Abstract

본 발명은 비수용성 여부를 판단하려는 물질의 화학구조 정보를 바탕으로 용해도 상수 (solubility parameter)를 계산하고, 정립된 정량적인 기준을 통해 비수용성 여부를 최종적으로 판단하며, 대상 물질이 실제로 합성되기 전에도 비수용성인지 아닌지를 판단할 수 있는 비수용성 물질 평가방법을 제공한다.

비수용성 물질 평가 방법 및 평가 시스템
본 출원은 2014년 12월 23일자 한국 특허 출원 제10-2014-0186680호에 기초한 우선권의 이익을 주장하며, 해당 한국 특허 출원의 문헌에 개시된 모든 내용은 본 명세서의 일부로서 포함된다.
본 발명은 비수용성 물질 평가 방법 및 평가 시스템에 관한 것이다.
유기화합물의 여러 물성의 정확한 값을 구체적으로 아는 것은 그 물질의 용도의 타당성을 검토하거나 합성 및 정제 과정을 설계하고 보관, 운반, 사용, 폐기의 방법과 조건을 설정하는 등, 생산과 소비의 전 과정에 걸친 제반 의사결정 사항들에 결정적이기 때문에 산업적으로나 학문적으로 매우 중요한 문제이다. 관심 있는 유기화합물의 관심 있는 물성의 값을 가장 정확하게 알 수 있는 방법은 역시 실험이겠으나 정제된 시료의 준비, 정확한 측정을 위한 환경의 구축 등 여러 가지 측면에서 상당한 비용과 시간이 드는 것이 사실이며 경우에 따라서는 불가능할 수도 있다. 따라서, 그 대안으로 오래전부터 많은 연구자들이 유기화합물의 여러 물성의 정확한 값을 예측하고자 노력을 기울여 왔다. 이처럼 물성 예측은 오랜 역사를 가지며 끊임없이 새로운 예측 방법들이 등장하여 현재는 물성별로 정확도와 적용범위 등이 서로 다른 여러 예측 모형들이 공존하고 있는 상황이다.
한편, 용액을 사용하는 공정을 바탕으로 한 신소재 개발 및 분리 방법 개발에서는 대상 물질인 용질이 물에 대해 용해되지 않는 특성인 비수용성인지 아닌지를 정확하게 판단하는 것은 매우 중요하다.
이와 같이, 비수용성 여부가 중요한 이유는 용액을 구성하는데 가장 빈번하게 사용되는 용매가 바로 물이기 때문이다. 또한 물에 잘 용해되는 수용성 물질은 외부에 배출되었을 경우 물과 분리가 잘 되지 않아 환경적인 문제를 발생시킬 수 있기 때문에 대상 물질의 비수용성 여부를 정확하게 판단해야 할 필요가 있다.
현재까지, 비수용성 물질 여부를 판단하기 위해서 사용되고 있는 방법은 대상 물질을 직접 물에 용해시키는 용해도 실험을 수행하는 것이다. 하지만 소재 개발에서 테스트되는 수많은 물질들에 대해 용해도 실험을 수행해서 비수용성 여부를 판단하는 것은 시간 및 비용적인 한계로 인해 불가능하다는 문제가 있다.
특히 대상 물질이 고가인 경우에 단지 비수용성 여부를 판단하기 위해 많은 양을 소모해야 하는 용해도 실험을 수행하는 것은 어려운 문제가 있다. 이뿐만 아니라, 현재의 실험 방법을 사용하기 위해서는 대상 물질이 반드시 실제로 합성이 되어 있어야 한다는 문제도 있다. 즉, 대상 물질이 합성되기 전에는 실험 방법을 적용할 수 없기 때문에 비수용성 여부를 전혀 판단할 수 없다는 문제가 있다.
상기의 문제점을 해결하기 위해서 본 발명은 대상 물질의 비수용성 여부를 실제 실험의 수행 없이도 정량적으로 명확하게 판단할 수 있는 새로운 방법인 비수용성 물질 평가방법을 제공하는 것을 목적으로 한다.
본 발명은 비수용성 여부를 판단하려는 물질의 화학구조 정보를 바탕으로 용해도 상수(solubility parameter)를 계산하고, 정립된 정량적인 기준을 통해 비수용성 여부를 최종적으로 판단하는 비수용성 물질 평가방법을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위해서, 본 발명은 (1) N개의 대상물질들을 선정하여, 상기 대상물질들의 화학 구조 정보를 생성하는 단계; (2) 상기 생성된 화학 구조 정보를 이용하여 비극성 용해도 상수(D), 극성 용해도 상수(P) 및 수소결합 용해도 상수(H)를 계산하는 단계; (3) 하기 수학식 1 및 하기 수학식 2로 상기 대상물질들의 비수용성 특성값을 계산하는 단계; 및 (4) 상기 계산된 대상물질들의 비수용성 특성값을 정량적 기준으로 평가하는 단계;를 포함할 수 있다.

본 발명의 다른 태양은 본 발명의 비수용성 물질 평가방법을 적용한 모듈을 포함하는 비수용성 물질 평가시스템을 제공한다.
또한, 본 발명의 또 다른 태양은 본 발명의 비수용성 물질 평가방법을 실행하기 위한 프로그램이 기록된 컴퓨터에서 판독 가능한 기록 매체를 제공한다.
본 발명은 대상 물질의 비수용성 여부를 실제 실험의 수행 없이도 정량적으로 명확하게 판단할 수 있는 비수용성 물질 평가방법을 제공한다.
특히 본 발명에서 제공하는 비수용성 물질 평가방법은 대상 물질이 실제로 합성되기 전에도 비수용성인지 아닌지를 판단할 수 있다.
나아가, 비수용성 물질을 실제 실험을 수행하지 않고도 명확하게 평가하는 본 발명은, 향후 용액공정을 이용한 소재 개발 및 관련 공정 문제점 해결과 효과적인 분리공정을 개발에 매우 유용하게 사용될 것으로 기대된다.
도 1은 본 발명의 비수용성 여부를 판단할 대상 물질들을 선정하여 각 대상물질들의 화학 구조 정보를 생성하는 단계의 일실시예의 순서도이다.
이하, 본 발명을 상세하게 설명한다. 하기의 구체적 설명은 본 발명의 일 실시예에 대한 설명이므로, 비록 한정적 표현이 있더라도 특허청구범위로부터 정해지는 권리범위를 제한하는 것은 아니다.
본 발명에서 비수용성 물질은 용질로서 용매인 물에 용해되지 않는 물질을 의미한다.
종래에는 비수용성 물질 여부를 판단하기 위해서 사용되고 있는 방법은 대상 물질을 직접 물에 용해시키는 용해성 실험을 수행하는 것이다. 하지만 소재 개발에서 테스트되는 수많은 물질들에 대해 용해성 실험을 수행해서 비수용성 여부를 판단하는 것은 시간 및 비용적인 한계로 인해 불가능하다는 문제가 있다.
이에 본 발명자들은 예의 노력한 바, 직접 대상물질을 합성하여 용해도 실험을 수행하지 않더라고 비수용성 물질 여부를 판단할 수 있는 방법을 발견하여 본 발명을 완성하게 되었다.
본 발명은 GIWiS(Gaugeable Indicator of Water-insoluble Signal)에 관한 것이다. 구체적으로 설명하면, 상기 GIWiS방법은 비수용성 여부를 판단하려는 물질의 화학구조 정보를 바탕으로 용해도 상수(solubility parameter)를 이용해서 정립된 정량적인 기준을 통해 비수용성 여부를 최종적으로 판단한다.
즉, 본 발명은 (1) N개의 대상물질들을 선정하여, 상기 대상물질들의 화학 구조 정보를 생성하는 단계; (2) 상기 생성된 화학 구조 정보를 이용하여 비극성 용해도 상수(D), 극성 용해도 상수(P) 및 수소결합 용해도 상수(H)를 계산하는 단계; (3) 하기 수학식 1 및 하기 수학식 2로 대상물질의 비수용성 특성값을 계산하는 단계; 및 (4) 상기 계산된 비수용성 특성값을 정량적 기준으로 평가하는 단계;를 포함할 수 있다.

이하 본 발명의 비수용성 물질 평가방법에 대해 설명한다.
먼저 (1) 단계에 대해 설명한다.
본 단계에서는 비수용성 여부를 판단할 대상 물질들을 선정하여 각 대상물질들의 화학 구조 정보를 생성하는 단계이다. 즉, 물에 대해서 용해되지 않는 특성인 비수용성 여부를 판단할 대상 물질 N개를 선정하고, 선정된 N개의 대상 물질에 대한 화학 구조 정보를 생성한다. 상기 화학 구조 정보는 각각의 대상 물질들이 구성하는 원자 및 분자 사이의 화학 결합에 대한 정보를 나타낸다.
본 발명의 바람직한 일실시예에 따르면, 상기 (1)단계에서 대상물질의 개수는 자연수이면 무관하다. 즉, 평가할 대상 물질의 개수는 몇 개라도 상관없이 수행가능하며, 예를 들어, N이 100 내지 1000이어도, 모두 수행 가능하다.
그리고 상기 화학 구조 정보는 통상적으로 사용 가능한 프로그램을 사용해서 생성할 수 있으나, 바람직하게는 Accelrys사에서 개발한 Material Studio Visualizer프로그램을 사용할 수 있다.
도 1은 본 발명의 바람직한 일실시예로, 상기 (1)단계의 순서를 나타낸 순서도이다. 즉, (a) 비수용성 여부를 판단할 대상 물질로 (b) 1,2,4-trichlorobenzene을 선정하면, (c) 상기 대상 물질의 화학 구조 정보를 Material Studio Visualizer를 사용해서 생성한다. 프로그램에 의해 생성된 화학 구조에서 (d) C로 나타낸 것은 방향족 링(aromatic ring)을 구성해 벤젠(benzene)을 구성하는 탄소 원자(carbon atom)를 나타내고, Cl로 및 H로 나타낸 것은 각각 염소 원자(chlorine atom) 및 수소 원자 (hydrogen atom)를 나타낸다.
다음 (2) 단계에 대해 설명한다.
본 단계에서는 생성된 화학 구조를 이용한 용해도 상수 계산한다. 즉, N개의 대상 물질들에 대해 생성된 화학 구조 정보를 바탕으로 각각의 대상 물질들의 용해도 상수 (solubility parameter)를 계산한다.
본 발명에서 계산을 통해 얻을 수 있는 용해도 상수 (solubility parameter)는 특성에 따라 3가지로 나누어질 수 있다. 첫번째로, 비극성(nonpolar) 특성으로 인한 비극성 용해도 상수 (Nonpolar dispersion solubility parameter,D), 두번째로는 극성(polar) 특성으로 인한 극성 용해도 상수(Polar solubility parameter, P) 그리고 세번째로 수소 결합 특성으로 인한 수소 결합 용해도 상수(Hydrogen bonding solubility parameter, H)로 나누어질 수 있다.
상기 용해도 상수를 계산할 수 있는 방법은 통상적으로 사용 가능한 프로그램이면 무엇이든 가능하나, 바람직하게는 C.M. Hansen이 개발한 HSPiP (Hansen Solubility Parameters in Practice) 프로그램을 사용해서 용해도 상수를 계산할 수 있다.
다음 (3) 단계에 대해 설명한다.
본 단계에서는 상기 대상 물질들에 대한 비수용성 특성값을 계산한다. 즉, N개의 대상 물질들 중에서 i번째 물질(Xi)에 대해 상기 (2)단계에서 계산한 3개의 용해도 상수를 이용해서 하기의 수학식 1 및 수학식 2에 나타낸 식으로 정의된 비수용성 특성값인 GIWiS1[Xi]와 GIWiS2[Xi]를 계산한다. 상기 i는 1 내지 N의 정수이다.

그리고 본 발명의 바람직한 일실시예에 따르면, 상기 εx는 환산계수(scaling factor)로 0이 아닌 실수 값인 경우 특별한 제한은 없지만, 바람직하게는 0.01 ~ 332.5의 실수일 수 있고, 가장 바람직하게는 242.2일 수 있다. 본 발명의 상기 수학식 1 및 수학식 2는 상기 환산계수(εx)를 통해서 값의 크기를 조절할 수 있다.
다음 (4) 단계에 대해 설명한다.
본 단계는 상기 (3)단계에서 계산된 비수용성 특성값을 정량적 기준을 통한 대상물질들의 비수용성 여부를 평가하는 것이다. 즉, N개의 대상 물질들의 비수용성 여부를 정립된 2개의 정량적 기준 1과 정량적 기준 2를 사용하여 평가하여 최종 판단한다.
구체적으로 설명하면, 비수용성 여부에 대한 평가 과정은 하기에 나타낸 것과 같이 두가지 단계로 수행될 수 있다. 첫번째 단계로 N개의 대상 물질 가운데 i번째 대상물질(Xi)에 대해 계산된 GIWiS1[Xi]와 GIWiS2[Xi]가 하기에 나타낸 정량적 조건 1을 만족시키는지 여부를 평가한다. 단, i는 1내지 N의 정수이다.
상기 정량적 조건 1은 0.90 < GIWiS1[Xi] 이고, GIWiS2[Xi] < 47.3인 것을 의미하며, 이를 만족시키는지를 평가한다. 만약, 상기 i번째 대상물질(Xi)이 상기 정량적 조건 1을 만족시키면, 상기 i번째 대상물질(Xi)은 비수용성 물질로 판단한다.
그리고 본 발명의 바람직한 일실시예에 따르면, 상기 i번째 대상물질(Xi)이 정량적 조건 1을 만족시키면, i < N 일 때, i+1번째 대상 물질(Xi + 1)이 정량적 조건 1을 만족시키는지를 평가한다. 그리고 계속 이와 같이 반복하여 평가하다가, i = N 이 되면, N개의 대상 물질들에 대해서 비수용성 여부 판단을 모두 마쳤기 때문에 평가 단계를 종료한다.
한편, 상기 i번째 대상물질(Xi)이 상기 정량적 조건 1을 만족시키지 않는 경우, 다음 두번째 단계로 평가한다. 본 발명의 바람직한 일실시예에 따르면, 상기 정량적 조건 1를 만족시키지 않으면, N개의 대상 물질 가운데 i번째 대상물질(Xi)에 대해 계산된 GIWiS1[Xi]와 GIWiS2[Xi]가 하기에 나타낸 정량적 조건 2를 만족시키는지 여부를 평가한다. i는 1 내지 N의 정수이다.
상기 정량적 조건 2는 1.03 < GIWiS1[Xi] 이고, GIWiS2[Xi]< 53.1인 것을 의미하며, 이를 만족시키는지 여부를 평가한다. 만약, 상기 i번째 대상물질(Xi)이 상기 정량적 조건 2를 만족시키면, 대상물질(Xi)은 비수용성 물질로 판단한다. 그리고 본 발명의 바람직한 일실시예에 따르면, 상기 대상물질(Xi)이 정량적 조건 2를 만족시키면, i < N 일 때, i+1번째 대상 물질(Xi + 1)을 상기 정량적 조건 1을 만족시키는지를 평가한다. 즉, i번째 대상물질(Xi)이 정량적 조건 2를 만족하여, 비수용성 물질이므로, 다음 대상물질의 비수용성 물질 평가를 수행한다. 이와 같이 반복하여 평가하다가, i = N 일 때에는, N개의 대상 물질에 대해서 비수용성 여부 판단을 모두 마쳤기 때문에 평가 단계를 종료한다.
한편, 상기 i번째 대상물질(Xi)이 정량적 조건 2를 만족시키지 않으면, 상기 i번째 대상물질(Xi)은 비수용성 물질이 아니다. 그리고 다음으로, i < N 일 때에는, i+1번째 대상 물질(Xi + 1)을 상기 정량적 조건 1을 만족시키는지를 평가한다. 즉, i번째 대상물질(Xi)이 정량적 조건 2를 만족하지 않아도, 계속해서 다음 대상물질의 비수용성 물질 평가를 수행한다. 이와 같이 반복하여 평가하다가, i = N 일 때에는, N개의 대상 물질에 대해서 비수용성 여부 판단을 모두 마쳤기 때문에 평가단계를 종료한다.
결론적으로, 본 발명의 상기 i번째 대상물질(Xi)이 상기 정량적 조건 1 및 상기 정량적 조건 2 모두를 만족시키지 않는 경우에는 i번째 대상물질(Xi)은 비수용성 물질로 판단하지 않는다. 그리고 상기 정량적 조건 2는 정량적조건 1이 놓칠 수 있는 비수용성 물질을 찾을 수 있다. 따라서, 상기 정량적 조건 1 및 정량적 조건 2는 서로 보완을 해주는 관계이다.
이와 같이 본 발명에서 제공하는 비수용성 물질 평가방법은 기존에 알려진 비수용성 물질에 대해 적용해 본 결과 비수용성 여부를 정확하게 평가하는 것을 확인해서 본 발명의 효용성을 확보하였다.
본 명세서는 본 발명의 비수용성 물질 평가방법을 적용한 모듈을 포함하는 비수용성 물질 평가시스템을 제공한다.
상기 비수용성 물질 평가시스템에 있어서 비수용성 물질 평가방법과 중복되는 구성에 대한 설명은 동일하다.
또한, 본 발명에서 모듈(module)이란 용어는 특정한 기능이나 동작을 처리하는 하나의 단위를 의미하며, 이는 하드웨어나 소프트웨어 또는 하드웨어 및 소프트웨어의 결합으로 구현할 수 있다.
본 발명의 또 다른 태양은 본 발명의 비수용성 물질 평가방법을 실행하기 위한 프로그램이 기록된 컴퓨터에서 판독 가능한 기록 매체를 제공한다.
이하 본 발명을 실시예에 기초하여 더욱 상세하게 설명하지만, 하기에 개시되는 본 발명의 실시 형태는 어디까지 예시로써, 본 발명의 범위는 이들의 실시 형태에 한정되지 않는다. 본 발명의 범위는 특허청구범위에 표시되었고, 더욱이 특허 청구범위 기록과 균등한 의미 및 범위 내에서의 모든 변경을 함유하고 있다. 또한, 이하의 실시예, 비교예에서 함유량을 나타내는 "%" 및 "부"는 특별히 언급하지 않는 한 중량 기준이다.
실시예
(1) 비수용성 여부를 판단할 대상 물질의 선정 및 화학 구조 정보를 생성
; 비수용성 여부를 판단할 대상 물질(Xi)로 하기에 나타낸 총 3종(N=3)을 물질을 선정하였다(i=1 내지 N). 그리고, 상기 총 3종의 물질의 화학 구조 정보를 Accelrys사에서 개발한 Material Studio Visualizer 프로그램을 사용해서 생성하였다. 그리고 하기 표 1에 3종 대상 물질(Xi)과 각각의 대상 물질에 대해 생성된 화학 구조 정보를 나타내었다.

(2) 생성된 화학 구조를 이용한 용해도 상수 계산
; 3종 대상 물질에 대해 생성된 화학 구조 정보를 바탕으로 용해도 상수(solubility parameter)를 계산하였다. 본 실시예에서 용해도 계산은 HSPiP(Hansen Solubility Parameters in Practice)를 이용해서 수행하였다. 이를 통해 각각의 대상 물질에 대해 3가지 서로 다른 특성을 나타내는 용해도 상수인 비극성 용해도 상수(nonpolar dispersion solubility parameter), 극성 용해도 상수(polar solubility parameter), 수소결합 용해도 상수(hydrogen bonding solubility parameter)를 계산하여, 각각의 대상 물질에 대해 계산된 용해도 값을 하기의 표 2에 나타냄.

(3) 대상 물질에 대한 비수용성 특성값 계산
; 총 3개의 각각의 대상 물질(Xi)에 대해 상기 (2)단계의 과정을 통해 계산된 3개의 용해도 상수를 이용해서 하기에 표시된 수학식 1 및 수학식 2로 정의된 비수용성 특성인 GIWiS1과 GIWiS2를 계산하였다.

(4) 정량적 기준을 통한 대상물질의 비수용성 여부 평가

- 정량적 조건 1: GIWiS1[Xi] > 0.90인 동시에 GIWiS2[Xi] < 47.3
- 정량적 조건 2: GIWiS1[Xi] > 1.03인 동시에 GIWiS2[Xi] < 53.1
상기 정량적 조건 1 및 정량적 조건 2에 대한 평가를 통해 최종적으로 판단된 비수용성 여부에 대한 결과는 표 4에 나타내었다.

표 4를 설명하면, 1,2,4-trichlorobenzene(X1)는 GIWiS1[X1]=1.33이고, GIWiS2[X1]=26.0으로 정량적 조건 1을 만족시키므로, 비수용성 물질로 최종 판단되었다.
그리고 Bicyclo[4.4.0]deca-1,3,5,7,9-pentene(X2)도 GIWiS1[X2]=1.25이고, GIWiS2[X2]=28.3으로 정량적 조건 1을 만족시키므로, 비수용성 물질로 최종 판단되었다. 또한, Pyrene(X3) 역시, GIWiS1[X3]=1.53이고, GIWiS2[X3]=22.1로 정량적 조건 1을 만족시킴으로써, 비수용성 물질로 최종 판단되었다.
비교예
2-부탄올(2-Butanol)을 대상 물질(X4)로 정하여, (1) 화학 구조 정보 생성 및 (2) 용해도 상수 계산은 상기 실시예와 동일하게 실시하였다. 계산된 상기 대상 물질(X4)인 2-부탄올(2-Butanol)의 비극성 용해도 상수, 극성 용해도 상수 및 수소 결합 용해도 상수는 각각 15.8, 5.7 및 14.5이었다.
그리고 대상 물질(X4)인 2-부탄올(2-Butanol)의 (3) 비수용성 특성값 계산도 상기 실시예와 동일하게 실시하였다.
마지막으로 대상 물질(X4)인 2-부탄올(2-Butanol)의 (4) 비수용성 여부 평가도 상기 실시예와 동일하게 실시하였다.
그 결과, 대상 물질(X4)인 2-부탄올(2-Butanol)은 GIWiS1[X4]=1.01이고, GIWiS2[X4]=64.9로 정량적 조건 1 및 정량적 조건 2를 모두 만족시키지 못하였다.
따라서 대상 물질(X4)인 2-부탄올(2-Butanol)은 비수용성 물질이 아닌 것으로 최종 판단되었다.
실험예
상기 실시예 및 비교예에서의 실시한 본 발명의 비수용성 평가방법의 타당성을 확인하기 위해 하기의 방법으로 확인하였다.
상기 4종 대상물질(X1, X2, X3 및 X4)를 1 기압 및 25℃에서 각각 100g의 물에 24시간 용해시켜 용해도를 측정하였다.
그 결과 실시예 대상물질인 3종(X1, X2 및 X3)은 모두 0.005g 이하로 용해되어 물에 대한 용해도가 0에 매우 가까운 값을 나타내는 것으로 확인하였다. 반면에, 비교예의 대상물질(X4)은 30g이 용해되어, 물에 대한 용해도가 매우 크다는 것을 확인하였다.
따라서 실시예의 3종 대상물질은 모두 비수용성 물질로 확인되었으며, 비교예의 1종의 대상물질은 비수용성 물질이 아닌 것으로 확인되었다.
결론적으로, 본 발명의 GIWiS를 이용한 비수용성 평가방법은 실제적인 용해도 측정 실험 없이도, 비수용성 물질을 정확하게 판단할 수 있다는 것을 알 수 있다.

Claims (9)

  1. (1) N개의 대상물질들을 선정하여, 상기 대상물질들의 화학 구조 정보를 생성하는 단계;
    (2) 상기 생성된 화학 구조 정보를 이용하여 상기 대상물질들의 비극성 용해도 상수(D), 극성 용해도 상수(P) 및 수소결합 용해도 상수(H)를 계산하는 단계;
    (3) 하기 수학식 1 및 하기 수학식 2로 상기 대상물질들의 비수용성 특성값을 계산하는 단계; 및
    (4) 상기 계산된 대상물질들의 비수용성 특성값을 정량적 기준으로 평가하는 단계;를 포함하는 비수용성 물질 평가방법;
