Description
본 발명은 항염증 활성을 갖는 펩티드 및 이를 포함하는 조성물에 관한 것이다.
염증은 외부의 물리적 자극, 각종 알레르기 유발 물질 접촉을 예로 들 수 있는 화학적 자극 또는 박테리아, 곰팡이 또는 바이러스와 같은 미생물의 침입 등에 의해 생체 조직이 손상되는 것을 막기 위한 생체의 방어 반응 중 하나이다.
염증 신호는 사이클로옥시게나제(COX) 경로 또는 리폭시게나제(LOX) 경로를 통하여 만들어지며, 프로스타글란딘, 트롬복산 등을 생성한다. 염증 신호가 전달되면 생체 내에서는 여러 가지 변화가 일어나는데, 그 중 하나로 염증이 필요한 부위의 혈관을 확장시켜 혈액 공급이 왕성하게 일어나도록 하여, 호중구 등 염증 반응에 필요한 혈액 세포가 집중 공급되는 현상을 들 수 있다. 그러나 이러한 생체의 방어 반응이 비정상적으로 과도하게 일어나면 여러 염증성 질환들이 발병하게 되므로, 이를 방지하고자 염증 신호 경로의 효소(예를 들어 COX-1, COX-2, 5-LOX, 12-LOX 등)를 억제하여 염증 신호 경로를 차단함으로써, 과도한 염증 반응을 억제할 수 있는 약물들이 개발되고 있다.
염증은 반응기간에 따라서 급성염증(acute inflammation, 즉각적 반응, 비특이적 반응, 수일~수주일), 만성염증(chronic inflammation, 지연된 반응, 특이성 반응, 수주일 이상), 아급성 염증(subacute inflammation, 급성과 만성염증의 중간단계, 다핵구와 단핵구의 혼합형 산출물의 특징)으로 나뉜다.
또한, 염증을 유발하는 인자에는 펩티드성 인자 이외에 프로스타그란딘, 루코트리엔, 혈소판활성인자와 같은 지질성 인자, 염증인자 합성효소, NO(nitric oxide)와 같은 자유라디칼, 여러 종류의 세포접착분자, 면역계, 응고인자 등이 관여한다.
현재까지 알려진 염증의 기전은 외부의 생물학적인 원인(세균, 바이러스, 기생충), 물리학적인 원인(기계적인 자극, 열, 방사선, 전기), 화학적 원인 등으로 인한 세포손상으로 히스타민 및 키닌 등이 방출되어 혈관확장, 모세혈관 투과성 증가 및 염증부위로 대식세포의 집결이 일어나고, 이에 따라서 감염부위의 혈류량 증가, 부종, 면역세포 및 항체 이동, 통증, 발열 등의 현상이 일어난다.
현재 사용되고 있는 염증의 치료제에는 이부프로펜과 같은 합성의약품, 항히스타민제, 스테로이드, 코티손, 면역억제제, 면역 항진제 등이 사용되고 있으나 치료효과가 일시적이거나 단순 증상완화, 과민반응, 면역체계 악화 등의 부작용이 많이 있고 염증의 근본적인 치료가 어렵다. ( KR2012-0130996A)

텔로미어(telomere)는 염색체의 말단에 반복적으로 존재하는 유전 물질로서, 해당 염색체의 손상이나 다른 염색체와의 결합을 방지한다고 알려져 있다. 세포가 분열할 때마다 텔로미어의 길이는 조금씩 짧아지는데, 일정한 횟수 이상의 세포 분열이 있게 되면 텔로미어는 매우 짧아지고, 그 세포는 분열을 멈추고 죽게 된다. 반면 텔로미어를 길게 하면 세포의 수명이 연장된다고 알려져 있으며, 그 예로 암세포에서는 텔로머라제(telomerase)라는 효소가 분비되어 텔로미어가 짧아지는 것을 막기 때문에, 암세포가 죽지 않고 계속 증식할 수 있다고 알려져 있다. 본 발명자들은 텔로머라제로부터 유래되는 수많은 펩티드들이 항염 활성을 가질 수 있음을 발견하고 본 발명을 완성하게 되었다.
본 발명의 일측면은 항염증 활성을 갖는 펩티드로서, 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 제공한다.
서열번호 1 내지 서열번호 14에 기재된 펩티드는 아래 표 1과 같다. 서열번호 15는 인간의 텔로머라제 전장 단백질의 서열이다. 서열번호 1은 텔로머라제로부터 유래된 펩티드로서 16개 아미노산으로 구성된 펩티드이다.
아래 표 1의 "이름"은 펩티드를 구별하기 위해 명명한 것이다. 본 발명의 다른 일측면에서, 서열 번호 2 내지 서열 번호 14에 기재된 펩티드 중 하나 이상의 펩티드는 텔로머라제에 포함된 펩티드 중 해당 위치의 펩티드를 선별해 합성한 "합성 펩티드"를 포함한다. 본 명세서에서 "pep"이라 함은 서열번호 2 내지 서열번호 14 중 어느 한 서열을 갖는 펩티드, 또는 그 서열과 80% 이상의 서열 상동성을 갖는 펩티드, 또는 상기 서열의 단편을 총칭하는 용어이다.
본 발명의 일측면은 항염증 활성을 갖는 펩티드로서, 서열번호 2 내지 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 코딩하는 폴리뉴클레오티드를 제공한다. 상기 폴리뉴클레오티드를 이용하여 펩티드를 대량 생산할 수 있다. 예컨대 펩티드를 코딩하는 폴리뉴클레오티드를 포함하는 벡터를 숙주세포에 넣어 배양함으로써, 펩티드를 대량 생산할 수 있다.
본 발명의 일측면에서, 아미노산 변화는 펩티드의 물리화학적 특성이 변경되도록 하는 성질에 속한다. 예를 들어, 펩티드의 열안정성을 향상시키고, 기질 특이성을 변경시키고, 최적의 pH를 변화시키는 등의 아미노산 변화가 수행될 수 있다.
본 발명의 일측면에서, 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드는, 30개 이하의 아미노산으로 구성될 수 있다.
본 발명의 일측면에서, 서열번호 2 내지 서열번호 14의 서열을 갖는 펩티드, 상기 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드는 텔로머라제, 구체적으로 인간(Homo sapiens) 텔로머라제에서 유래한 펩티드를 포함한다.

펩티드의 생물학적 특성에 있어서의 실재적인 변형은 (a) 치환 영역 내의 폴리펩티드 골격의 구조, 예를 들면 시트 또는 나선 입체 구조를 유지하는데 있어서의 이들의 효과, (b) 표적 부위에서의 상기 분자의 전하 또는 소수성을 유지하는데 있어서의 이들의 효과 또는 (c) 측쇄의 벌크를 유지하는데 있어서의 이들의 효과가 상당히 상이한 치환부를 선택함으로써 수행된다. 천연 잔기는 통상의 측쇄 특성에 기준하여 다음 그룹으로 구분된다:

펩티드의 다른 유형의 아미노산 변이체는 항체의 글리코실화 패턴이 변화된 것이다. 변화란 의미는 펩티드에서 발견된 하나 이상의 탄수화물 잔기의 결실 및(또는) 펩티드 내에 존재하지 않는 하나 이상의 글리코실화 부위의 부가를 나타낸다.
펩티드의 글리코실화는 전형적으로 N-연결되거나 O-연결된 것이다. N-연결된이란 탄수화물 잔기가 아스파라긴 잔기의 측쇄에 부착된 것을 말한다. 트리펩티드 서열 아스파라긴-X-세린 및 아스파라긴-X-트레오닌 (여기서, X는 프롤린을 제외한 임의의 아미노산임)은 탄수화물 잔기를 아스파라긴 측쇄에 효소적 부착시키기 위한 인식 서열이다. 따라서, 이들 트리펩티드 서열 중의 하나가 폴리펩티드에 존재함으로써, 잠재적인 글리코실화 부위가 생성된다. 본 발명의 일측면에 따른 서열번호 2 내지 서열번호 14의 서열을 갖는 펩티드, 상기 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드를 세포에 처리하면, 세포핵 내 HMGB1 단백질 양이 증가한다. 이는 상기 서열을 갖는 펩티드, 상기 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드가 우수한 염증 예방 또는 억제 효과를 가짐을 의미한다.
또한 본 발명의 일측면에 따른 서열번호 2 내지 서열번호 14의 서열을 갖는 펩티드, 상기 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드는 세포 내 독성이 낮아 생체 내 안정성이 높다는 장점을 가진다.

본 발명의 일측면에서는 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 0.01g/L 내지 1kg/L, 구체적으로 0.1g/L 내지 100g/L, 더 구체적으로 1 g/L 내지 10g/L 의 함량으로 포함할 수 있다. 상기 범위로 포함하는 경우 본 발명의 의도한 효과를 나타내기에 적절할 뿐만 아니라, 조성물의 안정성 및 안전성을 모두 만족할 수 있으며, 비용 대비 효과의 측면에서도 상기 범위로 포함하는 것이 적절할 수 있다.
본 발명의 일측면에 따른 조성물은 인간, 개, 닭, 돼지, 소, 양, 기니아피그 또는 원숭이를 포함하는 모든 동물에 적용될 수 있다.
본 발명의 일측면에서 조성물은 서열번호 2 내지 서열번호 14로 이루어진 군으로부터 선택되는 서열을 갖는 펩티드, 상기 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드를 유효 성분으로 포함하는 염증성 질환의 치료 또는 예방용 약학 조성물을 제공한다. 본 발명의 일측면에 따른 약학 조성물은 경구, 직장, 경피, 정맥 내, 근육 내, 복강 내, 골수 내, 경막 내 또는 피하 등으로 투여될 수 있다.
경구 투여를 위한 제형은 정제, 환제, 연질 또는 경질 캅셀제, 과립제, 산제, 액제 또는 유탁제일 수 있으나, 이에 제한되는 것은 아니다. 비경구 투여를 위한 제형은 주사제, 점적제, 로션, 연고, 겔, 크림, 현탁제, 유제, 좌제, 패취 또는 분무제일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일측면에 따른 약학 조성물은 필요에 따라 희석제, 부형제, 활택제, 결합제, 붕해제, 완충제, 분산제, 계면 활성제, 착색제, 향료 또는 감미제 등의 첨가제를 포함할 수 있다. 본 발명의 일측면에 따른 약학 조성물은 당업계의 통상적인 방법에 의해 제조될 수 있다.
본 발명의 일측면에 따른 약학 조성물의 유효 성분은 투여 받을 대상의 연령, 성별, 체중, 병리 상태 및 그 심각도, 투여 경로 또는 처방자의 판단에 따라 달라질 것이다. 이러한 인자에 기초한 적용량 결정은 당업자의 수준 내에 있으며, 이의 1일 투여 용량은 예를 들어 1 ng/kg/일 내지 10 mg/kg/일, 구체적으로는 10 ng/kg/일 내지 1 mg/kg/일, 더 구체적으로는 0.1 ㎍/kg/일 내지 100 ㎍/kg/일, 보다 더 구체적으로는 0.2 ㎍/kg/일 내지 20 ㎍/kg/일이 될 수 있으나, 이에 제한되는 것은 아니다. 본 발명의 일측면에 따른 약학 조성물은 1일 1회 내지 3회 투여될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일측면에서는 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 유효 성분으로 포함하는 피부 염증의 개선 또는 예방용 피부 외용제 조성물을 제공한다.
본 발명의 다른 일측면에서는 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 유효 성분으로 포함하는 피부 염증의 개선 또는 예방용 화장료 조성물을 제공한다.
본 발명의 일측면에 따른 피부 외용제 조성물 또는 화장료 조성물은 국소 적용에 적합한 모든 제형으로 제공될 수 있다. 예를 들면, 용액, 수상에 유상을 분산시켜 얻은 에멀젼, 유상에 수상을 분산시켜 얻은 에멀젼, 현탁액, 고체, 겔, 분말, 페이스트, 포말(foam) 또는 에어로졸의 제형으로 제공될 수 있다. 이러한 제형은 당해 분야의 통상적인 방법에 따라 제조될 수 있다.
본 발명의 일측면에 따른 화장료 조성물은 주 효과를 손상시키지 않는 범위 내에서, 바람직하게는 주 효과에 상승 효과를 줄 수 있는 다른 성분들을 포함할 수 있다. 또한 본 발명의 일측면에 따른 화장료 조성물은 보습제, 에몰리언트제, 계면 활성제, 자외선 흡수제, 방부제, 살균제, 산화 방지제, pH 조정제, 유기 또는 무기 안료, 향료, 냉감제 또는 제한(制汗)제를 더 포함할 수 있다. 상기 성분의 배합량은 본 발명의 목적 및 효과를 손상시키지 않는 범위 내에서 당업자가 용이하게 선정 가능하며, 그 배합량은 화장료 조성물 전체 중량을 기준으로 0.01 내지 5 중량%, 구체적으로 0.01 내지 3 중량%일 수 있다.
본 발명의 일측면에서 조성물은 서열번호 2 내지 서열번호 14로 이루어진 군으로부터 선택되는 서열을 갖는 펩티드, 상기 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드를 유효 성분으로 포함하는 염증 예방 또는 억제용 식품 조성물을 제공한다.
본 발명의 일측면에 따른 식품 조성물의 제형은 특별히 한정되지 않으나, 예를 들어, 정제, 과립제, 분말제, 액제, 고형 제제 등으로 제형화될 수 있다. 각 제형은 유효 성분 이외에 해당 분야에서 통상적으로 사용되는 성분들을 제형 또는 사용 목적에 따라 당업자가 어려움 없이 적의 선정하여 배합할 수 있으며, 다른 원료와 동시에 적용할 경우 상승 효과가 일어날 수 있다.
상기 유효 성분의 투여량 결정은 당업자의 수준 내에 있으며, 이의 1일 투여 용량은 예를 들어 구체적으로는 10 ng/kg/일 내지 1 mg/kg/일, 더 구체적으로는 0.1 ㎍/kg/일 내지 100 ㎍/kg/일, 보다 더 구체적으로는 0.2 ㎍/kg/일 내지 20 ㎍/kg/일이 될 수 있으나, 이에 제한되지 않으며, 투여하고자 하는 대상의 연령, 건강 상태, 합병증 등 다양한 요인에 따라 달라질 수 있다.
본 발명의 일측면은 일측면에서는 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드의 염증성 질환 예방 또는 치료 용도를 제공한다.
본 발명의 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드 또는 이를 포함하는 조성물을 대상에게 적용하는 것을 포함하는 대상의 염증성 질환 예방 또는 치료 방법을 제공한다.
본 발명의 일측면에서 조성물은 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드 또는 이를 포함하는 조성물 및 펩티드 또는 이를 포함하는 조성물의 투여량, 투여 경로, 투여 횟수 및 적응증 중 하나 이상을 개시한 지시서를 포함하는 염증성 질환 예방 또는 치료용 키트를 제공한다.
본 명세서에서 사용된 용어들은 특정 구체예들을 설명하기 위한 목적으로만 의도된 것이지 본 발명을 한정하고자 하는 의도가 아니다. 명사 앞에 개수가 생략된 용어는 수량을 제한하고자 하는 것이 아니라 언급된 명사 물품이 하나 이상 존재하는 것을 나타내는 것이다. 용어 "포함하는", "갖는", 및 "함유하는"은 열린 용어로 해석된다(즉, "포함하지만 이에 한정되지는 않는"의 의미).
수치의 범위를 언급하는 것은 단지 그 범위 내에 속하는 각각의 별개의 수치들을 개별적으로 언급하는 것을 대신하는 쉬운 방법이기 때문이며, 그것이 아님이 명시되어 있지 않는 한, 각 별개의 수치는 마치 개별적으로 명세서에 언급되어 있는 것처럼 본 명세서에 통합된다. 모든 범위의 끝 값들은 그 범위 내에 포함되며 독립적으로 조합 가능하다.
본 명세서에 언급된 모든 방법들은 달리 명시되어 있거나 문맥에 의해 명백히 모순되지 않는 한 적절한 순서로 수행될 수 있다. 어느 한 실시예 및 모든 실시예 또는 예시적 언어 (예컨대, "~과 같은")를 사용하는 것은, 청구범위에 포함되어 있지 않는 한, 단지 본 발명을 더 잘 기술하기 위함이지 본 발명의 범위를 제한하고자 함이 아니다. 명세서의 어떤 언어도 어떤 비청구된 구성요소를 본 발명의 실시에 필수적인 것으로 해석되어져서는 아니된다. 다른 정의가 없는 한, 본 명세서에 사용되는 기술적 및 과학적 용어들은 본 발명이 속하는 기술 분야에서 통상의 지식을 갖는 사람에 의해 통상 이해되는 것과 같은 의미를 갖는다.
본 발명의 바람직한 구체예들은 본 발명을 수행하기 위해 발명자에게 알려진 가장 최적의 모드를 포함한다. 바람직한 구체예들의 변이들이 앞선 기재를 읽으면 당업자에게 명백하게 될 수 있다. 본 발명자들은 당업자들이 그러한 변이를 적절히 이용하길 기대하고, 발명자들은 본 명세서에 기재된 것과 다른 방식으로 본 발명이 실시되기를 기대한다. 따라서, 본 발명은, 특허법에 의해 허용되는 것과 같이, 첨부된 특허청구범위에서 언급된 발명의 요지의 균등물 및 모든 변형들을 포함한다. 더욱이, 모든 가능한 변이들 내에서 상기 언급된 구성요소들의 어떤 조합이라도 여기서 반대로 명시하거나 문맥상 명백히 모순되지 않는 한 본 발명에 포함된다. 본 발명은 예시적인 구체예들을 참조하여 구체적으로 나타내어지고 기술되었지만, 당업자들은 하기 청구범위에 의해 정의되는 발명의 정신 및 범위를 벗어나지 않고서도 형태 및 디테일에서 다양한 변화가 행해질 수 있음을 잘 이해할 것이다 .
종양 괴사 인자 (TNF), 특히 TNF-α 는 염증성 세포로부터 방출되어 다채로운 세포 상해 반응, 면역 반응 및 염증 반응을 일으키는 것으로 알려져 있다. TNF-α 는 많은 염증 질환 및 자기 면역 질환의 발증이나 천연화 (遷延化) 에 관여하고, 또한 혈액 중에 방출되어 전신에 작용하면, 중증의 패혈증 및 패혈증성 쇼크를 일으키는 것이 알려져 있다. 이와 같이 TNF-α 는 생체의 면역계에 광범위하게 관련된 인자이기 때문에, TNF-α 를 억제하는 약제의 개발이 활발히 실시되고 있다. TNF-α 는 불활성형으로 생합성되고 프로테아제에 의해 절단되어 활성형이 되지만, 이 활성화에 관여하는 효소는 종양 괴사 인자 변환 효소 (TACE) 로 불리고 있다. 따라서 이 TACE 를 저해하는 물질은 TNF-α 에서 기인되는 질환, 병태, 이상 상태, 상태가 좋지 않음, 좋지 않은 자각 증상 등을 치료, 개선, 예방할 수 있다(KR2011-0060940A).
이하, 실시예 및 실험예를 들어 본 발명의 구성 및 효과를 보다 구체적으로 설명한다. 그러나 아래 실시예 및 실험예는 본 발명에 대한 이해를 돕기 위해 예시의 목적으로만 제공된 것일 뿐 본 발명의 범주 및 범위가 그에 의해 제한되는 것은 아니다.
실시예 1: 펩티드의 합성 및 항염 활성 측정
실험예 1: 펩티드의 합성
인간 텔로머라제로부터 선별된 서열번호 2 내지 서열번호 14의 펩타이드를 합성하였다.
펩티드들은 종래에 알려진 고상 펩티드 합성법에 따라 제조하였다. 구체적으로, 펩티드들은 ASP48S(Peptron, Inc., 대한민국 대전)를 이용하여 Fmoc 고상 합성법(solid phase peptide synthesis, SPPS)을 통해 C-말단부터 아미노산 하나씩 커플링함으로써 합성하였다. 다음과 같이, 펩티드들의 C-말단의 첫 번째 아미노산이 수지에 부착된 것을 사용하였다. 예컨대 다음과 같다:
NH2-Lys(Boc)-2-chloro-Trityl Resin
NH2-Ala-2-chloro-Trityl Resin
NH2-Arg(Pbf)-2-chloro-Trityl Resin
펩타이드 합성에 사용한 모든 아미노산 원료는 N-term이 Fmoc으로 보호(protection)되고, 잔기는 모두 산에서 제거되는 Trt, Boc, t-Bu (t-butylester), Pbf (2,2,4,6,7-pentamethyl dihydro-benzofuran-5-sulfonyl) 등으로 보호된 것을 사용하였다. 예컨대 다음과 같다:
Fmoc-Ala-OH, Fmoc-Arg(Pbf)-OH, Fmoc-Glu(OtBu)-OH, Fmoc-Pro-OH, Fmoc-Leu-OH, Fmoc-Ile-OH, Fmoc-Phe-OH, Fmoc-Ser(tBu)-OH, Fmoc-Thr(tBu)-OH, Fmoc-Lys(Boc)-OH, Fmoc-Gln(Trt)-OH, Fmoc-Trp(Boc)-OH, Fmoc-Met-OH, Fmoc-Asn(Trt)-OH, Fmoc-Tyr(tBu)-OH, Fmoc-Ahx-OH, Trt-Mercaptoacetic acid.
커플링 시약(Coupling reagent)으로는 HBTU[2-(1H-Benzotriazole-1-yl)-1,1,3,3-tetamethylaminium hexafluorophosphate] / HOBt [N-Hydroxxybenzotriazole] /NMM [4-Methylmorpholine] 를 사용하였다. Fmoc 제거는 20%의 DMF 중 피페리딘(piperidine in DMF)을 이용하였다. 합성된 펩타이드를 Resin에서 분리 및 잔기의 보호기 제거에는 절단 칵테일(Cleavage Cocktail) [TFA (trifluoroacetic acid) /TIS (triisopropylsilane) / EDT (ethanedithiol) / H2O=92.5/2.5/2.5/2.5] 를 사용하였다.
아미노산 보호기가 결합된 출발 아미노산이 고상 지지체에 결합되어 있는 상태를 이용하여 여기에 해당 아미노산들을 각각 반응시키고 용매로 세척한 후 탈보호하는 과정을 반복함으로써 각 펩티드를 합성하였다. 합성된 펩티드를 수지로부터 끊어낸 후 HPLC로 정제하고, 합성 여부를 MS로 확인하고 동결 건조하였다.
서열번호 1의 Pep 1(EARPALLTSRLRFIPK)을 예로 들어 구체적인 합성 과정을 설명하면 다음과 같다.
1) 커플링
NH2-Lys(Boc)-2-chloro-Trityl Resin 에 보호된 아미노산(8당량)와 커플링 시약 HBTU(8당량)/HOBt(8당량)/NMM(16당량) 을 DMF에 녹여서 첨가한 후, 상온에서 2시간 동안 반응하고 DMF, MeOH, DMF순으로 세척하였다.
2) Fmoc 탈보호
20%의 DMF 중의 피페리딘(piperidine in DMF) 을 가하고 상온에서 5분 간 2회 반응하고 DMF, MeOH, DMF순으로 세척하였다.
3) 1과 2의 반응을 반복적으로 하여 펩타이드 기본 골격 NH2-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(Boc)-2-chloro-Trityl Resin을 만들었다.
4) 절단(Cleavagge): 합성이 완료된 펩타이드 Resin에 절단 칵테일(Cleavage Cocktail) 을 가하여 펩타이드를 레진에서 분리하였다.
5) 얻어진 mixture에 Cooling diethyl ether를 가한 후, 원심 분리하여 얻어진 펩타이드를 침전시킨다.
6) Prep-HPLC로 정제 후, LC/MS로 분자량을 확인하고 동결하여 파우더로 제조하였다.
서열번호 2 내지 서열번호 14의 펩티드를 합성하는 방법은 상기 서열번호 1의 펩티드를 합성하는 방법과 같은 방법을 사용하되, 붙여주는 아미노산을 달리하였다.
실험예 2 :TNF α 저해 활성 검증
실험예 2-1 : 세포주 배양
건강한 사람에게서 혈액을 채혈 (50 ml)한 후 Biocoll Separating Solution (Biochrom AG, Berlin, Germany)를 사용하여 PBMC (peripheral blood mononuclear cells) 층을 회수하였다. 회수된 PBMC는 사람의 혈청 (20%)이 첨가된 RPMI 1640 배지에 부유하여 30분 가량 사람의 혈청을 코팅한 100-mm 폴리스티렌(polystyrene) 세포 배양 플레이트에 옮겨 2시간 가량 37℃, 5% 배양기에서 2시간 배양하였다. 그 후 바닥에 붙은 모노사이트(monocytes)들을 차가운 PBS로 떼어낸 후 24 웰 플레이트(well plate)에 웰 당 2 x 105 셀이 되도록 RPMI 1640 배지 (supplemented with penicillin-streptomycin; 100 mg/ml, human serum; 20%)에 부유시켜 실험하기 하루 전에 배양시켜 놓았다.
실험예 2-2: 사이토카인 분석(Cytokine assay)
서열번호 2 내지 서열번호 14의 각 펩티드들이 TNF-α에 대해 어떠한 영향을 미치는지 알아보기 위해 ELISA 실험을 수행하였다.
PBMC에서 유래한 모노사이트(monocytes)를 100ng/mL PMA와 함께 처리하여 24-웰 플레이트에 웰 당 2 x 105 셀이 되도록 실험 하루 전에 배양시켰다. 그 후 LPS (lipopolysaccharide; 10 ng/ml, Sigma)를 2 시간 처리하고 PBS로 두 번 씻어내었다. 이 후 OPTI-MEM 배지(Invitrogen/Life Technologies, Carlsbad, CA, USA)에 넣어 1 시간 동안 배양하였고, 그 후 OPTI-MEM에 서열번호 2 내지 서열번호 14의 각 펩티드를 0.1, 1μM로 각 well에 처리한 후 1시간 동안 배양하였다. 배양이 끝난 후 배지를 모아서 -80℃에 보관하고 이를 ELISA (R&D, Minneapolis, MN, USA) 키트 매뉴얼에 따라 TNF-α를 정량하였다. 구체적인 정량방법은 다음과 같다.
TNF 측정은 샌드위치(sandwich) ELISA법을 이용한다. 미리 코팅되어 있는 24 웰 플레이트에 TNFα primary antibody를 100ul 씩 넣고 4℃에서 overnight 한다. 다음날 0.5% tween 20 워시 용액으로 5분씩 3회 세척한 후 측정할 샘플과 standard 용액을 100ul씩 넣고 2시간 동안 상온에서 반응시킨다. 상기와 같이 wash한 후 HRP-conjugated secondary antibody를 각 100 ul씩 넣고 2시간동안 상온에서 반응시킨다. 이후 다시 wash하고 avidin/biotin을 첨가하여 발색시켜 흡광도를 측정한다. Standard의 흡광도로부터 표준곡선을 구하고 이를 이용하여 각 샘플의 흡광도로부터 TNFα 의 양을 정량한다.
세포 배양액의 TNF-α 레벨을 ELISA로 측정한 결과, LPS만을 처리한 비교군에 비해 서열번호 2 내지 서열번호 14의 펩티드가 사용된 각각의 펩티드 농도 또는 어느 한 농도에서 TNF- α의 레벨을 감소시킴을 확인하였다. (도 1 내지 도 13)
참고로 기출원된 서열번호 1의 펩티드 역시 세포 배양액의 TNF-α 레벨을 ELISA로 측정한 결과, LPS만을 처리한 비교군에 비해 사용된 두 가지의 농도 모두에서 TNF- α의 레벨을 감소시킴을 확인하였다. (도 14)
Claims (19)
- 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드.
- 제1항에 있어서, 상기 단편은 3개 이상의 아미노산으로 구성된 단편인 펩티드.
- 제1항에 있어서, 상기 펩티드는 30개 이하의 아미노산으로 구성된 펩티드.
- 제1항에 있어서. 상기 펩티드는 서열번호 2 내지 서열번호 14 중 어느 하나의 아미노산 서열로 이루어진 펩티드.
- 제1항에 있어서. 상기 펩티드는 인간의 텔로머라제로부터 유래된 것인 펩티드.
- 항염증 활성을 갖는 펩티드를 코딩하는 폴리뉴클레오티드로서,서열번호 2 내지 서열번호 14 중 어느 하나의 아미노산 서열을 포함하는 펩티드 또는 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 코딩하는 폴리뉴클레오티드.
- 제6항에 있어서, 상기 펩티드는 30개 이하의 아미노산을 코딩하는 폴리뉴클레오티드.
- 제6항에 있어서, 상기 펩티드는 인간의 텔로머라제로부터 유래된 것인 폴리뉴클레오티드.
- 서열번호 2 내지 서열번호 14 중 어느 하나의 아미노산 서열을 포함하는 펩티드,상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드를 유효성분으로 포함하는 항염증 조성물.
- 제9항에 있어서, 상기 펩티드는 30개 이하의 아미노산으로 구성된 것인 항염증 조성물.
- 제9항에 있어서, 상기 펩티드는 서열번호 2 내지 서열번호 14 중 어느 하나의 아미노산 서열로 이루어진 것인 항염증 조성물.
- 제9항에 있어서, 상기 펩티드는 인간의 텔로머라제로부터 유래된 것인 항염증 조성물.
- 제9항에 있어서, 상기 조성물은 염증성 질환의 치료 또는 예방용인 항염증 조성물.

- 제9항에 있어서, 상기 조성물은 염증의 개선 또는 예방을 위한 식품조성물인 항염증 조성물.
- 제9항 내지 제 17항 중 어느 한 항에 따른 항염증 조성물을 투여하는 것을 특징으로 하는 염증성 질환을 예방 또는 치료하는 방법.
- 서열번호 2 내지 서열번호 14 중 어느 하나 이상의 아미노산 서열을 포함하는 펩티드, 상기 아미노산 서열과 80%이상의 서열 상동성을 갖는 펩티드 또는 그 단편인 펩티드 또는 이를 포함하는 조성물; 및 펩티드 또는 이를 포함하는 조성물의 투여량, 투여 경로, 투여 횟수 및 적응증 중 하나 이상을 개시한 지시서를 포함하는 염증성 질환 예방 또는 치료용 키트.
