Abstract

본 발명은 PAUF를 포함하는 혈관신생 촉진용 조성물에 관한 것으로, 보다 구체적으로는 PAUF를 이용한 만성궤양, 지연성 상처치유 및/또는 심혈관계 질환의 치료용 약학적 조성물 등에 관한 것이다. 본 발명의 피에이유에프 단백질은 혈관내피세포를 자극하여 증식, 이동 및 분화를 촉진시킴으로서 혈관신생과정을 유도할 수 있는 효과를 보인다. 즉, 혈관신생촉진인자로서의 신규한 기능을 갖는 PAUF 단백질은 혈관 신생이 요구되는 다양한 질환의 치료제 개발에 유용하게 이용될 것으로 기대된다.

Description

피에이유에프를 포함하는 혈관신생 촉진용 조성물
본 발명은 PAUF를 포함하는 혈관신생 촉진용 조성물에 관한 것으로, 보다 구체적으로는 PAUF를 이용한 혈관신생이 요구되는 질환의 치료용 약학적 조성물 등에 관한 것이다.
신생혈관형성은 기존에 존재하는 미세혈관에서 새로운 혈관이 생성되는 일련의 과정을 말하며, 배아 발생, 상처의 치유 및 배란 등에는 필수적인 과정으로서 그 이외 상황에는 일어나지 않도록 체내에서 정교하게 조절되고 있다.
그러나 많은 병적 질환이 자율적으로 조절되지 않는 신생혈관형성에 의해 일어난다. 신생혈관이 관절을 침범하고 연골을 파괴시키는 류머티스 관절염, 망막 내 혈관의 형성이 수정체를 침범하여 시력을 상실하게 하는 당뇨병성 망막증, 그리고 신생 혈관의 생성이 종양의 증식과 종양 세포의 순환 및 다른 장기로의 전이를 용이하게 하는 등은 과도한 신생혈관형성이 병증을 악화시키는 주요한 원인이 되는 대표적인 사례다 (Folkman J., Nat. Med 1:27-31 , 1995).
한편, 발생 및 상처 치유, 기관 형성에 중요한 과정인 혈관 생성이 제대로 일어나지 않을 경우 역시 심각한 질환이 발생하게 된다. 예를 들어, 발생단계에서 혈관 형성이 되지 않을 경우 태반의 미발달을 초래하여 유산의 원인이 될 수 있으며, 조직의 궤양 및 허혈의 발생이 기관의 기능 이상을 유발시키고 더 나아가서는 사망에 이르게 한다. 최근 식생활의 변화, 영양 상태의 개선과 노년 인구의 증가에 따라 동맥경화, 심근경색 및 협심증, 뇌경색, 급성사지허혈과 같은 다양한 심혈관계 질환이 성인 사망률의 수위를 차지하는 심각한 질환으로 대두되고 있으나 뚜렷한 치료법이 정립되지 않은 상태이며, 이러한 허혈성 질환을 치료하기 위해 신생혈관형성를 유도하는 방법 또는 촉진하는 새로운 치료법 및 인자의 개발이 주목을 받고 있다 (김덕경 et al, 대한내분비학회, 16(3), 328-338, 2001). 국내외 제약회사들은 단백 요법, 유전자 요법 및 줄기세포를 이용한 신생혈관형성 유도하는 치료법을 개발하여 제시하고 있으며, 생체 내 단백질을 이용한 단백질 치료법의 경우 안정성과 효능의 측면에서 치료약물로의 개발 가능성이 높은 것으로 평가되고 있다.
신생혈관형성 과정은 혈관내피세포를 자극하는 혈관신생촉진인자의 작용에 의해 개시되며, 혈관 기저막의 분해, 혈관 내피세포의 이동, 증식 및 분화를 통해 혈관이 재구성되는 과정을 통해 새로운 모세혈관이 형성된다. 이러한 혈관생성에는 혈관 내피세포를 자극하는 여러 가지 성장 인자, 사이토카인, 지질대사물질들이 관여하고 있다. 주요한 혈관신생촉진인자로는 혈관내피성장인자(VEGF), 섬유아세포성장인자(FGF), 안지오포이에틴, 혈소판유래성장인자(PDGF) 등이 있으며, 이들 인자들을 신생혈관형성이 요구되는 다양한 질환에 적용하고자 하는 시도가 활발히 이루어지고 있다. 특히 심각한 허혈성 질환을 치료하기 위해 혈관신생촉진인자들을 투여하여 새로운 혈관의 생성을 유도함으로서 혈류의 증가를 통해 허혈 증상의 개선을 촉진시키는 방법이 시행되고 있다 (Timothy D. Henry et al., Circulation., 103, 1359-1365, 2003; Robert J Lederman et al., The Lancet., 15, 2053-2058, 2002). 이에 혈관성 조직 복구에 의해 증상이 개선될 수 있는 질환의 치료를 위해 보다 효과적이고 새로운 인자들의 발굴이 지속적으로 요구되고 있는 실정이다.
한편, 피에이유에프는 비 암화된 췌장조직에 비해 암화된 췌장조직에서 차별적으로 높게 발현되는 단백질로서 췌장암 세포의 성장, 이동 및 침윤을 유도하는 인자로 보고되고 있다 (Lee et al., Oncogene, 29(1), 56-67, 2010). 그러나 지금까지 피에이유에프 단백질의 혈관신생 촉진 효과에 대한 기능적 내용이 제안되거나 보고된 바가 없었다.
본 발명자들은 다양한 실험실적 및 생체 내 조건을 통하여 조사한 결과 피에이유에프 단백질은 혈관내피세포를 자극하여 혈관신생에서 요구되는 일련의 과정들을 활성화시킴으로서 새로운 혈관의 형성을 유도할 수 있다는 점을 발견하게 되었다. 이에 본 발명자들은 피에이유에프 단백질의 새로운 기능인 혈관신생촉진에 대한 효과를 밝히고 이를 이용하여 혈관신생이 요구되는 질환에 적용할 수 있는 치료용 조성물을 개발할 가능성을 확인함으로써 본 발명을 완성하게 되었다.
따라서, 본 발명은 신생혈관형성 작용을 촉진하여 혈관 신생이 요구되는 각종 질환의 치료용 조성물 및 이를 이용하여 혈관 신생이 요구되는 질환을 치료하는 방법 등을 제공하고자 한다.
구체적으로, 본 발명은 피에이유에프를 유효성분으로 포함하는 혈관신생 촉진용 조성물을 제공하고자 한다.
또한 본 발명은 피에이유에프를 유효성분으로 포함하는 혈관신생이 요구되는 질환의 치료용 약학적 조성물을 제공하고자 한다.
또한 본 발명은 약제학적으로 유효한 양의 피에이유에프 단백질을 유효성분으로 포함하는 약학적 조성물을 개체에 투여하는 단계를 포함하는, 혈관신생이 요구되는 질환의 치료 방법을 제공하고자 한다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
본 발명은 피에이유에프를 유효성분으로 포함하는 혈관신생 촉진용 조성물을 제공한다. 본 발명의 일 구현예로, 상기 혈관신생은 발생, 상처 치유 또는 기관 형성 과정에서 일어나는 것을 포함한다.
본 발명은 피에이유에프를 유효성분으로 포함하는 혈관신생이 요구되는 질환의 치료용 약학적 조성물을 제공한다. 본 발명의 일 구현예로, 상기 혈관신생이 요구되는 질환은 만성 궤양, 지연성 상처치유 또는 심혈관계 질환을 포함하며, 상기 심혈관계 질환은 동맥경화, 심근경색, 협심증, 뇌경색 또는 급성사지허혈을 포함할 수 있으나, 이에 한정되지 않는다. 본 발명의 다른 구현예로, 상기 피에이유에프 단백질은 혈관내피세포의 증식, 이동, 또는 분화를 증가시키는 것을 특징으로 한다. 본 발명의 또 다른 구현예로 상기 피에이유에프 단백질은 세포 내 신호전달인자의 활성을 조절함으로써 혈관신생을 촉진하는 것이다.
또한 본 발명은 약제학적으로 유효한 양의 피에이유에프 단백질을 유효성분으로 포함하는 약학적 조성물을 개체에 투여하는 단계를 포함하는 혈관신생이 요구되는 질환의 치료 방법을 제공한다.
본 발명에서 혈관신생이 요구되는 질환은 상술한 바와 같다.
본 발명의 피에이유에프 단백질은 혈관내피세포를 자극하여 증식, 이동 및 분화를 촉진시킴으로서 혈관신생과정을 유도할 수 있는 효과를 보인다.

피에이유에프 단백질과 관련된 기존 연구에 따르면, PAUF는 췌장암 세포의 성장, 이동 및 침윤을 유도하는 인자로 보고되고 있다. 그러나 본 발명자들은 PAUF 단백질이 실험실적 조건뿐만 아니라 생체 내 조건에서 혈관신생촉진 효과가 있음을 새로이 밝히고, 이에 기초하여 본 발명을 완성하게 되었다.
이에, 본 발명은 피에이유에프를 유효성분으로 포함하는 혈관신생 촉진용 조성물을 제공한다. 본 발명의 일 구현예로, 상기 혈관신생은 발생, 상처 치유 또는 기관 형성 과정에서 일어나는 것을 포함한다.
또한, 본 발명은 피에이유에프를 유효성분으로 포함하는 혈관신생이 요구되는 질환의 치료용 약학적 조성물을 제공한다.
상기 본 발명의 조성물은 PAUF 외에도, 기존 치료 활성 성분, 기타 보조제, 약제학적으로 허용가능한 담체 등의 성분을 추가로 포함할 수 있다. 상기 약제학적으로 허용가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 및 에탄올 등을 포함한다.
상기 본 발명의 혈관신생이 요구되는 질환은 만성 궤양, 지연성 상처치유 및 심혈관계 질환을 포함하며, 상기 심혈관계 질환은 동맥경화, 심근경색, 협심증, 뇌경색 및 급성사지허혈 등을 포함한다.
본 발명의 조성물은 혈관내피세포의 증식, 이동, 또는 분화를 증가시킴으로써 부작용 없이 혈관 신생을 촉진하고 질환을 치료하는 활성을 나타낸다. 본 발명의 조성물은 세포 내 신호전달인자의 활성을 조절함으로써 혈관 신생을 촉진할 수 있다. 특히, 본 발명의 조성물은 신생혈관형성 작용이 일어나지 않아 생기는 각종 혈관신생 질환의 치료 뿐만 아니라, 발생, 상처 치유 및 기관 형성에 있어서 효과적으로 작용할 수 있다.
이에, 본 발명은 약제학적으로 유효한 양의 피에이유에프 단백질을 유효성분으로 포함하는 약학적 조성물을 개체에 투여하는 단계를 포함하는 혈관신생이 요구되는 질환의 치료 방법을 제공한다.
본 발명에서 “개체”란 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는 인간 또는 비-인간인 영장류, 생쥐, 쥐, 개, 고양이, 말 및 소 등의 포유류를 의미한다. 또한 본 발명에서 “약제학적 유효량”은 투여되는 질환 종류 및 중증도, 환자의 연령 및 성별, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정되며 상기 요소를 모두 고려하여 부작용 없이 최대 효과를 얻을 수 있는 양으로, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 조성물은 목적 조직에 도달할 수 있는 한 투여방법에는 제한이 없다. 예를 들면, 경구 투여, 동맥 주사, 정맥 주사, 경피 주사, 비강 내 투여, 경기관지 투여 또는 근육 내 투여 등이 포함된다. 일일 투여량은 약 0.0001 내지 100mg/kg이고, 바람직하게는 0.001 내지 10mg/kg이며, 하루 일회 내지 수회 나누어 투여하는 것이 바람직하다.
본 발명의 일실시예에서는 PAUF 단백질이 혈관내피세포에 미치는 영향을 살펴보기 위해, M199배지에서 배양한 혈관내피세포에 PAUF 단백질과 항-PAUF 항체를 넣고 세포 증식, 세포 이동 효과, 세포 분화 및 세포 투과성을 측정하였다. 그 결과 PAUF는 농도의존적으로 혈관내피세포의 증식, 이동, 분화 및 투과성을 증가시킨다는 사실을 확인하였다(도 1 내지 도 4 참조).
본 발명의 또 다른 실시예에서는 생체 내에서 PAUF의 혈관 생성 촉진 효과를 관찰하기 위하여, 7주령의 C57BL/6 생쥐를 이용한 마트리젤 플러그 어세이를 수행한 결과, PAUF 단백질을 처리한 군에서 농도 의존적으로 신생혈관 생성이 증가되었음을 확인할 수 있었다(도 9 참조).
본 발명의 또 다른 실시예에서는 PAUF의 혈관신생효과를 실험동물 모델을 통해 확인하기 위하여, PAUF 과발현 및 발현 저해된 췌장암 세포주를 6주령의 누드 마우스 Balb/c에 이종 이식 또는 동소 이식한 동물 모델을 구축하고, 3주 후 혈관내피세포 표지자인 CD31을 면역조직화학법을 통하여 정량하였다. 그 결과, PAUF 발현의 증가와 감소에 따라 종양 내 혈관 생성의 증가와 감소가 나타난다는 것을 확인할 수 있었다(도 16 참조).
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1. PAUF 단백질의 혈관내피세포 성장 효과 : 세포 성장 어세이
혈관내피세포를 1 ㎖의 M199배지 내에 5 × 104 cells 씩 계수하여 24 세포 배양 웰에 각각 분주하여 1일 동안 배양하고, 무혈청 M199배지로 교체하였다. 6시간 후에 각 웰에 VEGF 0.05㎍/㎖와 PAUF 단백질 0, 0.1, 0.2, 0.5㎍/㎖을 첨가한 후 37℃ CO2 배양기에서 배양하였다. 배양 시작 48시간 후에 세포를 회수하여 트리판 블루 용액에 염색한 후 살아있는 세포의 수를 계수하여 세포의 증식을 측정한 결과를 도 1a에 나타내었다.
도 1a에 나타난 바와 같이, PAUF 단백질의 첨가량이 증가할수록 세포의 수가 증가되었으며, PAUF는 농도의존적으로 혈관내피세포의 증식을 유도하였다.
상기 결과가 PAUF 의존적 현상임을 확인하기 위해 PAUF에 대한 항체인 폴리클로날 항-PAUF 항체를 사용하여 동일한 어세이를 수행하였다. 구체적으로, 혈관내피세포를 24 세포 배양 웰에 웰 당 5 × 104 cells로 분주하여 1일 동안 배양하고, 무혈청 M199배지로 교체하였다. 6시간 후에 각각의 웰에 PAUF 단백질 0.5㎍/㎖와 항-PAUF 항체 300nM 또는 노말 토끼 항체를 함께 처리하여 37℃ CO2 배양기에서 배양하였다. 배양 시작 48시간 후에 세포를 회수하여 트리판 블루 용액에 염색 후 살아있는 세포 수를 계수하여 측정한 결과를 도 1b에 나타내었다.
도 1b에 나타난 바와 같이, PAUF 단백질에 대한 폴리클로날 항-PAUF를 처리함으로써 혈관내피세포증식이 저해되었음을 알 수 있었다.
실시예 2. PAUF의 혈관내피세포 이동 효과 : 세포 이동 어세이(migration assay)
혈관내피세포의 화학주성 이동성에 대한 PAUF의 영향을 알아보기 위해 트렌스웰(Corning Costar, 미국)을 사용하여 검정하였다.
구체적으로, 혈관내피세포를 분리하여 PBS(phosphate buffered saline)로 2회 씻어 준 후 1% 우태아 혈청(FBS) M199배지에서 1 × 106 cells/㎖의 농도로 최종 현탁하여 준비하고, 트랜스웰의 필터 하부 표면은 10㎍ 젤라틴으로 코팅 하였다. PAUF 단백질 0, 0.05, 0.1, 0.5㎍/㎖를 각각 VEGF 0.05㎍/㎖ 및 1% 우태아 혈청 M199배지와 혼합하여 600㎕씩 트랜스웰의 하부 챔버에 넣고, 그 위에는 트렌스웰을 올려놓은 후, 상기 준비한 혈관내피세포 100㎕ (1 × 105 cells)를 취하여 넣은 후, 이를 37℃ CO2 배양기에서 4시간 동안 배양하였다. 배양이 끝나면 트렌스웰의 필터 하부로 이동한 세포를 100% 메탄올로 고정, 헤마토실린으로 염색하고 필터의 상부에 남아 있는 세포는 면봉을 이용하여 제거하였다. 필터의 하부로 이동하여 염색된 세포의 사진은 최소 5개의 선택 면적에서 계수하여 이동성을 측정한 결과를 도 2a에 나타내었다.
도 2a에 나타난 바와 같이, PAUF는 농도의존적으로 혈관내피세포의 이동을 유도하였다.
혈관내피세포 이동능의 변화가 PAUF에 의존적이라는 것을 증명하기 위해 폴리클로날 항-PAUF 항체를 사용하여 상기와 동일한 실험을 수행하였다. 구체적으로, 혈관내피세포는 1% 우태아 혈청 M199배지에서 1 × 106 cells/㎖의 농도로 최종 현탁하여 준비하고, 10㎍ 젤라틴으로 하부 코팅된 트렌스웰에 혈관내피세포를 100㎕ (1 × 105 cells) 취하여 넣었다. 하부 챔버에는 PAUF 단백질 0.5㎍/㎖와 항-PAFUF 항체(300nM) 또는 노말 토끼 항체를 1% 우태아 혈청 M199배지와 혼합하여 넣어주었다. 트렌스웰을 하부 챔버 위에 올려놓은 후 37℃ CO2 배양기에서 4시간 동안 배양하였다. 배양 후 트렌스웰의 필터 하부로 이동한 세포를 100% 메탄올로 고정시키고 헤마토실린으로 염색하였다. 필터 위해 남아있는 이동하지 못한 세포는 면봉으로 제거하고 염색된 세포는 사진으로 찍어 최소 5개의 선택 면적에서 계수하여 이동성을 측정하한 결과를 도 2b에 나타내었다.
도 2b에 나타난 바와 같이, PAUF에 대한 항체를 처리함으로서 세포 이동성이 저해되었음을 확인할 수 있었다.
실시예 3. PAUF의 혈관내피세포 분화에 미치는 영향 : 관 형성 어세이
PAUF 단백질이 혈관내피세포의 분화를 유도하여 관 형성에 영향을 미칠 수 있는지 확인하기 위해 성장 인자 감소 마트리젤(10㎎/㎖)을 이용한 관 형성 어세이를 수행하였다.
혈관내피세포는 1% 우태아 혈청이 포함된 M199배지에서 12시간 동안 배양한 후 트립신을 처리하여 모으고 1% 우태아 혈청이 함유된 M199배지에 재현탁 하였다. 성장 인자 감소 마트리젤을 24 세포 배양 웰에 250㎕ 씩 각각 분주하고 37℃ CO2 배양기에서 중합화 하였다. 혈관내피세포는 중합화 된 마트리젤이 있는 웰에 웰당 1 × 105 cells의 농도로 넣고, VEGF 0.05㎍/㎖와 PAUF 단백질 0, 0.05, 0.1, 0.5㎍/㎖를 첨가하였다. 24시간 동안 37℃ CO2 배양기에서 배양한 후 배양물을 사진으로 찍고(× 40), Image J (National Institute of Health)공개 프로그램을 이용하여 형성된 튜브의 길이를 측정하여 그 결과를 도 3a에 나타내었다.
한편, PAUF 의존적 관 형성 효과임을 확인하기 위해 폴리클로날 항-PAUF 항체를 사용하여 동일한 어세이를 수행하였다. 구체적으로, 1% 우태아 혈청 M199배지에서 12시간 동안 배양된 혈관내피세포를 모아 동일한 배지로 재현탁 하였다. 250 ㎕의 성장 인자 감소 마트리젤을 24 세포 배양 웰에서 각각 중합화하고, 준비된 혈관내피세포는 웰당 1 × 105 cells의 농도로 분주하였다. 여기에 PAUF 단백질 0.5㎍/㎖와 항-PAFUF 항체 (300nM) 또는 노말 토끼 항체를 함께 넣어주고, 24시간 동안 37℃ CO2 배양기에서 배양하였다. 24시간 후 배양물을 사진으로 찍고 (× 40), Image J (National Institute of Health)공개 프로그램을 이용하여 형성된 튜브의 길이를 측정하여 그 결과를 도 3b에 나타내었다.
그 결과 도 3a에 의하면, PAUF는 농도의존적으로 혈관내피세포의 분화를 유도함으로서 관 형성을 증가시켰으며, 도 3b에 나타난 바와 같이, PAUF에 대한 항체를 처리하였을 때 이러한 현상이 저해됨을 확인할 수 있었다.
실시예 4. PAUF에 의해 유도된 혈관내피세포 투과성 효과 분석 어세이
PAUF가 혈관내피세포의 투과성 증가에 미치는 효과를 분석하기 위해 FITC(fluorescein isothiocyanate)-덱스트란을 이용한 투과성 어세이를 수행하였다.
구체적으로, 혈관내피세포는 트랜스웰의 상위 챔버에 놓고, 챔버의 필터가 완전히 세포로 덮일 때까지 배양한 후, 각각 0, 0.1, 0.2, 0.5㎍/㎖ 농도의 PAUF 단백질을 2시간 동안 처리한 다음, FITC-덱스트란을 상위 챔버에 첨가하였다. 30분이 지난 후 하부 트렌스웰로 확산된 FITC-덱스트란의 양을 형광계측기를 이용하여 측정한 결과를 도 4에 나타내었다.
도 4에 의하면, PAUF를 처리한 경우 농도의존적으로 혈관내피세포의 투과성이 증가되고 있음을 알 수 있다.
실시예 5. PAUF에 의해 유도된 VE-캐드헤린의 발현 양상 분석
혈관내피세포의 투과성 변화가 내피세포의 간극에 존재하는 VE-캐드헤린(vascular endothelial-cadherin)의 발현양상의 변화에 의한 것인지를 확인하기 위해 면역세포화학염색법을 시행하였다.
구체적으로, 혈관내피세포는 젤라틴으로 코팅된 공초점 현미경 관찰용 배양 접시에서 24시간 동안 배양한 후, 무혈청 M199배지로 세척하고, 동일 배지에서 6시간동안 배양하였다. 실험군으로 0.5㎍/㎖ 농도의 PAUF 단백질을 처리한 것, PAUF와 이에 대한 항체를 함께 처리한 것으로 하였으며, VEGF 0.05㎍/㎖을 처리한 군을 양성 대조군으로 설정하였다. 이들을 각각 2시간 동안 처리한 후 세포를 3.7% 포르말린으로 고정시켰다. 이를 PBS로 세척하고 0.1% Triton X-100과 3% BSA/PBS 용액으로 블로킹한 후 VE-캐드헤린에 대한 항체를 2시간 동안 처리하였다. 다시 FITC가 부착된 2차 항체를 1시간 동안 처리하고 DAPI(4',6-diamidino-2-phenylindole)로 핵을 염색하였다. 이 시료를 공초점 형광현미경으로 관찰한 결과를 도 5에 나타내었다.
도 5에 나타난 바와 같이, PAUF를 처리하였을 때 내피세포의 간극에 존재하는 VE-캐드헤린의 세포 접착기능이 감소하였으며, 이는 PAUF에 대한 항체를 처리하였을 때 저해됨을 확인할 수 있다.
실시예 6. PAUF에 의한 VE-캐드헤린과 Src의 인산화 조절 분석
VE-캐드헤린의 세포 접착성 기능의 저하로 인한 세포 투과성 증가의 기작은 VE-캐드헤린과 Src의 인산화 유도에 의존한다는 것이 알려져 있다. (Esser S. et al., Journal of Cell Sci., 1998, 111, 1853-1865) 이를 확인하기 위해 PAUF에 의한 VE-캐드헤린과 Src의 인산화가 조절되는지를 분석하였다.
구체적으로, 혈관내피세포는 젤라틴으로 코팅된 60mm 플레이트에 배양한 후 PBS로 세척하고, 다시 6시간 동안 무혈청 M199배지로 배양하였다. 이에 PAUF 단백질을 0, 0.01, 0.5㎍/㎖의 농도 구배로 처리하고, PAUF 단백질과 이에 대한 각각의 항체를 함께 처리하였다. 이러한 처리를 한 세포들을 모은 후 정량하고 포스포티로신 항체로 면역침강법을 시행하였다. 그 다음 VE-캐드헤린 및 Src에 대한 항체를 사용하여 웨스턴 블롯을 시행하여 인산화된 VE-캐드헤린 및 Src 단백질 수준의 양을 확인한 결과를 도 6a에 나타내었다.
도 6a에 나타난 바와 같이, PAUF를 처리하였을 때, VE-캐드헤린과 Src의 인산화가 이의 농도에 따라 증가하고 있으며, 도 6b에 나타난 바와 같이 PAUF의 항체에 의해 이러한 현상이 저해되었다.
실시예 7. 생체 내 조건에서 PAUF의 혈관 투과성 증가 효과
PAUF에 의한 혈관 투과성 증가를 생체 내 조건에서 확인하기 위해 에반스 블루 염료를 이용한 마일즈 어세이를 실시하였다.
구체적으로, 7주령의 C57BL/6 생쥐의 꼬리 정맥으로 에반스 블루를 주사하여 정맥을 통해 염료가 퍼지도록 하였다. 피부 내로 0, 1, 5㎍/㎖ 농도의 PAUF 단백질을 주입하고 30분간 방치한 후, 피부 표피를 벗겨내어 혈관의 투과성 변화에 의해 에반스 블루 염료가 누출되었는지를 확인하였으며, 그 결과를 도 7에 나타내었다.
도 7에 나타난 바와 같이, PAUF를 처리한 군에서 농도 의존적으로 혈관의 투과성이 증가되었다.
실시예 8. 생쥐 대동맥 환을 이용한 미세혈관 형성 어세이
신생혈관형성에 대한 PAUF의 영향을 생체 외 분석 방법인 생쥐의 대동맥 환을 이용한 미세혈관 형성 어세이를 실시하였다.
구체적으로, C57BL/6 생쥐 (7주령 내지 8주령)에서 동맥을 적출하여 1 mm 두께로 동맥 환 조직을 만든다. 마트리젤(10㎎/㎖) 150㎕로 코팅된 48 웰 플레이트에 준비된 동맥 환 조직을 올려놓고, 다시 그 위에 마트리젤을 올려 코팅함으로써 고정시킨다. 그 다음으로 0, 1, 5㎍/㎖ 농도의 PAUF 단백질을 혈청이 제거된 배지 (human endothelial serum-free medium)와 함께 혼합하여 최종 200㎕를 각 웰에 넣어 주고, 37℃ CO2 배양기에서 배양하였다. 5일 후 각 동맥 환 조직으로부터 발아 된 정도를 광학 현미경으로 관찰하여 사진으로 찍고 (× 40), 그 발아정도는 이중맹검법을 통하여 정량화하였으며, 그 결과를 도 8에 나타내었다.
도 8에 의하면, PAUF 단백질을 처리한 경우 농도 의존적으로 생쥐의 대동맥 환으로부터 미세혈관의 발아가 증가함을 확인할 수 있다.
실시예 9. 생쥐를 이용한 마트리젤 플러그 어세이
PAUF에 의한 혈관 생성 촉진을 생체 내 조건에서 확인하기 위해 생쥐를 이용한 마트리젤 플러그 어세이를 수행하였다.
구체적으로, 각각 0, 1, 5㎍/㎖ 농도의 PAUF 단백질과 헤파린(10units/㎖)을 포함하는 600㎕의 마트리젤을 7주령의 C57BL/6 생쥐에 피하주사를 통하여 주입하였다. 7일이 경과된 후, 생쥐의 피부에서 마트리젤 플러그를 적출하여 사진을 찍고 그 결과를 도 9에 나타내었다.
도 9에 의하면, PAUF 단백질이 처리되지 않은 PBS군에서는 신생혈관의 생성이 약한 반면, PAUF 단백질이 처리된 군에서는 농도 의존적으로 신생혈관의 생성이 증가되고 있음을 관찰할 수 있다.
또한 마트리젤 플러그 내에 존재하는 내피세포의 양을 측정하기 위해 이를 3.7%(w/v)의 포르말린에 고정시키고 15%와 30% 수크로스에 담그고 하루 동안 보관한 후, 꺼내어 OCT(optimal cutting temperature) 용액으로 냉동절편용 표본을 만들었다. 다음으로, 이를 5㎛ 두께로 절편하여 혈관내피세포의 마커인 CD31을 항체를 이용하여 염색한 후 공초점 형광현미경을 통해 관찰하였고, CD31에 대해 양성으로 염색된 세포의 수를 측정함으로서 정량화한 결과를 도 10에 나타내었다.
도 10에 의하면, PAUF 단백질이 농도별로 처리된 경우 CD31을 발현하는 내피세포가 처리되지 않은 대조군에 비해 높은 바, PAUF에 의한 신생혈관생성이 활발히 진행되었음을 확인할 수 있다.
실시예 10. PAUF에 의한 혈관내피세포 내 신호전달인자들의 활성 확인
PAUF에 의한 혈관 신생 촉진 현상이 혈관내피세포 내 주요한 신호전달인자들의 활성과 관련이 있음을 확인하고자 하였다.
구체적으로, 혈관내피세포를 젤라틴으로 코팅된 60mm 플레이트에 20% 우태아 혈청이 포함된 M199배지에서 배양 후 PBS로 세척하고 다시 무혈청 배지에서 6시간 동안 배양하였다. 이어서 0.5㎍/㎖ 농도의 PAUF를 각각 0, 5, 10, 20, 30, 45, 60, 90분 간 처리한 후 50mM Tris-HCl(pH7.4), 150mM NaCl, 1% Triton-X100, 1% DOC, 5mM EDTA, 50mM NaF, 1mM Na3Vo4, 프로티나아제 저해제로 구성된 RIPA 버퍼(radioimmunoprecipitation assay buffer)로 세포를 용해시켜 모았다. 다음으로, 0, 0.01, 0.1, 0.5, 1㎍/㎖ 농도의 PAUF를 혈관내피세포에 20분 간 처리한 후, RIPA 버퍼로 세포를 용해시켜 모았다. 이렇게 제조된 세포용해물을 정량하여 ERK(extracellular signaling related kinase), AKT(protein kinase B) 그리고 eNOS(endothelial nitric oxide synthase) 단백질의 인산화 정도를 확인하기 위해 웨스턴 블롯을 실시하였다. 이를 위해 정량된 세포 용해물 40 ㎍을 SDS-PAGE 로딩 완충액과 혼합한 후 가열하여 단백질을 변성시키고, 10% 아크릴아마이드 젤에서 전기영동하여 단백질을 크기별로 분리시켰다. 다음으로, 젤을 니트로셀룰로오스 막에 전이하였다. 니트로셀룰로오스 막은 5%(w/v) 탈지 우유를 포함한 TBST 버퍼(25 mM Tris, pH 7.5; 125 mM NaCl 및 0.1%(v/v) Tween-20)에서 1시간 동안 블로킹시켰다. ERK (Cell signaling), phospho ERK (Cell signaling), AKT (Cell signaling), phospho AKT (Cell signaling), eNOS (Cell signaling), phospho eNOS (Cell signaling), 또한 actin (Santa Cruz)에 대한 1차 항체는 3% BSA(bovine serum albumin) 용액에 1 : 1,000으로 희석하여 반응시켰고, 2차 항체는 HRP(horseradish peroxidase)가 달린 IgG를 1 : 5,000으로 사용하였다. 이 후 ECL(enhanced chemiluminescence, Milipore)에 반응시키고 X-ray (Agfa)에 감광시켜 확인한 결과를 도 11에 나타내었다.
도 11에 의하면, PAUF는 농도 및 시간 의존적으로 EKR, AKT 그리고 eNOS의 활성을 증가시키고 있음을 확인할 수 있다. 이는 PAUF에 의해 유도된 혈관 신생 촉진이 이러한 주요 인자들의 활성에 의해 나타날 수 있다는 것을 시사하고 있다.
실시예 11. 화학적 저해제 처리에 의한 혈관내피세포의 활성 변화 확인
혈관내피세포 활성 변화가 PAUF에 의해 활성화된 세포 내 신호전달인자에 의한 것임을 확인하기 위해, MEK(mitogen-activated ERK kinase) 저해제인 PD98059, PI3K(phosphoinositide 3-kinase) 저해제인 보르트마닌, 그리고 NOS(nitric oxide synthase) 생성 저해제인 L-NAME(L-NG-Nitroarginine methyl ester)을 처리하여 세포의 증식, 이동성, 관 형성능 및 세포 투과성 변화에 대해 알아보았다.
구체적으로, 세포의 증식 변화를 확인하기 위해 PAUF 단백질을 처리하기 1시간 전에 사용하고자 하는 화학적 저해제를 세포에 처리하고, PAUF 단백질을 처리하여 48시간 후 세포의 증식을 계수하여 정량화하였으며, 그 결과를 도 12에 나타내었다. 도 12에 나타난 바와 같이, PAUF에 의해 유도된 내피세포의 증식은 각 저해제를 처리함으로서 감소하였으며, 이를 통해 PAUF에 의한 내피세포의 증식 증가는 상기 세포 내 신호전달인자가 필수적으로 작용한다는 것을 알 수 있다.
또한 세포의 이동성 변화를 확인하기 위해 PAUF 단백질을 처리하기 1시간 전에 사용하고자 하는 화학적 저해제를 세포에 처리하고, 트렌스 웰을 이용하여 어세이를 실시하였으며, 그 결과를 도 13에 나타내었다.
도 13에 나타난 바와 같이, PAUF에 의해 유도된 내피세포의 이동성이 각 저해제를 처리함으로써 현저히 감소하였으며, 이를 통하여 PAUF에 의한 내피세포 이동성 증가는 상기 세포 내 신호전달인자가 필수적으로 작용한다는 것을 알 수 있다.
또한, 세포의 관 형성능에 대한 영향은 상기 화학적 저해제를 세포에 1시간 동안 처리한 후 PAUF 단백질을 처리하여 어세이를 수행하였으며, 그 결과를 도 14에 나타내었다.
도 14에 나타난 바와 같이, 각각의 저해제를 처리함으로써 PAUF에 의해 유도된 내피세포 분화 효과가 현저히 감소하였으며, 이를 통하여 PAUF에 의한 분화능 증가에 상기 세포 내 신호전달인자가 필수적으로 작용한다는 것을 알 수 있다.
이에 더하여, 화학적 저해제 처리 시 PAUF에 의해 유도되는 혈관내피세포 투과성 변화는 트렌스 웰에 가득 배양된 세포에 상기 화학적 저해제를 처리하고, 1시간 후 PAUF 단백질을 처리하여 FITC-덱스트란으로 투과성 정도를 확인하였으며, 그 결과를 도 15에 나타내었다.
도 15에 나타난 바와 같이, PAUF에 의해 유도된 혈관내피세포의 투과성 효과가 각각의 화학적 저해제를 처리함으로써 감소하였으며, 이는 PAUF에 의한 투과성의 증가는 상기 세포 내 신호전달인자의 신호전달경로가 주요하다는 것을 시사한다.
상기 결과는 PAUF가 이들 세포 내 신호 전달 인자를 활성화시킴으로서 혈관 신생에 주요한 과정을 증진시킨다는 것을 나타내고 있다.
실시예 12. 실험동물 모델에서 PAUF에 의한 신생혈관 생성 효과
PAUF의 혈관신생효과를 실험동물 모델을 통해 확인하였다.
구체적으로, PAUF 과발현 췌장암 세포주와 발현이 저해된 저해된 췌장암 세포주를 6주령의 누드 마우스 Balb/c에 이종이식 또는 동소 이식한 동물 모델을 구축하였다. 3주 후 누드 마우스를 희생하고 복개하고 종양을 적출하여 4% 포르말린으로 고정한 후 파라핀 블락을 제조하였다. 이를 5㎛ 두께로 박편한 후 혈관내피세포 표지자인 CD31을 확인하기 위해 면역조직화학법을 시행하고 정량하였으며, 그 결과를 도 16에 나타내었다.
도 16에 의하면, PAUF이 과발현된 세포주에서 발달한 종양의 경우 대조군에 비해 혈관의 생성이 향상되었다. 반면, PAUF 발현이 저해된 세포주에서 발달한 종양의 경우 대조군에 비해 혈관의 생성이 현저히 저하됨을 관찰할 수 있다. 이러한 결과는 PAUF가 동물모델에서 혈관신생유도를 조절하는 효과가 있음을 시사하고 있다.
혈관신생촉진인자로서의 신규한 기능을 갖는 PAUF 단백질은 혈관 신생이 요구되는 다양한 질환의 치료제 개발에 유용하게 이용될 것으로 기대된다.

Claims (11)

  1. 피에이유에프를 유효성분으로 포함하는, 혈관신생 촉진용 조성물.
  2. 제 1항에 있어서,
    상기 혈관신생은 발생, 상처 치유 또는 기관 형성 과정에서 일어나는 것인, 조성물.
  3. 피에이유에프를 유효성분으로 포함하는, 혈관신생 요구 질환의 치료용 약학적 조성물.
  4. 제 3항에 있어서,
    상기 혈관신생이 요구되는 질환은 만성 궤양, 지연성 상처치유, 또는 심혈관계 질환인, 조성물.
  5. 제 4항에 있어서,
    상기 심혈관계 질환은 동맥경화, 심근경색, 협심증, 뇌경색, 및 급성사지허혈로 이루어지는 군으로부터 선택되는 하나 이상인, 조성물.
  6. 제 3항에 있어서,
    상기 피에이유에프 단백질은 혈관내피세포의 증식, 이동, 또는 분화를 촉진시키는 것을 특징으로 하는, 조성물.
  7. 제 3항에 있어서,
    상기 피에이유에프 단백질은 세포 내 신호전달인자의 활성을 조절함으로써 혈관신생을 촉진하는 것인, 조성물.
  8. 제 7항에 있어서,
    상기 세포 내 신호전달인자는 ERK(extracellular signaling related kinase), AKT(protein kinase B), 및 eNOS(endothelial nitric oxide synthase)로 이루어지는 군으로부터 선택된 하나 이상인, 조성물.
  9. 약제학적으로 유효한 양의 피에이유에프 단백질을 유효성분으로 포함하는 약학적 조성물을 개체에 투여하는 단계를 포함하는, 혈관신생 요구 질환의 치료 방법.
  10. 제 9항에 있어서,
    상기 혈관신생이 요구되는 질환은 만성 궤양, 지연성 상처치유, 또는 심혈관계 질환인, 방법.
  11. 제 10항에 있어서,
