Abstract

L'invention porte sur un procédé de séquençage d'au moins une molécule d'acide nucléique matrice, le procédé comprenant la digestion de façon différentielle d'un certain nombre de copies de ladite ou desdites molécules matrices pour de cette manière produire des molécules matrices digérées, au moins une partie des molécules matrices digérées ayant des nombres différents de nucléotides enlevés d'au moins une extrémité de la molécule matrice ; le séquençage d'au moins une partie des molécules matrices digérées pour produire des lectures représentant la teneur de séquences d'au moins une partie d'extrémité des molécules matrices digérées ; et la production d'une séquence consensus pour ladite ou lesdites molécules matrices à l'aide de chevauchements entre les lectures.
