Abstract
リグノセルロースを含有する植物バイオマスが収容された反応槽に、-40℃~80℃の状態で液体アンモニアまたは液体アンモニアを含むアンモニアを供給した後、60℃~135℃かつ供給時の反応槽温度よりも高い温度まで昇温し、気相及び液相を含むアンモニアによる処理を行う(エステル結合の開裂)。その後、反応槽温度-40℃~40℃まで反応槽内を冷却し、液相のアンモニアの質量/植物バイオマスの乾燥質量が0.1~1.1となるように反応槽内に液体アンモニアまたは液体アンモニアを含むアンモニアを追加し、-40℃~40℃で液相を含むアンモニアによる処理を行う(結晶型転移)。
Description
本発明は、リグノセルロースを含有する植物バイオマスから糖を製造する際に用いられる酵素糖化用原料を製造する方法、前記原料から糖を製造する方法、並びに前記糖からエタノールを製造する方法に関する。
近年、地球温暖化対策の一環として、木質バイオマスや草本バイオマス等の、リグノセルロース(セルロース、ヘミセルロース、リグニンからなる複合体)を含む植物バイオマスからエタノールを製造し、各種燃料や化学原料として利用しようとする試みが広く行われている。リグノセルロースを含有する植物バイオマス(以下、単に「植物バイオマス」ということもある。)からのエタノールの製造は、例えば、収集した植物バイオマスを、糖化工程において単糖に分解した後、得られた糖を酵母等の微生物を利用した発酵によりエタノールに変換することにより行うことができる。
また、前記糖を、エタノール発酵だけでなく、各種の微生物を用いる発酵に供することにより、様々な化学原料、例えば有機酸等を製造することもできる。一例として、前記糖を乳酸発酵に供することにより、生分解性高分子の原料等に使用される乳酸を製造することができる。
植物バイオマスの糖化は、従来、濃硫酸を用いて行われることが多かったが、環境負荷低減の観点から、硫酸の使用量を少なくすることが望まれている。そこで、近年は、濃硫酸による糖化に代わる手段として、酵素を用いる糖化が広く研究されている。酵素による糖化は、環境に対する影響の観点から望ましい手段であるが、この酵素糖化のためには、セルロースに対して酵素を作用させやすくするとの目的から、植物バイオマスに前処理を施して、リグノセルロース中のセルロースとリグニンとを分離することが必要となる。
この植物バイオマスの前処理方法としては、従来、希硫酸、加圧熱水等による蒸煮処理などが一般的である(例えば、下記特許文献1~4参照。)。しかしながら、前述のように、環境負荷低減の観点から硫酸の使用は好ましくない。
一方、植物バイオマスをアンモニアで処理することにより、その化学的反応性又は生物化学的反応性が向上することが知られている(例えば、下記特許文献5、6、非特許文献1、2参照。)。
植物バイオマス中のリグノセルロースにあっては、リグニンとヘミセルロースとが共有結合(エステル結合等)により結合し、ヘミセルロースとセルロースとが水素結合を介して相互作用することにより、セルロースとリグニンとが強固に複合化されており、このリグニンが酵素のセルロースへの作用を阻害するとされる。そして、植物バイオマスをアンモニアにより処理することによって酵素糖化の効率が向上する作用機構のひとつは、前記リグニンとヘミセルロースとの間のエステル結合がアンモニアによりアミド化開裂する等により、リグニンの少なくとも一部がセルロースから分離されることに基づくとされる。以下、前記リグニンとヘミセルロースとの間のエステル結合等の開裂によるリグニンのセルロースからの分離に基づく酵素糖化の効率向上の作用を「化学的作用」という場合もある。なお、前記エステル結合のアミド化開裂反応は、アンモニア処理の処理温度が高いほど進行しやすい。
また、リグノセルロースをアンモニアで処理する際、上記リグニンとセルロースとの分離とは別に、アンモニアによるセルロースの結晶型の転移により酵素糖化の効率が向上することも知られている。すなわち、植物バイオマスを構成するセルロースは、主としてセルロースI型結晶として存在するが、このセルロースI型結晶がアンモニアと接触することにより、セルロースI型結晶よりも低い結晶密度を有し、酵素の作用をより受けやすいセルロースIII型結晶に転移することが知られている(例えば、下記特許文献7参照。)。以下、セルロース結晶型の転移に基づく酵素糖化の効率向上の作用を「物理的作用」という場合もある。
前記物理的作用は、セルロースI型結晶が液相あるいは超臨界状態にあるアンモニアと接触することにより発現することが知られている。一方、気相のアンモニアはセルロースに対して構造的観点から何ら影響を与えないとされる(非特許文献1参照。)。これは、気相のアンモニアではセルロースとの間でセルロース-アンモニア中間体を形成することができないため、セルロースI型結晶をセルロースIII型結晶に転移させることができないためと考えられる(非特許文献2参照)。
そのため、従来、前記植物バイオマスのアンモニア処理において、化学的作用と物理的作用の両方を同時に有効に利用しようとすると、高圧容器内に植物バイオマスとアンモニアとを導入し、化学的作用を発現させるためのリグニンとヘミセルロースとの間の結合の開裂反応を促進するために高圧容器内を加熱し、且つ、物理的作用を発現させるためのセルロース結晶の転移を進行させるために、高圧容器内でアンモニアを液相に保って処理を行うか、あるいは更に高温高圧の条件として、アンモニアを超臨界状態として処理を行うことが行われていた。

ところで、植物バイオマスのアンモニアによる処理においては、通常、処理済みの植物バイオマスからアンモニアを分離・回収して再使用する。このアンモニアの分離・回収、再使用の工程においては、アンモニアの精製処理を含め、液相のアンモニアを気化させる、あるいは気相のアンモニアを液化させるといった操作を要する。ここで、アンモニアは蒸発潜熱が極めて大きい物質であることから(1262kJ/kg(0℃、101.3kPaA))、これらの操作に要するアンモニア単位量当りのエネルギーが大きく、植物バイオマス単位量当りのアンモニア使用量が増加すると、前記アンモニアの分離・回収、再使用に要する消費エネルギーが増大し、プロセスの経済合理性を失わせることとなる。また、再生使用するとはいえ、多量のアンモニアを使用すること自体、コストの上昇を招く。従って、アンモニア処理工程を含む植物バイオマスからの酵素糖化用原料の製造、前記原料を用いる糖の製造、並びに前記糖からのエタノールの製造において、アンモニア処理におけるアンモニア使用量の低減は、プロセスの商業化にとって極めて重要な事項である。
しかしながら、従来の、アンモニア処理工程を含む植物バイオマスからの酵素糖化用原料の製造、前記原料を用いる糖の製造、並びに前記糖からのエタノールの製造においては、アンモニア処理におけるアンモニア使用量の低減については必ずしも十分な検討がなされていない。
さらに、本発明者らの検討によれば、従来法による場合、経済合理性を満たすまでにアンモニア使用量を低減することが非常に困難であることが判明した。
例えば、液相のアンモニアによる植物バイオマスの処理においては、アンモニアは通常、圧力容器内で気液混相となっている。この際に、圧力容器内に液相のアンモニアを所定量存在するようにしようとすると、容器内の温度が高温であるほど、気相部の圧力(当該温度におけるアンモニアの飽和蒸気圧)が上昇して蒸気密度が増加するため、気相のアンモニア量が増加して、容器内の全アンモニア量は増加する。すなわち、処理すべき植物バイオマスの単位量当たり必要となるアンモニアの量が増加してしまう。
一方、圧力容器内の温度を低くすると、容器内に存在する液相アンモニアの量を前記所定量とするために必要な全アンモニアの量は低減されるが、温度の低下に伴い、十分な化学的作用を発現させることが困難となる。仮にこの方法で十分な化学的作用を発現させようとすると、圧力容器内で極めて長時間処理するという、商業的実施に適さない操作が必要となる。
本発明は、前記事情に鑑みてなされたものであり、植物バイオマスに酵素糖化のために十分な反応性を付与することができ、かつ、アンモニアの使用量を十分に低減することが可能な酵素糖化用原料の製造方法、前記原料を用いて効率的に糖を製造する方法、並びに前記糖からエタノールを製造する方法を提供することを目的とする。
本発明者らは、前記目的を達成するために鋭意検討を行なった結果、まず、以下の知見を得た。
すなわち、植物バイオマスのアンモニアによる処理における物理的作用は、植物バイオマスと、植物バイオマスの量に対して特定の割合の量の液相のアンモニアとを接触させることにより効率的に発現させることができることが判明した。また、液相のアンモニアを用いた処理による物理的作用は、処理温度、処理時間の影響を殆ど受けず、低温・短時間でも十分に発現することが判明した。一方、アンモニア処理における化学的作用は、アンモニアが気相であっても発現することが判明した。
そして、本発明者らは、上記の知見に基づき、アンモニア使用量の低減という課題の解決を目的としつつ、アンモニア処理を複数回行うことを試みた。その結果、驚くべきことに、2種の特定のアンモニア処理を組み合わせることによって、植物バイオマスに酵素糖化のために十分な反応性を付与することができ、かつ、アンモニア使用量を飛躍的に低減させることができることを見出した。すなわち、1段目のアンモニア処理は、相対的に高温のアンモニアで処理を行い、エステル結合開裂反応を進行せしめるものである。また、2段目のアンモニア処理は、相対的に低温で液体アンモニアまたは液体アンモニアを含むアンモニアによる処理を行い、セルロースの結晶型の転移を進行せしめるものである。
さらに、本発明者らは、上記1段目のアンモニア処理において、気相アンモニアに加えて少量の液相アンモニアが存在する条件で処理を行うことにより、気相アンモニアのみで処理を行う場合に比較して、エステル結合の開裂がより効率的に進行するとの知見を得た。
ただし、植物バイオマスが収容された反応槽(圧力容器)への液体アンモニアまたは液体アンモニアを含むアンモニアの供給を、80℃を超える高温(特に処理温度、典型的には120℃)に保った状態で行うと、アンモニアの気化が進むために、容器内に液相アンモニアを存在させるためには多量のアンモニアを供給する必要がある。また、アンモニア分圧が上昇するために、反応槽の耐圧性能を高める必要があり、装置コストの上昇を招く。
そこで本発明者らは、さらに検討を重ねた結果、植物バイオマスが収容された反応槽を比較的低温(-40~80℃)に保ち、ここに液体アンモニアまたは液体アンモニアを含むアンモニアを供給し、その後処理温度まで反応槽温度を昇温することにより、前記高温で液体アンモニアまたは液体アンモニアを含むアンモニアを供給した場合に比較して、同一の処理温度、同一のアンモニア供給量であっても、圧力容器内に存在する液体アンモニア量が増加し、アンモニア分圧が低下することを見出し、本発明に到達した。
すなわち、本発明は、リグノセルロースを含有する植物バイオマスをアンモニアにより処理して酵素糖化用原料を得る酵素糖化用原料の製造方法であって、

前記第一工程においては、前記処理温度下で前記反応槽内に存在する液相のアンモニアの質量の前記植物バイオマスの乾燥質量に対する比が0.05~0.25となるように、前記反応槽に液体アンモニアまたは液体アンモニアを含むアンモニアを供給することが好ましい。前記条件を満たすことにより、効率的なエステル結合の開裂とアンモニアの使用量の低減との両立を有効に達成することができる。
また、本発明は、上記本発明の酵素糖化用原料の製造方法によって得られた酵素糖化用原料を酵素により糖化する工程を備える、糖の製造方法を提供する。
本発明の糖の製造方法においては、前記酵素糖化用原料の製造方法によって得られた糖化効率の向上した酵素糖化用原料を用いることで、酵素糖化により効率的に糖を製造することができる。
また、本発明は、前記糖の製造方法によって得られた糖を発酵させる工程を備える、エタノールの製造方法を提供する。
本発明のエタノールの製造方法においては、前記糖の製造方法によって得られた糖を用いることにより、出発原料の植物バイオマスから効率的にエタノールを製造することができる。
本発明によれば、従来における諸問題を解決することができ、化学的作用と物理的作用の両方を有効に利用して、酵素糖化の効率が十分に高められた酵素糖化用原料を与えることができ、従来よりも使用するアンモニアの量を低減することができ、更に高圧容器での処理時間を短縮することができる、リグノセルロースを含有する植物バイオマスのアンモニア処理による酵素糖化用原料の製造方法、前記原料を用いて効率的に糖を製造する方法、並びに前記糖からエタノールを製造する方法を提供することができる。
<第1実施形態:酵素糖化用原料の製造方法>
本発明の第1実施形態に係る酵素糖化用原料の製造方法は、リグノセルロースを含有する植物バイオマスをアンモニアにより処理して酵素糖化用原料を得る酵素糖化用原料の製造方法であって、

以下、本実施形態に係る酵素糖化用原料の製造方法について詳述する。
(植物バイオマス)
本実施形態の酵素糖化用原料の製造方法において原料として使用する植物バイオマスとしては、リグノセルロースを含有する限りにおいて特に制限はなく、目的に応じて適宜選択することができる。例えば、農業や林業等の生産活動に伴う残渣として得られる「廃棄物バイオマス」や、エネルギー等を得る目的で意図的に栽培して得られる「資源作物バイオマス」などを使用することができる。前記「廃棄物バイオマス」としては、例えば、廃建材、間伐材、稲わら、麦わら、もみ殻、バガスなどが挙げられ、また、前記「資源作物バイオマス」としては、例えば、セルロース類の利用を目的として栽培されるシラカバ、ユーカリ、ポプラ、アカシア、ヤナギ、スギ、スイッチグラス、ネピアグラス、エリアンサス、ミスカンサス、ススキ、リードカナリーグラスなどが挙げられる。また、植物バイオマスは、木に由来する「木質バイオマス」、草に由来する「草本バイオマス」などにも分類される。本発明においては、木質バイオマス及び草本バイオマス共に使用することができる。植物バイオマスは、1種単独で使用してもよいし、2種以上を併用してもよい。なお、未処理の植物バイオマスに含まれるリグノセルロースを構成するセルロースは、基本的にセルロースI型結晶の形態をとっている。
本実施形態の酵素糖化用原料の製造方法において原料として使用する植物バイオマスとしては、リグノセルロースを含有する限りにおいて特に制限はなく、目的に応じて適宜選択することができる。例えば、農業や林業等の生産活動に伴う残渣として得られる「廃棄物バイオマス」や、エネルギー等を得る目的で意図的に栽培して得られる「資源作物バイオマス」などを使用することができる。前記「廃棄物バイオマス」としては、例えば、廃建材、間伐材、稲わら、麦わら、もみ殻、バガスなどが挙げられ、また、前記「資源作物バイオマス」としては、例えば、セルロース類の利用を目的として栽培されるシラカバ、ユーカリ、ポプラ、アカシア、ヤナギ、スギ、スイッチグラス、ネピアグラス、エリアンサス、ミスカンサス、ススキ、リードカナリーグラスなどが挙げられる。また、植物バイオマスは、木に由来する「木質バイオマス」、草に由来する「草本バイオマス」などにも分類される。本発明においては、木質バイオマス及び草本バイオマス共に使用することができる。植物バイオマスは、1種単独で使用してもよいし、2種以上を併用してもよい。なお、未処理の植物バイオマスに含まれるリグノセルロースを構成するセルロースは、基本的にセルロースI型結晶の形態をとっている。
(植物バイオマスの粒子化工程)
植物バイオマスとしては、収集されたものをそのまま使用してもよいが、裁断や粉砕等によりある程度以下の大きさに粒子化してから使用することが、取り扱いの容易さ及びアンモニア処理の効率の観点から望ましい。この場合、粒子化後の植物バイオマスの粒子の大きさとしては特に制限はなく、粒子としての取り扱いやすさなどに応じて適宜選択することができるが、例えば、通過するメッシュの目開きとして、5mm以下が好ましく、3mm以下がより好ましい。前記メッシュの目開きの大きさが5mmを超えると、後述するアンモニア処理の効率が低下することがある。一方、単位操作としての粉砕はエネルギー効率が低く、植物バイオマスを微細な粒子にまで粉砕すると、粉砕に要するエネルギーが増大して、プロセスの経済合理性を損なうこととなる。従って、粉砕の消費エネルギー量は、例えば、植物バイオマス乾燥質量1kg当り1MJ以下が好ましく、粉砕に消費するエネルギー見合いで粒子の大きさを選択してもよい。
植物バイオマスとしては、収集されたものをそのまま使用してもよいが、裁断や粉砕等によりある程度以下の大きさに粒子化してから使用することが、取り扱いの容易さ及びアンモニア処理の効率の観点から望ましい。この場合、粒子化後の植物バイオマスの粒子の大きさとしては特に制限はなく、粒子としての取り扱いやすさなどに応じて適宜選択することができるが、例えば、通過するメッシュの目開きとして、5mm以下が好ましく、3mm以下がより好ましい。前記メッシュの目開きの大きさが5mmを超えると、後述するアンモニア処理の効率が低下することがある。一方、単位操作としての粉砕はエネルギー効率が低く、植物バイオマスを微細な粒子にまで粉砕すると、粉砕に要するエネルギーが増大して、プロセスの経済合理性を損なうこととなる。従って、粉砕の消費エネルギー量は、例えば、植物バイオマス乾燥質量1kg当り1MJ以下が好ましく、粉砕に消費するエネルギー見合いで粒子の大きさを選択してもよい。
粒子化に用いる装置としては特に制限はなく、目的に応じて適宜選択することができ、例えば、ウィレーミル、カッターミル、ハンマーミル、ピンミル等を用いることができる。
(植物バイオマスの乾燥工程)
収集された植物バイオマスは通常水分を保有している。この水分を保有した植物バイオマスをそのままアンモニア処理に供してもよいが、物理的作用を有効に利用して、得られる酵素糖化用原料の酵素糖化の効率を十分に向上させるためには、植物バイオマスを乾燥して使用することが好ましい。
収集された植物バイオマスは通常水分を保有している。この水分を保有した植物バイオマスをそのままアンモニア処理に供してもよいが、物理的作用を有効に利用して、得られる酵素糖化用原料の酵素糖化の効率を十分に向上させるためには、植物バイオマスを乾燥して使用することが好ましい。
植物バイオマスのアンモニア処理を行う系内に、植物バイオマスの量に対して一定以上の量の水が存在すると、結晶型転移に必要なセルロース-アンモニア中間体の生成が抑制される傾向にあると推定される。そのため、本実施形態の酵素糖化用原料の製造方法における結晶型転移工程において、系内の水分量が所定の水準を越えると、セルロースI型結晶のセルロースIII型結晶への転移が十分に進行しない。そこで、セルロース結晶の転移を進行させ、物理的作用を十分に発現させるためには、第二の工程における結晶型転移の際に系内の水分量を所定の水準以下とすることが好ましく、そのためには、植物バイオマスを乾燥し、その水分含有量を低減させてアンモニア処理に供することが好ましい。
第二工程において、植物バイオマス及びアンモニア中に含まれる水分量は下記式(1)を満たすことが好ましい。そのためには、植物バイオマスを第二工程に供するに際して、当該植物バイオマス中の水分含有量が少なくとも下記式(1)を満たすことが好ましい。

更に、第二工程においては、植物バイオマス及びアンモニア中に含まれる水分量が下記式(2)を満たすことが好ましい。そのためには、乾燥後の植物バイオマスを第二工程に供するに際して、当該植物バイオマス中の水分含有量が少なくとも下記式(2)を満たすことが好ましい。

植物バイオマスの乾燥方法としては特に制限はなく、目的に応じて適宜選択することができるが、大気下に高温で乾燥すると、植物バイオマスの組織が破壊されたり酸化されたりするため、例えば、天日乾燥、自然乾燥、100℃以下での通風による乾燥、減圧乾燥、ジメチルエーテルを用いた乾燥などが好ましい。
植物バイオマスの粒子化を行う場合、乾燥はその前であっても後であってもよいが、取り扱い上、粒子化の後が好ましい。
植物バイオマスの乾燥は、第一工程と第二工程との間に行ってもよいが、第一工程の前に行うことが好ましい。
(第一工程)
第一工程においては、まず、リグノセルロースを含有する植物バイオマスが収容された反応槽に、反応槽温度-40℃~80℃の状態で液体アンモニアまたは液体アンモニアを含むアンモニアを供給する。次いで、60℃~135℃の範囲内でありかつ供給時の反応槽温度よりも高い温度である処理温度まで反応槽温度を昇温する。そして、該処理温度下で気相及び液相を含むアンモニアによる植物バイオマスの処理を行い、リグノセルロースを構成するリグニンとヘミセルロースとの間のエステル結合の少なくとも一部を開裂させる。リグニン/ヘミセルロース間の結合としては、エステル結合の他にエーテル結合も存在するが、第一工程におけるエステル結合の開裂は、エーテル結合の開裂を伴うものであってもよい。
第一工程においては、まず、リグノセルロースを含有する植物バイオマスが収容された反応槽に、反応槽温度-40℃~80℃の状態で液体アンモニアまたは液体アンモニアを含むアンモニアを供給する。次いで、60℃~135℃の範囲内でありかつ供給時の反応槽温度よりも高い温度である処理温度まで反応槽温度を昇温する。そして、該処理温度下で気相及び液相を含むアンモニアによる植物バイオマスの処理を行い、リグノセルロースを構成するリグニンとヘミセルロースとの間のエステル結合の少なくとも一部を開裂させる。リグニン/ヘミセルロース間の結合としては、エステル結合の他にエーテル結合も存在するが、第一工程におけるエステル結合の開裂は、エーテル結合の開裂を伴うものであってもよい。
ここで、本実施形態の第一工程において、リグノセルロースを含有する植物バイオマスが収容された反応槽に、反応槽温度-40℃~80℃(好ましくは外気温~60℃)の状態で液体アンモニアまたは液体アンモニアを含むアンモニアを供給し、次いで、60℃~135℃(好ましくは80℃~130℃)の範囲内でありかつ供給時の反応槽温度よりも高い温度である処理温度まで反応槽温度を昇温する、という構成を採用しているのは、後述する本発明者らの独自の知見に基づくものである。
すなわち、まず、エステル結合の開裂は、必ずしも液相のアンモニアを必要とするものではなく、液相を含まない気相のアンモニアのみであっても進行し得るが、少量の液体アンモニアが存在する条件で処理を行うことにより、気相アンモニアのみで処理を行う場合に比較して、エステル結合の開裂をより効率的に進行させることができる。
ただし、植物バイオマスが収容された反応槽(圧力容器)への液体アンモニアまたは液体アンモニアを含むアンモニアの供給を、80℃を超える高温(特に処理温度、典型的には120℃)に保った状態で行うと、アンモニアの気化が進むために、反応槽内に液相アンモニアを存在させるためには多量のアンモニアを供給する必要がある。また、アンモニア分圧が上昇するために、反応槽の耐圧性能を高める必要があり、装置コストの上昇を招く。
そこで本実施形態の第一工程では、植物バイオマスが収容された反応槽を比較的低温(-40~80℃)に保ち、ここに液体アンモニアまたは液体アンモニアを含むアンモニアを供給し、その後処理温度まで反応槽温度を昇温する、という構成を採用している。かかる構成の採用により、前記高温で液体アンモニアまたは液体アンモニアを含むアンモニアを供給した場合に比較して、同一の処理温度、同一のアンモニア供給量であっても、圧力容器内に存在する液体アンモニア量が増加し、アンモニア分圧が低下するため、効率的なエステル結合の開裂とアンモニア消費量の低減とを両立することが可能となる。
前記第一工程における液体アンモニアまたは液体アンモニアを含むアンモニアの供給量は、本発明の効果が損なわれない限りにおいて特に制限されないが、処理温度下で反応槽内に存在する液相のアンモニアの質量の植物バイオマスの乾燥質量に対する比(液相のアンモニアの質量/植物バイオマスの乾燥質量)が、好ましくは0.05~0.25、より好ましくは0.06~0.15となるように、反応槽に液体アンモニアまたは液体アンモニアを含むアンモニアを供給することが好ましい。前記条件を満たすことにより、効率的なエステル結合の開裂とアンモニアの使用量の低減との両立を有効に達成することができる。
また、第一工程における全アンモニアの植物バイオマスに対する量は、第二工程における全アンモニアの植物バイオマスに対する量以下であることが好ましい。第一工程及び第二工程における全アンモニアの量が前記条件を満たすことにより、第一工程及び第二工程で使用するアンモニアの総量を低減することができる。また、このような条件を選択しても、エステル結合の開裂を進行させて、化学的作用を十分に発現させることができる。
第一工程を実施するひとつの好ましい態様は、反応槽として圧力容器を用い、比較的高いアンモニア分圧及び比較的高い温度、比較的短時間でアンモニア処理を行うものである。この態様により、設備コストが高額となる圧力容器の容量を小さくし、且つ生産性を高めることができる。
前記圧力容器を用いる第一工程の態様において、アンモニア分圧は、好ましくは0.5MPaA~4MPaA、より好ましくは1.0MPaA~2.0MPaAである。アンモニア分圧が0.5MPaA未満である場合には、エステル結合の開裂が十分な速度で進行しない傾向にある。一方、アンモニア分圧が4MPaAを超える場合には、アンモニア使用量の増大を招く傾向にある。
前記圧力容器を用いる第一工程の態様において、処理温度は60~135℃、好ましくは80~135℃、最も好ましくは100~135℃である。処理温度が60℃を下回る場合には、エステル結合開裂の進行に長時間を要し、容量が低減された圧力容器を用いる装置においては、生産性が低下する傾向にある。一方、処理温度が135℃を超える場合には液相のアンモニアを存在させるために必要なアンモニアの供給量が増大してしまう。
エステル結合開裂工程におけるリグニンとヘミセルロースとの間のエステル結合のアミド化開裂は、FT-IR分光分析により確認することができる。より具体的には、FT-IRスペクトルにおける1662cm-1付近の吸収は、エステル結合開裂工程後の植物バイオマス中に生成したアミド基に帰属され、エステル結合のアミド化開裂の指標とすることができる。さらに、リグニンの芳香環に帰属される吸収(1515cm-1付近)の面積に対するアミド基に帰属される吸収の面積の比を算出し、これをエステル結合のアミド化開裂度合いの指標とすることもできる。なお、本発明においては、Thermo社製のNICOLET380を用い、波数400~4000cm-1の範囲について反射法によるFT-IRスペクトルの測定を行った。
また、エステル結合開裂工程におけるリグニンとヘミセルロースとの間のエステル結合の開裂は、熱水によるヘミセルロースの抽出によっても確認することができる。アンモニア処理によりリグニンとの間のエステル結合が開裂したヘミセルロースは冷水には溶解しにくいが、熱水には溶解しやすい。一方、エステル結合開裂工程に供していない植物バイオマス中のヘミセルロースは熱水に抽出されない。そこで、エステル結合開裂工程に供した後の植物バイオマスを熱水により抽出し、植物バイオマス中に含まれる全ヘミセルロースの量に対する抽出されたヘミセルロースの量の割合(以下、「抽出率」という。)をリグニン/ヘミセルロース間のエステル結合の開裂の指標とすることを検討した。なお、リグニンとの間のエステル結合が開裂したヘミセルロースが冷水に溶解しないのは、ヘミセルロースとセルロースとの間の水素結合等が原因であると考えられる。
上記熱水によるヘミセルロースの抽出の具体的な手順は以下のとおりである。
すなわち、まず、エステル結合開裂工程に供した後の植物バイオマス試料0.5gを、20mLの50mM-酢酸緩衝液(pH4.5)中に懸濁させ、100℃の熱水浴中で10分間加熱して抽出を行う。これを過し、ろ液10mLにヘミセルラーゼを含む市販酵素剤をタンパク量として約1mg添加し、37℃、200rpmにて24時間振とうし、抽出されたヘミセルロースの酵素分解を行う。得られた酵素分解液中のキシロースの濃度を、Shodex SUGAR SP0810(商品名)カラムを用いた高速液体クロマトグラフィー法により定量する。一方、別途に原料植物バイオマスを硫酸糖化法により糖化処理し、生成するキシロースの量を同様に定量する。このキシロースは、実質的に原料植物バイオマス中に含まれる全ヘミセルロースが加水分解して生成したと見なすことができる。前記熱水抽出及び酵素糖化により得られるキシロース量を前記硫酸糖化法により得られる原料植物バイオマス中の全ヘミセルロース由来のキシロースの量で除すことにより、ヘミセルロースの抽出率を算出する。
すなわち、まず、エステル結合開裂工程に供した後の植物バイオマス試料0.5gを、20mLの50mM-酢酸緩衝液(pH4.5)中に懸濁させ、100℃の熱水浴中で10分間加熱して抽出を行う。これを過し、ろ液10mLにヘミセルラーゼを含む市販酵素剤をタンパク量として約1mg添加し、37℃、200rpmにて24時間振とうし、抽出されたヘミセルロースの酵素分解を行う。得られた酵素分解液中のキシロースの濃度を、Shodex SUGAR SP0810(商品名)カラムを用いた高速液体クロマトグラフィー法により定量する。一方、別途に原料植物バイオマスを硫酸糖化法により糖化処理し、生成するキシロースの量を同様に定量する。このキシロースは、実質的に原料植物バイオマス中に含まれる全ヘミセルロースが加水分解して生成したと見なすことができる。前記熱水抽出及び酵素糖化により得られるキシロース量を前記硫酸糖化法により得られる原料植物バイオマス中の全ヘミセルロース由来のキシロースの量で除すことにより、ヘミセルロースの抽出率を算出する。
(第二工程)
前記第一工程後、まず、反応槽温度-40℃~40℃まで反応槽内を冷却する。次いで、液相のアンモニアの質量の植物バイオマスの乾燥質量に対する比(液相のアンモニアの質量/植物バイオマスの乾燥質量)が0.1~1.1となるように、反応槽内に液体アンモニアまたは液体アンモニアを含むアンモニアを追加する。そして、液相を含むアンモニアによる植物バイオマスの処理を行い、リグノセルロースを構成するセルロースの少なくとも一部の結晶型をI型からIII型に転移させて酵素糖化用原料を得る。
前記第一工程後、まず、反応槽温度-40℃~40℃まで反応槽内を冷却する。次いで、液相のアンモニアの質量の植物バイオマスの乾燥質量に対する比(液相のアンモニアの質量/植物バイオマスの乾燥質量)が0.1~1.1となるように、反応槽内に液体アンモニアまたは液体アンモニアを含むアンモニアを追加する。そして、液相を含むアンモニアによる植物バイオマスの処理を行い、リグノセルロースを構成するセルロースの少なくとも一部の結晶型をI型からIII型に転移させて酵素糖化用原料を得る。
なお、本実施形態において、第一工程においてエステル結合開裂を行った後、第二工程において結晶型転移を行うのは、次の理由による。
すなわち、エステル結合が開裂していない植物バイオマスについて結晶型転移を行うと、結晶型転移により一旦生成したセルロースIII型結晶が水分と接触することにより、セルロースI型結晶へと再度転移しやすい。一方、エステル結合開裂によりリグニンとヘミセルロースとの間のエステル結合の少なくとも一部が開裂されていると、結晶型転移により生成したセルロースIII型結晶は比較的安定となり、水分との接触によりセルロースI型結晶へ再度転移することが抑制される。
すなわち、エステル結合が開裂していない植物バイオマスについて結晶型転移を行うと、結晶型転移により一旦生成したセルロースIII型結晶が水分と接触することにより、セルロースI型結晶へと再度転移しやすい。一方、エステル結合開裂によりリグニンとヘミセルロースとの間のエステル結合の少なくとも一部が開裂されていると、結晶型転移により生成したセルロースIII型結晶は比較的安定となり、水分との接触によりセルロースI型結晶へ再度転移することが抑制される。
第二工程においては、リグノセルロースを含有する植物バイオマスを液相を含むアンモニアにより処理し、リグノセルロースを構成するセルロースの少なくとも一部の結晶型をI型からIII型に転移させる。なお、本発明において、「リグノセルロース」とは、未処理の植物バイオマスに含まれるリグニンとヘミセルロースとセルロースの複合体だけでなく、エステル結合開裂工程により、リグニンとヘミセルロースとの間のエステル結合の少なくとも一部が開裂した前記複合体、及び、未処理の植物バイオマスを結晶型転移工程に供し、セルロースI型結晶の少なくとも一部がIII型結晶に転移した前記複合体を含むものとする。
第二工程においては、アンモニアが液相を含むことが必要である。これは、セルロースI型結晶の少なくとも一部をIII型結晶に転移させるためには、セルロース-アンモニア中間体の生成が必要であり、該中間体の生成にはセルロースが液相のアンモニアと接触することが必要となるためである。従って、液相のアンモニアが存在すれば、セルロースI型結晶の少なくとも一部をIII型結晶に転移させることができる。
なお、第二工程における液相を含むアンモニアとは、通常は気液混相のアンモニアである。これは、圧力容器内を、気相部をもたず液体アンモニアで充満された状態とすることは困難であり、また、そのようにした場合には、使用するアンモニアの量が、物理的作用を最大限に利用するために必要な最小量よりも増加する傾向にあることから好ましくない。
第二工程において、結晶型転移の処理温度は-40℃~40℃であり、その処理温度において液相のアンモニアの質量の植物バイオマスの乾燥質量に対する比(液相のアンモニアの質量/植物バイオマスの乾燥質量)が0.1~1.1となるように、反応槽内に液体アンモニアまたは液体アンモニアを含むアンモニアを追加する。
なお、第二工程において、結晶型転移のために必要な液相のアンモニア量は温度の影響を殆ど受けない。一方、アンモニアが気液混相にある場合、気相のアンモニア量は温度によって変化し、低温ほど減少する。更に結晶型転移の進行も温度の影響を殆ど受けず、低温においても速やかに進行する。よって、アンモニア使用量を低減する観点から、結晶型転移工程における温度は低温であることが好ましい。他方、アンモニアをより低温に冷却するほど冷凍機の消費エネルギーが増加する。よって、低温にすることによるアンモニア使用量の低減と、冷凍機の消費エネルギーの増加とのバランスから、処理温度並びにその処理温度での液相のアンモニアの質量/植物バイオマスの乾燥質量の比を決定することが好ましい。具体的には、結晶型転移の処理温度は、-40℃~40℃であり、好ましくは-10℃~20℃である。また、液相のアンモニアの質量/植物バイオマスの乾燥質量の比は、前記のとおり0.1~1.1であり、好ましくは0.4~0.8である。なお、結晶型転移の処理温度は、-40℃~40℃の範囲内である限りにおいて、第一工程後に反応槽温度を冷却したときの温度と同一であっても異なっていてもよい。
第二工程におけるアンモニア分圧は特に限定されず、処理温度において液相のアンモニアが存在するために必要な分圧であればよい。その分圧は当該温度におけるアンモニアの飽和蒸気圧であり、当然処理温度によって変化するが、処理温度が-40~40℃である場合の好ましいアンモニア分圧として、0.07~1.6MPaAを例示することができる。
第二工程において、結晶型転移の処理時間は特に限定されない。液相を含むアンモニア処理による結晶型転移は速やかに進行することから、処理時間は例えば0.01~1時間である。
第二工程において、植物バイオマス及びアンモニア中に含まれる水分量は下記式(1)を満たすことが好ましい。

更に、第二工程においては、植物バイオマス及びアンモニア中に含まれる水分量が下記式(2)を満たすことが好ましい。

更に、原料バイオマスとしてエリアンサスを用いた例においては、植物バイオマスの乾燥質量に対して約40質量%以上の液相のアンモニアを存在せしめることにより、全てのセルロースI型結晶をIII型結晶に転移させることができる。従って、ほぼ全てのI型結晶をIII型結晶に転移せしめて物理的作用を最大限に利用しつつ、使用するアンモニア量を最小量とするためには、結晶型転移工程に存在する液相アンモニアの量を植物バイオマスの乾燥質量に対して約35~約50質量%とすることが好ましい。なお、前記の好ましい植物バイオマスの量に対する液相アンモニア量の比率は植物バイオマスの種類、セルロースの含有率、アンモニア処理に使用する装置及びその運転条件等により変化するため、実験により最適な液相アンモニア量を決定することが好ましい。
「植物バイオマスの乾燥工程」に関する説明において述べたように、結晶型転移工程において系内に存在する水分量が増加すると、セルロースI型結晶のIII型結晶への転移が抑制される傾向にある。そして、セルロースI型結晶のIII型結晶への転移を促進するためには、植物バイオマス中及びアンモニア中に存在する水分量は前記式(2)を満たすことが好ましい。前記水分量が前記式(2)を満たさない場合は、植物バイオマスを液相を含むアンモニアで処理しても、セルロースI型結晶のIII型結晶への転移が殆ど進行しない傾向にある。
更に、植物バイオマス中及びアンモニア中に存在する水分量は下記式(3):

植物バイオマス中及びアンモニア中に存在する水分量は前記式(3)を満たすことにより、リグノセルロースを構成するセルロースにおいて、セルロースI型結晶の全てがIII型結晶に転移する傾向にあり、物理的作用を十分に発現せしめることができる。
第二工程におけるセルロースI型結晶からIII型結晶への転移は、処理前後の植物バイオマスについてX線回折(XRD)分析を行うことによって確認することができる。より具体的には、XRDパターンにおいて、2θ=16°付近(101)及び2θ=22°付近(020)に検出されるピークがI型結晶に帰属されるピークであり、2θ=12°付近(101)及び2θ=21°付近(020)に検出されるピークがIII型結晶に帰属されるピークであり、これらのピークの有無により、各結晶型の有無を判定することができる。なお、本発明者らは、リグノセルロースを含有する植物バイオマスに気相のアンモニアによる処理を行った場合、処理後の植物バイオマスについてのXRDパターンにおいて、III型結晶に帰属されるピークが検出されないこと、すなわち気相のアンモニアによる処理では結晶型転移が起こらないことを併せて確認している。
以上の、本発明の第1実施形態に係る酵素糖化用原料の製造方法によれば、植物バイオマスに酵素糖化のために十分な反応性を付与することができ、かつ、アンモニアの使用量を十分に低減することが可能となる。
なお、前記第一工程は、ヘミセルロースとリグニンとの間のエステル結合を開裂するアンモニア処理以外の処理、例えば、所謂アルカリ蒸解処理、あるいは加圧熱水による蒸煮処理等に置き換えても、同様に酵素糖化の効率を向上することができる。但し、これらの工程においては多量の水を使用するため、処理後の植物バイオマスが多くの水を包含する。前述のように、本実施形態における結晶型転移工程においては、植物バイオマスが所定の割合を超える水分を含む場合には、セルロースI型結晶のIII型結晶への転移が阻害される傾向にあり、同工程の前に植物バイオマスの乾燥を行う必要がある。多量の水分を含む植物バイオマスの乾燥には多大なエネルギーを要することから、前記アンモニア処理以外の工程によりエステル結合開裂を行う方法は、経済合理性を損なう傾向にある。
<第2実施形態:糖の製造方法>
本発明の第2実施形態に係る糖の製造方法は、前記第1実施形態に係る酵素糖化用原料の製造方法により得られた酵素糖化用原料を酵素糖化する酵素糖化工程を少なくとも含み、必要に応じて更にその他の工程を含む。

以下、好ましい実施形態に沿って、本発明の糖の製造方法について詳細に説明する。
(酵素糖化工程)
本実施形態の糖の製造方法に係る酵素糖化工程は、前記酵素糖化用原料と酵素とを接触させることにより、バイオマスを構成するセルロース、及びヘミセルロースを加水分解して単糖類を得る工程である。

前記酵素糖化工程に用いられる酵素糖化の方法としては、酵素を用いる限りにおいて特に制限はなく、公知の方法を適宜選択することができる。硫酸等を用いる化学的な糖化方法を用いた場合には、過分解により単糖の収率が低下する傾向にあること、糖化に続く工程である発酵工程において阻害作用をもつ物質が生成し易い傾向にあること、及び硫酸等の環境負荷物質の排出が生じるなどの問題があるのに対して、酵素を用いる糖化方法においては、温和な条件を選択することが可能であり、前記の問題を生じ難い傾向にある。
前記酵素糖化工程において使用する酵素としては特に制限はなく、目的に応じて適宜選択することができ、例えば、セルラーゼ、セロビアーゼ(β-グルコシダーゼ)などが挙げられる。また、これら酵素を適当な担体又はマトリックスに固定化した固定化酵素を使用することもできる。
前記酵素糖化工程における酵素の使用量としては特に制限はなく、目的に応じて適宜選択することができるが、例えば、前記酵素糖化用原料中の固形分乾燥質量1gに対して、0.001mg~100mgが好ましく、0.01mg~10mgがより好ましく、0.1mg~1mgが更に好ましい。前記酵素の使用量が、前記酵素糖化用原料中の固形分乾燥質量1gに対して、0.001mg未満であると、酵素糖化が不十分となることがあり、100mgを超えると、糖化阻害が起こることがある。一方、前記酵素の使用量が前記更に好ましい範囲内であると、酵素の使用量に対して得られる糖の量が多い点で有利である。
前記酵素糖化工程における温度としては特に制限はなく、目的に応じて適宜選択することができるが、10℃~70℃が好ましく、20℃~60℃がより好ましく、30℃~50℃が更に好ましい。前記温度が、10℃より低い温度であると、酵素糖化が十分に進行しないことがあり、70℃を超えると、酵素が失活することがある。一方、前記温度が、前記更に好ましい範囲内であると、酵素の使用量に対して得られる糖の量が多い点で有利である。
前記酵素糖化工程におけるpHとしては特に制限はなく、目的に応じて適宜選択することができるが、例えば、3.0~8.0が好ましく、3.5~7.0がより好ましく、4.0~6.0が更に好ましい。前記pHが、3.0未満、又は8.0を超えると、酵素が失活することがある。一方、前記pHが、前記更に好ましい範囲内であると、酵素の使用量に対して得られる糖の量が多い点で有利である。
前記酵素糖化工程により、単糖としては、前記酵素糖化用原料に含まれるセルロースからはグルコースが得られる。また、前記酵素糖化用原料に含まれるヘミセルロースからはグルコース、ガラクトース、マンノースといった六炭糖及びキシロース、アラビノースといった五炭糖が生成する。
(その他の工程)
上記の酵素糖化工程により得られる単糖を含む糖液は、そのまま後述する発酵工程に供してもよいが、例えば、糖液のpHを調整する工程、糖の濃度を調整する工程などを施すことにより、発酵により適した糖液としてもよい。

本実施形態の糖の製造方法により得られる糖は、後述するエタノールの製造方法に用いるだけでなく、その他の物質、例えば乳酸等の有機酸の製造の原料として用いることもできる。
<第3実施形態:エタノールの製造方法>
本発明の第3の実施形態に係るエタノールの製造方法は、前記第2実施形態に係る糖の製造方法により得られた糖を発酵する発酵工程(エタノール発酵工程)を少なくとも含み、必要に応じて更にその他の工程を含む。

以下、本実施形態に係るエタノールの製造方法について詳述する。
(発酵工程)
本実施形態のエタノールの製造方法に係る発酵工程は、前記本発明の糖の製造方法により得られた糖を含む糖液に、エタノール発酵微生物を添加し、エタノール発酵を行う工程である。

前記エタノール発酵微生物としては特に制限はなく、目的に応じて適宜選択することができるが、酵母、ザイモモナス・モビリス等のザイモモナス属の細菌等が好ましく、酵母がより好ましい。
前記酵母としては特に制限はなく、目的に応じて適宜選択することができるが、サッカロマイセス・セルビシエ等のサッカロマイセス属の酵母が好ましい。但し前述のように、前記植物バイオマスを構成するヘミセルロースからは、酵素糖化によりキシロース、アラビノースといった五炭糖が生成するが、サッカロマイセス属の天然酵母は五炭糖を資化してエタノールを産生する能力をもたない。このため、六炭糖だけでなくヘミセルロース由来の五炭糖も有効に利用してエタノールに変換するためには、五炭糖を資化してエタノールを産生する能力を有する酵母(ペントース資化酵母)を使用することも好ましく行われる。前記ペントース資化酵母としては特に制限はなく、目的に応じて適宜選択することができるが、ピキア・スティピティス、カンジダ・シハタエ等が好ましい。六炭糖及び五炭糖を効率的にエタノールに変換するためには、サッカロマイセス属の酵母と、前述のペントース資化酵母とを組み合わせて使用する方法も好ましく採用される。この場合、サッカロマイセス属の酵母と前述のペントース資化酵母を共存させて発酵を行なってもよいし、まずサッカロマイセス属の酵母により糖液中のグルコースを資化させ、その後前述のペントース資化酵母により五炭糖を資化させてもよい。
前記発酵工程に用いる酵母は、天然の酵母であってもよいし、遺伝子組換え酵母であってもよい。特に、六炭糖と五炭糖の両方の資化能を有する遺伝子組換え酵母を用いることにより、効率的にセルロース及びヘミセルロース由来の六炭糖及び五炭糖の両方をエタノールに変換することができる。
前記発酵工程における前記酵母の使用量、糖以外の添加物、発酵温度、pH、発酵時間等の条件としては特に制限はなく、公知の条件を適宜選択して用いることができるが、pHは4~7、発酵温度は20℃~37℃程度が好ましい。
また、耐熱性の酵母を用いて、通常よりも高い温度で発酵を行なうことで、冷却のための設備を必要とせず、また雑菌の繁殖を抑制して効率的に発酵を行なうこともできる。前記耐熱性の酵母としては例えば、クロイベロマイセス・マルキシアナス等のクロイベロマイセス属に属する耐熱性酵母が挙げられる。これらの耐熱性酵母を使用する場合は、発酵の温度は37℃以上50℃以下程度とすることができる。
前記発酵工程としては、前述の酵素糖化工程と発酵工程とを同時に行う、所謂並行複発酵法を採用してもよい。この並行複発酵法を採用することにより、前記酵素糖化工程と発酵工程とを単一の工程として実施することができ、簡略化された工程によってエタノールを製造することが可能となる。前記並行複発酵としては、前記本実施形態の酵素糖化用原料の製造方法によって得られた酵素糖化用原料に、酵素糖化のための酵素、及び、酵素糖化により生成する糖をそのまま反応系内でエタノール発酵させるための微生物を添加し、酵素糖化及びエタノール発酵を行う。
(その他の工程)
本発明に係るエタノールの製造方法は、前記発酵工程において得られたエタノールを含む培地からエタノールを分離・精製する精製工程を更に備えることが好ましい。前記精製工程により、エタノールは発酵培地中に含まれる種々の物質から分離・精製され、また濃縮される。前記分離・精製の方法としては特に制限はなく、目的に応じて適宜選択することができるが、例えば、まず発酵培地を、菌体等の固形分を遠心分離及び/又はろ過などにより固液分離し、エタノールを含む水溶液を回収し、その後、該水溶液を蒸留、膜分離などの方法によりエタノールを濃縮、精製する方法が好ましい。
本発明に係るエタノールの製造方法は、前記発酵工程において得られたエタノールを含む培地からエタノールを分離・精製する精製工程を更に備えることが好ましい。前記精製工程により、エタノールは発酵培地中に含まれる種々の物質から分離・精製され、また濃縮される。前記分離・精製の方法としては特に制限はなく、目的に応じて適宜選択することができるが、例えば、まず発酵培地を、菌体等の固形分を遠心分離及び/又はろ過などにより固液分離し、エタノールを含む水溶液を回収し、その後、該水溶液を蒸留、膜分離などの方法によりエタノールを濃縮、精製する方法が好ましい。
本実施形態のエタノールの製造方法によれば、前記酵素糖化用原料を用いて得られた糖を用いることで、効率的にエタノールを製造することができる。前記エタノールの製造方法により得られたエタノールは、例えば、燃料用エタノール、工業用エタノールなどとして好適に利用可能である。
以下に本発明の実施例を説明するが、本発明は、これらの実施例に何ら限定されるものではない。
[実施例1]
<酵素糖化用原料の製造>
(植物バイオマス)
リグノセルロースを含有する植物バイオマスとしてエリアンサスを用いた。収穫したエリアンサスを目開き3mmのスクリーンで粒度を制御しながらカッターミルを用いて粉砕し、エリアンサス粒子を得た。該粒子の平均粒子径はレーザー回折法(装置:レーザー回折粒子径測定装置LMS-2000e(セイシン企業社製))で測定したメジアン径(d50)として678μmであった。このエリアンサス粒子を減圧乾燥器にて、40℃、5kPaAの減圧下に一昼夜乾燥した。得られたバイオマスの含水率は、バイオマスの全質量を基準として0.9質量%であった。
(第一工程)
撹拌装置を備えた反応槽(内容積2000Lの圧力容器)に上記のバイオマスを乾燥質量として50kg充填し、反応槽を真空ポンプにて-0.095MPaGまで減圧した後、窒素を0MPaGまで供給した。次いで、反応槽内を40℃まで昇温した後、バイオマスを攪拌した状態にてポンプを用いて液体アンモニアを含むアンモニアを18.3kg供給した。アンモニアの供給後、反応槽を120℃まで昇温し、2.0時間のアンモニア処理を行った。なお、前記処理時の反応槽のアンモニア分圧は1.1MPaAで、温度・圧力条件及びアンモニアの供給量との対比から、前記条件において、5.34kgのアンモニアが液化状態で存在し、その液化状態のアンモニアのバイオマスに対する比率は0.103であった。
(第二工程)
前記第一工程の後、反応槽を20℃程度まで冷却し、10℃における液化状態のアンモニアのバイオマスに対する比率が0.6になるように液体アンモニアを追加供給した後、バイオマスを攪拌した状態にて11.0℃で20分処理した。処理終了後にアンモニアの排気を行った。排気後、反応槽を真空ポンプにて-0.095MPaGまで減圧しバイオマス内に残留しているアンモニアを除去した。アンモニアの除去終了後、反応槽を窒素にて大気圧まで加圧し、反応槽より酵素糖化用原料Aを抜き出した。
<糖の製造(酵素糖化)>
酵素糖化用原料Aを用い、以下の操作により、酵素糖化反応を行った。
内容積50mLの遠心管に、乾燥固形分として500mgとなるように精秤した酵素糖化用原料Aを取り、試料濃度5%(乾燥質量/体積)、酵素としてAccellerase DUET(登録商標)(商品名、ジェネンコア社製)を酵素糖化用原料に対して125%(体積/乾燥質量)、pH4.5(酢酸緩衝液)となるように酵素糖化反応液を調製した。これを50℃の恒温室にて、回転振とう機(200回転/分)を用いて24時間転倒振とうして酵素糖化反応を行った。反応後、遠心分離によって得られた上澄み液中のグルコース濃度を、グルコースCIIテストワコー(商品名、和光純薬社製)を用いて測定し、酵素糖化率の尺度としてグルコース収率を算出した。
なお、グルコース収率は下記式(4)で定義される。
グルコース収率(%)=[酵素糖化反応液中のグルコース量/(酵素糖化原料の乾燥質量×全グルコース化率/100)]×100 (4)
ここで、全グルコース化率(%)は(バイオマス原料を別途化学的に完全に加水分解したときに得られるグルコースの量/バイオマス原料の乾燥質量)×100によって求められるものであり、バイオマス原料基準のグルコースの理論収率に相当する。
以上得られた結果をまとめて表1に示す。
<エタノールの製造>
酵素糖化原料としてバイオマスAを用い、上記の<糖の製造(酵素糖化)>における遠心管を用いた酵素糖化に相当する条件(但し酵素糖化原料固形分濃度15g/100mL)で酵素糖化を行なって糖液を得た。得られた糖液中のグルコース濃度は4.6g/100mL、キシロース濃度は2.6g/100mLであった。
次に、容量50mLの三角フラスコ中にて、前記糖液15mLに、キシロース資化能を付与したサッカロマイセス酵母を光学密度OD600が20となるよう植菌し、30℃にて、140rpmで振とう培養して発酵を行なった。グルコースは発酵開始後4時間後に全て消費され、キシロースは48時間後に全て消費された。エタノール収率は66%であった。
<酵素糖化用原料の製造>
(植物バイオマス)
リグノセルロースを含有する植物バイオマスとしてエリアンサスを用いた。収穫したエリアンサスを目開き3mmのスクリーンで粒度を制御しながらカッターミルを用いて粉砕し、エリアンサス粒子を得た。該粒子の平均粒子径はレーザー回折法(装置:レーザー回折粒子径測定装置LMS-2000e(セイシン企業社製))で測定したメジアン径(d50)として678μmであった。このエリアンサス粒子を減圧乾燥器にて、40℃、5kPaAの減圧下に一昼夜乾燥した。得られたバイオマスの含水率は、バイオマスの全質量を基準として0.9質量%であった。
(第一工程)
撹拌装置を備えた反応槽(内容積2000Lの圧力容器)に上記のバイオマスを乾燥質量として50kg充填し、反応槽を真空ポンプにて-0.095MPaGまで減圧した後、窒素を0MPaGまで供給した。次いで、反応槽内を40℃まで昇温した後、バイオマスを攪拌した状態にてポンプを用いて液体アンモニアを含むアンモニアを18.3kg供給した。アンモニアの供給後、反応槽を120℃まで昇温し、2.0時間のアンモニア処理を行った。なお、前記処理時の反応槽のアンモニア分圧は1.1MPaAで、温度・圧力条件及びアンモニアの供給量との対比から、前記条件において、5.34kgのアンモニアが液化状態で存在し、その液化状態のアンモニアのバイオマスに対する比率は0.103であった。
(第二工程)
前記第一工程の後、反応槽を20℃程度まで冷却し、10℃における液化状態のアンモニアのバイオマスに対する比率が0.6になるように液体アンモニアを追加供給した後、バイオマスを攪拌した状態にて11.0℃で20分処理した。処理終了後にアンモニアの排気を行った。排気後、反応槽を真空ポンプにて-0.095MPaGまで減圧しバイオマス内に残留しているアンモニアを除去した。アンモニアの除去終了後、反応槽を窒素にて大気圧まで加圧し、反応槽より酵素糖化用原料Aを抜き出した。
<糖の製造(酵素糖化)>
酵素糖化用原料Aを用い、以下の操作により、酵素糖化反応を行った。
内容積50mLの遠心管に、乾燥固形分として500mgとなるように精秤した酵素糖化用原料Aを取り、試料濃度5%(乾燥質量/体積)、酵素としてAccellerase DUET(登録商標)(商品名、ジェネンコア社製)を酵素糖化用原料に対して125%(体積/乾燥質量)、pH4.5(酢酸緩衝液)となるように酵素糖化反応液を調製した。これを50℃の恒温室にて、回転振とう機(200回転/分)を用いて24時間転倒振とうして酵素糖化反応を行った。反応後、遠心分離によって得られた上澄み液中のグルコース濃度を、グルコースCIIテストワコー(商品名、和光純薬社製)を用いて測定し、酵素糖化率の尺度としてグルコース収率を算出した。
なお、グルコース収率は下記式(4)で定義される。
グルコース収率(%)=[酵素糖化反応液中のグルコース量/(酵素糖化原料の乾燥質量×全グルコース化率/100)]×100 (4)
ここで、全グルコース化率(%)は(バイオマス原料を別途化学的に完全に加水分解したときに得られるグルコースの量/バイオマス原料の乾燥質量)×100によって求められるものであり、バイオマス原料基準のグルコースの理論収率に相当する。
以上得られた結果をまとめて表1に示す。
<エタノールの製造>
酵素糖化原料としてバイオマスAを用い、上記の<糖の製造(酵素糖化)>における遠心管を用いた酵素糖化に相当する条件(但し酵素糖化原料固形分濃度15g/100mL)で酵素糖化を行なって糖液を得た。得られた糖液中のグルコース濃度は4.6g/100mL、キシロース濃度は2.6g/100mLであった。
次に、容量50mLの三角フラスコ中にて、前記糖液15mLに、キシロース資化能を付与したサッカロマイセス酵母を光学密度OD600が20となるよう植菌し、30℃にて、140rpmで振とう培養して発酵を行なった。グルコースは発酵開始後4時間後に全て消費され、キシロースは48時間後に全て消費された。エタノール収率は66%であった。

酵素糖化用原料Aに代えて酵素糖化用原料Bを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表1に示す。

第一工程におけるアンモニア供給時の反応槽温度を60℃にしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Bを得た。なお、120℃における反応槽のアンモニア分圧は1.3MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.096であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Bを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表1に示す。
[実施例3]

第一工程におけるアンモニア供給時の反応槽温度を80℃にしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Cを得た。なお、120℃における反応槽のアンモニア分圧は1.3MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.092であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Cを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表1に示す。

<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Dを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表1に示す。

酵素糖化用原料Aに代えて酵素糖化用原料Eを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。

<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Fを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。

第一工程において、反応槽にバイオマスを乾燥質量として75kg充填したこと、アンモニア供給時の反応槽温度を80℃にしたこととアンモニアの供給量を16.4kgにしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Gを得た。なお、120℃における反応槽のアンモニア分圧は1.1MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.061であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Gを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表1に示す。
<酵素糖化用原料の製造>
第一工程において、反応槽にバイオマスを乾燥質量として75kg充填したこと、アンモニア供給時の反応槽温度を80℃にしたこととアンモニアの供給量を16.4kgにしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Gを得た。なお、120℃における反応槽のアンモニア分圧は1.1MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.061であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Gを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表1に示す。

第一工程におけるアンモニア供給時の反応槽温度を120℃にしたこととアンモニアの供給量を13.0kgにしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Hを得た。なお、120℃における反応槽のアンモニア分圧は1.2MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.014であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Hを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表2に示す。

第一工程における液体アンモニア供給時の反応槽温度を110℃にしたこととアンモニアの供給量を13.0kgにしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Iを得た。なお、120℃における反応槽のアンモニア分圧は1.2MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.015であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Iを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表2に示す。

第一工程における液体アンモニア供給時の反応槽温度を110℃にしたこととアンモニアの供給量を13.0kgにしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Iを得た。なお、120℃における反応槽のアンモニア分圧は1.2MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.015であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Iを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表2に示す。
[比較例3]

第一工程におけるアンモニア供給時の反応槽温度を110℃にしたこととアンモニアの供給量を13.9kgにしたこと以外は実施例1と同様にして、第一及び第二工程を実施し、酵素糖化用原料Jを得た。なお、120℃における反応槽のアンモニア分圧は1.2MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.034であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Jを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表2に示す。

[比較例5]
<酵素糖化用原料の製造>
第一工程におけるアンモニア供給時の反応槽温度を120℃にしたこととアンモニアの供給量を13.0kgにしたことと120℃の気体状態でアンモニアを供給したこと以外は実施例1と同様にして、第一及び第二工程及を実施し、酵素糖化用原料Lを得た。なお、120℃における反応槽のアンモニア分圧は1.2MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.014であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Lを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表2に示す。
<酵素糖化用原料の製造>
第一工程におけるアンモニア供給時の反応槽温度を120℃にしたこととアンモニアの供給量を13.0kgにしたことと120℃の気体状態でアンモニアを供給したこと以外は実施例1と同様にして、第一及び第二工程及を実施し、酵素糖化用原料Lを得た。なお、120℃における反応槽のアンモニア分圧は1.2MPaAで、液化状態のアンモニアのバイオマスに対する比率は0.014であった。
<糖の製造(酵素糖化)>
酵素糖化用原料Aに代えて酵素糖化用原料Lを用いたこと以外は実施例1と同様にして、酵素糖化反応を行い、グルコース収率を算出した。
以上得られた結果をまとめて表2に示す。
表1に示す通り、実施例1~7によれば、アンモニアの使用量を低減しつつ、酵素糖化効率に優れる酵素糖化用原料を製造することが可能であり、また、得られた酵素糖化用原料を用いて酵素糖化を行うことにより、効率的に糖を製造することが可能であり、更に得られた糖を用いて発酵を行うことにより、効率的にエタノールを製造することができることが明らかとなった。
Claims (4)
- リグノセルロースを含有する植物バイオマスをアンモニアにより処理して酵素糖化用原料を得る酵素糖化用原料の製造方法であって、
リグノセルロースを含有する植物バイオマスが収容された反応槽に、反応槽温度-40℃~80℃の状態で液体アンモニアまたは液体アンモニアを含むアンモニアを供給し、次いで、60℃~135℃の範囲内でありかつ供給時の反応槽温度よりも高い温度である処理温度まで反応槽温度を昇温し、該処理温度下で気相及び液相を含むアンモニアによる前記植物バイオマスの処理を行い、リグノセルロースを構成するリグニンとヘミセルロースとの間のエステル結合の少なくとも一部を開裂させる第一工程と、
前記第一工程後、反応槽温度-40℃~40℃まで前記反応槽内を冷却し、液相のアンモニアの質量の前記植物バイオマスの乾燥質量に対する比(液相のアンモニアの質量/植物バイオマスの乾燥質量)が0.1~1.1となるように前記反応槽内に液体アンモニアまたは液体アンモニアを含むアンモニアを追加し、-40℃~40℃で液相を含むアンモニアによる前記植物バイオマスの処理を行い、リグノセルロースを構成するセルロースの少なくとも一部の結晶型をI型からIII型に転移させて酵素糖化用原料を得る第二工程と、
を備える酵素糖化用原料の製造方法。 - 前記第一工程において、前記処理温度下で前記反応槽内に存在する液相のアンモニアの質量の前記植物バイオマスの乾燥質量に対する比が0.05~0.25となるように、前記反応槽に液体アンモニアまたは液体アンモニアを含むアンモニアを供給する、請求項1に記載の酵素糖化用原料の製造方法。
- 請求項1又は2に記載の酵素糖化用原料の製造方法によって得られた酵素糖化用原料を酵素により糖化する工程を備える、糖の製造方法。
- 請求項3に記載の糖の製造方法によって得られた糖を発酵させる工程を備える、エタノールの製造方法。
