Abstract
An optical system and a fibre scanning system 14 for a confocal microscopy systemwhich may be adapted to employ nonlinear opticaleffects like 2-photon fluorescence and second harmonic generation imaging are disclosed. The opticalsystemcomprises a first lens arrangement (10a), a second lens arrangement (10b) and an optical fibre (4). The first and second lens arrangements (10a,b) form an optical entity (7) being arranged such that light emitted from the optical fibre (4) enters the optical entity (7) on a first side and is transmitted to a second side. Therein, the optical entity (7) comprises an optical magnification m and a numerical aperture NA2 on the second side wherein the optical entity satisfies the condition |m * NA
2| < 0.1. Thereby, the optical system can be adapted, e.g. by using a tiltable scanning photonic crystal fibre (4) that is moved in front of the optical entity (7) and a liquid lens having a variable focus, for the acquisition of a three-dimensional volume scan of a sample (8) to be examined.
