Abstract

Disclosed is a generally applicable method for improving the stability of a protein molecule, including thermal stability, chemical stability against a modifying agent and resistance against a protease, compared with the intrinsic stability of the protein molecule, without deteriorating the original functions of the protein molecule. The method is for producing a useful mutant form of a protein, and comprises the steps of: selecting, as a target site for mutation, an amino acid residue located on a partial segment whose three-dimensional structure is stabilized by the interaction among adjacent amino acid residues located on the amino acid sequence for the protein, by using a three-dimensional structure coordinate data from the protein; selecting, as other amino acid residue by which the target site for mutation is to be substituted, an amino acid residue having high frequency of occurrence in a conformation that is the same as or similar to the conformation of the partial segment; and substituting the target site for mutation by the selected amino acid residue.
