Definitions

  • the present disclosure relates generally to nucleic acid arrays, and more specifically to nucleic acid arrays that can be treated to prevent reuse.
  • the present disclosure provides methods for degrading arrays.
  • the methods involve treating an array with a degradative agent, where one or more polynucleotide probes of the array include a nucleotide analog that is susceptible to chemical modification by the degradative agent.
  • the array may be a microarray and/or a spatially- addressable array.
  • the nucleotide analog may be a base-modified nucleotide (e.g., deoxyuridine monophosphate (dUMP) or deoxyuridine triphosphate (dUTP)).
  • the degradative agent may be a glycosylase (e.g., uracil DNA glycosylase).
  • One embodiment provided is a method of preparing an array in which the probes contain one or more nucleotide analogs, which upon modification prevent or minimize the ability to reuse the array.
  • the probes on the array are exposed to an buffer solution that is designed to facilitate the removal or degradation of the probes from the array.
  • the buffer solution may contain, for example, a degradative agent that interacts with and/or modifies at least some, and preferably substantially all, of the nucleotide analogs present in the probes.
  • uracil DNA glycosylase refers to any enzyme that catalyzes the hydrolysis of the N-glycosylic bond between the uracil and sugar, leaving an apyrimidinic site in uracil-containing single or double-stranded DNA. It will be appreciated that once sufficient degradation of the probes has been achieved, the probes can then be degraded from the array to prevent reuse of the array. In certain embodiments, the chemically modified probes are degraded and/or removed from the array using heat or protease treatment, or other methods well know to those of skill in the art. As discussed above, the degrading conditions are sufficiently stringent to remove substantially all or at least some portion of the probes from the array.
