  • Non-Patent Documents 1 and 2 There is a risk of failing to output a certain difference, and the fact is that it is close to semi-quantitative rather than quantitative. This is due to the development of means to properly correct the error factors that affect the data obtained in the ChIP-seq method, the acquisition of appropriately corrected quantitative data by that means, and multiple experiments. Appropriate quantitative comparative analysis with appropriately corrected quantitative data indicates that the observed variation is necessary and important to understand that the variation is biologically meaningful.
  • Patent Document 2 As a method for quantifying a target protein contained in a biological sample, a method using an internal standard peptide has been developed (Patent Document 2). In this method, a peptide that binds to a target protein is synthesized, and a known amount is added to a biological sample as an internal standard. For example, by evaluating the interaction between a transcription factor that controls the expression of an oncogene and its binding sequence by chromatin immunoprecipitation, the prediction and evaluation of a disease state, and further the therapeutic effect can be examined. Moreover, the relationship between transcription control by a DNA binding protein such as a transcription factor and a disease can be examined by comparing using a sample derived from a patient and a sample derived from a healthy person. Furthermore, the relationship between transcriptional control by a DNA-binding protein such as a transcription factor and gene mutation can be examined by comparing a subject having a gene mutation and a subject having no mutation using each sample.

Description

Rev. Genom. Human Genet. (2008) vol.9, p.387-402およびPersson et al., Chem. Soc. Rev. (2010) vol.39, p.985-999)。 他にもNanopore(Oxford Nanopore Technologies)等が知られている。
 ウイルスベクターpMX-puro(東京大学医科学研究所のDr.T.Kitamuraより分与を受けた。)

 (a):ビオチン化酵母DNA(濃度10ng/μL)の添加量(μL)

 Irf4とその類似因子Irf8の両方を欠損するマウス(dKO、Dr.R.Sciammas(Houston Methodist)より供与された。)
<2-2.ChIP-Seq>
