤 시스플라틴 을 처리하였다. 그 다음 세포에서 RNA를 추출하여 real-time qPCR을 실시하였다.
PCR에 사용된 프라이머와 프로브 서열은 하기 표 2와 같으며 모두 TIB MOLBIOL (Berlin, Germany)에서 제작되었다. LightCycler Real-time PCR Detection system (Roche Diagnostics, Basel , Switzerland) 장비를 사용하여 각각 530 and 705 nm 파장에서 mRNA레벨을 확인하였다.

<실시예 3> 자궁경부암 세포에서 시스폴라틴과 HPV E6/E7 siRNA가 세포 사멸에 미치는 효과
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA가 TP53의 활성과 세포 사멸에 미치는 효과를 확인하기 위하여 다음과 같이 실험을 실시하였다.
6-웰 플레이트에 HeLa와 CaSki 세포를 배양하여 형광물질이 표시된 GFP-TP53 백터를 이용하여 형질전환 시켜 안정적인 세포주를 구축하였다. GFP-TP53 백터는 puormycin (Sigma-Aldr ich, St . Louis , MO) 내성 마커를 가지고 있는 lent ivi s시 스템을 활용하였다.
안정적으로 구축된 세포주에 시스플라틴 (CDDP)과 HPV E6/E7 siRNA를 처리하 였다. 그 다음 각' 웰의 살아있는 암세포를 타임랩스 기법을 이용하여 IncuCyte HD system (Essen Instruments , Ann Arbor , Michigan)장비를 통해 5일 동안 촬영하였 다. 또한 각 HPV E6/E7 siRNA에 대한 세포의 증식속도와 TP53을 의미하는 GFP의 개 수 및 강도는 Incucyte ZOOM software (Essen Bioscience)를 사용하여 분석하였다. 그 결과 도 3에서 나타난 바와 같이, Hela 세포에서 siRNA와 시스플라틴을 함께 처리한 경우에 시간이 지날수록 세포 증식속도가 감소하였고, GFP가 결합된 TP53의 개수가 더 증가하는 것으로 나타났다 (도 3A, 3B) . 또한 CaSki 세포에서도 siRNA와 시스플라틴을 함께 처리한 경우에 시간이 지날수록 세포 증식속도가 감소 하였고, GFP가 결합된 TP53의 개수가 더 증가하는 것으로 나타났다 (도 3C) .
<실시예 4>
자궁경부암 세포에서 시스폴라틴과 HPV E6/E7 siRNA의 결합에 반응하는 TP53 및 E2F
자궁경부암 세포에서 시스플라틴 (CDDP)과 HPV E6/E7 siRNA의 결합에 TP53과 E2F의 변화를 확인하기 위하여 다음과 같이 실험을 실시하였다.
96웰 플레이트에서 Hela 세포를 배양하여 GFP가 결함된 TP53과 RFP가 결합된 E2F로 형질전환 시켰고, HPV E6/E7 siRNA 또는 시스플라틴을 처리하였다. 그 다음 HPV E6/E7 siRNA 침묵 효과가 나타나는 Hela 세포에서 시간에 따른 GFP-TP53과 RFP-E2F의 변화를 공초점 현미경 (confocal microscopy)을 이용하여 촬영하였다. 빨 간색은 Ex/Em = 565nm/650nm, 녹색은 Ex/Em = 495nm/545nm에서 촬영하였다. 또한 각 신호의 평균 강도는 12시간부터 24시간까지 20분마다 촬영한 안정된 세포의 타 임랩스 공초점 이미지를 이용하여 관찰하였다. 그 결과 도 4에 나타난 바와 같이, HPV E6/E7 siRNA를 처리하기 전에 비하여 처리한 후에 GFP가 결합된 TP53의 발현량이 증가하고 시간이 지날수톡 초록색 신호 가 강해지는 것을 확인할 수 있었다. GFP가 결합된 TP53의 발현량의 경우, 19~21시 간에서 증가하며, RFP가 결합된 E2F의 발현량의 경우는 반대로 감소하는 것을 확인 할 수 있었다 (도 4A) .
한편, 시스플라틴 (CDDP)과 HPV E6/E7 siRNA의 병용치료의 경우, 12~14시간에 서 GFP가 결합된 TP53의 발현량이 증가하며, RFP가 결합된 E2F의 발현량의 경우는 반대로 감소하는 것을 확인할 수 있었다 (도 4B) .
<실시예 5>
HPV 양성 자궁경부암 세포에서 3증 혼합물의 치료 효과 <실시예 5-1>
in vitro 실험에서 HPV 양성 자궁경부암 세포에 미치는 3중 혼합물의 치료 효과
자궁경부암 세포에서 시스플라틴 (CDDP) , HPV E6/E7 siRNA 및 항암제를 처리 하였을 때 HPV 양성 암세포에 나타나는 치료 효과를 확인하기 위하여 다음과 같이 실험을 실시하였다.
Hela 세포에 HPV E6/E7 siRNA, 시스플라틴, 항암제인 파클리탁셀 (pacl itaxel , PTX)을 처리하였다. 그 다음 세포 용해물을 수득하여 웨스턴 블랏을 실시하였고, RNA를 추출하여 RT_qPCR을 실시하였다. 프라이머와 프로브 서열은 다 음과 같으며 모두 TIB M0LBI0L (Berl in, Germany)에서 제작되었다.

LightCycler Real-t ime PCR Detect ion system (Roche Diagnost ics , Basel , Switzer land) 장비를 사용하여 각각 530 and 705 nm 파장에서 mRNA레벨을 확인하였 다.
또한 GFP가 결합된 TP53을 이용하여 IncuCyte HD system (Essen Instruments , Ann Arbor , Michigan)장비를 통해세포 증식 비율, GFP 개수 및 강도 를 측정하였다. 또한 Hela 세포에 HPV E6/E7 siRNA, 시스플라틴 및 시스플라틴+파 클리탁샐 (pacl i taxel ) 또는 그들의 흔합물을 처리한 뒤, FACS .assay를 통해 세포 주기 분석 및 각 단계 (G0-G1 , S 및 G2/M)에서 세포의 비율 (% eel Is)을 측정하였다. 그 결과를 도 5A 내지 5D에 나타냈다.
Hela 세포에 HPV E6/E7 siRNA, 시스플라틴, 파클리탁셀을 처리한 경우에 시 간에 따라 TP53의 단백질 발현량이 증가하였으나, 18-E6와 18-E7의 단백질 발현량 이 감소하는 것으로 나타났다 (도 5A) . 또한, Hela 세포에 HPV E6/E7 siRNA 001 (SP, 20 nM) , 시스폴라틴 (5 μ Μ) , 파클리탁셀 (10 nM)을 함께 처리한 경우에 CDKN1A의 mRNA 발현량이 증가하는 것을 확인하였다 (도 5B) . 또한 GFP-TP53으로 형 질전환시킨 Hela 세포에 3가지 흔합물을 처리한 경우에 세포 증식이 억제되고, GFP 의 개수가 증가하며, GFP의 강도가 강해지는 것으로 나타났다 (도 5C) . 한편, 대조 군에 비하여 Hela 세포에 HPV E6/E7 siRNA Pool (SP, 20 nM) , 시스플라틴 (5 μ Μ) , 파클리탁셀 (10 nM)을 처리한 경우에 Sub-Gl기 세포의 비율이 높고, G0/G1기 세포 의 비율이 낮은 것으로 나타났다. 이를 통해, HPV E6/E7 siRNA, 시스플라틴, 파클 리탁셀 흔합물은 G1기와 G2/M기 세포주기를 억제하는 것을 확인할 수 있었다.
【산업상 이용가능성】
백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 이를 필요로하는 개체에 병용 또는 순차적으로 투여하는 것을 특징으로 하 는 본 발명의 방법에 따르면, 낮은 농도의 백금계 항암제를 처리하여도 세포 내 p53의 증가된 발현 수준을 오랜 시간동안 유지할 수 있어 암 세포의 사멸을 효과적 으로 유도할 수 있을 뿐만 아니라, 백금계 항암제의 투여에 따른 약물 부작용을 최 소화 할 수 있어 암의 예방 또는 치료제 개발에 유용하게 사용될 수 있어 산업상 이용가능성이 매우 우수하다.

백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiquitin ligase)에 대한 siRNA를 이를 필요로 하는 개체에 병용 또는 순차적으로 투여하는 것을 특징으로 하는 세포에서 증가된 수준 (elevated level)의 p53을 유지시키는 방법.
【청구항 2]
제 1항에 있어서, 상기 백금계 항암제는 시스플라틴 (cis- diamminedichloroplatinum [II]), 카보플라틴 (carboplat in), 옥살리플라틴 (oxaliplatin), 네다플라틴 (nedaplatin) , 피코플라틴 (picoplatin), 트리플라틴 테 트라나이트레이트 (triplatin tetranitrate), 사트라플라틴 (satraplat in) 및 이들 의 흔합물로 이루어진 군에서 선택되는 것을 특징으로 하는 방법.
【청구항 3】
제 1항에 있어서, 상기 유비퀴틴 리가아제는 인유두종바이러스 (Human
Papillomarius, HPV)의 E6/E6-AP 복합체, E6, E6-AP, 인간 HDM2, Pirh2 및 C0P1 로 이루어진 군에서 선택된 것임을 특징으로 하는 방법.

제 1항에 있어서, 상기 세포는 상기 개체의 암세포인 것을 특징으로 하는 방 법.

제 1항에 있어서, 상기 방법은 상기 세포의 사멸을 유도하는 것을 특징으로 하는 방법 .

제 5항에 있어서, 상기 세포의 사멸은 아품토시스 (apoptosis) 경로를 통해 이루어지는 것을 특징으로 하는 방법.

제 1항에 있어서, 상기 siRNA는 서열번호 1 내지 10으로 이루어진 군에서 선 택되는 것을 특징으로 하는 방법
【청구항 8】
백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 유효성분으로 포함하는, 세포에서 p53의 수준을 증가된 수준 (elevated level )으로 유지시키는 것에 의한 세포 사멸 촉진용 조성물.

제 8항에 있어서, 상기 세포는 암세포인 것을 특징으로 하는 조성물.
【청구항 10】
백금계 항암제 및 p53에 대한 유비퀴틴 리가아제 (ubiqui t in l igase)에 대한 siRNA를 유효성분으로 포함하는, 암세포에서 p53의 수준을 증가된 수준 (elevated level )으로 유지시키는 것에 의한 암 예방 또는 치료용 약학적 조성물.
【청구항 111
(a) 인유두종바이러스 (Human Papi 1 lomarius , HPV)의 E6 및 E7에 대한 siRNA 를 자궁경부암 세포주에 형질도입하는 단계;
(b) 상기 (a) 단계에서 형질도입된 세포주에 자궁경부암 치료제 후보물질을 처리하는 단계;
(c) 상기 치료제 후보물질올 처리한 직후 내지 48시간 동안 일정한 시간 간 격마다 세포 내 p53(tumor protein 53)의 발현수준을 측정하는 단계;
(d) 치료제 후보물질을 처리하지 않은 세포와 비교해 세포 내 p53(tumor protein 53)의 발현증가 지속시간이 연장된' 물질을 선별하는 단계를 포함하는 자궁 경부암 예방 또는 치료제 스크리닝 방법.
【청구항 12]
제 11항에 있어서, 상기 (d) 단계 이후에 (e) 선별된 물질의 효과를 세포 또 는 동물에서 확인하는 단계를 추가로 포함하는 것올 특징으로 하는 스크리닝 방법.

제 11항에 있어서, 상기 단계 (c)에서 p53의 발현은 RT-PCR, 경쟁적 RT- PCRCcompetitive RT-PCR) , 실시간 RT-PCR(Real-t ime RT-PCR) , RNase 보호 분석법 (RPA:RNase protection assay) , 노던 블랏팅 (northern blotting), DNA 마이크로어 레이 칩 분석법, 웨스턴 블럿팅 (Western Blotting), 효소면역분석법 (enzyme- 1 inked i匪 unosorbent assay) , 방사능면역분석법 (RIA), 방사면역확산법, 오우크테로니 면 역 확산법, 로케트 면역 전기영동, 면역조직화학분석법, 면역침전법 (immunoprecipitation), 보체 고정 분석법, 유세포 분석법 (FACS) 및 단백질 칩 분 석법으로 이루어진 군에서 선택된 방법으로 측정하는 것을 특징으로 하는 스크리닝 방법 ·

제 11항에 있어서, 상기 siRNA는 서열번호 1 내지 10으로 이루어진 군에서 선 택되는 것을 특징으로 하는 스크리닝 방법.
