Abstract

본 발명은, 일반적인 바이오디젤 생산방법인 화학적 반응에 따른 고온 고압공정이 갖는 과다 에너지 사용 문제 및 다량의 폐수 발생 문제를 해결하고자, 화학 촉매 대신에 지방분해효소인 리파제(lipase)를 이용하여 생물학적 바이오디젤을 보다 효율적으로 생산할 수 있도록 고안된 연속 반응기 및 바이오디젤 생산 방법을 제공한다.

Description

고정화 효소를 이용한 연속식 바이오디젤 생산 반응기 및 바이오디젤 생산 방법
본 발명은 디젤 대체 연료인 바이오디젤 또는 지방산 알킬 에스테르를 생산하기 위한 연속식 바이오디젤 생산 반응기 및 이를 이용한 바이오디젤 생산 방법에 관한 것이다. 보다 구체적으로, 일반적인 바이오디젤 생산방법인 화학적 반응에 따른 고온 고압 공정이 갖는 과다 에너지 사용 문제 및 다량의 폐수 발생 문제를 해결하고자, 화학 촉매 대신에 지방분해효소인 리파제(lipase)를 이용하여 생물학적 바이오디젤을 보다 효율적으로 생산할 수 있도록 고안된 연속 반응기 및 바이오디젤 생산 방법에 관한 것이다.
트리아실글리세롤 리파제(triacylglycerol lipase, EC 3.1.1.3)는 트리아실글리세롤을 분해하는 카르복시 에스터 하이드롤레이즈(carboxylic ester hydrolase)로서 세제, 유지화합물, 식품 및 정밀화학제품 산업 등 다양한 산업에서 활용되는 생물촉매(biocatalyst)이다. 최근 원유고갈이나 지구온난화 문제로 원유를 대체하는 바이오 에너지 연구가 활발하며 특히 수송용 연료로서 경유를 대체할 수 있는 동, 식물성 유지 유래의 바이오디젤이 전 세계적으로 활용되고 있다.
이러한 바이오디젤의 생산 방법은 산이나 알칼리 촉매를 사용하는 화학적인 방법이 현재 주로 이용되고 있지만 한편으로는 생산공정 자체가 고 에너지 요구성 공정이며 중화를 위한 세척과정 중에 발생하는 폐수 등으로 인한 2차 환경오염 문제를 안고 있다(Pizarro 등, 2003, Pceess Biochem, 38,1077).
뿐만 아니라 바이오디젤 생산원가의 70% 이상을 차지하는 것이 원료의 단가인데 이러한 원료단가를 낮추기 위해서는 식용인 아닌 저급의 유지를 사용할 필요성이 있다. 그러나 폐식용유나 팜 슬러지 오일과 같은 저급 유지의 특성상 자유지방산(free fatty acid) 의 함량이 매우 높아 산가(acid value)가 매우 높다. 이처럼 산가가 높은 원료를 이용하여 기존 알칼리 촉매 방식을 사용하게 되면 비누화 반응으로 인해 비누가 형성되고 이로 인해 반응공정 효율이 낮아질 뿐만 아니라 심할 경우 공정 자체가 불가능해 진다. 이러한 저급유지의 자유지방산을 제거하기 위해서 1차 반응으로 산촉매를 사용하여 자유지방산을 먼저 제거하고 난 원료를 다시 알칼리 촉매로 2차 반응을 해야 하는 다단계 방식을 사용해야 하는 문제점이 있다.
향후 대규모의 바이오디젤 수요에 대비한 바이오디젤의 양산을 위해서는 친환경적인 바이오디젤 전환공정이 필요하므로 다양한 미생물 유래의 생물촉매인 리파제가 바이오디젤 전환반응에 활용되고 있다. 특히 효소를 이용한 바이오디젤 전환은 폐식용유나 자유지방산 함량이 높은 저급 원료 등 다양한 종류의 원료를 이용할 수 있고 주로 상온에서 반응이 일어나기 때문에 바이오디젤 전환을 위한 에너지 비용이 적고 특히 중성 pH에서 반응이 되므로 중화를 위한 폐수의 발생이 없다는 장점이 있다. 또한 바이오디젤의 회수가 용이하며 부산물로서 고순도, 고품질의 글리세린을 얻을 수 있다(Fjerbaek 등, 2009, Biotechnol. Bioeng, 102, 1298).
그러나 효소공정은 화학적 방법에 비해 상대적으로 반응속도가 느리며 촉매의 생산비용이 높고 장기적인 활성 유지가 어려운 단점이 있어서 상용화의 저해 요인이 되고 있다(Zhang 등, 2003, Bioresour. Technol., 89, 1 ; Fukuda 등, 2001, J. Biosci. Bioeng., 92, 405 ; Haas 등 2006, Bioresour. Technol., 97, 671). 따라서 효소를 이용한 바이오디젤의 생산을 위해서는 고활성의 효소를 저비용으로 대량생산하는 기술이 절대적으로 필요하다.
바이오디젤 생산을 위한 리파제로서 박테리아, 효모 및 곰팡이 등 다양한 미생물 유래의 리파제가 이용되고 있다(Fjerbaek 등, 2009, Biotechnol. Bioeng, 102, 1298). 효모 캔디다 앤탁티카 유래의 리파제 B(Candida antarctica lipase B, CalB)는 이성질체 전환반응이나 폴리에스터 합성, 바이오디젤 전환반응 등에서 활용되는 효율적인 생물촉매이며(Anderson 등, 1998, Biocatal. Biotransform. 16, 181; Tan 등 2010, 28, 628) 특히 노보자임스(Novozymes)사에서 판매하는 고정화 효소인 Novozym435는 바이오디젤 전환반응용 효소로 많이 연구되고 있다. Novozym435는 곰팡이 발현시스템을 이용하여 대량 분비생산하고 아크릴릭 레진에 고정화한 효소로서 매우 고가에 판매되고 있어서 실제 바이오디젤 생산공정에 적용하기에는 비용측면에서 상당한 무리가 있다. 따라서 효소활성 개량을 통해서 효소비용을 절감하려는 연구와 재조합 방법으로 대량생산 하려는 연구가 무수히 진행되고 있다. 분자진화 기술을 이용한 효소의 활성을 개량(Zhang 등, 2003, Protein Eng. 16, 599; Kim 등, 2007, J Microbiol Biotechnol. 17, 1308; Sandstr등, 2009, Protein Eng Des Sel. 22, 413) 하였으며 효소의 저가생산을 위해서는 다양한 재조합 미생물을 이용하였다. CalB의 재조합 생산을 위해서 대장균(Blank 등, 2006, J. Biotechnol, 125, 474, Liu 등, 2006, Appl Microbiol Biotechnol. 72, 1024), 사카로마이세스 세레비지에(Zhang 등, 2003, Protein Eng, 16, 599), 피키아 파스토리스(Rotticci-Mulder 등, Protein Exp. Purif. 21, 386), 및 아스퍼질러스 오라이제(Hoegh 등, Can. J. Bot., 73, S869) 등 다양한 숙주세포가 사용되었다.
한편, 본 발명자들은 효모표면 발현 방법을 이용하여 CalB를 분자진화하고 스크리닝하여 고활성의 효소 CalB14를 개발(한국등록특허 제10-0475133호)한 바 있으며 이를 효모 단백질융합인자 기술을 이용하여 재조합 대량생산(한국등록특허 제 10-0626753호)하였고 이를 이용하여 생물학적 친환경 바이오디젤 생산에 적용한 바 있다(한국등록특허 제10-0697310호). CalB14 효소의 추가적인 개량을 통해 효소활성을 더욱 증진시키고 효소비용을 절감하기 위하여 CalB14를 분자진화하여 CalB1422를 개발하여 기존 효소에 비해 바이오디젤 전환반응에 매우 효율적임을 확인하고 특허를 출원한 바 있다(한국공개특허 제10-2011-0121244호).
본 발명에서는 재조합 CalB1422와 같은 리파제 효소를 이용하여 바이오디젤을 대량생산하기 위하여 연속적으로 바이오디젤 생산이 가능한 연속 생산 반응기를 제작하였다. 이를 이용하여 자유지방산의 함량이 매우 높은 저급의 유지를 원료로 하고 연속적으로 바이오디젤을 효소적 방법으로 생산할 수 있는 시스템을 개발, 본 발명을 완성하였다.

본 발명의 목적은 고정화 효소를 사용하여 기질의 지속공급을 통해 연속적으로 바이오디젤을 생산하는 장치 및 생산 방법을 제공하는 것이다.
본 발명의 다른 목적은 효소학적 방법으로 높은 전환율로 바이오디젤을 연속적으로 장기간 생산할 수 있는 장치 및 생산 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 바이오디젤 반응에 사용된 고정화 효소를 손쉽게 회수하고 반복적으로 사용할 수 있는 장치 및 생산 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 자유지방산 함량이 높아 높은 산가(acid value)를 가진 폐유지들을 손쉽게 바이오디젤로 전환시킬 수 있는 장치 및 생산 방법을 제공하는 것이다.
상기와 같은 과제를 해결하기 위한 본 발명에 따른 바이오디젤 생산 반응기는 메쉬를 포함하는 고정화 효소통이 위치하며, 상기 고정화 효소통 에 고정화 효소가 위치하는 반응기, 유지 및 알코올을 각각 상기 반응기에 공급하는 유지 공급부와 알코올 공급부 및 상기 반응기에서 생성된 바이오디젤을 회수하는 바이오디젤 회수부를 포함하는 것을 특징으로 한다.
상기 유지 공급부로부터 공급된 유지 및 알코올 공급부로부터 공급된 알코올이 상기 고정화 효소통 내에 유입되어 효소촉매반응이 이루어지며, 이에 따라 생산된 바이오디젤이 상기 바이오디젤 회수부에서 회수되는 것이 바람직할 수 있다.
상기 연속식 바이오디젤 생산 반응기는 동력부재 및 상기 동력부재에 의해 동력을 전달받아 회전 가능하며, 상기 고정화 효소통에 고정되는 회전축을 더 포함하는 것이 바람직할 수 있다.
상기 연속식 바이오디젤 생산 반응기는 글리세롤 트랩(glycerol trap)을 더 포함하며, 상기 반응기에서 생성된 글리세롤은 바이오디젤 중 일부와 함께 상기 글리세롤 트랩으로 유동하며, 상기 글리세롤 트랩으로 유동된 글리세롤과 바이오디젤 중 글리세롤이 침전하고 바이오디젤은 분리되어 상기 반응기에 재유입되는 것이 바람직할 수 있다.
상기 반응기에 바이오디젤 배출관이 위치하며, 상기 바이오디젤 배출관은 상기 반응기 내측에 위치하는 상기 고정화 효소통의 하단에 위치하며, 그리고 상기 바이오디젤 배출관은 상하 두 개로 브랜치(brach)되어, 상부의 관은 상기 바이오디젤 회수부와 유체소통하고, 하부의 관은 상기 글리세롤 트랩과 유체소통하는 것이 바람직할 수 있다.
상기 유지 공급부 및 상기 알코올 공급부는 각각 펌프에 의하여 각각 상기 반응기와 유체 소통하며, 상기 펌프는 연동식 펌프(peristaltic pump)인 것이 바람직할 수 있다.
상기 반응기는 캡과 몸체를 포함하며, 상기 몸체의 외벽 내부에는 물이 유동하는 워터재킷이 위치하는 것이 바람직할 수 있다.
상기 워터재킷은 항온수조와 유체 소통하며, 상기 항온수조의 온도가 조절 가능하도록 하는 것이 바람직할 수 있다.
또한, 상기 연속식 바이오디젤 생산 반응기를 이용한 연속식 바이오디젤 생산 방법을 제공한다.
상기 연속식 바이오디젤 생산방법은 상기 고정화 효소통에 상기 고정화 효소를 위치시킨 후 이를 상기 반응기 내에 결합시키는 단계, 상기 유지 공급부로부터 상기 반응기에 유지를 공급하는 단계, 상기 고정화 효소통을 교반시키는 단계, 상기 알코올 공급부로부터 상기 반응기에 알코올을 공급하는 단계 및 상기 반응기에서 바이오디젤이 생산되고, 상기 바이오디젤 회수부가 상기 생산된 바이오디젤을 회수하는 단계를 포함한다.
상기 생산된 바이오디젤을 회수하는 단계 이후에 기 설정된 수치 이상의 전환율로 바이오디젤이 생산된 경우 상기 유지 공급부 및 상기 알코올 공급부로부터 상기 반응기에 유지 및 알코올을 추가로 공급하는 단계를 더 포함하며, 이후, 상기 반응기에 유지 및 알코올을 더 공급하는 단계와 생산된 바이오디젤을 회수하는 단계가 반복되는 것이 바람직할 수 있다.
상기 반응기에 유지 및 알코올을 공급하는 단계의 유지 및 알코올의 공급속도가, 상기 생산된 바이오디젤을 회수하는 단계의 바이오디젤의 회수속도에 상응하는 것이 바람직할 수 있다.
상기 고정화 효소통 내에 위치하는 상기 고정화 효소는 리파제(lipase)인 것이 바람직할 수 있다.
상기 유지 및 상기 알코올의 비율은, 12:1인 것이 바람직할 수 있다.
상기 유지의 산가(acid value)는 20 내지 130인 것이 바람직할 수 있다.
상기 고정화 효소의 양은 유지량의 10%(w/v)인 것이 바람직할 수 있다.
본 발명에 따른 연속식 바이오디젤 생산 반응기 및 생산 방법에 따라, 종래의 화학적 방법의 바이오디젤 생산기술과 비교하여 에너지 효율이 높고 친환경적인 방법으로 바이오디젤을 생산할 수 있다. 특히 기존의 화학적 방법은 자유지방산의 농도가 높은 고산가 저급 원료를 사용하기 위해서 복잡한 전처리단계가 필요하였지만 본 발명에 따른 연속식 바이오디젤 생산 반응기 및 생산 방법에서는 전처리 없이 그대로 바이오디젤을 생산할 수 있다.
또한, 바이오디젤 생산 시 발생하는 부산물인 글리세롤(glycerol)의 순도가 높고 분리정제가 용이하여 다양한 용도로 이용이 가능하다.
또한, 바이오디젤 반응에 있어서 필요한 알코올(예를 들어, 메탄올)과 유지를 지속적으로 유입해주면서 그 반응으로 생산된 바이오디젤을 연속적으로 생산하고 추출할 수 있다.
실제로, 반응 볼륨 1리터의 반응기에서 생산해내는 바이오디젤의 양은 시간당 약 33ml로서 유입되는 유지는 이 중 약 90%이고, 동시에 유입되는 알코올은 이중 약 10%를 차지하는데, 본 발명에 따른 연속식 바이오디젤 생산 반응기를 이용하여 진행한 결과 약 100일 동안 평균 88% 이상의 바이오디젤 전환율을 유지함을 확인하였다(실시예 1 및 도 9 참조).
또한, 자유지방산의 함량이 많은 높은 산가의 유지를 복잡한 과정 및 처리 없이 친환경적인 효소 반응만으로 바이오디젤로 전환하여 생산할 수 있다. 이는 폐식용유 또는 팜슬러지유 등의 자유지방산의 함량이 매우 많아 산가가 높아진 유지를 효과적으로 바이오디젤로 전환하는 것을 의미한다. 종래 방법에 따르면, 폐식용유와 팜슬러지유와 같은 경우에 높은 산가를 띄고 있는데 화학촉매를 사용하는 화학적 바이오디젤 반응을 하게 되면 유지 내 자유지방산과 알칼리성의 화학 촉매 간에 비누화 반응이 일어나 바이오디젤로의 전환반응이 이루어질 수 없었다. 이로 인하여 때문에 1차적으로 자유지방산을 제거하는 산 촉매 화학반응이 이루어지고 2차적으로 염기 촉매 화학반응을 하게 되는데 이들 반응 공정 중에 다량의 오염물 생성이 불가피한 실정이다. 하지만 본 발명에 따른 연속식 바이오디젤 생산 반응기 및 생산 방법을 사용할 경우 이와 같이 높은 산가를 띄고 있는 유지를 사용해도 별도의 처리 없이 바이오디젤로의 전환이 용이하게 된다.
실제로 본 발명에 따른 연속식 바이오디젤 생산 반응기에 높은 산가를 띈 유지를 사용한 결과, 유지 내 자유지방은 바이오디젤 반응이 진행되면서 급격하게 하락하였고, 95% 이상의 전환율에 도달하면서 산가 역시 반비례적으로 5% 내의 낮은 위치까지 도달하였다. 이로써 효소를 사용하여 연속식 반응기에서 연속적으로 바이오디젤을 생산하는 공정에도 적용 가능하다는 것을 확인하였다.

이하, 도면과 실시예를 통하여 본 발명을 더욱 상세하게 설명하기로 한다. 이들 실시 예는 단지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시 예에 의해 제한되는 것으로 해석되지는 않는다.
1. 연속식 바이오디젤 생산 반응기의 설명
도 1 내지 도 5를 참조하여 본 발명에 따른 연속식 바이오디젤 생산 반응기를 상세히 설명한다.
도 1에 도시된 바와 같이, 본 발명에 따른 연속식 바이오디젤 생산 반응기는, 반응기(100), 동력부재(200), 항온수조(300), 유지 공급부(400), 알코올 공급부(500), 글리세롤 트랩(600) 및 바이오디젤 회수부(700)를 포함한다.
반응기(100)의 내부에는 고정화 효소통(150)이 위치하며, 그 내부에는 고정화 효소가 위치하여 효소촉매반응이 이루어지고 바이오디젤이 생산된다. 아래에서 상술한다.
동력부재(200)는 회전축(210)에 동력을 전달하고 회전축(210)은 고정화 효소통(150)에 고정되어, 동력부재(200)의 동작에 의하여 고정화 효소통(150)이 회전하면서 교반 작용을 돕는다.
항온수조(300)는, 반응기(100)의 몸체(120) 외벽에 위치하는 워터재킷(121)과 유체소통함으로써, 반응기(100)의 온도를 일정하게 유지시키는 기능을 한다.
상기 반응기(100) 내부의 반응온도는 40~42℃ 인 것이 바람직하다.
유지 공급부(400)는 펌프(410)에 의하여 바이오디젤 생산을 위한 유지를 반응기(100)에 공급한다. 또한, 상기 반응기(100)에 공급되는 유지는 천연유지, 가공유지, 폐유지 또는 이의 조합으로 이루어진 것이 바람직사용 가능하나, 폐유지인 것이 특히 바람직하다.
알코올 공급부(500)는 펌프(510)에 의하여 바이오디젤 생산을 위한 알코올을 반응기(100)에 공급한다. 또한, 상기 반응기(100)에 공급되는 알코올은 메탄올, 에탄올, 프로판올, 부탄올 또는 이의 조합으로 이루어진 것이 사용 가능하다.
글리세롤 트랩(600)에는 반응기(100)에서 생산되는 부산물인 글리세롤과 바이오디젤 일부가 유입되는데, 비중을 이용하여 이중 글리세롤을 분리하는 기능을 한다. 글리세롤이 분리된 바이오디젤은 반응기(100)에 재유입된다. 펌프(610)에 의하여 유체소통한다. 아래에서 상술한다.
바이오디젤 회수부(700)는 펌프(710)에 의하여 반응기(100)에서 생산된 바이오디젤을 회수한다.
전술한 펌프들(410, 510, 610, 710)은 연동식 펌프(peristaltic pump)인 것이 바람직하다. 특히, 유지 공급부(400) 및 알코올 공급부(500)와 연동하는 펌프(410, 510)가 연동함으로써 유지와 알코올을 일정 비율로 연속적으로 혼합 공급할 수 있어서 바람직하다. 이에 따라 유지 및 알코올의 속도와 동일한 속도로 바이오디젤이 생성되어 회수될 수 있다.
도 1 내지 도 4를 참조하여, 반응기(100)와 그 내부에 위치하는 고정화 효소통(150)을 상술한다. 도 2에서는 설명을 위하여 고정화 효소통(150)이 반응기(100) 외부에 도시된다.
반응기(100)는 반응기 캡(110)과 반응기 몸체(120)로 이루어진다.
반응기 캡(110)에는 동력부재(200)에 의하여 동력을 전달받는 회전축(210)이 고정화 효소통(150)에 삽입될 수 있도록 축공(112)이 위치한다. 또한, 유지 공급부(400)로부터 유지를 공급받기 위한 유지 유입관(114), 알코올 공급부(500)로부터 알코올을 공급받기 위한 알코올 유입관(115) 및 글리세롤 트랩(600)으로부터 글리세롤이 분리된 바이오디젤을 재유입받기 위한 바이오디젤 유입관(116)이 위치한다.
반응기 몸체(120)의 외벽에는 워터재킷(121)이 위치한다. 반응기 몸체(120)의 외벽에는 워터 유출관(122) 및 워터 유입관(123)이 위치하고, 이들은 항온수조(200)와 유체소통한다. 이를 통하여 반응기(100) 내부의 온도가 항온수조(200)의 온도 조절에 의하여 용이하게 제어 가능하다.
반응기 몸체(120)의 내측에는 전술한 바와 같이 고정화 효소통(150)이 위치하며, 고정화 효소통(150)의 하단에 바이오디젤 배출관(126)이 위치한다. 바이오디젤 배출관(126)은 상하의 두 개의 관으로 브랜치(branch)되어, 상부의 관은 글리세롤이 침전되지 않은 바이오디젤인바 바이오디젤 회수부(700)와 유체소통하고, 하부의 관은 글리세롤이 침전되어 있을 것인바 글리세롤 트랩(600)과 유체소통한다. 즉, 바이오디젤 회수부(700)로는 글리세롤이 거의 없는 바이오디젤이 유동할 것이며, 글리세롤 트랩(600)으로는 글리세롤과 바이오디젤 일부가 유동할 것이다.
고정화 효소통(150)은 반응기(100) 내부의 비어있는 공간에 위치한다.
고정화 효소통(150)은 고정화 효소통 캡(151)과 고정화 효소통 몸체(152)로 이루어진다. 특히, 고정화 효소통 몸체(152)는 스테인레스 재질의 다공성 철망인 메쉬 형태인 것이 바람직하다. 이에 따라, 고정화 효소는 고정화 효소통(150) 내에 머물고 기질인 유지와 알코올은 통과할 수 있으며, 산물의 유출 또한 용이하여 연속반응을 가능하게 한다.
고정화 효소통(150)은 회전축(210)에 의해 회전하며 반응기 내부의 기질을 지속 혼합한다.
도 5를 참조하여 글리세롤 트랩(600)을 상술한다.
전술한 바와 같이, 글리세롤 트랩(600)은 반응기(100)의 바이오디젤 배출관(126)의 하부의 관과 유체소통하여, 반응기(100)에서 생성된 글리세롤이 바이오디젤 일부와 함께 유입된다.
글리세롤 및 바이오디젤 유입관(620)을 통하여 유입되는데, 유입된 글리세롤 및 바이오디젤은 비중 차이로 인하여 분리되어, 글리세롤만이 글리세롤 트랩(600)의 하부에 침전될 것이다.
이에 따라 바이오디젤이 분리되는데, 글리세롤 트랩(600)의 중간 즈음에 바이오디젤 유출관(630)의 말단을 위치시킴으로써 바이오디젤을 추출할 수 있으며, 추출된 바이오디젤은 전술한 바와 같이 다시 반응기(100)에 재유입된다.
2. 연속식 바이오디젤 생산 반응기를 이용한 바이오디젤 생산방법
<고정화 효소의 준비>
먼저, 리파제 활성이 개선된 변이리파제 CalB1422를 본 발명에 따른 방법에 사용하도록 대량 생산하기 위하여, pYGa-ST3-CalB1422 재조합 벡터를 Y2805△gal80 균주에 형질전환 하여 5ℓ 용량의 발효조에서 유가식 배양을 수행하였다. 배양에 들어가기 전에 50mℓ의 최소 액체배지(0.67% 아미노산이 결여된 효모기질, 0.5% 카사미노에시드, 2% 포도당)에 초기배양한 후 이를 200mℓ의 YPD 액체배지(1% 효모추출물, 2% 펩톤, 2% 포도당)에서 배양하여 활성화시킨 후 발효조의 본 배양액에 접종하여 30℃에서 48시간 동안 배양하였다. 발효 배지로 분비된 리파제를 확인하기 위하여 시간별 배양 상등액을 취하여 SDS-PAGE 분석하였다(도 6). CalB1422 유전자 크기로부터 유추되는 단백질 크기(33 kD)보다 큰 50 kD 단백질로 분비 생산되었는데, 이는 단백질분비융합인자 TFP3의 pro 서열이 융합된 형태로 분비 생산되지만 리파제의 고유활성은 유지됨을 확인한 바 있다. 48시간의 발효 배양 후 약 3~5 g/ℓ의 리파제가 배지로 분비 생산되었음을 확인하였다. pNPP 측정법을 이용하여 발효 시간별 리파제 효소의 활성을 분석한 결과 48시간의 발효 배양 후 약 250,000 U/L의 활성을 확인하였다(도 6). 재조합 대량생산된 CalB1422를 이용하여 바이오디젤 생산용 고정화 촉매를 개발하기 위해서, 고정화 흡착담체로서 Lewatit VP OC 1600 (Bayer)을 이용하여 고정화하였다. 효소를 고정화하기 전에 담체 100g을 담체가 충분히 잠길 정도양의 메탄올에 60분간 세척한 후 다시 동량의 50mM 트리스 완충용액(pH 7.5)에 25℃에서 1시간씩 2회 세척하여 전처리 하였다. 효소액은 레진 그람 당 효소 3000Unit을 첨가하여 진탕배양기에서 25℃, 200rpm의 속도로 17시간 고정화시켰다. 고정화 시간은 상등액에 존재하는 초기 효소활성과 일정시간이 경과한 후 잔여 효소활성을 비교하여 더 이상 잔여 활성이 변화가 없을 때까지 고정화 반응을 진행하였다(도 7). 또한 도면 7에서 보는 바와 같이 고정화 후 상등액에 남아 있는 단백질량을 확인한바 대부분의 효소 단백질이 담체에 흡착고정화 되었음을 확인할 수 있었다. 고정화 반응이 끝난 담체는 동량의 상기 완충용액으로 고정화된 담체를 2~3회 반복 세척하였다. 세척된 고정화 효소는 상온에서 진공펌프를 이용하여 완전 건조시키고 4℃의 냉장에 보관하였다.
<실시예 1>
산가가 낮은 대두유를 기질로 사용하였다. 우선 반응기(100)에 기질인 대두유를 900ml 넣고 고정화 효소통(150)에 고정화 효소를 사용된 기질 무게의 10%에 해당하는 효소(100g)를 투입, 반응기(100)에 채워 효소가 기질 속으로 잠길 수 있도록 한 후 반응기(100)의 온도를 40℃로 유지하였다. 반응시작 1시간 후부터 외부에서 메탄올을 시간당 17~20ml을 연동펌프(510)를 이용하여 지속적으로 공급하면서 바이오디젤 전환율을 시간별로 측정하였다(도 8). 반응시작 6~7시간 후에 바이오디젤 전환율이 95%에 달했으며 이때부터 기질과 메탄올을 12:1의 비율로 연동펌프(410, 510)를 이용하여 지속적으로 공급하여 연속반응을 유도하였다. 생산된 바이오디젤을 회수하는 펌프(710)의 유속은 기질로 사용되는 유지와 메탄올 공급량의 합과 동일한 비율로 조정하였다. 연속적으로 생산되어 회수되는 바이오디젤의 양은 시간당 약 30ml 정도 였다.
반응 중간에 기질 및 메탄올의 유입과 생산되어 회수되는 바이오디젤 간의 균형이 유지될 수 있도록 하였다. 생산된 바이오디젤을 반응기 외부의 바이오디젤 회수부(700)에서 수거하는 동시에 일부는 글리세롤 트랩(600)으로 흘려보냈다. 글리세롤 트랩(600)으로 들어간 바이오디젤은 글리세롤과의 비중 차이에 의한 침전작용으로 침전 및 분리시켰다. 글리세롤이 제거된 바이오디젤은 다시 반응기(100)로 들어가고 침전된 글리세롤은 간단히 회수할 수 있었다. 생산된 바이오디젤의 전환율 분석에는 가스 크로마토그래피(GC chromatograph : Agilent GC 7890A)를 사용하였다. 분석방법은 기질로 사용된 식용 대두유 내에 함유되어 있는 여러 지방산 중 약 11%를 함유하고 있는 팔미트산(C16:0)의 최종 함유량을 기준으로 분석하였다. 이러한 연속식 반응 공정을 통해 약 100일간 바이오디젤 연속반응을 수행한 결과 생성된 바이오디젤의 전환율을 평균 90%를 유지한 채 효소의 활성이 100일 이상 지속될 수 있음을 확인하였다(도 9).
<실시예 2>
실시예 1의 바이오디젤 반응에 사용했던 기질은 식용의 대두유로서 산가 1.0이하, 자유지방산의 함량이 0.5%이하인 고 정제된 식물성 유지였다. 하지만 이를 대량의 바이오디젤 반응에 사용하게 됐을 때 원료인 식물성 오일의 가격이 만만치 않게 된다. 따라서 현재 주로 사용되고 있는 화학적 생산방법의 바이오디젤에서는 폐식용유 등의 자유지방산의 함량이 높은 기질을 사용하고 있다. 하지만 전술한 바와 같이 이러한 자유지방산 함량이 높은 기질을 화학적 방법으로 반응하기 위해서는 1차적으로 산을 사용하여 자유지방산을 제거하는 등의 추가적인 공정이 필수적으로 사용되어야 한다. 그러나 이러한 공정 중에는 이물질 및 폐수 등의 발생으로 환경오염적인 문제가 나타날 수 있다. 이에 반해 효소를 촉매로 사용하는 생물학적 바이오디젤 반응에는 이러한 자유지방산의 함량은 반응 상에 큰 영향을 주지 않는다는 것이 최대 장점이다. 따라서 실시예 2는 인위적으로 산가 및 자유지방산의 함량이 조정된 기질들을 생물학적 바이오디젤 반응에 사용할 때 과연 정제된 기질을 사용했을 때와 차이가 있는 가에 대한 것이다.  
인위적으로 조정된 산가가 70 및 90으로 비교적 높은 2종의 식물성 오일을 기질로 사용하여 생물학적 바이오디젤 반응을 수행하였다.
유지를 제외한 다른 실험 환경은 실시예 1과 유사하기에 상세한 설명은 생략한다.
산가 70의 오일은 약 35%의 자유지방산과 65%의 트리글리세리드(triglyceride)로 구성되어 있다. 자유지방산 및 바이오디젤의 전환율은 가스 크로마토그래피 장비를 이용하여 분석하였으며 산가변화의 분석은 페놀프탈레인(phenolphthalein)지시약법을 이용해 확인하였다. 그 결과 도 10에서 보는 바와 같이 산가 70 및 90의 유지 모두가 반응이 진행됨에 따라 기질의 산가는 전환율과 반비례적으로 급격히 낮아 졌으며 기질 내 남아있던 자유지방산 또한 대부분이 반응에 사용되어 사라진 것을 확인할 수 있었다. 이에 따라 자유지방산의 함량이 높은 기질이라도 효소를 사용한 바이오디젤 전환 반응에는 거의 영향을 주지 않는다는 것을 확인할 수 있었다(도 10).
<실시예 3>
실시예 2에서 볼 수 있듯이 효소를 이용한 생물학적 반응으로 산가 70과 90의 유지를 별다른 처리나 추가적인 공정 없이 전환율 95%이상의 바이오디젤로 전환하여 생산할 수 있었다. 또 다른 식물성 기질로 팜 산업 부산물인 팜 슬러지 오일(palm sludge oil)은 팜유의 착유공정중에 나오는 다양한 폐수로부터 회수되는데 처리과정에 따라 산가가 매우 다양하게 나타난다. 그동안 지방산이 함유된 식물성 오일이지만 자유지방산의 함량이 매우 높아 화학적 바이오디젤 전환반응에 적합하지 않았던 팜 슬러지 오일을 이용하여 생물학적 바이오디젤 반응에 적용하였다.
본 실시예에서는 산가가 최소 100이상이며 자유지방산이 약 60~70%에 달하는 저급의 팜 슬러지 오일을 확보하여 효소를 이용한 생물학적 바이오디젤 전환 반응을 수행하였다. 확보된 팜 슬러지오일은 높은 자유지방산 함유 외에도 수분(약 5~10%) 및 기타 불순물(약 2~5%)의 함유로 인해 바이오디젤 반응에 바로 사용하기에는 다소 문제가 있어서 1차적으로 간단한 정제과정을 거쳤다. 방법은 80~90℃로 유지되는 dry oven에서 일정시간 동안 보관하여 포화지방산이 다량 함유돼 상온에서 고체상태인 유지를 액체의 오일상태로 녹임과 동시에 함유되어 있는 수분을 증발시키고 유지보다 비중이 무거운 기타 불순물들은 침전시켜 순수 유지만을 얻었다. 우선 산가 100의 팜 슬러지오일을 알칼리 촉매를 사용한 화학적 바이오디젤 전환반응을 수행하였다. 그 결과 기질이 알칼리촉매와 접촉하자마다 급격하게 굳어져 고체화 되는 비누화 반응(saponification)을 일으켜 반응이 전혀 이루어지지 않았다. 다음으로 1차 산 촉매를 사용하고 2차 염기촉매를 사용하는 화학적 방법의 바이오디젤 전환반응을 수행하였다. 그 결과 염기촉매만을 사용했을 때와 다르게 1차 산 촉매를 사용한 반응에서 대부분의 자유지방산이 제거되었고 2차 염기촉매를 사용하여 95%이상의 바이오디젤 전환반응을 할 수 있었다. 그러나 반응 중에 기타 불순물 및 오염된 폐수가 발행하였다. 마지막으로 앞선 실시예들에 사용했던 고정화 효소를 사용하여 실험을 수행하였다. 본 발명에서 개발된 동일한 반응기(100) 및 반응조건을 사용하여 생물학적 바이오디젤 전환 반응을 수행하였다. 팜 슬러지오일의 특성상 상온에서는 고체로 굳어지기 때문에 액체로 유지될 수 있는 온도인 50℃로 반응기(100)의 온도를 유지시켰다. 그 결과 산가 100이상, 자유지방산 함유량 60% 이상의 저급유지를 기질로 사용했음에도 반응시간 14시간 만에 전환율 95%이상까지 도달하였다(도 11). 100이상이었던 산가는 반응 후 4시간 만에 절반 이하로 떨어졌고 최종 산가는 10미만까지 도달하였다. 앞선 정제된 기질을 사용했을 때보다 반응시간이 좀 더 걸렸는데 이는 첨가되는 메탄올이 50℃의 반응기 온도에 의해 첨가되는 양 모두가 반응에 사용되지 못하고 일부가 증발되는 문제로 인해 반응이 다소 늦어졌다고 해석할 수 있겠으나, 연속식 반응은 충분히 양호하게 수행됨을 확인하였다.
<실시예 4>
산가 100이상의 저급유지인 팜 슬러지오일을 효소촉매를 이용한 생물학적 바이오디젤 전환반응이 95%이상까지 가능함을 확인함에 따라 본 발명에서 개발된 반응기를 이용하여 연속적 바이오디젤 반응을 수행하였다. 반응 조건은 기질 900ml과 기질무게의 10%인 고정화 효소 100g을 반응기(100)내 기질에 충분히 잠길 수 있도록 하여 반응을 수행하였다. 유입되는 기질과 메탄올의 비율은 전과 동일한 12:1로 하였고 외부로 회수되는 바이오디젤은 유입과 유출의 균형이 맞도록 조정하였다. 그 외의 조건들은 앞선 실시예들과 동일하다.
최초 0에서 95%이상까지의 전환반응은 약 10시간에 거쳐 무난하게 진행되었다. 95%이상의 전환율에 도달 후 기질과 메탄올의 유입, 생성된 바이오디젤과 글리세롤의 유출의 균형을 맞추면서 연속식 반응을 수행하였다. 그 결과 연속반응이 시작되고 최초 95%였던 전환율이 점차 하락하는 추세가 나타났다(도 12).
이상, 본 명세서에는 본 발명을 당업자가 용이하게 이해하고 재현할 수 있도록 도면에 도시한 실시예를 참고로 설명되었으나 이는 예시적인 것에 불과하며, 당업자라면 본 발명의 실시예로부터 다양한 변형 및 균등한 타 실시예가 가능하다는 점을 이해할 것이다. 따라서 본 발명의 보호범위는 특허청구범위에 의해서 정해져야 할 것이다.

  12. 제 11 항에 있어서,
    상기 (f) 단계의 유지 및 알코올의 공급속도가, 상기 (e) 단계의 바이오디젤의 회수속도에 상응하는 것을 특징으로 하는,
    연속식 바이오디젤 생산 방법.
  13. 제 9 항에 있어서,
    상기 고정화 효소통(150) 내에 위치하는 상기 고정화 효소는 리파제(lipase)인 것을 특징으로 하는,
    연속식 바이오디젤 생산 방법.
  14. 제 9 항에 있어서,
    상기 유지 및 상기 알코올의 비율은, 12:1 인 것을 특징으로 하는,
    연속식 바이오디젤 생산 방법.
  15. 제 9 항에 있어서,
    상기 유지의 산가(acid value)는 20 내지 130인 것을 특징으로 하는,
    연속식 바이오디젤 생산 방법.
  16. 제 9 항에 있어서,
    상기 고정화 효소의 양은 유지량의 10%(w/v)인 것을 특징으로 하는,
    연속식 바이오디젤 생산 방법.
  17. 제 13 항에 있어서,
    상기 리파제는 캔디다 앤트악티카 유래의 CalB인 것을 특징으로 하는,
