Abstract
본 발명의 일 측면에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치는 분석 대상인 시료가 주입되는 시료 주입구와 시약이 주입되는 시약 주입구와 시료와 시약을 혼합하기 위하여 상기 시료 주입구 및 상기 시약 주입구와 연통된 반응채널과 상기 반응채널과 연통된 광 입사구와 상기 광 입사구에서 이격 배치된 검출구가 형성된 제1기판과, 상기 광 입사구와 마주하며 광원이 빛이 투과하는 광 입사부와 상기 검출구와 마주하며 광원의 빛이 투과하는 검출부를 구비하며 상기 제1기판에 결합되는 제2기판, 및 상기 광 입사구에 일측 단부가 연결되며, 상기 검출구에 타측 단부가 연결되는 흐름셀을 포함하는 미세유체칩과, 상기 광 입사부와 마주하도록 배치된 광원과, 상기 검출부와 마주하도록 배치된 검출기와, 상기 광원과 상기 검출기 사이에 배치된 차단벽, 및 상기 미세유체칩과 연결되며 시료를 산화분해하여 상기 미세유체칩으로 공급하는 산화분해부재를 포함한다.
Description
본 발명은 연속 미세 유체 장치에 관한 것으로서 보다 상세하게는 총질소 또는 총인 분석용 연속 미세 유체 장치에 관한 것이다.
질소와 인은 농업용 비료, 사람이나 가축의 분뇨, 합성세제 등으로부터 주로 발생되며 수중으로 다량 유입될 경우 부영양화, 연안의 적조현상, 어류 독소, 용존산소 결핍 등을 야기하므로 수질모니터링에 있어서 가장 중요한 항목이다. 특히 아질산성 질소는 체내에 주입되었을 경우 헤모글로빈의 산소운반능력을 저하시켜 유아청색증을 유발하며, 아마이드나 아민과 결합하여 발암물질인 N-니트로소 화합물을 생성함에 따라 미국환경보호국에서는 음용수 내의 아질산성 질소의 농도를 1ppm 이하로 규제하고 있다. 그리고 질산성 질소가 수중이나 체내에서 아질산성 질소로 환원됨에 따라 음용수 내의 질산성 질소의 농도를 10ppm 이하로 규제하고 있다. 수중의 질소와 인의 농도는 인간활동, 기후변화, 계절, 태양복사, 온도, 해류, 용존산소량 등에 의해 급격하게 변화하므로 고밀도의 빠른 분석을 통해 수질오염에 대처해야 한다.
상용화된 총질소와 총인 분석용 검출 장치는 대부분 감도와 선택성이 우수한 흡광광도법을 기초로 한 것으로 크기(대형냉장고 크기)가 커서 이동 측정이 불가능하며, 수변에 설치한 가건물 내에 장치를 비치하여 측정하므로 많은 장소에 설치가 어렵다(국내등록특허 제10-0718261호, 제10-0877810호, 제10-0584935호). 그리고 수 mL/min의 빠른 유속으로 분석함에 따라 시료와 시약의 소모 및 폐수발생량이 커서 잦은 유지보수가 요구되며, 시료 전처리에 10분~30분이 소요되어 분석시간이 길어짐에 따라(국내등록특허 제10-0875629호) 급격한 수질변화를 모니터링하기에 적합하지 않다. 상기한 문제점들을 해결하기 위해 적은 양의 시료와 시약을 사용하여 빠른 분석이 가능한 수 cm2 크기의 미세유체칩을 기반으로 한 소형의 흡광 검출 장치가 개발되었다. 미세유체칩을 기반으로 한 흡광 검출 장치는 시료와 시약 소모량및 폐수발생량이 적고 소비 전력량이 작아 잦은 유지보수가 필요하지 않으며, 크기가 작아 휴대가 가능하고 수중 부표 등을 이용해 원하는 지역에 많은 수의 설치가 가능함에 따라 고밀도 분석에 적합하다.
흡광 검출 방법에서 흡광도(A)는 아래의 [식 1]과 같이 비어의 법칙(Beer's law)에 의하여 표현된다.

즉, 흡광도(A)는 몰흡광계수(ε)(L/(mol·cm)), 광 경로 길이(b)(cm), 몰농도(C)(mol/L)에 비례한다. 몰흡광계수는 분석물질 또는 발색반응 후의 산물에 의해 결정되는 것이므로 흡광 검출감도를 향상시키기 위해서는 긴 광 경로 길이를 갖는 장치를 제작해야 한다. 또한, 넓은 범위의 농도를 갖는 분석물질을 측정하기 위해서는 광 경로 길이의 자유로운 조절이 필요하다.
현재까지 개발된 흡광 검출을 이용한 질소와 인 분석용 연속 미세 유체 장치는 무기성 질소와 인을 분석하는 것에 한정되어 있으며, 다음과 같은 문제점들에 의해 분석물질에 대한 고감도의 정확한 정량이 어렵다.
첫 번째 수 cm2 크기의 미세유체칩 내에 미세유체채널을 흡광 검출을 위한 흐름셀로 사용함에 따라 광 경로 길이를 연장하는 것에 한계가 있으며, 광 경로 길이의 조절이 어렵다.
두 번째 유리나 투명한 고분자 등을 사용하여 제작한 미세유체칩 내에 흡광검출을 위한 흐름셀로 사용되는 직선의 미세유체채널의 양쪽 끝에 광원과 검출기가 각각 연결된 광학섬유를 마주보는 구조로 배열함으로써(미국등록특허 제0180963호, Anal. Chim. Acta 1999, 382, 113) 외부의 빛과 광원의 산란광(이하 "미광(stray light)"이라 한다.)이 검출기로 직접 유입됨에 따라 검출감도가 떨어지고, 검량곡선이 직선성을 보이는 농도범위가 줄어든다. 또한, 미세유체칩 내에 흡광 검출을 위한 흐름셀과 광학장치들을 정교하게 배열해야 함에 따라 제작이 어렵고 복잡하며, 상기 미광을 최소화하기 위해 소형의 집광렌즈나 슬릿 등의 추가적인 광학장치의 사용이 요구된다.
세 번째 외부 표준물질을 이용한 검량곡선법(분석물질로 제조한 표준시료의 농도에 따른 흡광도로부터 얻은 직선의 관계식을 이용하여 시료 내에 분석물질을 정량하는 방법)을 이용하여 분석물질을 정량함에 따라 실제시료와 표준시료의 조성의 차이에 의해 분석오차가 발생할 수 있다. 더 자세히 설명하면 상기 분석오차는 실제시료 내에 존재하는 분석물질을 제외한 다른 물질들(매트릭스)에 의한 분석신호의 변화(매트릭스 효과)로부터 발생한다. 특히, 강하구와 같이 조석에 의해 0~30‰ 염도가 변하는 시료들은 분석오차가 더욱 심각하다.
종래의 상용화된 질소와 인 분석용 흡광 검출 장치도 외부 표준물질을 이용한 검량곡선법을 정량법으로 사용하고 있어 매트릭스에 의한 분석오차가 발생할 수 있다.
본 발명은 적은 양의 시료 및 시약을 사용하여 고효율, 고감도 분석이 가능한 총질소 또는 총인 분석용 연속 미세 유체 장치를 제공하고자 한다.
본 발명의 일 측면에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치는 분석 대상인 시료가 주입되는 시료 주입구와 시약이 주입되는 시약 주입구와 시료와 시약을 혼합하기 위하여 상기 시료 주입구 및 상기 시약 주입구와 연통된 반응채널과 상기 반응채널과 연통된 광 입사구와 상기 광 입사구에서 이격 배치된 검출구가 형성된 제1기판과, 상기 광 입사구와 마주하며 광원의 빛이 투과하는 광 입사부와 상기 검출구와 마주하며 광원의 빛이 투과하는 검출부를 구비하며 상기 제1기판에 결합되는 제2기판, 및 상기 광 입사구에 일측 단부가 연결되며, 상기 검출구에 타측 단부가 연결되는 흐름셀을 포함하는 미세유체칩과, 상기 광 입사부와 마주하도록 배치된 광원과, 상기 검출부와 마주하도록 배치된 검출기와, 상기 광원과 상기 검출기 사이에 배치된 차단벽, 및 상기 미세유체칩과 연결되며 시료를 산화분해하여 상기 미세유체칩으로 공급하는 산화분해부재를 포함한다.
여기서 상기 산화분해부재는 열 또는 빛을 발생시키는 반응막대와 상기 반응막대에 감겨지며, 시료가 이동하는 통로를 제공하는 튜브를 포함할 수 있다.
또한, 상기 반응막대는 자외선을 방출하는 자외선 램프로 이루어질 수 있다.
또한, 상기 산화분해부재에는 시료를 공급하는 시료 주입관과 산화제를 공급하는 산화제 주입관이 연결 설치될 수 있다.
또한, 상기 산화분해부재에는 표준물첨가시료를 상기 산화분해부재로 공급하는 표준물첨가시료 주입관이 연결 설치될 수 있다.
또한, 상기 산화분해부재에는 카드뮴-구리 컬럼이 연결 설치되고, 상기 카드뮴-구리 컬럼은 튜브와 상기 튜브 내에 채워진 카드뮴 입자를 포함할 수 있다.
또한, 상기 카드뮴 입자는 산으로 처리된 후, 황산구리 용액으로 활성화될 수 있다.
또한, 상기 카드뮴-구리 컬럼에는 카드뮴의 침전을 방지하는 완충용액을 공급하는 완충용액 주입관이 연결 설치될 수 있다.
또한, 상기 완충용액은 이미다졸을 포함할 수 있다.
또한, 상기 제1기판에는 상기 검출구와 연결된 용액 배출채널 및 상기 용액 배출 채널에 연결된 용액 배출구가 형성될 수 있다.
또한, 상기 흐름셀의 양측 단부는 상기 제2기판을 향하도록 설치될 수 있다.
또한, 상기 제1기판과 제2기판은 빛을 흡수할 수 있는 색을 갖는 물질로 이루어지거나, 빛을 흡수할 수 있는 색으로 착색된 구조로 이루어질 수 있다.
또한, 상기 입사부와 검출부는 빛이 투과할 수 있도록 투명한 소재로 이루어질 수 있다.
또한, 상기 광 입사구 및 상기 검출구에는 상기 흐름셀이 삽입되는 체결부가 형성될 수 있다.
또한, 상기 검출부와 상기 검출기 사이에는 상기 광원에서 출사되는 빛의 반값 반폭보다 더 작은 반값 반폭을 갖도록 빛을 투과시키는 간섭 필터가 설치될 수 있다.
또한, 상기 광원과 상기 검출기 및 상기 차단벽은 하우징 내에 설치되고, 상기 미세유체칩은 상기 하우징 상부에 배치되며, 상기 하우징의 상면에는 상기 광원의 상부에 위치하는 입사채널 및 상기 검출기의 상부에 위치하는 검출채널이 형성되고, 상기 간섭 필터는 상기 검출채널과 상기 검출기 사이에 간섭 필터가 설치될 수 있다.
본 발명은 흡광 검출을 위한 흐름셀로 액체 코어 도파관을 사용하여 광 경로 길이의 연장이 쉽고, 상기 액체 코어 도파관을 미세유체칩의 광 입사구와 검출구에 설치된 체결부에 착탈 결합하여 용이하게 설치할 수 있어 광 경로 길이의 조절이 가능하다. 따라서 다양한 수환경에 적용하여 넓은 농도범위의 총질소와 총인을 고감도로 검출할 수 있다.
또한, 하우징 내에 차단벽을 사이에 두고 병렬로 배치되어 있는 광원 및 검출기의 상부에 빛을 흡수하는 재료로 제작한 미세유체칩을 고정 설치하여 미광을 효과적으로 차단할 수 있다.
또한, 미세유체칩은 광원의 빛이 통과하는 부분만 투명하고 하우징 내에 광원의 높이를 자유롭게 조절할 수 있어 소형의 슬릿이나 집광렌즈 등의 추가적인 광학장치의 사용이 불필요하다.
또한, 산화분해부재는 테프론 튜브의 얇은 벽을 통해 빛을 효율적으로 이용할 수 있고, 내부부피가 매우 작아 시료의 열 및 물질전달 속도가 빠르기 때문에 수 분 내에 복잡한 구조를 가져 분해가 쉽지 않은 다양한 질소 및 인 화합물의 산화분해가 가능하다.
또한, 정량법으로 표준물첨가법을 사용하여 다양한 매트릭스가 존재하는 시료, 특히 염도가 높은 해수나 염도가 계속적으로 변화하는 강하구 내 총질소와 총인의 농도를 정확하게 높은 재현성으로 분석할 수 있다.

도 1은 본 발명의 제1 실시예에 따른 미세유체칩의 상판을 도시한 평면도이고, 도 2는 본 발명의 제1 실시예에 따른 미세유체칩의 하판을 도시한 평면도이며, 도 3은 본 발명의 제1 실시예에 따른 미세유체칩을 도시한 단면도이다.
도 1 내지 도 3을 참조하여 설명하면, 본 실시예에 따른 미세유체칩(30)은 상판(제1기판)(10)과 하판(제2기판)(20) 및 상판(10)에 결합된 흐름셀(31)을 포함한다.
상판(10)은 사각판 형상으로 이루어지며, 상판(10)에는 시료 주입구(11)와 시약 주입구(12)가 형성되어 있다. 또한, 시료 주입구(11)에는 시료 주입관(53)과의 결합을 위한 체결부(33)가 형성되며, 시약 주입구(12)에는 시약 주입관(52)과의 결합을 위한 체결부(32)가 형성되어 있다.
또한, 상판(10)에는 시료채널(16), 시약채널(17), 반응채널(18), 및 광 입사구(13)가 형성된다. 시료 주입구(11)에는 시료채널(16)이 연결 형성되어 시료는 시료채널(16)을 통해서 이동하며, 시약 주입구(12)에는 시약채널(17)이 연결 형성되어 시약은 시약채널(17)을 통해서 이동한다. 시료채널(16)과 시약채널(17)에는 반응채널(18)이 연결 형성되며, 반응채널(18)에서 시약과 시료는 혼합된다. 반응채널(18)은 중첩 배치되며 일측 단부와 타측 단부가 교호적으로 연결되어 사행형상으로 형성된다. 반응채널(18)의 한쪽 끝에는 광 입사구(13)가 연결 형성되며 광 입사구(13)에는 흐름셀(31)이 결합될 수 있도록 체결부(34)가 설치된다.
시약은 시료에서 특정 성분을 검출하기 위한 것으로서 검출하고자 하는 성분과 반응하여 특정한 색을 갖도록 한다.
본 실시예에서는 시료가 시약과 반응하여 특정한 색을 나타내는 것으로 예시하고 있으나, 시료의 종류에 따라서는 시약과의 반응 없이 시료 자체가 특정한 파장의 빛을 흡수할 수 있다. 이 경우에는 시약 주입구(12)와 반응채널(18) 없이 시료 주입구(11)가 광 입사구(13)와 직접 연통된다.
또한, 상판(10)에는 흐름셀(31)과 연결된 검출구(14)와 검출구(14)에 연결된용액 배출채널(19), 및 용액 배출채널(19)과 연결된 용액 배출구(15)가 형성된다. 용액 배출채널(19)은 흐름셀(31)을 통해서 유입된 시료 및 시약이 이동하는 통로가 되며, 용액 배출구(15)에는 사용된 시료 및 시약을 배출시키기 위한 별도의 저장 용기가 연결 설치될 수 있다.
검출구(14)에는 흐름셀(31)이 결합될 수 있도록 체결부(35)가 형성되어 있으며, 용액 배출구(15)에는 배출관이 결합될 수 있도록 체결부(36)가 형성되어 있다. 검출구(14)와 광 입사구(13)는 이격 배치되며, 흐름셀(31)을 통해서 연결된다. 체결부들(32, 33, 34, 35, 36)은 폴리머 또는 튜브 등으로 이루어지며, 관 또는 튜브가 용이하게 착탈 결합될 수 있도록 홀을 갖는다.
상판(10)은 빛을 흡수할 수 있는 소재로 이루어지며, 특히 검정색의 폴리디메틸실록산(PDMS)으로 이루어질 수 있다. 다만 상판(10)은 빛을 흡수할 수 있는 다양한 색을 갖는 물질로 이루어지거나, 빛을 흡수할 수 있는 색으로 착색될 수 있다.
한편, 하판(20)에는 광 입사구(13)와 대응되는 하부에 광 입사부(22)가 형성되고, 검출구(14)와 대응되는 하부에 검출부(23)가 형성된다. 하판(20)은 검정색 PDMS으로 이루어지며, 광 입사부(22)와 검출부(23)는 빛을 통과시킬 수 있도록 각각 투명 PDMS으로 채워진다. 광 입사부(22)와 검출부(23)는 검정색 PDMS에 홀을 형성한 후, 투명 PDMS를 주입하여 형성할 수 있다. 다만, 하판(20)이 착색된 경우에는 광 입사부(22)와 검출부(23)만 착색되지 아니하고 투명하게 형성된다.
상기한 바와 같이 상판(10)과 하판(20)이 빛을 흡수할 수 있도록 형성되면, 외부에서 또는 광원에서 입사된 미광을 흡수하여 흡광 검출감도를 향상시킬 수 있다.
이하에서는 상판(10)과 하판(20)의 제조 방법에 대하여 설명한다. 준비된 실리콘 웨이퍼 상에 포토레지스트를 스핀 코팅한 후, 포토 마스크를 설치한 상태에서 자외선 노광하여 형상을 전사한다. 이후에 현상하여 양각 패턴을 형성한다.
양각 패턴이 형성된 실리콘 웨이퍼 위에 검정색 PDMS를 붓고 가교결합 후에 웨이퍼에서 분리시키면 음각이 형성된 검정색 PDMS의 상판을 얻을 수 있다. 하판(20)도 상판(10)과 동일한 방법으로 제조한다.
상판(10)과 하판(20)은 결합되는 부분을 아크 방전으로 표면처리한 후, 상판(10)의 광 입사구(13)와 검출구(14)의 중심에 하판(20)의 광 입사부(22)와 검출부(23)의 중심이 일치하도록 결합한다.
흐름셀(31)은 유입구와 유출구를 구비하여 용액을 이동시킴과 동시에 빛이 이동할 수 있는 셀을 의미하며, 연속적으로 흡광도를 검출할 수 있게 한다. 본 실시예에 따른 흐름셀(31)은 테프론 계열의 액상 코어를 갖는 도파관으로 이루어진다. 흐름셀(31)로서 액체 코어 도파관이 적용됨에 따라 전반사를 통해서 경로를 구부리더라도 빛을 유입구에서 유출구로 이동시킬 수 있다.
또한, 흐름셀(31)은 유리, 실리콘, 폴리머 소재로 이루어진 군에서 선택되는 어느 하나로 이루어질 수 있다. 흐름셀(31)은 관 형상으로 이루어지며, 시료와 시약이 혼합된 용액이 흐름셀(31)을 통해서 광 입사구(13)에서 검출구(14)로 이동한다. 또한, 흐름셀(31)의 한쪽 끝으로 입사한 빛은 용액을 따라 이동하면서 전반사하여 다른 쪽 끝으로 이동한다.
흐름셀(31)의 일측 단부를 체결부(34)에 삽입하고, 흐름셀(31)의 타측 단부를 체결부(35)에 삽입하여 광 입사구(13)와 검출구(14)를 흐름셀(31)과 연결한다. 상기한 바와 같이 체결부(34, 35)를 형성하면 흐름셀(31)을 상판(10)과 용이하게 결합시킬 수 있다. 흐름셀(31)은 양쪽 단부가 하판(20)을 향하도록 체결부(34, 35)에 삽입 설치된다. 특히, 흐름셀(31)은 양쪽 단부가 평행하도록 설치되어, 흐름셀(31)의 양쪽 단부는 동일한 방향을 향한다.
이에 따라 흐름셀(31)은 굽어져 대략 U자 형태로 이루어질 수 있다. 또한, 흐름셀(31)은 O자 형태로 감겨진 후, 양쪽 단부가 평행하도록 결합부에 삽입될 수 있다.
상기한 바와 같이 흐름셀(31)을 상판에 결합 설치하면, 광 경로를 연장하여 흡광도를 고감도로 측정할 수 있다. 또한, 흐름셀(31)을 굽혀서 양단이 동일한 방향을 향하도록 설치되므로 흐름셀(31)의 길이를 연장하더라도 미세유체칩의 길이가 증가하지 아니하여 컴팩트한 미세유체칩을 제조할 수 있다.
시료 및 시약은 반응채널(18)과 광 입사구(13), 흐름셀(31), 검출구(14), 용액 배출채널(19)을 거쳐 용액 배출구(15)로 이동하여 유체가 흐르는 미세유체칩을 형성할 수 있다.
또한, 본 실시예와 같이 흐름셀(31)의 양쪽 단부가 평행하게 배치되면 검출기(43)로 입사되는 미광을 차단하여 정밀한 측정이 가능하다. 이는 광원과 검출기가 일직선 상에 있지 아니하기 때문이다.
도 4는 본 발명의 제1 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치의 구성도이다.
도 4를 참조하여 설명하면, 본 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치는 외형을 이루는 하우징(41)과 하우징(41) 내에 삽입 설치되며 광 입사부(22)의 아래에 배치된 광원(42), 검출부(23)의 아래에 배치된 검출기(43), 및 미세유체칩(30)을 포함한다.
한편, 미세유체칩(30)에는 시료 및 시약이 저장된 공급부(51)가 연결 설치되는데, 공급부(51)에는 연동펌프가 설치되어 미세유체칩(30)으로 시료 및 시약을 공급한다. 공급부(51)에는 시약 주입관(52) 및 시료 주입관(53)이 연결 설치된다.
하우징(41)은 대략 직육면체의 상자 형태로 이루어지며, 하우징(41)에는 광원(42)의 상부에 위치하는 입사채널(45)과 검출기(43)의 상부에 위치하는 검출채널(46)이 형성된다. 입사채널(45)은 광원(42)과 광 입사부(22) 사이에 위치하여 광원(42)에서 출사된 빛을 광 입사부(22)로 전달하는 통로가 되며, 검출채널(46)은 검출기(42)와 검출부(23) 사이에 위치하여 검출부(23)에서 검출기(43)로 빛을 전달하는 통로가 된다.
하우징(41)은 빛을 흡수할 수 있도록 검정색으로 착색된 금속으로 이루어지며, 흐름셀(31)을 갖는 미세유체칩(30)은 하우징(41)의 상면에 고정 설치된다. 이에 따라 광원(42)에서 배출된 빛은 직접 검출기(43)로 이동하지 못하고, 흐름셀(31)을 거쳐서 검출기(43)로 이동한다.
광원(42)은 발광다이오드로 이루어지며, 광원(42)은 분석물질과 시약의 종류에 따라 최대흡수파장을 고려하여 다양하게 선택될 수 있다. 검출기(43)는 광전자증배관으로 이루어지며, 흐름셀(31)에서 출사된 빛에서 특정 파장의 세기를 검출한다. 검출기(43)와 검출채널(46) 사이에는 간섭 필터(44)가 설치된다. 간섭 필터(44)는 입사되는 빛을 좁은 반값 반폭으로 투과시켜 검량곡선의 직선성을 향상시키는 역할을 한다. 간섭 필터(44)는 광원(42)에서 출사된 빛의 반값 반폭보다 더 작은 반값 반폭으로 빛을 투과시킨다.
광원(42)에는 전원(61)이 연결 설치되고, 검출기(43)에도 전원(62)이 연결 설치된다. 또한, 검출기(43)에는 검출기(43)에서 발생된 신호를 가공 처리하는 디지털 멀티미터(63)가 연결 설치되고, 디지털 멀티미터(63)는 가공된 신호를 컴퓨터(64)로 전달한다.
광원(42)과 검출기(43) 사이에는 차단벽(47)이 설치되는 바, 차단벽(47)은 하우징(41)의 상면에서 하면으로 이어져 배치된다. 상기한 차단벽(47)에 의하여 광원(42)에서 발생된 미광이 검출기(43)로 유입되는 것을 차단할 수 있다.
또한, 광원(42)과 검출기(43)는 차단벽을 사이에 두고 측방향으로 이격 배치되어 광원(42)에서 출사되는 미광이 검출기(43)로 유입되는 것을 원천적으로 차단할 수 있다. 종래에는 광원과 검출기가 일직선 상에 배치되어 광원에서 발생된 빛이 검출기로 직접 유입되는 것을 원천적으로 차단하지 못하여 미광이 유입되는 문제가 있다. 광원에서 발생된 빛은 검출대상 용액을 거쳐서만 검출기로 유입되어야 고감도의 검출을 할 수 있으나, 종래와 같이 광원의 빛이 직접 검출기로 유입되면 흡광 감도가 저하된다.
그러나 본 실시예에 따르면 흐름셀(31)이 굽혀져 양단이 평행하게 설치되므로 광원(42)과 검출기(43)가 일직선 상이 아닌 병렬로 배치되므로 미광의 유입을 차단할 수 있을 뿐만 아니라 총질소 또는 총인 분석용 연속 미세 유체 장치를 컴팩트하게 구성할 수 있다. 이에 따라 본 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치는 휴대 가능한 구조로 이루어지며, 생명과학, 환경오염물질의 실시간 환경모니터링, 질병진단. 조합화학 등의 분야에 적용이 가능하다.
[실험예 1]
상기한 총질소 또는 총인 분석용 연속 미세 유체 장치를 이용하여 아질산성 질소 성분을 검출한다. 질소는 생물체의 필수영양소로 주로 인간활동에 의해 발생한 다양한 오염원으로 인해 부영양화를 유발하며 이로 인해 적조현상, 물고기 떼죽음 등의 결과를 초래한다. 또한 아질산성 질소는 혈액 중의 헤모글로빈(hemoglobin)과 반응하여 산소운반기능이 없는 혈색소인 메테모글로빈(methemoglobin)을 생성함에 따라 산소부족현상으로 인한 유아청색증의 원인이 된다. 따라서 질소는 수질을 좌우하는 대표적인 오염물질로 분류되어 있다. 수중에 존재 하는 질소의 농도는 다양한 형태의 유,무기성 질소를 아질산성 질소의 형태로 전환하여 그리스법(Griess법)으로 흡광도를 통해 정량한다. 상기 그리스법에 의한 흡광 검출법은 국내 수질오염공정시험기준과 미국환경보호국에서 수중 질소 정량분석의 표준방법으로 채택한 신뢰성이 높은 분석법이다.
산성조건에서 아질산이온은 술파닐아마이드의 아민기와 반응하여 디아조늄염을 생성하며, 디아조늄염이 N-(1-나프틸)-에틸렌디아민 디하이드로클로라이드와 결합하여 생성된 붉은 색의 아조 염료는 540nm에서 최대흡수도를 가진다. 그리스법을 이용한 아질산성 질소의 정량분석을 위해 광원(42)으로 최대세기파장이 526nm인 녹색 발광다이오드가 적용되고, 검출기(43)는 광전자증배관을 사용하며, 아조 염료의 최대흡수파장에 맞추어 540nm의 빛을 10nm의 반값 반폭으로 투과하는 간섭 필터(44)를 적용한다. 미세유체칩에 8cm 길이의 흐름셀(내경: 250㎛, 외경: 500㎛)을 적용 설치한다.
그리스법을 위한 발색시약은 85% 인산 10mL에 0.2g의 술파닐아마이드와 0.02g의 N-(1-나프틸)-에틸렌디아민 디하이드로클로라이드를 녹인 다음 증류수로 최종부피를 100mL로 하여 제조한다. 그리고 아질산나트륨을 증류수에 녹여 1000ppm의 질소를 포함하는 아질산성 질소 표준시료를 제조한 다음 희석하여 5ppb, 10ppb, 50ppb, 100ppb, 150ppb, 200ppb, 300ppb의 질소를 포함하는 아질산성 질소 표준시료를 각각 제조한다.
도 5는 미세유체칩(30)에 상기 제조한 발색시약과 바탕시료인 증류수 그리고 서로 다른 농도의 아질산성 질소 표준시료를 순차적으로 주입하여 얻은 검출신호이다. 각 용액의 주입 시 사용된 연동펌프의 유속은 10㎕/min이다.
[실험예 2]
흡광도는 비어의 법칙에 따라 농도에 비례하므로 이상적인 경우 시료의 농도에 대한 흡광도는 직선성을 가지며, 일차함수로 표현이 가능하다. 그러나 여러 가지 원인들에 의해 직선성을 보이는 농도범위가 줄어든다. 본 실험예에서는 미광이 흡광도에 어떠한 영향을 미치는지 알아보기 위해 미세유체칩의 종류에 따른 아질산성 질소의 흡광 검출 결과를 비교한다.
도 6은 미세유체칩의 상판(10)과 하판(20) 모두를 검정색 PDMS로 제작한 것과 투명 PDMS로 제작한 것을 총질소 또는 총인 분석용 연속 미세 유체 장치에 적용하여 얻은 각각의 검량곡선을 도시한 것이다. 총질소 또는 총인 분석용 연속 미세 유체 장치는 아조 염료의 최대흡수파장에 맞추어 540nm의 빛을 10nm의 반값 반폭으로 투과하는 간섭 필터(44)를 사용하여 다색 파장에 의해 검량곡선이 직선성을 보이는 농도범위가 줄어드는 문제점을 최소화하였다.
도 6에 도시한 (1)은 검정색 PDMS가 미광을 흡수하여 검출기로 들어가는 것을 최소화함에 따라 5ppb~300ppb의 농도범위에서 직선성을 보인다. 이와는 달리 도 6에 도시한 (1')은 미광에 의해 직선성을 보이는 농도범위가 줄어든다. 더 자세하게 설명하면 미세유체칩에서 흡광 검출 흐름셀로 입사된 빛이 시료에 의해 흡수되는 정도는 동일하나, 투명 미세유체칩을 사용하였을 때 미광에 의해 검출기로 들어가는 빛이 검정색 미세유체칩을 사용하였을 때보다 많기 때문에 시료의 흡광도가 감소하여 (1')은 직선성을 보이는 농도범위가 줄어든다.
또한, 시료의 농도가 증가함에 따라 직선성에서 벗어나는 정도가 커지는 이유는 시료의 농도가 증가함에 따라 투과도는 감소하나, 모든 농도범위에서 검출기로 들어가는 미광은 동일하기 때문이다. 즉, 시료의 농도가 높아짐에 따라 검출기로 들어가는 모든 빛에 대한 미광의 분율이 커지기 때문이다.
[실험예 3]
본 실험예 3에서는 다색 파장이 흡광도에 어떠한 영향을 미치는지 알아보기 위해 실험예 1의 아질산성 질소 분석용 총질소 또는 총인 분석용 연속 미세 유체 장치에 간섭 필터(44)를 사용한 것과 사용하지 않은 것을 이용하여 각각의 검량곡선을 얻어 도 7에 도시하였다. 상기한 서로 다른 총질소 또는 총인 분석용 연속 미세 유체 장치는 미광에 의해 검량곡선이 직선성을 보이는 농도범위가 줄어드는 문제점을 최소화하기 위해 검정색 PDMS로 제작한 미세유체칩을 사용한다.
아질산성 질소는 그리스법에 의해 생성된 아조 염료의 흡광도를 최대흡수파장인 540nm에서 측정하여 정량분석이 가능하다. 따라서 넓은 농도범위에서 검량곡선이 직선성을 갖도록 하기 위해서는 광원으로 540nm의 단색 파장을 사용하거나, 아조 염료의 최대흡수도가 변하지 않는 540nm 주변에 좁은 띠너비(bandwidth)를 갖는 파장의 빛을 사용해야 한다.
본 실험예에서 사용한 간섭 필터(44)는 아조 염료의 최대흡수파장인 540nm 의 파장을 10nm의 좁은 반값 반폭으로 투과하는 것을 사용한다. 그리고 광원(42)으로 사용한 녹색 발광다이오드의 최대세기파장은 526nm이고, 반값 반폭은 47nm이다.
도 7에 도시한 (2)는 상기 간섭 필터(44)를 통과한 10nm의 좁은 반값 반폭을 가지는 540nm 의 빛이 검출기(43)로 들어감에 따라 5ppb~300ppb의 농도범위에서 직선성을 가진다. 이와는 달리 도 7에 도시한 (2')는 간섭 필터(44)를 사용하지 않아 526nm의 빛이 47nm의 넓은 반값 반폭으로 시료에 조사된다. 따라서 시료가 아조 염료의 최대흡수도보다 낮은 흡수도를 갖는 파장의 빛을 흡수함에 따라 흡광도가 감소하여 직선성을 보이는 농도범위가 줄어든 것을 알 수 있다.
이하에서는 도 8을 참조하여 본 발명의 제2 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치에 대해서 설명한다.
도 8은 본 발명의 제2 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치의 구성도이다.
도 8을 참조하여 설명하면, 본 제2 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치(102)는 외형을 이루는 하우징(41)과 하우징(41) 내에 삽입 설치되며 광 입사부(22)의 아래에 배치된 광원(42), 검출부(23)의 아래에 배치된 검출기(43), 및 미세유체칩(30), 산화분해부재(72), 및 카드뮴-구리 컬럼(73)을 포함한다.
한편, 미세유체칩(30)에는 시료 및 시약이 저장된 공급부(71)가 연결 설치되는데, 공급부(71)에는 연동펌프가 설치되어 미세유체칩(30)으로 시료 및 시약을 공급한다. 공급부(71)에는 시료 주입관(74), 표준물첨가시료 주입관(75), 산화제 주입관(76), 완충용액 주입관(77), 및 시약 주입관(78)이 연결 설치된다.
산화분해부재(72)는 유기성 질소를 포함하는 다양한 질소 화합물을 질산성 질소로 산화분해한다. 산화분해부재(72)는 열 또는 빛을 발생시키는 반응막대(72a)와 튜브(72b)를 포함하며, 튜브(72b)는 원기둥 형태의 소형 반응막대(72a)에 밀착하여 감겨진다. 반응막대(72a)는 자외선을 발생시키는 자외선 램프로 이루어질 수 있으며, 자외선 램프는 최대세기파장이 254nm인 자외선을 방출할 수 있다. 또한 반응막대(72a)는 열선이 내장된 히터로 이루어질 수도 있다.
반응막대(72a)가 자외선 램프로 이루어지는 경우, 반응막대에서 발생된 자외선에 의하여 산화제가 분해되어 산화반응이 이루어질 수 있다. 또한 반응막대(72a)가 히터로 이루어지는 경우, 튜브에 열을 공급하여 산화반응을 가속시킬 수 있다.
한편, 튜브(72b)는 시료가 이동하는 통로를 제공하며, 모세관으로 이루어질 수 있다. 또한 튜브(72b)는 불소 수지인 테프론(Teflon)으로 이루어질 수 있으며 유리, 고분자, 금속 등으로 이루어질 수도 있다.
튜브(72b)가 테프론 모세관으로 이루어지고 자외선 램프에 감겨질 경우, 벽 두께가 얇아 광투과거리를 최소화함으로써 빛을 효율적으로 이용할 수 있고, 내부 부피가 수 십 ㎕로 작음에 따라 시료의 열 및 물질전달 속도가 빠르기 때문에 수 분 내에 다양한 질소 화합물의 광산화분해가 가능하다.
또한, 튜브(72b)는 반응막대(72a)에서 발생하는 열을 직접적으로 받기 때문에 산화분해 효과를 극대화할 수 있다. 튜브(72b)의 입구에는 시료 주입관(74), 표준물첨가시료 주입관(75), 산화제 주입관(76)이 연결 설치된다. 시료 주입관(74)으로는 분석을 하기 위해 채취된 시료가 주입되고, 표준물첨가시료 주입관에는 표준물첨가법을 적용하기 위한 표준물첨가시료가 주입된다.
표준물첨가법이라 함은 분석하고자 하는 성분과 같은 물질의 일정량을 시료 용액에 넣은 다음, 전체를 분석하고 여기에서 분석하고자 하는 성분을 정량하는 분석법으로서 화학적 방해나 간섭에 의한 분석 결과의 부정확성을 보정하기 위해 사용된다. 표준물첨가시료는 분석하고자 하는 성분을 포함하는 물질 또는 증류수로 이루어질 수 있다.
산화제 주입관(76)으로는 산화제가 주입되는데 산화제는 붕산 완충용액과 이에 용해된 과황산칼륨을 포함할 수 있다. 또한 산화제는 황산용액과 이에 용해된 과황산칼륨을 포함할 수 있다.
한편, 튜브(72b)의 출구와 완충용액 주입관(77)은 카드뮴-구리 컬럼(73) 입구에 연결 설치된다. 이에 따라 산화분해반응을 마친 시료는 완충용액과 함께 카드뮴-구리 컬럼(73)으로 주입된다.
한편, 카드뮴-구리 컬럼(73)은 질산성 질소를 아질산성 질소로 환원하며, 튜브에 구리로 활성화된 카드뮴 입자가 채워질 수 있다. 또한, 카드뮴-구리 컬럼(73)은 카드뮴 입자가 튜브를 빠져나가는 것을 막기 위해 카드뮴 입자보다 내경이 작은 튜브로 양 끝이 막혀진 구조로 이루어진다.
카드뮴-구리 컬럼(73)의 출구는 가공시료 주입관(79)과 연결되며 가공시료 주입관(79)은 미세유체칩(30)의 시료 주입구(11)에 설치된 체결부(33)에 연결 설치된다. 이에 따라 카드뮴-구리 컬럼(73)을 통해서 아질산성 질소로 환원된 시료는 시료 주입구(11)로 이동한다.
하우징(41)은 대략 직육면체의 상자 형태로 이루어지며, 하우징(41)에는 광원(42)의 상부에 위치하는 입사채널(45)과 검출기(43)의 상부에 위치하는 검출채널(46)이 형성된다. 입사채널(45)은 광원(42)과 광 입사부(22) 사이에 위치하여 광원(42)에서 출사된 빛을 광 입사부(22)로 전달하는 통로가 되며, 검출채널(46)은 검출기(42)와 검출부(23) 사이에 위치하여 검출부(23)에서 검출기(43)로 빛을 전달하는 통로가 된다.
하우징(41)은 빛을 흡수할 수 있도록 검정색으로 착색된 금속으로 이루어지며, 흐름셀(31)을 갖는 미세유체칩(30)은 하우징(41)의 상면에 고정 설치된다. 이에 따라 광원(42)에서 배출된 빛은 직접 검출기(43)로 이동하지 못하고, 흐름셀(31)을 거쳐서 검출기(43)로 이동한다.
광원(42)은 발광다이오드로 이루어지며, 광원(42)은 분석물질과 시약의 종류에 따라 최대흡수파장을 고려하여 다양하게 선택될 수 있다. 검출기(43)는 광전자증배관으로 이루어지며, 흐름셀(31)에서 출사된 빛에서 특정 파장의 세기를 검출한다. 검출기(43)와 검출채널(46) 사이에는 간섭 필터(44)가 설치된다. 간섭 필터(44)는 입사되는 빛을 좁은 반값 반폭으로 투과시켜 검량곡선의 직선성을 향상시키는 역할을 한다. 간섭 필터(44)는 광원(42)에서 출사된 빛의 반값 반폭보다 더 작은 반값 반폭으로 빛을 투과시킨다.
광원(42)에는 전원(61)이 연결 설치되고, 검출기(43)에도 전원(62)이 연결 설치된다. 또한, 검출기(43)에는 검출기(43)에서 발생된 신호를 가공 처리하는 디지털 멀티미터(63)가 연결 설치되고, 디지털 멀티미터(63)는 가공된 신호를 컴퓨터(64)로 전달한다.
광원(42)과 검출기(43) 사이에는 차단벽(47)이 설치되는 바, 차단벽(47)은 하우징(41)의 상면에서 하면으로 이어져 배치된다. 상기한 차단벽(47)에 의하여 광원(42)에서 발생된 미광이 검출기(43)로 유입되는 것을 차단할 수 있다.
또한, 광원(42)과 검출기(43)는 차단벽을 사이에 두고 측방향으로 이격 배치되어 광원(42)에서 출사되는 미광이 검출기(43)로 유입되는 것을 원천적으로 차단할 수 있다.
[실험예 4]
총질소는 무기성 질소와 유기성 질소의 합으로 수중에 질소를 정확하게 정량하기 위해서는 다양한 형태로 존재하는 질소 화합물을 흡광 검출이 가능한 형태로 모두 전환해야 한다. 제2 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치(102)는 산화분해부재(72)를 이용하여 수중의 다양한 질소 화합물을 질산성 질소로 산화분해하고, 산화분해부재(72)에서 생성된 질산성 질소를 카드뮴-구리 컬럼(73)을 통해 아질산성 질소로 환원한다. 상기 환원된 아질산성 질소를 발색시약과 반응하여 생성된 아조 염료의 흡광도를 총질소 또는 총인 분석용 연속 미세 유체 장치(102)를 통해 검출하고, 이를 통해 총질소를 정량한다.
본 실험예에서는 카드뮴-구리 컬럼(73)의 성능을 확인하기 위해 본 제2 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치(102)로 질산성 질소 표준시료를 아질산성 질소로 환원하여 그 환원율을 확인한다.
산화분해부재(72)는 원기둥 형태의 반응막대(72a)에 산화분해 반응기인 튜브(72b)(내경: 200㎛, 외경: 360㎛)이 감긴 구조로 이루어진다. 튜브(72b)는 테프론으로 이루어지며 1m의 길이를 갖는다. 여기서 반응막대(72a)는 자외선 램프로 이루어지며 자외선 램프는 최대세기파장이 254nm이고, 길이가 5.30cm이며, 직경이 0.65cm인 자외선 램프로 이루어질 수 있다. 또한, 튜브(72b)는 고온에서 사용 가능한 캡톤(Kapton) 테이프에 의하여 반응막대(72a)에 고정된다.
산화분해부재(72)는 튜브(72b)의 벽 두께인 80μm의 짧은 광투과거리를 통해 빛을 효율적으로 이용할 수 있고, 내부부피가 31μL로 작음에 따라 시료의 열 및 물질전달 속도가 빠르기 때문에 수 분 내에 다양한 질소 화합물의 산화분해가 가능하다. 또한, 반응막대(72a)에서 발생하는 열을 직접적으로 받기 때문에 산화분해 효과를 극대화 할 수 있다. 산화분해 과정에서 사용되는 산화제는 2m mol의 과황산칼륨을 0.1M 붕산 완충용액(pH 9.0) 100mL에 녹여 제조하며, 산화제 주입관(76)을 통해 산화분해부재(72)로 주입된다.
산화분해부재(72)는 시료 주입관(74), 표준물첨가시료 주입관(75), 산화제 주입관(76)을 통해 2㎕/min의 유속으로 반응에 필요한 시약과 시료를 공급받는다.
한편, 테프론 튜브(72b)의 출구와 완충용액 주입관(77)은 티 커넥터의 두 입구에 각각 연결 설치되며, 티 커넥터의 출구는 카드뮴-구리 컬럼(73)의 입구와 연결 설치되어 산화분해 반응을 마친 시료는 완충용액과 함께 카드뮴-구리 컬럼(73)으로 주입된다.
상기한 카드뮴-구리 컬럼(73)은 내경이 1mm이고 길이가 5cm인 튜브와 튜브 내에 채워진 활성화된 카드뮴 입자를 포함한다. 카드뮴 입자는 318~847㎛ 크기로 이루어지며, 산으로 처리된 후 80mM 황산구리 용액으로 활성화된다.
카드뮴 입자가 튜브를 빠져나가는 것을 막기 위해 튜브의 양 쪽 끝에 카드뮴 입자보다 내경이 작은 튜브(내경: 100㎛, 외경 1/16inch)을 연결하며, 카드뮴-구리 컬럼(73)의 입구는 산화분해부재(72)의 출구와 연결 설치된다. 카드뮴-구리 컬럼(73)의 출구는 가공시료 주입관(79)과 연결되며 가공시료 주입관(79)은 미세유체칩(30)의 시료 주입구(11)에 설치된 체결부(33)에 연결 설치된다.
한편, 카드뮴-구리 컬럼(73)을 활성화하고, 환원반응 중에 생성될 수 있는 카드뮴 침전이 컬럼을 막는 것을 방지하기 위하여 완충용액을 사용하며, 완충용액은 이미다졸로 이루어질 수 있다. 이미다졸 완충용액은 5m mol의 이미다졸(1,3-디아조-2,4-사이클로펜타디엔)과 5mM 황산구리 용액 100㎕를 증류수에 녹여 pH 7.8로 조절한 후, 최종부피를 100mL로 하여 제조하며, 완충용액 주입관(77)을 통해 5.5㎕/min 유속으로 카드뮴-구리 컬럼(73)으로 주입한다.
상기한 방법에 따라 시료, 표준물첨가시료 및 산화제는 산화분해부재(72)로 주입되어 시료 내 존재하는 다양한 질소 화합물이 질산성 질소로 전환된다. 산화분해부재(72)에서 전환된 질산성 질소는 완충용액 주입관(77)으로 주입된 이미다졸 완충용액과 함께 카드뮴-구리 컬럼(73)으로 주입되어 카드뮴-구리 입자 사이를 통과하면서 충분한 혼합과 환원반응이 이루어지며, 반응시간은 30초이다.
주입된 질산성 질소는 카드뮴-구리 컬럼(73)을 통해 아질산성 질소로 환원되며, 환원된 아질산성 질소는 미세유체칩(30)의 시료 주입구(11)로 주입된다. 미세유체칩(30)으로 주입된 시료는 시약 주입관(78)으로 11㎕/min 의 유속으로 주입된 발색시약과 반응하여 생성된 아조 화합물의 흡광도를 검출한다. 발색시약은 실험예 1에서 사용한 발색시약을 증류수로 4배 희석하여 사용한다. 질산성 질소와 아질산성 질소 표준시료는 질산나트륨과 아질산나트륨을 각각 증류수에 용해하여 제조한 1000ppm의 질소를 포함하는 표준시료를 제조한 다음, 희석하여 4ppm의 질소를 포함하는 질산성 질소와 아질산성 질소 표준시료를 제조한다.
본 실험예에서는 질산성 질소의 환원율을 확인하기 위해 시료 주입관(74)으로 증류수(바탕시료), 4ppm의 질소를 포함하는 아질산성 질소 표준시료와 질산성 질소 표준시료를 순차적으로 주입하여 각각의 검출신호를 얻는다. 그리고 시료 주입관(74)을 통해 주입된 아질산성 질소가 산화분해부재(72)에서 질산성 질소로 산화되므로 산화제 주입관(76)으로 산화제 대신 증류수를 주입하고, 산화분해부재(72)의 반응막대(72a)는 켜지 않는다. 또한, 본 실험예는 질소의 정량을 위한 것이 아니므로, 표준물첨가시료 주입관(75)에는 증류수를 주입한다.
도 9는 상기한 방법으로 얻은 바탕신호와 4ppm의 질소를 포함하는 아질산성 질소 표준시료와 질산성 질소 표준시료의 검출신호이다. 각각의 검출 신호를 흡광도로 계산하고, 4ppm의 질소를 포함하는 아질산성 질소 표준시료의 흡광도를 기준으로 하여 질산성 질소의 환원율을 계산한다. 동일한 실험을 세 번 반복하여 얻은 질산성 질소의 아질산성 질소로의 평균 환원율은 101%(상대표준편자 0.85%)로 확인되었다. 본 실험예의 결과로부터 카드뮴-구리 컬럼(73)은 30초의 짧은 시간 동안 질산성 질소 아질산성 질소로 100% 환원함으로써 총질소 분석시간을 최소화할 수 있음을 알 수 있다.
[실험예 5]
실험예 5에서는 본 발명의 제2 실시예의 산화분해부재(72)를 이용하여 수중에 존재하는 다양한 질소 화합물을 질산성 질소로 산화분해하여 전환율을 확인한다. 총질소는 무기성 질소(질산성 질소, 아질산성 질소, 암모니아성 질소)와 유기성 질소로 나뉜다. 따라서 본 실험예는 무기성 질소 화합물과 유기성 질소 화합물 각각의 전환율을 확인하기 위해 두 개의 실험을 수행한다. 본 실험을 위해 실험예 4의 총질소 또는 총인 분석용 연속 미세 유체 장치의 표준물첨가시료 주입관(75), 산화제 주입관(76), 완충용액 주입관(77), 시약 주입관(78)에는 실험예 4에서 기술한 방법으로 제조한 용액을 각각의 주입관을 통해 주입한다. 시료 주입관(74)으로 주입한 질소 화합물 표준시료는 산화분해부재(72)에서 5분 동안 반응하여 질산성 질소로 전환되고, 전환된 질산성 질소는 실험예 4에서 확인한 바와 같이 카드뮴-구리 컬럼(73)에서 아질산성 질소로 100% 환원된다. 본 실험예에서 사용되는 질소 화합물인 염화암모늄, 요소, 글라이신(glycine)과 에틸렌디아민테트라아세트산(ethylenediaminetetraacetic acid, EDTA)의 표준시료는 각각 증류수에 용해하여 1000ppm의 질소를 포함하는 표준시료를 제조한 다음, 희석하여 4ppm의 질소를 포함하는 표준시료를 제조한다.
도 10은 시료 주입관(74)으로 증류수(바탕시료), 4ppm의 질소를 포함하는 질산성 질소, 아질산성 질소, 염화암모늄 표준시료를 연속적으로 주입하여 얻은 검출신호이며, 도 11은 시료 주입관(74)으로 증류수(바탕시료), 4ppm의 질소를 포함하는 질산성 질소, 요소, 글라이신과 에틸렌디아민테트라아세트산 표준시료를 연속적으로 주입하여 얻은 검출신호이다. 각각의 검출신호를 흡광도로 계산하고, 4ppm의 질소를 포함하는 질산성 질소 표준시료의 흡광도를 기준으로 하여 질소 화합물의 전환율을 계산한다. 상기한 실험을 각각의 질소화합물 표준시료에 대하여 세 번 반복하여 얻은 전환율과 상대표준편차는 표 1에 나타내었다.

표 1의 결과로부터 본 발명의 산화분해부재(72)는 복잡한 구조를 가져 분해가 쉽지 않은 유기성 질소를 포함하는 다양한 질소 화합물을 수 분 내에 질산성 질소로 산화분해할 수 있음을 알 수 있다. 따라서, 상기한 산화분해부재(72)와 카드뮴-구리 컬럼(73)은 연속흐름상에서 수중의 다양한 질소 화합물을 아질산성 질소로 100%(±2%) 전환함에 따라 정확하고 재현성(2% 이하의 상대표준편차)이 높은 총질소 정량이 가능하다.
[실험예 6]
염도가 높은 해수나 조석에 의해 시간과 위치에 따라 염도가 변화하는 강하구 내에 총질소를 흡광광도법으로 정확하게 정량하기 위해서는 시료 내에 염분을 포함한 다양한 매트릭스에 의한 분석오차를 보정해야 한다. 그리고 흡광 분석에 필요한 바탕시료는 분석물질을 제외한 모든 물질의 조성이 동일한 시료이므로 실제시료의 경우 바탕시료를 준비하는 것은 어려우며, 특히 강하구와 같이 염도가 시간과 위치에 따라 변하는 경우, 바탕시료를 준비하는 것은 불가능하다. 따라서 특정 염도의 바탕시료를 사용하여 염도가 변화하는 실제시료 내 총질소를 오차 없이 분석할 수 있어야 한다. 본 실험예에서는 실험예 4의 총질소 또는 총인 분석용 연속 미세 유체 장치에 염분을 포함하는 총질소 표준시료를 주입하고, 바탕시료로 증류수를 사용하여 총질소의 정확한 정량이 가능한 지 확인한다.
본 실험예에서는 총질소 표준시료와 표준물첨가시료로 다양한 질소화합물 중에 가장 안정한 질산성 질소를 사용한다. 총질소 표준시료와 표준물첨가시료는 질산나트륨을 증류수에 용해하여 제조한 1000ppm의 질소를 포함하는 총질소 표준시료를 제조한 다음, 희석하여 수 ppm의 질소를 포함하는 총질소 표준시료와 표준물첨가시료를 각각 제조한다. 30‰의 염분을 포함하는 총질소 표준시료는 5M(293‰)의 염화나트륨 표준용액을 증류수로 희석하여 60의 염화나트륨 용액을 제조하고, 2ppm, 8ppm의 질소를 포함하는 총질소 표준시료와 각각 1:1로 희석하여 30‰의 염화나트륨과 1ppm, 4ppm의 질소를 포함하는 총질소 표준시료를 제조한다. 염화나트륨이 해수의 염류 중 85% 이상을 차지하므로 본 실험예에서 염도를 조절하는 데 사용한다.
표준물첨가법으로 총질소를 분석하기 위해 상기한 총질소 또는 총인 분석용 연속 미세 유체 장치에 다음과 같은 순서로 용액을 주입한다. 먼저 바탕신호를 얻기 위해 시료 주입관(74)와 표준물첨가시료 주입관(75)으로 증류수를 주입하고, 산화제 주입관(76), 완충용액 주입관(77) 및 시약 주입관(78)으로 실험예 4에서 기술한 방법으로 제조한 용액을 각각의 주입관을 통해 주입한다. 바탕신호를 얻은 후 시료 주입관(74)으로 총질소 표준시료를 주입하여 검출신호를 얻는다. 그리고 표준물첨가시료 주입관(75)으로 1ppm, 2ppm, 3ppm의 질소를 포함하는 총질소 표준물첨가시료를 순차적으로 15분씩 주입하여 각각의 검출신호를 얻는다. 상기한 본 실험예에 사용되는 모든 용액은 실험예 4에서와 같은 유속으로 주입한다. 상기한 방법으로 얻은 검출신호로부터 흡광도를 계산하여 표준물첨가시료의 농도에 대한 흡광도의 추세선을 그리고, 이를 외삽하여 얻은 x 절편의 값에 -1을 곱하여 총질소의 농도를 알 수 있다.
도 12a와 도 13a는 상기한 방법으로 얻은 염분을 포함하지 않은 1ppm, 4ppm의 질소를 포함하는 총질소 표준시료 각각의 표준물첨가에 의한 검출신호이며, 도 14a와 도 15a는 30‰의 염화나트륨과 1ppm, 4ppm의 질소를 포함하는 총질소 표준시료 각각의 표준물첨가에 의한 검출신호이다. 그리고 도 12b, 도 13b, 도 14b 및 도 15b는 도 12a, 도 13a, 도 14a 및 도 15a의 검출신호를 표준물첨가시료의 농도에 대한 흡광도로 나타낸 그래프이다.

표 2는 상기 4개의 총질소 표준시료에 대하여 상기한 방법으로 실험을 세 번 반복하여 얻은 총질소의 검출농도와 상대오차(4.4% 이하), 그리고 상대표준편차 (0.830% 이하)를 나타낸 것이다. 표 2의 결과로부터 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 염분이 없는 표준물첨가시료를 사용하여 높은 염도를 가지는 시료 내에 총질소의 농도를 정확하고 높은 재현성으로 분석할 수 있음을 알 수 있다.
[실험예 7]
실험예 7에서는 실제시료 내 총질소의 농도를 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치로 분석하고, 그 결과를 상용화된 자외선-가시광선 흡광광도계로 분석한 결과와 비교한다. 실제시료는 바다와 강하구에서 채수하였으며, 염도가 변화하는 강하구에서는 동일한 위치에서 만조와 간조 때 각각 시료를 채수하였다. 모든 시료의 염도는 염도계로 측정하였다. 본 제2 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치를 이용한 실제시료의 총질소 분석방법은 실험예 6에 기술한 바와 같다. 비교분석법은 국내 수질오염공정시험기준과 미국환경보호국에서 수중 질소 정량분석의 표준방법으로 채택한 신뢰성이 높은 분석법으로 채수한 시료를 산화제와 혼합하고 고압증기멸균기를 이용하여 0.13MPa, 120℃에서 30분간 분해한 다음, 분해과정이 끝난 시료를 카드뮴-구리 컬럼(73)에 통과시켜 아질산성 질소로 환원한다. 환원된 아질산성 질소는 표준물첨가법을 이용하여 자외선-가시광선 흡광광도계로 분석하여 총질소를 정량한다.

표 3은 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치와 자외선-가시광선 흡광광도계로 채수한 실제시료 내 총질소의 농도를 세 번 반복하여 분석한 결과를 나타낸다. 상기한 두 가지 방법으로 검출한 총질소의 농도가 거의 유사하고, 2% 이하의 낮은 상대표준편차를 보이는 것으로부터 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 정확하고 재현성이 높은 총질소 정량이 가능함을 알 수 있다.
본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 수중에 존재하는 총질소를 분석하기 위한 산화분해부재(72), 카드뮴-구리 컬럼(73), 총질소 또는 총인 분석용 연속 미세 유체 장치를 소형화하고, 분석에 필요한 각각의 과정이 연속흐름상에서 효율적으로 이루어져 빠른 시간에 정확하고 재현성이 높은 총질소 분석이 가능하다. 그리고 표준물첨가법을 사용하여 염도가 높은 해수나 염도가 계속적으로 변화하는 강하구뿐만 아니라 다양한 매트릭스가 존재하는 시료 내 총질소를 정확하게 분석할 수 있다. 또한, 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 광 경로 길이의 조절이 가능함에 따라 넓은 농도범위의 총질소를 검출할 수 있어 대부분의 수환경에 적용이 가능하다.
이하에서는 도 16을 참조하여 본 발명의 제3 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치에 대해서 설명한다.
도 16은 본 발명의 제3 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치의 구성도이다.
도 16을 참조하여 설명하면, 본 제3 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치(103)는 시료 내 다양한 인 화합물을 무기인산염으로 산화하기 위한 산화분해부재(82)를 포함한다.
외형을 이루는 하우징(41)과 하우징(41) 내에 삽입 설치되며 광 입사부(22)의 아래에 배치된 광원(42), 검출부(23)의 아래에 배치된 검출기(43), 및 미세유체칩(30), 및 산화분해부재(82)를 포함한다.
한편, 미세유체칩(30)에는 시료 및 시약이 저장된 공급부(81)가 연결 설치되는데, 공급부(81)에는 연동펌프가 설치되어 미세유체칩(30)으로 시료 및 시약을 공급한다. 공급부(81)에는 시료 주입관(84), 표준물첨가시료 주입관(85), 산화제 주입관(86), 및 시약 주입관(88)이 연결 설치된다.
산화분해부재(82)는 원기둥 형태의 반응막대(82a)에 산화분해 반응기인 튜브(82b)(내경: 200㎛, 외경: 360㎛)이 감긴 구조로 이루어진다. 튜브(82b)는 테프론으로 이루어지며 1m의 길이를 갖는다. 여기서 반응막대(82a)는 자외선 램프로 이루어지며 자외선 램프는 최대세기파장이 254nm이고, 길이가 5.30cm이며, 직경이 0.65cm인 자외선 램프로 이루어질 수 있다. 또한, 튜브(82b)는 고온에서 사용 가능한 캡톤(Kapton) 테이프에 의하여 반응막대(82a)에 고정된다.
산화분해부재(82)는 튜브(82b)의 벽 두께인 80㎛의 짧은 광투과거리를 통해 빛을 효율적으로 이용할 수 있고, 내부부피가 31㎕로 작음에 따라 시료의 열 및 물질전달 속도가 빠르기 때문에 수 분 내에 다양한 인 화합물을 무기인산염으로 산화분해가 가능하다. 또한, 반응막대(82a)에서 발생하는 열을 직접적으로 받기 때문에 산화분해 효과를 극대화 할 수 있다.
한편, 튜브(82b)의 출구는 가공시료 주입관(89)과 연결되며 가공시료 주입관(89)은 미세유체칩(30)의 시료 주입구(11)에 설치된 체결부(33)에 연결 설치된다. 이에 따라 산화분해부재(82)에 의하여 산화된 시료는 시료 주입구(11)로 이동한다.
한편, 시료의 분석을 위한 미세유체칩(30), 하우징(41) 및 검출기(43)는 상기한 제1 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치와 동일한 구조로 이루어지므로 동일한 구조에 대한 중복 설명은 생략한다.
[실험예 8]
총인은 유기성 인과 무기인산염의 합으로 수중에 인을 정확하게 정량하기 위해서는 다양한 형태로 존재하는 인 화합물을 흡광 검출이 가능한 무기인산염으로 모두 전환해야 한다. 본 발명의 제3 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치(103)는 수중의 다양한 인 화합물을 산화분해부재(82)를 이용하여 무기인산염으로 전환한 다음, 전환된 무기인산염을 산성조건에서 몰리브덴산 암모늄 및 메타바니딘산 암모늄과 반응하여 생성된 몰리브덴황의 최대흡수파장인 370nm에서 흡광도를 측정해 총인의 정량이 가능하다. 몰리브덴황법은 발색시약이 1년 동안 안정해 현장적용성이 높으며 잦은 유지보수를 필요로 하지 않는다.
본 제3 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치는 광원(42)으로 최대세기파장이 370nm인 발광다이오드가 적용되고, 흡수파장에 맞추어 370nm의 빛을 10nm의 반값 반폭으로 투과하는 간섭 필터(44)를 적용한다.
본 실험예에서는 상기한 총질소 또는 총인 분석용 연속 미세 유체 장치(103)에 1ppm의 인을 포함하는 무기인산염과 다양한 유기성 인 화합물을 연속적으로 주입하여 얻은 검출신호로부터 전환율을 확인한다. 무기인산염 표준시료와 표준물첨가시료는 인산이수소나트륨을 증류수에 용해하여 1000ppm의 인을 포함하는 무기인산염 표준시료를 제조한 다음, 희석하여 수 ppm의 인을 포함하는 무기인산염 표준시료와 표준물첨가시료를 각각 제조한다. 산화분해과정에서 사용되는 산화제는 10m mol의 과황산칼륨을 0.36M 황산용액 10mL에 녹여 제조하며, 산화제 주입관(86)을 통해 산화분해부재(82)로 주입한다. 상기한 방법으로 제조한 표준시료, 표준물첨가시료, 그리고 산화제는 2.5㎕/min의 유속으로 산화분해부재(82)로 주입된다. 산화분해부재(82)에서 전환된 무기인산염은 가공시료 주입관(89)를 통해서 미세유체칩(30)의 시료 주입구(11)로 주입된다.
몰리브덴황법을 위한 발색시약은 37wt% 염산 95mL에 0.7143g의 몰리브덴산 암모늄과 0.0358g의 메타바니딘산 암모늄을 녹인 다음 증류수로 최종부피를 1000mL로 하여 제조한다. 총질소 또는 총인 분석용 연속 미세 유체 장치(103)로 주입된 시료는 시약 주입관(88)을 통해 7.5㎕/min의 유속으로 미세유체칩(30)의 시약 주입구(12)로 주입된 발색시약과 반응하여 생성된 몰리브덴황의 흡광도를 측정하여 총인을 정량한다. 본 실험예에서 사용되는 인 화합물인 포도당-6-인산(glucose-6-phosphate), 아데노신인산(adenosine monophosphate), 피트산(phytic acid)의 표준시료는 각각 증류수에 용해하여 제조한 1000ppm의 인을 포함하는 표준시료를 제조한 다음, 희석하여 1ppm의 인을 포함하는 표준시료를 제조한다.
도 17은 시료 주입관(84)에 증류수(바탕시료), 1ppm의 인을 포함하는 무기인산염, 포도당-6-인산, 아데노신인산, 피트산 표준시료를 순차적으로 주입하여 얻은 검출신호이다. 각각의 검출신호를 흡광도로 계산하고, 1ppm의 인을 포함하는 무기인산염 표준시료의 흡광도를 100%로 기준하여 인 화합물의 전환율을 계산한다. 상기한 실험을 각각의 인 화합물 표준시료에 대하여 세 번 반복하여 얻은 전환율과 상대표준편차는 표 4에 나타내었다.

표 4의 결과로부터 본 발명의 산화분해부재(82)는 연속흐름상에서 복잡한 구조를 가져 분해가 쉽지 않은 유기성 인을 포함하는 다양한 인 화합물을 수 분 내에 무기인산염으로 100%(±0.854%) 전환함에 따라 정확하고 재현성(2% 이하의 상대표준편차)이 높은 총인 정량이 가능하다.
[실험예 9]
본 실험예에서는 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치에 무기인산염 표준시료를 주입하여 총인의 정확한 정량이 가능한지 확인한다. 표준물첨가법으로 총인을 분석하기 위해 실험예 8의 총질소 또는 총인 분석용 연속 미세 유체 장치에 다음과 같은 순서로 용액을 주입한다.
본 실험예에서 총인 표준시료와 표준물첨가시료로 무기인산염을 사용한다. 먼저 바탕신호를 얻기 위해 시료 주입관(84)과 표준물첨가시료 주입관(85)으로 증류수를 주입하고, 산화제 주입관(86)과 시약 주입관(88)으로 산화제와 발색시약을 각각 주입한다. 상기한 본 실험예에 사용되는 모든 용액과 각각의 용액을 주입하는 유속은 실험예 8에서와 같다. 바탕신호를 얻은 후 시료 주입관(84)으로 총인 표준시료를 주입하여 검출신호를 얻는다. 그리고 표준물첨가시료 주입관(85)으로 0.5ppm, 1ppm, 1.5ppm의 인을 포함하는 총인 표준물첨가시료를 순차적으로 15분씩 주입하여 각각의 검출신호를 얻는다.
상기한 방법으로 얻은 검출신호로부터 흡광도를 계산하여 표준물첨가시료의 농도에 대한 흡광도의 추세선을 그리고, 이를 외삽하여 얻은 x 절편의 값에 ?1을 곱하여 총인의 농도를 알 수 있다.
도 18a는 상기한 방법으로 얻은 1ppm의 인을 포함하는 총인 표준시료의 표준물첨가에 의한 검출신호이며, 도 18b는 도 18a의 검출신호를 표준물첨가시료의 농도에 대한 흡광도로 나타낸 그래프이다.

표 5는 총인 표준시료에 대하여 상기한 방법으로 실험을 세 번 반복하여 얻은 검출된 총인의 농도와 상대오차(1.62% 이하), 그리고 상대표준편차(0.800% 이하)를 나타낸 것이다. 표 5의 결과로부터 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 정확하고 높은 재현성으로 분석할 수 있음을 알 수 있다.
[실험예 10]
실험예 10에서는 실제시료 내 총인의 농도를 실험예 9와 같은 방법으로 분석하고, 그 결과를 상용화된 자외선-가시광선 흡광광도계로 분석한 결과와 비교한다. 상기 실제시료는 축산농가에서 채수하였으며, 총질소 또는 총인 분석용 연속 미세 유체 장치를 이용한 실제시료의 총인 분석방법은 실험예 9에 기술한 바와 같다. 비교분석법은 국내 수질오염공정시험기준과 미국환경보호국에서 수중 인 정량분석의 표준방법으로 채택한 신뢰성이 높은 분석법으로 채수한 시료를 산화제와 혼합하여 고압증기멸균기를 이용하여 0.13MPa, 120℃에서 30분간 분해한 다음, 분해과정이 끝난 시료를 표준물첨가법을 이용하여 자외선-가시광선 흡광광도계로 분석하여 총인을 정량한다.

표 6은 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치와 자외선-가시광선 흡광광도계로 채수한 각 실제시료 내 총인의 농도를 세 번 반복하여 분석한 결과를 나타낸다. 상기한 두 가지 방법으로 검출한 총인의 농도가 거의 유사하고, 2.5% 이하의 낮은 상대표준편차를 보이는 것으로부터 본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 정확하고 재현성이 높은 총인 정량이 가능함을 알 수 있다.
본 발명의 총질소 또는 총인 분석용 연속 미세 유체 장치는 수중에 존재하는 총인을 분석하기 위한 산화분해부재(82), 총질소 또는 총인 분석용 연속 미세 유체 장치를 소형화하고, 분석에 필요한 각각의 과정이 연속흐름상에서 효율적으로 이루어져 빠른 시간에 정확하고 재현성이 높은 총인 분석이 가능하다. 또한, 본 실시예에 따른 총질소 또는 총인 분석용 연속 미세 유체 장치는 광 경로 길이의 조절이 가능함에 따라 넓은 농도범위의 총인을 검출할 수 있어 대부분의 수환경에 적용이 가능하다.
상기에서는 본 발명의 바람직한 실시예에 대하여 설명하였지만, 본 발명은 이에 한정되는 것이 아니고 특허청구범위와 발명의 상세한 설명 및 첨부한 도면의 범위 안에서 여러 가지로 변형하여 실시하는 것이 가능하고 이 또한 본 발명의 범위에 속하는 것은 당연하다.
Claims (16)
- 분석 대상인 시료가 주입되는 시료 주입구와 시약이 주입되는 시약 주입구와 시료와 시약을 혼합하기 위하여 상기 시료 주입구 및 상기 시약 주입구와 연통된 반응채널과 상기 반응채널과 연통된 광 입사구와 상기 광 입사구에서 이격 배치된 검출구가 형성된 제1기판과,상기 광 입사구와 마주하며 광원의 빛이 투과하는 광 입사부와 상기 검출구와 마주하며 광원의 빛이 투과하는 검출부를 구비하며 상기 제1기판에 결합되는 제2기판, 및상기 광 입사구에 일측 단부가 연결되며, 상기 검출구에 타측 단부가 연결되는 흐름셀을 포함하는 미세유체칩;상기 광 입사부와 마주하도록 배치된 광원;상기 검출부와 마주하도록 배치된 검출기;상기 광원과 상기 검출기 사이에 배치된 차단벽; 및상기 미세유체칩과 연결되며 시료를 산화분해하여 상기 미세유체칩으로 공급하는 산화분해부재;를 포함하는 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 산화분해부재는 열 또는 빛을 발생시키는 반응막대와 상기 반응막대에 감겨지며, 시료가 이동하는 통로를 제공하는 튜브를 포함하는 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 반응막대는 자외선을 방출하는 자외선 램프로 이루어진 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제2항에 있어서,상기 산화분해부재에는 시료를 공급하는 시료 주입관과 산화제를 공급하는 산화제 주입관이 연결 설치된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제4항에 있어서,상기 산화분해부재에는 표준물첨가시료를 상기 산화분해부재로 공급하는 표준물첨가시료 주입관이 연결 설치된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 산화분해부재에는 카드뮴-구리 컬럼이 연결 설치되고, 상기 카드뮴-구리 컬럼은 튜브와 상기 튜브 내에 채워진 카드륨 입자을 포함하는 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제6항에 있어서,상기 카드륨 입자는 산으로 처리된 후, 황산구리 용액으로 활성화된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제6항에 있어서,상기 카드뮴-구리 컬럼에는 카드뮴의 침전을 방지하는 완충용액을 공급하는 완충용액 주입관이 연결 설치된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제8항에 있어서,상기 완충용액은 이미다졸을 포함하는 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 제1기판에는 상기 검출구와 연결된 용액 배출채널 및 상기 용액 배출 채널에 연결된 용액 배출구가 형성된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 흐름셀의 양측 단부는 상기 제2기판을 향하도록 설치된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 제1기판과 제2기판은 빛을 흡수할 수 있는 색을 갖는 물질로 이루어지거나, 빛을 흡수할 수 있는 색으로 착색된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제12항에 있어서,상기 입사부와 검출부는 빛이 투과할 수 있도록 투명한 소재로 이루어진 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 광 입사구 및 상기 검출구에는 상기 흐름셀이 삽입되는 체결부가 형성된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제1항에 있어서,상기 검출부와 상기 검출기 사이에는 상기 광원에서 출사되는 빛의 반값 반폭보다 더 작은 반값 반폭을 갖도록 빛을 투과시키는 간섭 필터가 설치된 총질소 또는 총인 분석용 연속 미세 유체 장치.
- 제15항에 있어서,상기 광원과 상기 검출기 및 상기 차단벽은 하우징 내에 설치되고, 상기 미세유체칩은 상기 하우징 상에 배치되며, 상기 하우징의 상면에는 상기 광원의 상부에 위치하는 입사채널 및 상기 검출기의 상부에 위치하는 검출채널이 형성되고, 상기 간섭 필터는 상기 검출채널과 상기 검출기 사이에 간섭 필터가 설치된 총질소 또는 총인 분석용 연속 미세 유체 장치.
