Abstract
Description
乳がんの発生メカニズムは、未だに系統だった解明がなされていない。このため、乳がんの治療は、現時点では、原則的に外科的ながん組織の摘出に頼らざるを得ず、化学療法や放射線療法は、副次的な治療法に留まっている。乳がんの中でも、特にエストロゲン受容体の発現が陰性かつプロゲステロン受容体の発現が陰性かつERBB2(erythroblastic leukemia viral oncogene homolog 2)タンパク質の発現が陰性である、いわゆるトリプルネガティブタイプと呼ばれる種類のものは、進行が早く、また予後不良であることから、有効な治療法の開発が待たれている。
近年において、正常なヒト細胞を遺伝子操作することによるインビトロ用のがんモデルシステムの開発が進んでいる。腫瘍組織から樹立された従来型のセルラインは、未知の遺伝子異常を含んでいる場合は、遺伝子変化による腫瘍化の研究に適していなかったが、上記のようなインビトロ用の癌モデルシステムのいくつかについては、特定の遺伝子変化による腫瘍化への影響を直接的に解明するのに有用である。例えば、非特許文献1は、正常な乳腺上皮細胞へヒトTelomere Reverse Transcriptase(hTERT)遺伝子、SV40T抗原遺伝子及びHRASV12遺伝子を導入することによって、初めて乳腺上皮細胞のがん化に成功した報告である。また、Kendall,S.D.et alは、正常な乳腺上皮細胞にhTERT遺伝子、TP53変異遺伝子、Cyclin D1遺伝子、Cdk4変異遺伝子、HRASV12遺伝子及びc−Myc遺伝子を導入することにより、乳がん細胞を作製することに成功している(非特許文献2)。さらに、別の遺伝子の組合せとしてhTERT遺伝子、TP53変異遺伝子、HRASV12遺伝子及びPIK3CA変異遺伝子を正常ヒト乳腺上皮細胞に導入し、これを癌化させた例も報告されている(非特許文献3)。
しかしながら、このような研究が進められているにも関わらず、現時点において、実際に患者から採取される乳がん組織と病理学的に高い類似性を示す乳がん細胞を人工的に作製できたという報告は殆ど存在しない。特に、トリプルネガティブ型乳がん細胞の作製については、未だに成功例が報告されていない。
現時点において、がん細胞の研究及び抗がん剤の開発には、がん患者から採取したがん細胞株を使用しているが、患者から採取されたがん細胞株の殆どは、不特定の遺伝子損傷を有するために、細胞株間で遺伝子の変異の状態にばらつきがある。従って、このような細胞を特定の分子又はシグナル経路の研究に用いた場合、得られた実験結果を異なる細胞株に応用することができない。また、このような細胞は、不特定の遺伝子損傷を有するため、がん化に対する個々の遺伝子の関与を評価するための実験に用いることができない場合がある。

本発明者らは、正常なヒト乳腺上皮細胞に遺伝子操作を施すことにより、前記細胞を腫瘍化させることに成功した。このようにして得られた腫瘍細胞は、正常細胞に遺伝子操作を行って腫瘍化させたものであるため、遺伝子の変異の状態が特定されている。本発明者らにより得られたこれらの腫瘍細胞を詳細に観察したところ、ヒト乳がん細胞、特にトリプルネガティブ型乳がん細胞の組織学的特徴を示すことが判明した。以上のことから本発明に至った。
すなわち、本発明は以下に関する。
[1] 正常乳腺上皮細胞に、以下の処理(1)及び(2)を施すことにより、腫瘍細胞を作製する方法。
(1)p53の機能損失
(2)v−Src遺伝子の導入
[2] サイクリン依存性キナーゼ遺伝子の強制発現がさらに施される、前記[1]に記載の方法。
[3] 前記サイクリン依存性キナーゼ遺伝子が、Cdk4遺伝子である、前記[2]に記載の方法。
[4] 正常乳腺上皮細胞に、以下の処理(1)~(3)を施すことにより、腫瘍細胞を作製する方法。
(1)p53の機能損失
(2)EGFR変異遺伝子の導入
(3)c−Myc遺伝子の導入
[5] 前記腫瘍細胞が、エストロゲン受容体の発現が陰性かつプロゲステロン受容体の発現が陰性かつERBB2の発現が陰性である、前記[1]~[4]のいずれか1つに記載の方法。
[6] 前記正常乳腺上皮細胞が哺乳類、霊長類またはげっ歯類由来の細胞である、前記[1]~[5]のいずれか1つに記載の方法。
[7] 前記[1]~[6]のいずれか1つに記載の方法により製造された腫瘍細胞。
[8] 以下の工程を含む、腫瘍の治療剤のスクリーニング方法。
(a)前記[7]に記載の腫瘍細胞と候補物質とを接触させる工程
(b)腫瘍細胞の増殖阻害効果を検出する工程
[9] 前記[7]に記載の腫瘍細胞が移植された担癌モデル動物。
[10] 以下の工程を含む、腫瘍の治療剤のスクリーニング方法。
(a)前記[9]に記載の担癌モデル動物と候補物質とを接触させる工程
(b)腫瘍細胞の増殖阻害効果を検出する工程
本発明の方法により作製される腫瘍細胞は、正常細胞に遺伝子操作を行って腫瘍化させたものであるため、遺伝子の変異の状態が特定されている。従って、本発明により、乳がんの発達における個々の遺伝子の関与の評価に非常に有用なモデルシステムが提供される可能性を有している。また、本発明の方法により作製される腫瘍細胞は、トリプルネガティブ型乳がんの表現型を示すので、これまで治療が困難とされてきたトリプルネガティブ型乳がんの治療法及び治療薬の開発に非常に有用である可能性を有している。
なお、本明細書において引用した全ての文献、および公開公報、特許公報その他の特許文献は、参照として本明細書に組み込むものとする。また、本明細書は、2010年7月7日に出願された本願優先権主張の基礎となる日本国特許出願(特願2010−154744号)の明細書及び図面に記載の内容を包含する。
1.腫瘍細胞の作製方法
本発明は、その第一の態様において、正常乳腺上皮細胞に、以下の処理(1)及び(2)を施すことにより、腫瘍細胞を作製する方法を提供する。なお、処理(1)、(2)の順番は特に限定されない。
(1)p53の機能損失
(2)v−Src遺伝子の導入
上記態様において、本発明の方法は、上記の処理に加え、処理(3)として、サイクリン依存性キナーゼ遺伝子を強制発現させる工程を含んでいてもよい。
本発明は、その第二の態様において、正常乳腺上皮細胞に、以下の処理(1)~(3)を施すことにより、腫瘍細胞を作製する方法を提供する。なお、処理(1)~(3)の順番は特に限定されない。
(1)p53の機能損失
(2)EGFR変異遺伝子の導入
(3)c−Myc遺伝子の導入
本発明の方法は、TERT遺伝子を導入することなく、正常乳腺上皮細胞を腫瘍化させることを特徴とする(図2参照)。外来遺伝子の発現によって正常細胞から腫瘍細胞を作製する方法において、TERT遺伝子を導入することなく腫瘍細胞の作製に成功した例は、未だ報告されていない。
本発明において、「正常乳腺上皮細胞」とは、乳腺上皮に由来する正常細胞であれば特に限定されるものではない。乳腺はたとえばヒト、サル、マウスなどの哺乳動物で発達していることが知られている。本発明において、「正常乳腺上皮細胞」とは、好ましくは哺乳類の乳腺上皮又は乳管上皮に由来する正常細胞であり、より好ましくは霊長類又はげっ歯類の乳腺上皮又は乳管上皮に由来する正常細胞であり、さらに好ましくはヒトの乳管上皮又は乳管上皮に由来する正常細胞である。かかる正常細胞のうち、さらに好ましくは、乳管の外層を構成する筋上皮に由来する正常細胞(Myoepithelial cell)である(図1参照)。「正常細胞」とは、健康な状態、即ち、検出可能な疾患又は異常を有しない状態の細胞を意味し、「正常ヒト乳腺上皮細胞」とは、健常人のヒトの乳腺上皮細胞など検出可能な疾患又は異常を有しない状態にあるヒトの乳腺上皮細胞である。以下において、「ヒト乳腺上皮細胞」を「HME細胞(Human Myoepithelial cell)」と呼ぶ。
HME細胞は、ヒトから採取したものを用いてもよく、あるいは市販の細胞(例えば、Lonza社(Walkersville,MD,USA)から販売される細胞)を用いてもよい。
「p53の機能喪失」とは、p53タンパク質がその本来の生物学的機能を有しないことを意味する。
p53タンパク質は、TP53遺伝子によりコードされ、TP53遺伝子は、DNA修復タンパク質の活性化、細胞周期の制御、アポトーシスの誘導に関与するp53タンパク質をコードする遺伝子であり、その機能異常が種々の癌の発生と関連している(Lane,D.P.(1992)Nature 358:15−16)ことが知られている。
p53の機能を喪失させる方法としては、種々の方法が適用でき、p53タンパク質が機能を喪失するのであれば特に限定されないが、例えば、p53タンパク質の中和抗体の添加、前記抗体をコードする遺伝子の細胞導入、p53タンパク質をコードする遺伝子(以下、「TP53遺伝子」という)のノックアウト、RNA干渉によるTP53遺伝子のノックダウン(Sato M et al(Cancer Res.66,(2006)2116−2128)参照)、ドミナントネガティブTP53遺伝子の強制発現等が挙げられる。本発明において好ましいp53の機能を喪失させる方法としては、ドミナントネガティブTP53遺伝子(以下、p53CT遺伝子ともいう)の遺伝子導入である。ドミナントネガティブTP53遺伝子のヌクレオチド配列は、Shaulian,E.らの文献(Mol.Cell.Biol.,Dec 1992;12:5581)を参照することで得られる。p53タンパク質の機能喪失に際して使用するTP53遺伝子又はp53タンパク質は、好ましくは哺乳動物由来の遺伝子又はタンパク質であり、より好ましくは霊長類由来の遺伝子又はタンパク質であり、さらに好ましくは、ヒト由来の遺伝子又はタンパク質である。
なお、p53の機能を喪失させる代わりに、カスパーゼ−3、−8~10、12等他のアポトーシス誘導タンパク質の機能を喪失させてもよい。この場合もp53の機能喪失と同様の方法で当該タンパク質の機能を喪失させることが可能である。
「v−Src遺伝子」は、レトロウィルスの一種であるRous sarcoma virusに由来するがん関連遺伝子として発見されたものであり、その配列はMayer BJ et al.(J Virol.1986 Dec;60(3):858−67.)に記載されている。
「サイクリン依存性キナーゼ(Cdk)遺伝子」は、細胞周期の回転に関与するタンパク質をコードする遺伝子であり、そのファミリーは、Cdk1~13から構成される。本発明において正常乳腺上皮細胞に導入するCdk遺伝子は、Cdk1~13のいずれをコードする遺伝子であってもよいが、好ましくは、Cdk4をコードする遺伝子(Cdk4遺伝子)を用いることができる。Cdk4は、Cyclin D1の結合パートナーであり、Cdk4遺伝子の変異が各種腫瘍内で検出されている(Zuo,L.et al.,(1996).Nature Genet.12,97−99)。Cdk遺伝子のヌクレオチド配列は、NCBIデータベースに登録されているものを使用すればよい。本発明において、Cdk遺伝子は、好ましくは哺乳動物由来のCdk遺伝子であり、より好ましくは霊長類由来のCdk遺伝子であり、さらに好ましくはヒト由来のCdk遺伝子である。
「EGFR遺伝子」は、上皮成長因子受容体タンパク質をコードする遺伝子であり、EGFR遺伝子の発現上昇は肺がん、乳がん、大腸がん、胃がん、脳腫瘍など多くのがんで認められる。(Sharma SV et al.,Nature Rev Cancer.2007;7(3):169−81)。
本発明において、「EGFR変異遺伝子」は、野生型が変異したEGFR遺伝子であり、例えば、790番目のスレオニン残基がメチオニンに、858番目のロイシン残基がアルギニンに置換されたEGFRT790M.L858R変異タンパク質(配列番号10)をコードする遺伝子が挙げられる。EGFR変異遺伝子のヌクレオチド配列は、NCBIデータベースに登録されている野生型の遺伝子配列を基に、McCoy,M.S.et al(Mol.Cell.Biol.,(1984),4,1577−1582)、Samuels,Y et al(Science,(2004),304,554)又はScott、K.D.et al(Cancer Res,(2007),67,5622−5627)に記載の方法により作製可能である。本発明において、EGFR遺伝子は、好ましくは哺乳動物由来のEGFR遺伝子であり、より好ましくは霊長類由来のEGFR遺伝子であり、さらに好ましくはヒト由来のEGFR遺伝子である。
「c−Myc遺伝子」は、種々の遺伝子の発現調節及びDNA複製に関与するDNA結合因子タンパク質、即ち転写因子タンパク質をコードする遺伝子であり、c−Myc遺伝子の発現異常は、種々のがんとの関連性が示唆されている(Dominguez−Sola,D.et al.,(2007).Nature 448,445−451)。c−Myc遺伝子のヌクレオチド配列は、NCBIデータベースに登録されているものを使用すればよい。本発明においては、c−Myc遺伝子は、好ましくは哺乳動物由来のc−Myc遺伝子であり、より好ましくは霊長類由来のc−Myc遺伝子であり、さらに好ましくは、ヒト由来のc−Myc遺伝子である。
後述する実施例において使用する各遺伝子の各ヌクレオチド配列及びペプチド配列を下記表1に示すが、これらに限定されない。
本発明において、遺伝子をHME細胞に導入する場合、当該遺伝子を適切な発現カセットとして発現ベクターに挿入し、該ベクターでHME細胞を形質転換すればよい。適切な発現カセットは、少なくとも以下の(i)~(iii)を構成要素として含む。
(i)HME細胞で転写可能なプロモーター;
(ii)該プロモーターにin−frameに結合した遺伝子;及び
(iii)RNA分子の転写終結及びポリアデニル化シグナルをコードする配列
HME細胞で転写可能なプロモーターの例としては、CMV、CAG、LTR、EF−1α、SV40プロモーター等が挙げられるが、これらに限定されるものではない。
前記発現ベクターは、前記発現カセットの他に、形質転換されたHME細胞をセレクションするための選択マーカー発現カセットを有していてもよい。選択マーカーの例としては、ネオマイシン耐性遺伝子、ハイグロマイシンBホスホトランスフェラーゼ遺伝子等のポジティブセレクションマーカー、LacZ、GFP(Green Fluorescence Protein)及びルシフェラーゼ遺伝子などの発現レポーター、単純ヘルペスウイルスチミジンキナーゼ遺伝子(HSV−TK)、ジフテリア毒素Aフラグメント(DTA)等のネガティブセレクションマーカー等が挙げられるが、これらに限定されない。
形質転換されたHME細胞は、上記マーカーにより容易に選択することができる。例えば、ネオマイシン耐性遺伝子をマーカーとして導入した細胞であれば、G418を加えた培地中で培養することにより、一次セレクションを行うことができる。また、ターゲティングベクターがGFP等の蛍光タンパク質の遺伝子をマーカーとして含む場合には、薬剤耐性によるセレクションに加えて、FACS(Fluorescence Activated Cell Sorter)等を用いた蛍光タンパク質発現細胞のソーティングを行ってもよい。
HME細胞への遺伝子の導入に使用可能な発現ベクターは、例えば細胞へ遺伝子導入可能な発現ベクターが挙げられ、市販されているものを用いてもよい。例としてpEGFP−C1TM(Clontech)、pCMV−HATM(Clontech)、pMSCVpuroTM(Clontech)、pEF−DEST51TM(Invitrogen)、pCEP4TM(Invitrogen)、ViraPower II Lentiviral Gateway SystemTM(Invitrogen)等が挙げられる。発現ベクターは、エレクトロポレーション、マイクロインジェクション、リン酸カルシウム法、リポフェクション法、ウィルス感染等、公知の遺伝子導入法によりHME細胞へ導入することができる。遺伝子導入法の詳細については、「Sambrook & Russell,Molecular Cloning:A Laboratory Manual Vol.3,Cold Spring Harbor,Laboratory Press 2001」等を参照することができる。
2.腫瘍細胞
本発明において「腫瘍細胞」とは、in vivoで自立的に過剰増殖する細胞を意味し、上記の「腫瘍細胞の作製方法」において作製された腫瘍細胞のみならず、後述する「担癌モデル動物」から採取される腫瘍細胞であってもよく、あるいは、当該担癌モデル動物から採取される腫瘍細胞をin vitroで培養した細胞であってもよい。
腫瘍細胞の例としては、乳がん腫に含まれる細胞が挙げられる。本発明のある実施態様では、腫瘍細胞の好ましい形態として、乳がん細胞が挙げられる。腫瘍細胞は、より好ましくは、エストロゲン受容体(Estrogen Receptor:ER)、プロゲステロン受容体(Progesteron Receptor:PgR)及びERBB2(別称は「HER2」)タンパク質の全てのマーカーについて陰性のトリプルネガティブ型乳がん細胞である。
細胞の腫瘍化を確認する場合には、被検細胞を適切なモデル動物に皮下注射して腫瘍塊形成を観察することにより確認することができる。
モデル動物の例としては、ヒトを除く哺乳動物が好ましく、特に免疫抑制された哺乳動物が好ましい。免疫抑制された哺乳動物の例としては、ヌードラット及びヌードマウスが挙げられるが、これらに限定されるものではない。
細胞の腫瘍化を確認するための別の方法としては、被検細胞をソフトアガー上で培養し、そのコロニー形成を観察する方法(ソフトアガーコロニー形成アッセイ)が挙げられる。具体的には、TP53変異遺伝子及びv−Src遺伝子を導入したHME細胞を一定の細胞濃度に調製した上でソフトアガー培地に播種し、細胞増殖速度を観察する方法が挙げられる。ソフトアガーコロニー形成アッセイの詳細については、以下の文献を参照できる:Tanaka,S.et al.,Proc.natl.Acad.Sci.USA.94:2356−2361,1997。
本発明において得られる腫瘍細胞(以下、「本発明の腫瘍細胞」)は、非常に高い増殖能を有することを特徴とする。図4Aに示すように、本発明の腫瘍細胞は、ヒト乳がん細胞株MDA−MB231細胞(ATCC HTB−26)と比べて顕著に高い増殖能を有する。また、ある実施態様において、本発明の腫瘍細胞をモデル動物に移植して得られる腫瘍組織は、ER(−)、PgR(−)、ERBB2(−)であり、たとえばヒト由来の乳腺上皮細胞を用いた場合にはER(−)、PgR(−)、HER2(−)であり、トリプルネガティブ型の乳がん細胞の形態を示す(図5、6及び8)。
ヒトのトリプルネガティブ型の乳がんで観察される腫瘍細胞では、ホルモン受容体であるER及びPgRが発現しておらず、また、受容体型チロシンキナーゼであるHER2も発現していないため、ホルモン療法又は化学療法による治療効果が期待できず、最も治療が困難な種類の乳がんであると考えられている。
本発明の腫瘍細胞は、いずれも腫瘍形成率及び増殖能が高い。特に、モデル動物に本発明の腫瘍細胞を移植して腫瘍を形成させた場合、乳腺(管)上皮細胞が属する動物種において自然発生し得る腫瘍組織と病理学的に酷似した腫瘍をモデル動物で形成する。例えば、ヒト由来の乳腺上皮細胞を移植した場合には、ヒトであるがん患者から採取されたがん組織、特に、トリプルネガティブ型乳がん組織と病理学的に酷似した腫瘍をモデル動物で形成する。従って、本発明の腫瘍細胞は、トリプルネガティブ型乳がんの治療法又は治療薬の開発に非常に有用である可能性を有している。
3.担癌モデル動物
本発明において「担癌モデル動物」とは、前記腫瘍細胞をヒトを除くモデル動物に移植して腫瘍塊が形成された動物を意味する。モデル動物の例としては、ヒトを除く哺乳動物が好ましく、マウス、ラット、ブタ、イヌ、サル、ハムスター、ウサギなどが挙げられるが、これらに限定されるものではない。このようなモデル動物のうち、哺乳動物が好ましく、さらに霊長類又はげっ歯類に属する動物が好ましい。これらのうち、特に免疫抑制された哺乳動物が好ましく、さらには免疫抑制された霊長類又はげっ歯類に属する動物が好ましい。免疫抑制された哺乳動物は、通常の哺乳動物にシクロスポリン等の免疫抑制剤を投与することにより作製することもできるが、好ましくは、遺伝的バックグラウンドにより先天的に免疫が抑制された哺乳動物が好ましい。先天的に免疫が抑制された哺乳動物の例としては、ヌードラット及びヌードマウスが挙げられるが、これらに限定されるものではない。
モデル動物への前記腫瘍細胞の移植方法は特に限定されない。移植されるモデル動物に応じて従来から利用されている方法を適宜選択すればよい。マウス以外に移植した事例として例えば、Genetic induction of tumorigenesis in Swine Oncogene 26,1038−1045(11 September 2006)等を参照できる。移植した腫瘍細胞の再摘出の容易性の観点からは、皮下注射又は腹腔内注射による移植が好ましく、一方で、解剖学的な観点からは、同所移植が好ましい。
4.抗腫瘍剤のスクリーニング方法
本発明は、腫瘍の治療剤のスクリーニング方法を提供する。
本発明のスクリーニング方法の第一の態様として、以下の工程を含む、腫瘍の治療剤のスクリーニング方法を提供する。
(a)本発明の腫瘍細胞と候補物質とを接触させる工程
(b)腫瘍細胞の増殖阻害効果を検出する工程
また、本発明のスクリーニング方法の第二の態様として、以下の工程を含む、腫瘍の治療剤のスクリーニング方法を提供する。
(a)本発明の腫瘍細胞が移植された担癌モデル動物と候補物質とを接触させる工程
(b)腫瘍細胞の増殖阻害効果を検出する工程
上記において、「腫瘍細胞と候補物質とを接触させる」または「腫瘍細胞が移植された担癌モデル動物と候補物質とを接触させる」とは、該候補物質が、腫瘍細胞表面の分子と相互作用を生じさせる程度に接近するか、該分子と結合するか、又は該腫瘍細胞内に取り込まれる条件を調整することを意味する。腫瘍細胞が、たとえば培養細胞などの腫瘍細胞である場合、該細胞が接している培養培地に該候補物質を一定濃度以上で添加することにより、該細胞と候補物質とを接触させることができる。一方、腫瘍細胞が動物体内に移植されている場合、すなわち、腫瘍細胞が移植された担癌モデル動物の場合、該動物に候補物質を一定量で投与することにより、該腫瘍細胞と候補物質とを接触させることができる。この場合、投与経路は、候補物質の投与に一般的に採用されている経路であれば、特に限定はされないが、具体例としては、経口、舌下、経鼻、経肺、経消化管、経皮、点眼、静脈内注射、皮下注射、筋肉内注射、腹腔内注射、局所注射、外科的移植が挙げられ、好ましくは経口投与、腹腔内注射及び静脈内注射である。
候補物質の例としては、既に抗腫瘍効果が確認されている薬剤及び抗腫瘍作用の可能性がある化合物、ポリペプチド、核酸、抗体及び低分子化合物等が挙げられる。このような候補物質の具体例としては、代謝拮抗剤(例えば、5−フルオロウラシル(5−FU))、葉酸代謝拮抗薬(例えば、ジヒドロプテロイン酸シンターゼ阻害薬であるスルファジアジン及びスルファメトキサゾール、ジヒドロ葉酸レダクターゼ阻害薬(DHFR阻害薬)であるメソトレキセート、トリメトプリム、ピリメタミン)、ピリミジン代謝阻害薬(例えば、チミジル酸シンターゼ阻害薬である5−FU、フルシトシン(5−FC))、プリン代謝阻害薬(例えば、IMPDH阻害薬である6−メルカプトプリン及びそのプロドラッグであるアザチオプリン)、アデノシンデアミナーゼ(ADA)阻害薬)(例えば、ペントスタチン)、リボヌクレオチドレダクターゼ阻害薬(リボヌクレオチドレダクターゼ阻害薬であるヒドロキシウレア)、ヌクレオチドアナログ(プリンアナログであるチオグアニン、リン酸フルダラビン及びクラドリビン、ピリミジンアナログであるシタラビン及びゲムシタビン)、L−アスパラギナーゼ、アルキル化剤(例えば、ナイトロジェンマスタードであるシクロホスファミド、メルファラン及びチオテパ、白金製剤であるシスプラチン、カルボプラチン及びオキサリプラチン、ニトロソウレアであるダカルバシン、プロカルバシン及びラニムスチン)抗腫瘍性構成物質(ザルコマイシン、マイトマイシンC、ドキソルビシン、エピルビシン、ダウノルビシン、ブレオマイシン)、トポイソメラーゼ阻害剤(イリノテカン、ノギテカン、ドキソルビシン、エトポシド、レボフロキサシン、シプロフロキサシン)、微小管重合阻害剤(ビンブラスチン、ビンクリスチン、ビンデシン)、コルヒチン、微小管脱重合阻害薬(パクリタキセル、ドセタキセル)、分子標的剤(トラスツズマブ、リツキシマブ、イマチニブ、ゲフィチニブ、ボルテゾミブ、エルロチニブ)、デキサメサゾン等のステロイド薬、フィナステリド、アロマターゼ阻害剤及びタモキシフェン並びにこれらの組合せが挙げられるが、以上に限定されるものではない。
腫瘍細胞の増殖阻害効果については、同一の細胞数を同一の細胞密度で培養ディッシュに播種した同一条件の培養系、あるいは同一の細胞数を同一の細胞密度で移植した同一系統の担癌モデル動物を2系用意し、一方の系(サンプル)の細胞に前記候補物質を接触させ、他方の系(コントロール)の細胞には前記候補物質を接触させずに、両系の腫瘍細胞の増殖を観察し、一定期間経過後に前記2系のそれぞれに含まれる腫瘍細胞数を測定し、比較することで増殖阻害効果を確認することができる。
本発明の腫瘍細胞を培養細胞など腫瘍細胞の形態でスクリーニングに用いる場合、上記サンプル細胞に候補物質を接触させた後に、サンプル細胞の細胞数とコントロール細胞の細胞数とをセルカウンター等で測定し、それぞれの細胞数を比較して腫瘍増殖効果を確認することができる。
本発明の腫瘍細胞をモデル動物に移植した形態でスクリーニングに用いる場合、すなわち、腫瘍細胞が移植された担癌モデル動物の形態でスクリーニングに用いる場合、サンプル系のモデル動物に候補物質を投与した後に、サンプル系及びコントロール系の動物から腫瘍組織を取り出し、該サンプル系及びコントロール系の腫瘍組織に含まれる細胞数を測定及び比較することができる。あるいは、前記腫瘍組織を取り出して、該腫瘍組織の体積をサンプル系とコントロール系の間で比較して、腫瘍増殖効果を確認することもできる。腫瘍の体積は、以下の式により求めることができる。
腫瘍体積=ab2/2 (a:横幅、b:長さ)
あるいは、サンプル系及びコントロール系の動物に候補物質を投与した後に、腫瘍細胞の移植箇所に対し、計測可能な腫瘍塊が形成された割合をサンプル系とコントロール系の間で比較して、腫瘍増殖効果を確認することもできる。
モデル動物は、項目「3.担癌モデル動物」で述べたモデル動物と同様である。モデル動物への移植方法は特に限定されないが、移植した腫瘍細胞の再摘出の容易性から、皮下注射又は腹腔内注射が好ましい。
サンプル系の腫瘍細胞の増加率が、コントロール系の腫瘍細胞の増加率よりも小さい場合には、用いた候補物質は、抗腫瘍効果を有するものと判断することができる。あるいは、本発明の腫瘍細胞に関し、既に一定の条件下での増加率について複数のデータが存在する場合には、そのデータを統計処理して得られる平均値、標準偏差等から導き出される基準値と比較して判断してもよい。
腫瘍の増殖率の平均値、標準偏差等は、種々の統計方法によって得ることができるが、具体的には、培養細胞であれば、播種した時点の初期細胞数及び細胞密度をパラメーターとし、あるいは移植細胞であれば使用したモデル動物の移植時の体重及び移植した腫瘍細胞の細胞数をパラメーターとして、IBM SSPS Statistics 18(SSPS)等の統計解析ソフトでtwo−way ANOVA解析することにより求めることができる。本発明の方法の実施により得られた腫瘍細胞の増殖率を新たなデータとして統計解析用の母集団に加えて母数を大きくすることにより、さらに解析の精度を向上させることができる。
本発明の腫瘍細胞は、患者から採取された乳がん細胞と病理学的に酷似した特徴を示すことから、本発明のスクリーニング方法で腫瘍細胞の増殖抑制効果が確認された抗腫瘍剤は、実際にがん患者、特に乳がん患者(例えば、トリプルネガティブ型乳がん)の治療に用いた場合にも抗腫瘍効果を奏すると期待される。
以下、実施例を用いて本発明を詳細に説明するが、本発明は実施例に記載された態様に限定されるものではない。
以下の記載において、HME細胞に導入した遺伝子の種類に応じて、作製した各腫瘍細胞を下記の表に示すように表記する。
細胞培養
HME細胞は、30歳の白人女性に由来する正常ヒト乳腺上皮細胞をLonza社(Walkersville,MD,USA)から購入した。この細胞を、コラーゲンコーティングしたディッシュ上で、Lonza社から提供された種々の増殖因子を添加した血清不含MEGM medium(MEGM Bullet Kit;Lonza)中にて培養した。細胞は、湿潤インキュベーター内で低酸素環境下(3% O2及び5% CO2)で37℃にて維持し、これを本実施例において使用するHME細胞として使用した。
レトロウィルスベクターの作製及び該レトロウィルスベクターのHME細胞への導入
表1に示した遺伝子のうち、v−Src、Cdk4、ドミナントネガティブTP53、EGFRT790.L858R及びc−Mycをそれぞれ以下の表に示すベクターに組み込むことにより、レトロウイルス発現用ベクターを作製した。
上記ウィルスベクターに感染した細胞は、puromycin、bleocineの存在下で2週間培養することによりセレクションを行った。いずれの薬剤でセレクションを行う場合でも、培養細胞は、感染細胞のポリクローナルな増殖集団から選択した。
なお、Ecotropic receptor(Eco VR)を発現させたHME細胞は以下の手順で作製した。
以下のプライマーを用いてMouse Ecotropic retrovirus receptor(Slc7al,NM_007513)の全コーディング領域を含むcDNAをRT−PCRによりクローニングした。
上記RT−PCRの増幅産物をpCX4hygベクター(GenBank Accession No:AB086387)に組み込んだ。該ベクターと、Takara Bio社(Shiga,Japan)より購入したpGP及びpE−Amphoプラスミドとを293T細胞に導入することで、ウイルスベクターを作製し、該ウィルスベクターをHME細胞に感染させ、hygromycinの存在下で2週間培養することにより感染細胞のセレクションを行った。
イムノブロット法
タンパク質の測定、SDS−PAGE及びイムノブロッティングは、先の文献(Akagi,T.et al.,(2002).Mol.Cell Biol.22,7015−7023)の記載に従って行った。免疫反応陽性タンパク質のシグナルは、SuperSignal WestFemto reagent(Pierce社)を用いてケミルミネセンスにより可視化した。抗Src抗体、抗リン酸化Src(Tyr416)抗体及び抗p53抗体は、Cell Signaling Technology社より、抗リン酸化チロシン抗体(4G10)はMillipore社より入手した。
異種移植片増殖実験
マウスを用いて異種移植片増殖実験を行った。具体的には、表1に示す遺伝子の組合せを発現させたHME細胞を1x106個含む細胞懸濁液(single−cell−suspension)を、50% MATRIGELTM(BD Bioscience,San Jose,CA,USA)に懸濁し、週齢6又は7のメスの胸腺欠損ヌードマウス(BALB/c nu/nu;Japan SLC,Hamamatsu,Japan)または、NOD−SCIDマウスのわき腹に皮下注射した。移植後、目視で腫瘍塊の形成の有無を確認した。腫瘍塊の形成に際しては、移植後12週経過時に指でつまめる程度の体積の腫瘍塊の形成が認められない場合には、陰性と判断した。また、移植後、ノギスを用いて腫瘍の寸法を測定し、以下の式に基づいて腫瘍の体積を計算し、造腫瘍能として見積もった。

また、各がん関連遺伝子が導入されたHME細胞の移植箇所に対し、計測可能な腫瘍塊が形成された割合を腫瘍形成率として算出し、造腫瘍能として見積もった。
組織学的分析及び免疫組織化学染色
異種移植片増殖実験において、移植した異種移植片を、移植後20日目に取り出した。当該移植片を、ホルマリン固定及びパラフィン包埋した後に切片化し、通常のプロトコルに従ってヘマトキシリン・エオジン(H&E)染色を行った。先の文献(Sasai,K.et al.,(2008).Am.J.Surg.Pathol.32,1220−1227)の記載に従い、以下の抗体を用いて免疫組織化学染色を行った:サイトケラチン5/6(CK5/6;DAKO,M7237)、エストロゲン受容体(ER;DAKO,M7040)、プロゲステロン受容体(PgR;DAKO,M3569)、HER2(DAKO,K5204)。
具体的には、以下の手順で免疫組織化学染色を行った。厚さ4μmの組織切片をキシレンで脱パラフィン化した後にエタノールで脱水した。圧力鍋にて、10mMクエン酸バッファー(pH6.0)中で2分間加熱することにより、抗原を賦活させた。0.01% Tween20含有リン酸干渉生理食塩水(PBST)で組織切片を再水和させ、0.3%過酸化水素とインキュベートさせることで、内在性のペロキシダーゼを不活化させた。組織切片を、適切な希釈濃度の1次抗体で4℃にて一晩インキュベートし、PBSTで洗浄した後、室温にてEnvision Dual Link solution(Dako,Glostrup,Denmark)で30分間インキュベートした。次に、ジアミノベンジジン(Dako)で切片を処理して抗原抗体反応部位を可視化させ、ヘマトキシリン処理を90秒間行って核染色を行った。組織切片のスライドをEntellan Neu reagent(Merck,Whitehouse Station,NJ)でマウントした後にカバーガラスで封入し、観察に供した。
実験結果
[1] HME細胞へ導入した遺伝子の発現確認
イムノブロッティングを行い、HME細胞へ導入された遺伝子の発現を確認した。HME/453/v−Src細胞及びHME/53/v−Src細胞に導入したドミナントネガティブTP53遺伝子及びv−Src遺伝子が発現していることを、抗リン酸化Src抗体及び抗p53抗体を用いた抗体抗原反応により確認した(それぞれ図3A及び図3B)。また、v−Src遺伝子の機能により、チロシンのリン酸化が増強していることを抗リン酸化チロシン抗体により確認し、目的とする遺伝子が導入され、かつ発現していることを確認した(図3C)。
[2] 作製された腫瘍細胞の造腫瘍能の確認
作製された腫瘍細胞をヌードマウスに皮下移植し、移植された腫瘍片の造腫瘍能を確認した(図4、7及び8)。
図7に示すように、HMEC細胞に対してドミナントネガティブTP53遺伝子の導入が行われた細胞(HME/53細胞)及びドミナントネガティブTP53遺伝子及びCdk4遺伝子の導入が行われた細胞(HME/53/Cdk4細胞)では、腫瘍塊の形成が確認されなかった。
図4及び7に示すように、HMEC細胞に対してドミナントネガティブTP53遺伝子及びv−Src遺伝子の導入が行われた細胞(HME/53/Cdk4/v−Src細胞及びHME/53/v−Src細胞)では、指でつまめる程度以上の腫瘍塊の形成が確認され、100%の腫瘍形成率を示した。また、図4A及び図4Bに示すように、HME/53/Cdk4/v−Src細胞及びHME/53/v−Src細胞は、従来利用されているヒト乳がん細胞株MDA−MB231細胞と比べて短期間でより大きな腫瘍を形成することから、高い増殖能を有することが明らかになった。
図7に示すように、ドミナントネガティブTP53遺伝子が導入されたHMEC細胞(HME/53細胞)に対し、EGFRT790.L858R遺伝子又はc−Myc遺伝子のいずれかを単独で導入したHME/53/EGFRT790.L858R及びHME/53/c−Myc細胞では、腫瘍塊が形成されなかった。
一方、ドミナントネガティブTP53遺伝子、EGFRT790.L858R遺伝子及びc−Myc遺伝子の組み合わせを導入したHME/53/EGFRT790.L858R/c−Myc細胞では、腫瘍塊の形成が確認され、100%の腫瘍形成率を示した。
[3] 作製された腫瘍細胞の表現型の確認
異種移植片増殖実験により得られた腫瘍組織の表現型を確認するために種々のがんマーカーで免疫染色し、観察した。HME/53/v−Src細胞の皮下移植により形成した腫瘍の組織切片を、臨床で診断に使用されているマーカーで染色したところ、図5で示したようにER(−),pgR(−),HER2(−)であり、且つベーサル型の乳癌のマーカーであるCK5/6(+)であったことから、HME/53/v−Src細胞は、最も治療の困難な種類の乳癌であるトリプルネガティブ型且つベーサル型の乳癌と病理学的に酷似した腫瘍を形成することが判明した。また、HME/53/Cdk4/v−Src細胞の皮下移植により形成した腫瘍の組織切片も、図6に示すようにER(−),pgR(−),HER2(−)であったことから、同様にトリプルネガティブ型の乳癌と病理学的に酷似した腫瘍を形成することが示された。また、HME/53/EGFRT790.L858R/c−Myc細胞の皮下移植により形成した腫瘍の組織切片も、図8に示すように、ER(−),pgR(−),HER2(−)であり、且つベーサル型の乳癌のマーカーであるCK5/6(+)であったことからHME/53/EGFRT790.L858R/c−Myc細胞は、最も治療の困難な種類の乳癌であるトリプルネガティブ型且つベーサル型の乳癌と病理学的に酷似した腫瘍を形成することが判明した。
以上の結果が示すように、正常な乳腺上皮細胞に、TP53変異遺伝子とv−Src遺伝子の組み合わせ、TP53変異遺伝子とv−Src遺伝子とCdk4遺伝子の組み合わせ又はTP53変異遺伝子とEGFR2変異遺伝子とc−Myc遺伝子の組み合わせを遺伝子導入することにより、従来では腫瘍細胞の作成に必須とされていたhTERT遺伝子を用いずに腫瘍細胞を作製することが可能となった。また、本発明では、ヒト乳がん患者から採取される乳がん組織と酷似したヒト乳がんモデル、特にトリプルネガティブ型乳がんモデルを作製することが可能となった。

Claims (10)
- 正常乳腺上皮細胞に、以下の処理(1)及び(2)を施すことにより、腫瘍細胞を作製する方法。
(1)p53の機能損失
(2)v−Src遺伝子の導入 - サイクリン依存性キナーゼ遺伝子の強制発現がさらに施される、請求項1に記載の方法。
- 前記サイクリン依存性キナーゼ遺伝子が、Cdk4遺伝子である、請求項2に記載の方法。
- 正常乳腺上皮細胞に、以下の処理(1)~(3)を施すことにより、腫瘍細胞を作製する方法。
(1)p53の機能損失
(2)EGFR変異遺伝子の導入
(3)c−Myc遺伝子の導入 - 前記腫瘍細胞が、エストロゲン受容体の発現が陰性かつプロゲステロン受容体の発現が陰性かつERBB2の発現が陰性である、請求項1~4のいずれか1項に記載の方法。
- 前記正常乳腺上皮細胞が哺乳類、霊長類またはげっ歯類由来の細胞である、請求項1~5のいずれか1項に記載の方法。
- 請求項1~6のいずれか1項に記載の方法により製造された腫瘍細胞。
- 以下の工程を含む、腫瘍の治療剤のスクリーニング方法。
(a)請求項7に記載の腫瘍細胞と候補物質とを接触させる工程
(b)腫瘍細胞の増殖阻害効果を検出する工程 - 請求項7に記載の腫瘍細胞が移植された担癌モデル動物。
- 以下の工程を含む、腫瘍の治療剤のスクリーニング方法。
(a)請求項9に記載の担癌モデル動物と候補物質とを接触させる工程
(b)腫瘍細胞の増殖阻害効果を検出する工程
