Abstract

The invention provides a measuring system and to a method for reconstructing a histological volume from a stack of consecutive sectional images of a tissue sample. After the tissue sample has been provided, a reference image of the cross-sectional surface of the tissue sample is recorded and stored. A histological layer is removed from the cross-sectional surface of the tissue sample by means of a microtome and mounted on a substrate. A sectional image of the histological layer mounted on the substrate is then recorded. The recorded sectional image is geometrically aligned with the stored reference image and the aligned sectional image is associated with at least one further recorded sectional image of a histological layer in order to combine sectional images which have been associated with each other and aligned to form the histological volume.
