After overnight cultivation at 30°C and 280 rpm in a rotary shaker, cells were harvested, and inoculated at a starting cell density of 0.5 g L -1 for an anaerobic growth test at 32°C using an Alcohol Fermentation Monitor (Applikon, Delft, The Netherlands) with stirring speed 250 rpm.

  • As medium 400 ml mineral medium supplemented with 7.5 % glucose, 3.8 % xylose, 0.5% arabinose, 0.25 % galactose, 0.1 % mannose was used. During the experiment, biomass, sugar levels, glycerol, acetic acid and ethanol were monitored every four hours.
  • xylose levels were already decreased after 8 hours into the fermentation at glucose concentrations of 50 g L -1 , whereas for the other strains, DS65307-2T, DS65307-4T, DS65307-7T and DS65307 (reference strain), xylose concentrations started to decrease at lower glucose concentrations (10 g L-1 at 16 hours of fermentation).
  • DS65307-1 Tand DS65307-3T exhibited a higher degree of glucose and xylose co-fermentation as compared to DS65307 at high glucose concentrations (higher than > 15 g L -1 ).
  • HXT1 1 (N366T) is stably integrated into the genome at multiple HXT loci having two consequences: a) the deletion of multiple native HXT genes, b) expression of HXT11 (N366T) under control of the promoter of the deleted HXT gene it has replaced.

Colonies that have lost GFP fluorescence or pink/red-coloration are verified for marker removal by colony PCR with the appropriate primers.

Abstract

L'invention concerne une cellule de levure comprenant au moins une voie de conversion fonctionnelle du pentose en partant du pentose vers le produit de fermentation, où la cellule de levure comprend une ou plusieurs constructions transporteur, où la construction transporteur comprend des séquences d'ADN : a) un promoteur de transporteur b) un cadre de lecture ouvert d'un transporteur de pentose qui remplace un transporteur d'hexose connecté au promoteur de transporteur dans la cellule de levure d'origine naturelle et; c) un codon de terminaison, où le promoteur de transporteur est régulé par la régulation cellulaire induite par la signalisation SNF3 et/ou RGT2.
