Abstract

본 발명은 연속적인 파장의 백색광 대신 레드(Red), 그린(Green), 블루(Blue) 좁은 영역의 파장을 가지는 세가지 단색광을 섞어 백색광을 만들고, 형광물질에 적합한 형광여기광원을 추가하여 혼합광을 만드는 형광영상용 광원을 제공하고, 이를 통해 일반영상과 형광 영상을 동시에 촬영할 수 있는 형광 영상 시스템에 관한 것으로, 본 발명에 따른 형광 영상 시스템은 레드(Red), 그린(Green), 블루(Blue)의 단색광을 혼합하여 백색광을 생성하는 단색광원 및 형광영상을 위한 형광여기광원을 포함하며, 단색광과 형광이 혼합된 혼합광을 제공하는 혼합광원 및 상기 혼합광에서 가시광과 형광의 파장을 분리하여, 일반 영상, 형광영상과 혼합 영상을 촬영하는 영상촬영장치를 포함하는 것을 기술적 특징으로 한다.

Description

형광 영상 시스템
본 발명은 연속적인 파장의 백색광 대신 레드(Red), 그린(Green), 블루(Blue) 좁은 영역의 파장을 가지는 세가지 단색광을 섞어 백색광을 만들고, 형광물질에 적합한 형광여기광원을 추가하여 혼합광을 만드는 형광영상용 광원을 제공하고, 이를 통해 일반영상과 형광 영상을 동시에 촬영할 수 있는 형광 영상 시스템에 관한 것이다.
일반적으로 내시경, 복강경 및 영상장치들은 적절한 광원과 물체에 반사된 빛을 획득하는 렌즈와 센서(카메라)로 구성된다. 백색광 광원에서 조사된 빛이 물체에 반사되고 이를 검출하는 촬영장치가 일반적인 내시경, 복강경 및 기타 영상시스템이다. 여기에 추가하여 조사된 특정 파장의 빛이 물체에서 특정 파장의 형광을 일으키고 이 형광을 촬영하는 형광촬영장비는 질환을 특이적으로 진단하는 데 도움이 되기 때문에 점차 널리 사용되고 있다.
형광을 일으키는 물질은 생체내부에 존재하는 경우와 외부에서 투여하는 경우가 있다. 생체에 존재하는 형광물질에 의한 형광은 자가형광(autofluorescence)라 하며 생체의 상태나 질환에 따라 이러한 자가형광물질이 변화할 수 있어 자가형광의 변화로 이어지며, 이를 측정하면 병의 진단에 도움이 될 수 있다. 외부에서 필요에 따라 체내로 주사하는 형광도 생체의 상태 등에 따라 분포가 변화하므로 이 형광의 변화를 측정하여 이용할 수 있다.
내시경, 복강경 및 형광을 활용하는 영상장치에서, 형광영상은 일반적으로 보조적인 정보만을 제공하고, 오히려 일반반사광영상이 기본적인 영상이다. 따라서 일반반사광영상이 더 많이 사용되고 형광영상만 필요한 경우는 드물다. 이상적인 경우 형광영상을 촬영하는 경우도 일반반사광영상이 동시에 촬영되는 것이 유리하다. 예를 들어 종양을 절제하는 수술에서 형광영상장치를 이용하는 경우를 고려하면, 종양을 보여주는 형광영상을 참조하더라도 종양이 놓여있는 주변조직의 혈관이나 다른 정상조직을 상태를 동시에 보아야 안전하게 종양을 떼어낼 수 있는 것이다.
종래 다중모드 광원과 다중모드카메라를 이용하여 칼라일반영상, 반사영상, 형광영상을 얻는 경우 mirror등을 이용하여 광원과 광로를 바꾸는 기계적 방법이나 미국 등록특허 제 7,722,534호에 개시된 바와 같이, 빔스플리터 등을 이용하는 광학적 방법으로 각 영상 혹은 모드별로 따로따로 얻고 있다.
각 영상을 따로 얻는 경우, 기계적 방법의 경우 구조가 복잡해지고 기계적인 부분의 안전성이나 내구성이 좋지 못할 가능성이 있고, 시분할로 영상을 따로따로 얻으므로 수광량이 줄어들게 된다. 광학적인 방법에서도 빔스플리터 등을 이용하는 경우 역시 빛을 나누므로 수광량이 줄어들게 된다. 기존의 방법이 이런 복잡한 방식을 이용하는 것은 연속적인 파장을 이용하는 백색광을 일반영상의 광원으로 이용하는 한, 백색광의 파장과 형광영상의 방출광의 파장이 겹쳐서 물리적으로 일반영상과 형광영상을 동시에 획득하는 것은 불가능하기 때문이다.
즉, 종래 일반영상과 형광영상을 동시에 얻는 것이 불가능한 근본적인 원인은 일반영상광원이 약350 ~ 750 nm 사이의 가시광선영역 파장의 빛을 모두 포함하고 경우에 따라 750 nm이상의 적외선영역의 빛도 모두 포함하기 때문이다. 피사체에서 방출되는 형광의 파장의 빛이 일반영상광원의 반사광과 섞이기 때문이다.
본 발명은 상기와 같은 종래의 문제점을 해결하기 위하여 제안된 것으로, 본 발명에 따른 형광 영상 시스템의 목적은 연속적인 파장의 백색광 대신에 레드, 그린, 블루의 좁은 파장영역의 단색광을 혼합하여 혼합백색광을 생성하고, 여기에 형광물질에 적합한 형광여기광원을 추가한 광원을 만들고, 이 광원을 피사체에 조사하여 발생하는 반사광과 형광은 일반적인 연속적 파장의 빛이 아니라, 조사된 단색광과 형광 파장이 혼합된 것이므로, 단색광 미러 등 광학적 방법으로 분리 가능하므로, 일반 영상과 형광 영상이 동시에 촬영 가능한 시스템을 제공할 수 있는 데 있다.
본 발명에 따른 형광 영상 시스템은 레드(Red), 그린(Green), 블루(Blue)의 단색광을 혼합하여 백색광을 제공하고, 혼합된 백색광에 형광을 제공하기 위한 형광여기광원이 일체형으로 형성되어, 백색혼합광과 형광여기광이 함께 제공되는 혼합광원, 상기 혼합광원에서 상기 레드(Red), 그린(Green), 블루(Blue)의 단색광 중 적어도 어느 하나의 단색광의 파장을 분리하는 단색광 미러, 상기 형광 파장을 분리하여 형광영상 촬영장치에 전달하는 형광 미러, 상기 단색광 미러에서 분리된 단색광으로 일반 영상을 촬영하는 일반영상 촬영 장치 및 상기 형광 미러에서 방출된 형광으로 형광 영상을 촬영하는 형광영상 촬영장치를 포함하는 것을 특징으로 한다.
또한, 본 발명에 따른 혼합광원, 일반영상 촬영장치와 형광영상 촬영장치는 기본적으로 레드(Red), 그린(Green), 블루(Blue)의 단색광 파장을 각각 분리하는 레드광 미러, 그린광 미러, 블루광 미러를 포함하고, 이들 앞, 뒤, 혹은 사이에 형광파장에 따라 형광미러를 포함하는 것을 특징으로 한다. 하지만 형광의 파장에 따라 각각의 단색광미러는 일부 생략되거나 통합하여 구성할 수 있다.
또한, 특수한 예로 본 발명에 따른 형광 영상 시스템에 있어서, 단색광 미러는 상기 형광의 파장이 가시광 보다 장파장인 경우, 레드 광의 파장을 분리하는 레드광 미러로 형성되고 그린광 미러, 블루광 미러는 생략할 수 있는 것을 특징으로 한다.
또한, 특수한 예로 본 발명에 따른 형광 영상 시스템에 있어서, 단색광 미러는 상기 형광의 파장이 블루광 보다 단파장인 경우, 형광의 파장을 분리한 형광 미러로 형성되고, 레드광 미러, 그린광 미러, 블루광 미러는 생략할 수 있는 것을 특징으로 한다.
본 발명에 따른 형광 영상 시스템은 연속적인 파장의 백색광 대신에 레드, 그린, 블루의 단색광을 혼합하여 백색광을 생성하고, 형광물질에 적합한 형광여기광원을 추가한 혼합광원과 촬영시 레드, 그린, 블루의 단색광과 형광을 분리하여 동시에 촬영함으로써, 일반적인 백색광이 포함하는 380~780nm 영역의 연속적인 파장과 형광의 파장이 겹쳐지는 현상을 방지할 수 있으며, 이를 통해 기존의 시분할 장치나 기계적 장치 없이 일반 영상과 형광 영상이 동시에 촬영 가능한 시스템을 제공할 수 있는 효과가 있다.
또한, 백색광을 만드는 단색광은 필요에 따라 변화시킬 수 있어 거의 제한없이 형광여기광원의 파장을 정할 수 있어 모든 파장의 형광물질에 제한없이 사용할 수 있는 시스템을 구성할 수 있고, 광학장치의 정밀도에 따라 제한될 수 있기는 하지만 동시에 사용가능한 형광물질의 개수도 제한되지 않아 다수개의 다중스펙트럼영상촬영(multispectral)도 가능하다.
또한, 복수 개의 다이크로익 미러와 필터를 혼합 배치하여, 다수 개의 형광 영상을 동시에 획득할 수 있어 복강경 등에 한정되지 않고, 광의료기기 전반에 적용이 가능한 장점이 있다.

이하, 본 발명에 따른 형광 영상 시스템을 실시하기 위한 구체적인 내용을 설명하면 다음과 같다.
본 발명에 따른 형광 영상 시스템은 크게 혼합광원(50) 및 영상촬영장치(100)으로 구성되며, 상기 혼합광원(50)은 상기 레드(Red), 그린(Green), 블루(Blue)의 단색광을 혼합하여 백색광을 생성하는 단색광원 및 형광영상을 위한 형광여기광원을 포함하며, 단색광과 형광이 혼합된 혼합광을 제공한다.
도 1은 본 발명에 따른 상기 혼합광원(50)의 구성을 나타내는 도면으로, 컨트롤러(10)와 연결되어, 구동되는 레드광원(1), 그린광원(2), 블루광원(3)에서 각각 레드(R), 그린(G), 블루(B) 단색광을 생성하며, 본 발명에 있어서, 상기 레드광원(1), 그린광원(2), 블루광원(3)는 좁은 파장 영역을 갖는 레이저(laser)나 LED를 이용하여, 단색광을 얻거나 연속된 파장의 백색광에 광학필터를 추가하여, 좁은 파장 영역의 RGB 단색광을 얻는다.
보다 자세히 설명하면, 도 2에 도시된 바와 같이, 자외선과 적외선의 사이에 끼어있는 380~780nm의 파장의 빛을 인간이 인식하게 된다. 인간의 눈은 이 중에도 레드, 그린, 블루의 빛을 선택적으로 느낀다고 여겨지며, 이 색의 조합을 통해 모든 색을 뇌에서 인식할 수 있다. 또한, 도 3에 도시된 CIE 색좌표상에서 세가지 색상의 좌표로 만들어지는 삼각형내의 빛은 세가지 색상의 빛의 조합을 이용하여 얻을 수 있다. 즉 레드, 그린, 블루의 광은 파장이 일부 달라도 적절한 조합을 통하여 백색광을 만들 수 있다. 2개이상의 레드, 그린, 블루의 광 조합을 이용하는 경우 형광여기파장, 형광방출파장과 겹치지 않는 백색광광원을 만들 수 있는 것이다.
다양한 백색광을 구현하기 위해 여러 종류의 단색 LED 칩을 이용하는 경우, 반치폭인 20nm이하로 매우 좁은 단색광원이어서 이들 조합을 통해 다양한 색좌표를 구현할 수 있고 따라서 백색광도 얻을 수 있다. 일반적으로 R+G+B LED를 이용하는 백색을 만드는 경우 연색성이 나쁘지는 않으나 이 발명에서는 좁은 영역의 단색광원을 활용하므로 일반적인 경우에 비해 고 연색성(Color Rendering Index)의 자연백색을 확보하기는 어려울 수 있다. 하지만 경우에 따라 3개이상의 단색광을 혼합하는 등 적절한 단색광을 혼합한다면 연색성을 향상시킬 수 있고 백색광과 함께 교대로 이용하는 방법으로 단점을 보완할 수 있다.
레드, 그린, 블루 광은 고정된 파장범위를 가지지는 않으며, 이들을 적절히 혼합하면 백색을 얻을 수 있다. 본 발명에 따른 혼합광원(50)은 590-750 nm의 레드광, 480 -570 nm의 그린광, 370-460nm 블루광을 조합하여 백색광을 생성하며, 다음과 같은 다양한 조합을 가질 수 있다.
조합 실시예1 - BGR 460nm + 510nm + 610nm
조합 실시예2 - BGR 480nm + 540nm + 700nm
또한, 본 발명에 있어서, 상기 혼합광원(50)은 상기 레드(Red), 그린(Green), 블루(Blue)의 단색광으로 백색광을 혼합 시, 색온도를 조절하기 위한 강도 조절 장치를 더 포함하는 것이 바람직하다.
상기 레드광원(1), 그린광원(2), 블루광원(3)에서 발생한 단색광은 다이크로익 미러로 구현된 단색광 미러(21, 22, 23)를 통해 반사되어, 혼합장치(51)에 전달된다.
또한, 본 발명에 따른 혼합광원(50)은 특정 형광 물질에 적용되는 적어도 하나 이상의 형광여기광원(41, 42, 43, 44)을 더 포함하며, 상기 형광여기광원(41, 42, 43, 44)는 각각 광학필터를 장착하며, 여기광 파장에 따라 광원측 형광 미러(31, 32, 33)를 배치한다.
이러한 본 발명에 따른 혼합광원으로 연속적인 파장의 백색광 대신 레드, 그린, 블루 좁은 영역의 파장을 가지는 세가지 단색광을 섞어 백색광을 만들고, 형광물질에 적합한 형광여기광원을 추가하여 백색광과 형광여기광을 혼합한 혼합광을 제공할 수 있는 것이다.
상기 영상촬영장치(100)는 상기 혼합광원(50)의 혼합광에서 가시광과 형광의 파장을 분리하여, 일반 영상, 형광 영상, 혼합 영상을 촬영하며, 본 발명에 따른 상기 영상촬영장치(100)는 단색광 미러, 촬영장치측 형광 미러, 일반영상 촬영 장치 및 형광영상 촬영 장치를 포함한다.
상기 단색광 미러는 상기 혼합광원(50)에서 전달된 혼합광에서 상기 레드(Red), 그린(Green), 블루(Blue)의 단색광 중 적어도 어느 하나의 단색광의 파장을 분리하며, 일반영상의 촬영 장치의 위치에 따라 복수 개로 형성될 수 있다.
본 발명의 실시예에서 상기 단색광 미러는 한 쌍의 블루광 미러(71, 72), 그린광 미러(73, 74), 레드광 미러(75, 76)을 포함하여, 각 단색광의 파장에 따라 분리하여 반사하게 된다.
또한, 본 발명에 따른 단색광 미러는 형광의 파장에 따라 각각의 단색광미러는 일부 생략되거나 통합하여 구성할 수 있다.
상기 촬영장치측 형광 미러(61, 62, 63)는 형광 파장을 분리하여 형광영상 촬영장치에 전달하며, 본 발명의 실시예에서는 총 3개의 형광 미러(61, 62, 63)로 구성하였다.
상기 일반영상 촬영 장치(81)은 상기 블루광 미러(71, 72), 그린광 미러(73, 74), 레드광 미러(75, 76)에서 분리된 단색광을 혼합하여 일반 영상을 촬영하며, 상기 형광영상 촬영장치(82, 83, 84, 85)는 상기 형광 미러(61, 62, 63)에서 방출된 형광으로 형광 영상을 촬영하며, 적용되는 형광 물질의 수에 따라 복수 개로 형성된다.
도 5는 본 발명에 따른 형광 영상 시스템의 제1사용 상태를 나타내는 도면으로, 구체적으로 인도시아닌그린(indocyanine green)에 적용되는 형광 영상과 일반 영상을 촬영하는 경우에 해당할 수 있다.
상기 인도시아닌그린의 여기 광원 파장은 780 nm 이며, 방출 파장은 850 nm 으로, 형광파장이 가시광보다 장파장이며, 이 경우, 상기 영상촬영장치(100)은 단일의 레드광 필터(75)만을 구비할 수 있으며, 상기 레드광 필터(75)를 통해 R, G, B가 혼합된 백색광은 일반영상 촬영장치(81)에 전달되며, 상기 레드광 필터(75)에서 방출된 형광은 형광영상 촬영장치(85)에 전달되어, 일반영상과 형광영상을 동시에 촬영할 수 있다.
도 6은 본 발명에 따른 형광 영상 시스템의 제2사용 상태를 나타내는 도면으로, 형광의 방출 파장이 레드광과 그린광 사이로 형광 미러(84)는 레드광 미러(75) 및 그린광 미러(73) 사이에 위치한다. 상기 백색광 조합 실시예2로 시스템을 구성하는 경우, 5-ALA에 적용되는 형광 영상과 일반 영상을 동시에 촬영한다. 5-ALA는 여기 파장이 380~440 nm 로 여기 광원은 상기 혼합광원에서 블루 광원(3)보다 짧은 파장구간에 배치되고, 방출 파장은 635 nm 로 형광 미러(63)는 레드광 미러(75)와 그린광 미러(73)사이 파장 구간에 배치된다.
도 7은 본 발명에 따른 형광 영상 시스템의 제3사용 상태를 나타내는 도면으로, 구체적으로 플루오레세인(fluorescein)에 적용되는 형광 영상과 일반 영상을 동시에 촬영하는 경우에 적용될 수 있다. 상기 플루오레세인은 여기 광원 파장이 494 nm 로, 상기 백색광 조합 실시예2로 시스템을 구성하는 경우 여기 광원은 상기 혼합광원에서 그린 광원(2)과 블루 광원(3) 사이에 배치되고, 방출 파장은 521 nm 로 형광 미러(62)는 그린광 미러(73)과 블루광 미러(71) 사이에 배치된다.
도 8은 본 발명에 따른 형광 영상 시스템의 제4사용 상태를 나타내는 도면으로, 형광의 방출 파장이 블루광보다 짧은 파장의 형광만을 이용하는 경우 이처럼 형광 미러(61) 이외의 단색광 미러는 모두 생략가능하다.
도 9는 본 발명에 따른 형광 영상 시스템에 있어서, 두 가지 형광 물질에 적용되는 혼합광원(50) 및 영상촬영장치(100)의 개념을 나타내는 도면으로, 예를 들어 5-ALA와 인도시아닌그린에 적용되는 형광 형상과 일반영상을 동시에 촬영할 수 있다.
이상에서 설명한 바와 같이, 본 발명에 따른 형광 영상 시스템을 적용 시, 연속적인 파장의 백색광 대신에 레드, 그린, 블루의 단색광을 혼합하여 백색광을 생성하고, 형광물질에 적합한 형광여기광원을 추가함으로써, 백색광과 형광의 파장이 겹쳐지는 현상을 방지할 수 있으며, 이를 통해 일반 영상과 형광 영상이 동시에 촬영 가능한 시스템을 제공할 수 있다.
이상 본 발명의 실시예로 설명하였으나 본 발명의 기술적 사상이 상기 실시예로 한정되는 것은 아니며, 본 발명의 기술적 사상을 벗어나지 않는 범주에서 다양한 본 발명에 따른 형광 영상 시스템으로 구현할 수 있다.

  1. 레드(Red), 그린(Green), 블루(Blue)의 단색광을 혼합하여 백색광을 생성하는 단색광원 및 형광영상을 위한 형광여기광원을 포함하며, 단색광과 형광이 혼합된 혼합광을 제공하는 혼합광원 및
    상기 혼합광에서 가시광과 형광의 파장을 분리하여, 일반 영상, 형광영상과 혼합 영상을 촬영하는 영상촬영장치를 포함하는 것을 특징으로 하는 형광 영상 시스템.
  2. 제1항에 있어서,
    상기 형광의 파장이 가시광 구간인 경우,
    상기 영상촬영장치는,
    상기 혼합광원에서 레드(Red), 그린(Green), 블루(Blue)의 단색광 파장을 각각 분리하는 레드광 미러, 그린광 미러, 블루광 미러를 포함하는 단색광 미러;
    상기 형광 파장을 분리하여 형광영상 촬영장치에 전달하는 형광 미러;
    상기 단색광 미러에서 분리된 단색광을 혼합하여 일반 영상을 촬영하는 일반영상 촬영 장치 및
    상기 형광 미러에서 분리된 형광으로 형광 영상을 촬영하는 형광영상 촬영장치를 포함하는 것을 특징으로 하는 형광 영상 시스템.
  3. 제1항에 있어서,
    상기 형광의 파장이 가시광 보다 장파장인 경우,
    상기 영상촬영장치는,
    상기 혼합광원에서 레드 광보다 짧은 파장의 빛을 모두 분리하는 레드광 미러로 형성된 단색광 미러;
    상기 레드광 미러에서 분리된 가시광으로 일반 영상을 촬영하는 일반영상 촬영 장치 및
    상기 레드광 미러에서 방출된 형광으로 형광 영상을 촬영하는 형광영상 촬영장치를 포함하는 것을 특징으로 하는 형광 영상 시스템.

    상기 형광의 파장이 블루광 보다 단파장인 경우,
    상기 영상촬영장치는,
    상기 혼합광원에서 블루광보다 짧은 형광과 백색광을 분리하는 형광영상 촬영장치에 전달하는 형광 미러;
    상기 형광 미러에서 방출된 가시광으로 일반 영상을 촬영하는 일반영상 촬영 장치 및
    상기 형광 미러에서 분리된 형광으로 형광 영상을 촬영하는 형광영상 촬영장치를 포함하는 형광 영상 시스템.
  5. 제1항에 있어서,
    상기 혼합광원은
    590-750 nm의 레드광, 480 -570 nm의 그린광, 370-460nm 블루광을 조합하여 백색광을 생성하는 것을 특징으로 하는 형광 영상 시스템.
  6. 제1항에 있어서,
    상기 형광여기광원은
    인도시아닌그린(indocyanine green), 플루오레세인(fluorescein), 5-ALA 중 적어도 어느 하나 이상의 형광 물질에 적용되는 것을 특징으로 하는 형광 영상 시스템.
  7. 제1항에 있어서,
    상기 혼합광원은
    상기 레드(Red), 그린(Green), 블루(Blue)의 단색광으로 백색광을 혼합 시, 색온도를 조절하기 위한 강도 조절 장치를 더 포함하는 것을 특징으로 하는 형광 영상 시스템.
