PORTAMUESTRAS PARA MICROANALISIS DE RAYOS X CON MICROSCOPÍA ELECTRÓNICA DE BARRIDO
D E S C R I P C I Ó N
OBJETO DE LA INVENCIÓN
El objeto principal de Ia presente invención es un nuevo portamuestras para microanálisis de rayos X que evita interferencias entre las señales de Ia muestra y las señales del propio portamuestras. El portamuestras de Ia invención es especialmente útil para el análisis de muestras biológicas o de pequeño espesor.
ANTECEDENTES DE LA INVENCIÓN
El término microanálisis en microscopía electrónica se refiere a Ia identificación y análisis químico de volúmenes pequeños de materia. Con el uso del microanálisis de rayos X (XRMA) es posible obtener el contenido químico cualitativamente (elementos atómicos presentes a Ia muestra) y cuantitativamente (en masa), utilizando detectores de rayos X acoplados a microscopios electrónicos.
En sus inicios, el microanálisis se aplicaba al análisis de material de origen geológico para posteriormente utilizarse en biología con muestras de células en cultivo. El cambio de naturaleza de Ia muestra no es trivial, ya que normalmente en geología se analizan minerales con un número atómico (Z) elevado y alta densidad, mientras que las muestras biológicas se caracterizan por ser poco densas, tener bajos Z (al estar básicamente constituidas por C, H, O, N, P y S), y ser muy delgadas (esta técnica se aplica básicamente al estudio del contenido elemental de células).
El XRMA puede llevarse a cabo en dos tipos de microscopios
electrónicos: barrido (MEB o SEM, según sus siglas en inglés) y transmisión (MET o TEM, según sus siglas en inglés). También en un microscopio de transmisión-barrido (STEM, en su denominación inglesa). La preparación de las muestras es muy diferente en cada tipo de microscopio, ya que los objetivos que persiguen también Io son. El microanálisis de rayos X se puede realizar con dos espectrómetros: espectrómetro de dispersión de longitud de onda (WDS, según sus siglas inglesas) y espectrómetro de dispersión de energías (EDS, según sus siglas en inglés). Este último es el que se utiliza generalmente en los SEM. En un SEM se pretende estudiar Ia morfología superficial (topografía) de Ia muestra, por Io tanto ésta se coloca sobre un apoyo rígido, los más comunes de 12 a 15 mm de diámetro, y se recubre con algún metal (C, Au, Au-Pd) para dar conductividad a Ia muestra y aumentar Ia resolución de Ia imagen. Las muestras son relativamente gruesas, apareciendo fenómenos de absorción, dispersión y fluorescencia de rayos X que complican Ia identificación y cuantificación de los elementos presentes. Para cuantificar espectros obtenidos con análisis en SEM ("denominados en inglés "bulk analysis"), normalmente se aplica Ia corrección ZAF.
En un microscopio TEM se pueden distinguir estructuras dentro de Ia muestra, a partir de diferencias en Ia densidad de los electrones que llegan al detector después de atravesar Ia muestra. Ésta debe ser tan delgada como se pueda, y se coloca sobre una rejilla, normalmente de Al, de 3 mm de diámetro. Si se asume que Ia capa analizada es suficientemente delgada, entonces los efectos de absorción y fluorescencia de rayos X se pueden desestimar, de manera que Ia intensidad de los rayos X emitidos y de los rayos X que abandonan Ia muestra es Ia misma. Este es el criterio para el análisis de capa fina ("thin film"), que se puede llevar a cabo en TEM, pero no en SEM.
Las metodologías existentes hasta el presente para el análisis de células individuales utilizando SEM - EDS (por ejemplo, Sigee y Levado, 2000, "Ce// surface elemental composition of Microcystis aeruginosa: high-Si and low-Si subpopulations within the water column of a eutrophic lake", J. Plankton Res.
22 (11 ): 2137-2153) no son válidas cuando se trata de medir elementos ligeros (C, N y O), ya que las células son colocadas en filtros de policarbonato. Al contener el filtro mayor cantidad de C y O que las células, Ia señal proveniente del filtro es mucho más importante que Ia de Ia célula, quedando ésta muy diluida. La contribución del filtro también sería muy importante en el caso de utilizar filtros de otros materiales, como por ejemplo de fibra de vidrio (alta contribución en Si), o de plata (algunos picos característicos de Ia plata (Ag) interfieren con los elementos ligeros). Además, Ia densidad del volumen analizado aumenta, afectando tanto al análisis como a Ia forma del espectro de microanálisis resultante.
La utilización de otros soportes que normalmente se utilizan en las observaciones por SEM, como soporte de Al, de Cu-Zn, o de Be, también presentan inconvenientes en el análisis de células individuales:
• Oxidación del soporte (las células se encuentran en un medio líquido).
• Aparición de picos característicos y contribución en radiación de fondo por parte de los elementos que conforman el soporte. • Toxicidad de algunos soportes (Be).
Otros investigadores han creado un soporte de boro (Choél et al., 2005, "Quantitative determination of low-Z elements in single atmospheric partióles on boron substrates by automated scanning electrón microscopy-energy- dispersive X-ray spectrometry" , Anal. Chem. 77:5686-5692), que con las condiciones adecuadas del detector puede actuar como el soporte de Berilio, es decir, sin aportar picos característicos, pero participando en Ia radiación continua, absorción y dispersión de rayos X. Teniendo en cuenta que el detector de rayos X funciona en condiciones óptimas cuando recibe como máximo 1000 cuentas (rayos X)/s, resulta importante evitar tanto los picos característicos que contaminen Ia muestra (provenientes de las chapas y filtros), como picos de radiación continua que no provengan de Ia muestra que
nos interesa analizar. Eso es especialmente importante cuando Ia muestra contiene algún elemento de interés en muy baja concentración. Una aproximación a Ia técnica resultante de Ia utilización de este nuevo portamuestras se halla en un estudio previo de Segura-Noguera ("Relació entre Ia distribució de nutrients i oxígen dissolt i Ia composició elemental de fitoplancton a Ia Mar Catalana (N-O Mar Mediterránia). DESCRIPCIÓN DE LA INVENCIÓN
El objetivo de Ia presente invención es un portamuestras para microanálisis de rayos X con microscopía electrónica de barrido que permite conseguir espectros que contienen solamente los elementos de interés de Ia muestra, sin interferencias causadas por el material del soporte. Este portamuestras es especialmente útil en el análisis de muestras muy finas o muestras biológicas, aunque también se puede emplear para el análisis de partículas ambientales, minerales, cristales, y en general cualquier partícula u organismo o material que por su pequeño grosor sea atravesado por el haz de electrones del microscopio electrónico de barrido en el momento de su análisis. Además, el nuevo portamuestras permite utilizar las correcciones que se utilizan habitualmente en Ia microscopía electrónica de transmisión, ya que los fenómenos de absorción y fluorescencia se pueden despreciar, como se ha explicado en el apartado anterior.
De acuerdo con un aspecto de Ia invención, el portamuestras comprende una base sobre Ia que descansan unas columnas de apoyo que soportan una estructura de soporte para sostener Ia rejilla sobre Ia que se coloca Ia muestra.
La estructura de soporte puede ser de cualquier forma, siempre que sostenga Ia rejilla de un modo adecuado para colocar Ia muestra sobre ella. Por ejemplo, Ia estructura de soporte podría estar constituida simplemente por una
o varias ranuras dispuestas en el extremo superior de las columnas de apoyo para sujetar Ia rejilla, o bien por estructuras de forma cuadrada, triangular u otras. Sin embargo, de acuerdo con una realización particular de Ia invención, Ia estructura de soporte tiene una forma circular cuyo extremo superior está cerrado por una tapa que tiene un orificio central, y que sirve de apoyo para Ia rejilla. En realizaciones preferidas de Ia invención, el diámetro externo de esta estructura de soporte circular es de entre 11 mm. y 20 πm, mientras que el diámetro del orificio central de Ia tapa es de entre 7 mm. y 10 mm. Preferentemente, Ia estructura de soporte circular es un cilindro hueco cuya altura está entre 3 mm. y 6 mm.
La rejilla se fija a Ia tapa, preferiblemente empleando unos medios de sujeción, de modo que Ia muestra queda en el centro del orificio central de Ia tapa. Los medios de fijación pueden ser de cualquier tipo, como por ejemplo lengüetas, pinzas, ranuras, piezas en forma de tapón agujereado, u otros cualesquiera conocidos en Ia técnica.
La base puede tener cualquier forma, siempre que cumpla el objetivo de proporcionar un apoyo estable al portamuestras, aunque en una realización particular de Ia invención se trata de una base circular con un diámetro de entre 35 mm. y 45 mm., y preferiblemente con un espesor de entre 3 mm. y 6 mm.
Las columnas de apoyo pueden ser cualquier número y tener cualquier forma siempre que sostengan el cilindro superior por encima de Ia base en una posición adecuada para el análisis de Ia muestra mediante un microscopio electrónico. En una realización particular de Ia invención se utilizan tres columnas cilindricas de entre 1 ,5 mm. y 5 mm. que se apoyan en Ia periferia de Ia base y se doblan en ángulo recto para sostener el cilindro superior. Preferiblemente, su altura es de entre 23 mm. y 27 mm.
El portamuestras descrito puede estar fabricado en cualquier material, aunque es recomendable que dicho material no coincida con ninguno de los
materiales presentes en Ia muestra a analizar. En cualquier caso, y de acuerdo con una realización preferida de Ia invención, el material del portamuestras es aluminio.
Aunque no se describe específicamente en el presente documento, se entiende que el portamuestras de Ia invención comprende además un medio de acoplamiento adecuado para su fijación al microscopio electrónico que se vaya a emplear.
DESCRIPCIÓN DE LOS DIBUJOS
Para complementar Ia descripción que se está realizando y con objeto de ayudar a una mejor comprensión de las características de Ia invención, de acuerdo con un ejemplo preferente de realización práctica de Ia misma, se acompaña como parte integrante de dicha descripción, un juego de dibujos en donde con carácter ilustrativo y no limitativo, se ha representado Io siguiente:

REALIZACIÓN PREFERENTE DE LA INVENCIÓN
Se describe a continuación un ejemplo de un portamuestras (1 ) de acuerdo cono Ia invención haciendo referencia a las figuras adjuntas.
En este ejemplo, el portamuestras (1 ) comprende una base (2) de aluminio de 40 mm. de diámetro y 5 mm. de espesor, y que además tiene una rosca central de 4 mm. de diámetro (no mostrada en las figuras) para su fijación a soportes de microscopios electrónicos de barrido de Ia marca Hitachi.
Sobre Ia base (2) hay tres columnas (3a, 3b, 3c) de aluminio, equidistantes, de 4 mm. de diámetro y 24 mm. de altura. A esa altura las columnas (3a, 3b, 3c)
se doblan en ángulo recto hacia el centro del portamuestras (1 ) para sujetar Ia estructura (4) de soporte, que en este ejemplo es un cilindro sobre el cual se dispone una rejilla de 10 mm de diámetro (no mostrada en las figuras).
El cilindro, también de aluminio, tiene un diámetro externo de 15 mm. y una altura de 6 mm, estando cerrado en su extremo superior por una tapa (5) con un orificio central (6) de 9 mm. de diámetro. La rejilla que contiene las muestras queda sujeta a Ia tapa (5) mediante unos medios de fijación, que en este ejemplo comprenden un tapón agujereado de aluminio (no mostrada en las figuras) que encaja sobre Ia tapa (5). Las dimensiones del tapón agujereado de este ejemplo son: diámetro exterior 16 mm., diámetro interior 9 mm. y altura 3 mm.
Claims
1. Portamuestras (1 ) para el microanálisis de rayos X con microscopía electrónica de barrido, caracterizado porque comprende una base (2) sobre Ia que descansan unas columnas (3a, 3b, 3c) que soportan una estructura (4) de soporte que sostiene una rejilla sobre Ia que se dispone Ia muestra.
2. Portamuestras (1 ) de acuerdo con Ia reivindicación 1 , caracterizado porque Ia estructura (4) de soporte tiene forma circular cuyo extremo superior tiene una tapa (5) con un orificio central (6).
3. Portamuestras (1 ) de acuerdo con Ia reivindicación 2, caracterizado porque el diámetro externo de Ia estructura (4) de soporte circular es de entre 11 mm. y 20 mm.
4. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones 2-3, caracterizado porque el diámetro del orificio central (6) de Ia tapa (5) es de entre 7 mm. y 10 mm.
5. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones 2-4, caracterizado porque Ia estructura (4) de soporte circular es un cilindro hueco.
6. Portamuestras (1 ) de acuerdo con Ia reivindicación 5, caracterizado porque el cilindro tiene una altura de entre 3 mm. y 6 mm.
7. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones anteriores, caracterizado porque Ia base (2) es circular con un diámetro de entre 35 mm. y 45 mm.
8. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones anteriores, caracterizado porque Ia base (2) tiene un espesor de entre 3 mm. y 6 mm.
9. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones anteriores, caracterizado porque las columnas (3a, 3b, 3c) tienen entre 1 ,50 mm. y 5 mm. de diámetro.
10. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones anteriores, caracterizado porque las columnas (3a, 3b, 3c) tienen entre 23 mm. y 27 mm. de altura.
11. Portamuestras (1 ) de acuerdo con cualquiera de las reivindicaciones anteriores, caracterizado porque está hecho de aluminio.
