発明が解決しょうとする課題
[0008] 本発明は、 LPS等の TLRファミリ一と IL-1受容体ファミリ一に共通のシグナル伝達に 使用される細胞内因子アダプター分子 MyD88の機能を細胞内でシグナル特異的に 制御する手法を提供することを目的とする。
課題を解決するための手段
[0009] IL- 1/1い 18/TLRシグナル伝達における TIRドメインの重要な役割にもかかわらず、 本願の優先日(2007年 8月 1日)の時点で、構造データは受容体の TIRに関するものし 力利用できなかった。細胞内アダプタ一分子の TIRについては、そのいずれについ ても原子構造の決定はなされていな力つた。したがって、これらのアダプターの TIRド メインの構造情報は、相互作用機構をより正確に理解するために必須である。そこで 、 IL-1R/IL-18R/TLRシグナル伝達のより良い理解への一歩として、本発明者らは、 溶液 NMR分光法を用いてヒト MyD88の TIRドメインの溶液構造を決定した。この構造 は、 IL-1R/IL-18R/TLRシグナル伝達における TIR含有アダプタ一タンパク質の生物 学的機能をさらに研究するために必要な基礎を提供する。また、本発明者らは、培養 細胞を用いて MyD88 TIRの一連の機能分析を実施した。これらの分析は、 MyD88が I L- 18および TLR4シグナル伝達経路で機能するために決定的な数個のアミノ酸残基 を決定することを可能とした。興味深いことに、 MyD88 TIRの 3つの共通な相互作用 部位が同定され、そして 1つのさらなる部位は IL- 18シグナル伝達にのみ機能した。
[0010] MyD88は deathドメインと Toll Interleukin - 1 Receptor homology (TIR)ドメインからな る 2ドメイン蛋白である力 S、 deathドメインを欠失させ TIRドメイン (148- 296)のみ細胞に 発現させると、 IL-18を含む IL- 1ファミリ一や TLRのシグナル非特異的ドミナントネガテ イブインヒビタ一として働くことが知られている。この MyD88 TIRドメインのリシン 214とリ シン 238とアルギ-ン 269をァラニンに置換した変異体 MyD88 TIR K214Aと MyD88 TI R K238Aと MyD88 TIR R269Aを作製し、その機能を培養細胞上において、 NF- κ Β Luciferase reporter gene assayで検討した結果、 TLR4を介する LPSからのシグナルは 遮断できるが、 IL-18からのシグナルは遮断できないことがわ力 た。本発明は、これ らの知見に基づいて、完成されたものである。なお、本願の優先日(2007年 8月 1日) 以降に、 Rossiらにより解析されたヒト MyD88TIRの構造力 PROTEIN DATA BANKに 登録された(登録日: 2007年 6月 29日、公開日: 2007年 10月 23日、登録番号: 2js7)が 、その公開は本願の優先日の後であった。
本発明の要旨は以下の通りである。

[0015] 本発明は、細胞内シグナル伝達タンパクの機能的変異導入箇所の情報を提供する IL-18は、 IFN- γの産生を誘発する等、細胞性免疫機能において重要な分子であ ることがわ力つている力 そのシグナル伝達経路は図 8のように LPSを認識する受容 体である TLR4等と共通している。 LPSは、グラム陰性菌の細胞膜に由来する物質で あるが、生体内においてエンドトキシンショックを誘発する等、毒性を発揮する物質で ある。本発明者らが作製した MyD88 TIR K214A、 K238A、 R269A変異体は、 IL-18シ グナル伝達は阻害しないが、 LPSによる TLR4シグナル伝達は阻害する。従って、この 変異体分子は、 IL- 18による細胞性免疫機能を保持したまま、 LPSによる直接のェン ドトキシンショック等の病態発症を抑えることが期待できる。
発明の効果
[0016] 従来では、 LPSによる毒性の治療には、エンドトキシン除去ゃ抗炎症薬等での治療 が行われていたが、新たな治療アプローチとして、遺伝子レベルで細胞に組換え蛋 白を発現させることで、 IL - 18による細胞性免疫機能を維持したまま LPSによる炎症性 サイト力インの産生を制御することが可能となった。
また、逆に、この MyD88 TIRドメインの K214A、 K238及び R269を標的とする低分子 やペプチドは、 IL-18に特異的な作用をもたらす可能性がある。
本明細書は、本願の優先権の基礎である日本国特許出願、特願 2007-200776の明 細書おょぴ Zまたは図面に記載される内容を包含する。

発明を実施するための最良の形態
[0018] 以下、本発明の実施の形態についてより詳細に説明する。
1. MyD88 TIR変異体タンパク質
本発明は、以下の (a)、(b)若しくは (c)の MyD88 TIR変異体タンパク質又はその塩を 提供する。

[0022] 本明細書において、「リポ多糖刺激のシグナルを阻害する」とは、ほ乳類細胞にお レ、て、リポ多糖刺激による転写因子の活性化及び、その結果としてのサイト力イン産 生等の細胞の反応の抑制を意味する。リポ多糖刺激のシグナルを阻害するか否かは 、リポ多糖を認識する受容体を持ったほ乳類細胞に転写因子の結合認識部位の配 列を組み込んだレポ一ター遺伝子べクタ一を形質転換により導入した後、リポ多糖刺 激を加え、レポ一ター遺伝子の発現量を比較検討することにより、調べることができる 。例えば、リポ多糖を認識する受容体を持ったほ乳類細胞に、目的のタンパク質をコ —ドする配列と転写因子の結合認識部位の配列を組み込んだレポ一タ一遺伝子べ クタ一を形質転換により導入した後、リポ多糖刺激を加え、レポ一ター遺伝子の発現 量を測定し、目的のタンパク質をコードする配列を組み込んでいない形質転換細胞 におけるレポーター遺伝子の発現畺と比較して、発現量が減少していれば、リポ多糖 刺激のシグナルを阻害すると判定することができる。
[0023] また、「インタ一ロイキン 18刺激のシグナルを阻害する」とは、ほ乳類細胞において 、インターロイキン 18による転写因子の活性化及び、その結果としてのサイト力イン産 生 (特にインターフェロン Ί;)等の細胞の反応の抑制を意味する。インターロイキン 18 刺激のシグナルを阻害する力否かは、インタ一ロイキン 18を認識する受容体を持った ほ乳類細胞に転写因子の結合認識部位の配列を組み込んだレポーター遺伝子べク ターを形質転換により導入した後、リポ多糖刺激を加え、レポ一ター遺伝子の発現量 を比較検討することにより、.調べることができる。例えば、インタ一ロイキン 18を認識す る受容体を持ったほ乳類細胞に、目的のタンパク質をコードする配列と転写因子の 結合認識部位の配列を組み込んだレポーター遺伝子ベクターを形質転換により導 入した後、リポ多糖刺激を加え、レポーター遺伝子の発現量を測定し、目的のタンパ ク質をコードする配列を組み込んでレ、なレ、形質転換細胞におけるレポ一タ一遺伝子 の発現量と比較して、発現量が減少していれば、インターロイキン 18刺激のシグナル を阻害すると判定することができる。
[0024] (c)のタンパク質について、配列番号 3、 5又は 7のヌクレオチド配列力 なる DNAに 相補的な DNAとストリンジェン卜な条件下でハイブリダィズする DNAは、配列番号 3、 5 又は 7のヌクレオチド配列力 なる DNAに相補的な DNAの全部又は一部と少なくとも 8 0% (好ましくは、少なくとも 95%、より好ましくは、少なくとも 98%)の同一性があるとよい。 ノ、イブリダィゼ一シヨンはストリンジェン卜な条件下で行われる。核酸二本鎖又はハイ プリッドの安定性は、融解温度 Tm (プローブが標的 DNA^ら解離する温度)で表され る。この融解温度はストリンジェントな条件を定義するために用いられる。 1%のミスマツ チにより Tm力 S1°C低下すると仮定すると、ハイブリダィゼーシヨン反応の最終洗浄の 温度を低くしなければならない。例えば、プローブと 95%以上の同一性を有する配列 を求める場合には、最終洗浄温度を 5°C低くしなければならない。実際、 1%のミスマツ チにっき、 0.5〜1.5での間で1¾が変ゎることになる。ストリンジェン卜な条件の例として は、 5x SSC/5xデンハルト溶液 /1.0% SDS中 68°Cでハイブリダィズさせ、 0.2x SSC/0.1 %SDS中室温で洗浄することである。中程度にストリンジェントな条件の例としては、 3x SSC中 42°Cで洗浄することである。塩濃度や温度は、プローブと標的核酸との同一性 の最適なレベルを達成するために変更されうる。このような条件に関するさらなる指針 として、 Sambrook et al., 1989, Molecular Cloning, A Laboratory Manual,し old Spnn g Harbor Press, N.Y.; and Ausubel et al. (eds.), 1995, Current Protocols in Molecul ar Biology, (John Wiley & Sons. N.Y.) at Unit 2.10を参照されたい。
[0025] 本発明の MyD88 TIR変異体タンパク質としては、配列番号 8のアミノ酸からなるタン パク質 (MyD88 TIR K214A)、配列番号 4のアミノ酸からなるタンパク質 (MyD88 TIR K 238Α)、配列番号 6のアミノ酸からなるタンパク質 (MyD88 TIR R269A)などが好ましい 本発明の MyD88 TIR変異体タンパク質は、ヒト由来の野生型 MyD88 TIRタンパク質 の変異体であっても、ヒト以外の動物由来の野生型 MyD88 TIRタンパク質の変異体 であってもよレ、。
[0026] 本発明の MyD88 TIR変異体タンパク質の塩は、薬理学的に許容される塩であるとよ ぐ特に、薬理学的に許容される酸付加塩が好ましい。薬理学的に許容される酸付 加塩としては、無機酸 (例えば、塩酸、リン酸、臭化水素酸、硫酸)との塩、有機酸 (例 えば、酢酸、ギ酸、プロピオン酸、フマル酸、マレイン酸、コハク酸、酒石酸、クェン酸 、リンゴ酸、蓚酸、安息香酸、メタンスルホン酸、ベンゼンスルホン酸)との塩などを例 示することができる。
[0027] 本発明の MyD88 TIR変異体タンパク質及ぴその塩は、 IL-18の作用は遮断せずに 、 LPSの作用を特異的に遮断することが可能である。従って、本発明の MyD88 TIR変 異体タンパク質及びその塩は、 Toll様受容体及び IL-1/18シグナル伝達に関与する 疾患、具体的には、 LPSが誘発する疾患(例えば、エンドトキシンショック、 SIRS (syste mic inflammatory response syndrome)等の予防及び/又は治療に用いることができ る。また、 Toll様受容体及び IL- 1/18シグナル伝達の研究などを目的とする試薬とし て利用することもできる。
[0028] 本発明の MyD88 TIR変異体タンパク質及ぴその塩は、公知の方法によって製造す ることができる。例えば、後述の 2に記載のようにして MyD88 TIR変異体タンパク質を コードする DNAを得、得られた DNAを適当な発現ベクターに組み込んだ後、適当な 宿主に導入し、組換え蛋白質として生産させることにより、 yD88 TIR変異体タンパク 質を製造することができる(例えば、西鄉薰、佐野弓子共訳、 CURRENT PROTO COLSコンパクト版、分子生物学実験プロトコ一ル、 I、 II、 III、丸善株式会社:原著、 Ausubel, F. M.等, Short Protocols in Molecular Biology, Third ition, John Wiley & Sons, Inc. , New Yorkを参照のこと)。

また、当業者であれば、本発明の MyD88 TIR変異体タンパク質をドメインとして含む 機能性タンパク質(例えば、 MyD88、 TIRAP(Mal), TRIF(TICAM - 1)、 TRA (TICAM-I 1)、 SARMなど)を製造することが可能であり、このようなタンパク質も本発明の範囲に 含まれるものとする。

実施例
[0057] 以下、実施例に基づいて本発明を詳細に説明するが、本発明はこれらの実施例に 限定されるものではない。
〔実施例 1〕
材料および方法
サンプルの調製
ヒト MyD88の TIRドメイン (148- 296)をコードする遺伝子は、ヒトリンパ球から mRNAを 抽出し、逆転写酵素で cDNAを合成し、さらに PCR法を利用して増幅した。これを pGE X-5X-3 (Amersham)プラスミドベクター中にクローン化した。このプラスミドを大腸菌 B L-21 (DE3)株(Novagen)に形質転換した。この大腸菌のシングルコロュ一を選択し、 安定同位体元素で標識した栄養豊富な増殖培地 (Silantes)に接種した。 A600力 -0.5に達するまで大腸菌を 37°Cで培養した。次に培養物の温度を 25°Cまで下げ、ィ ソプロピル- β _D-チォガラクトシド (IPTG)を最終濃度が 1 mMとなるように添加すること によりタンパク質発現を誘導した。誘導 16時間後に 4°Cで 6,000 X gで 10分間遠心して 集菌し、細胞ペレットを精製まで- 80°Cで保存した。この細胞ペレットを溶菌バッファ —[20 mM Tris HC1; 0.4 M C1, 1 mM EDTA (pH 8.0), 0.05% 2—メルカプトェタノ一 ルおよび 1 mM Pefabloc (Roche)]に再懸濁し、超音波処理によって溶菌した。細胞 破砕物および不溶物を遠心(4°C、 6,000 X g、 10分間)により除去した。ダルタチオン - S-トランスフェラーゼ (GST)融合タンパク質として発現した MyD88の TIRドメインをまず ァフィ二ティークロマトグラフィーにより精製し、そして Factor Xaプロテアーゼ (Pharma cia)を用いた消化により GSTタグを除去した。次に、 GSTタグと MyD88の TIRドメインに 分離したタンパク質を Sephacryl S-100 HR 26/60カラム(Pharmacia)および 20 mM リン酸カリウムバッファー(pH: 6.0) (150 mM KC1, 0.1 raM EDTAおよび 10 mM DTT 含有)を用いてゲル濾過した。 MyD88タンパク質の単量体性 TIRドメインをプールし、 1 0 kDa Microsep microconcentrator (Amicon)を用いた限外濾過により濃縮した。 Mon o-Sカラム (Pharmacia)を用いたイオン交換クロマトグラフィーにより、さらに精製を行つ た。この精製プロトコ一ルを用いて、 15Nで標識した、および 13C、 15Nで二重標識した、 MyD88タンパク質の野生型 TIRドメインを調製した。タンパク質収量は、栄養豊富な増 殖培地 1リットルから約 1.0 mgであった。これらのタンパク質サンプノレは凝集体を形成 する傾向があるように思われたので、これらのタンパク質のサンプルバッファ一を 20 m リン酸カリウムバッファー(pH: 6.0) (0.1 mM EDTA, 10 mM DTT, 50 mM重水素 化 L-アルギニンおよび 50 mM重水素化 L-グノレタミン酸含有)と置換した (引用文献 # 1)。最終的に NMR実験に用いたタンパク質サンプルの最終濃度は、約 0.3 mMであつ た。

全ての NMRスペクトルは、 CryoProbeを備えた Bruker DRX500または DRX800分光 計を用いて 25°Cで記録した。タンパク質の主鎖および側鎖1 H, 13Cおよび 15N共鳴の 帰属を決定するため、三重共鳴 NMRを実施した。構造計算のための距離拘束は、 3 D 15N edited NOESYおよび 3D 13C edited NOESY実験 (混合時間 150 msec)より得た。 NMRPipe.ソフトウエアパッケージ (弓 I用文献 #2)を用レ、て NMRスぺクトルをプロセシング し、 Sparky (引用文献 #3)を用いて分析した。 ^- Ν定常状態ヘテロ NOE値を得るため に用いたパルスシーケンスは、以前に記述されている (引用文献 #4)。

溶液 NMRを用いた MyD88 TIRドメインの構造決定
TIRドメイン間の配列保存は、 3個の短いモチーフ(図 1の Boxl、 2および 3 29)に絞ら れ、そしてこれらのモチーフに焦点をあてた機能研究が各 TIRドメインについて行わ れている (引用文献 #10、 11、 17、 18、 19、 20)。 TIRドメインの構造的コアは、保存され た (F/Y) DAアミノ酸モチーフ力も始まる (引用文献 #19)。本研究においては、溶液 NM Rによる構造決定のために、ヒト MyD88の残基 148- 296からなる構築物が作製された。 ヒト MyD88の TIRドメイン (残基 157- 296)の立体構造を図 2に示す。最も低い全ェネル ギーを有する 20個の構造の統計値を表 1に要約する。上記 20個の構造の主鎖重原 子(N、 C cおよび C,)の距離二乗平均根偏差値 (rmsd)は 0.45オングストロームである (残基 157- 185、 188- 194および 203- 295を用いて重ね合わせた)。 N末端の 9個のァ ミノ酸 (148 - 156)は、この領域における中程度〜長距離範囲の NOESYクロスピークの 欠失によって示される、ランダムコイル傾向を示した。よって、これらの残基は図から 除外した。 2D Ή-Ν HSQCスペルトルにおいて、 G201を除く D195から C203までの残 基についてアミドクロスピークが欠失しており、これは BB-ループと対応している(図 1) 。側鎖プロトンの一部が観察され、この BB-ループ領域に帰属された力 NOESYクロ スピークの数は不十分で、この領域にっレ、ては構造収束 (convergence)が比較的悪 かった。 15Nヘテロ核 NOE実験は、これらの欠失した残基 G201、 V204および S206 の近くに存在する残基のアミド基が比較的低い NOE値を示す(それぞれ 0.67、 0.55お よび 0.67)ことを示した。これは、 BB-ループがおそらく堅固なコンホメ一シヨンをとつて おらず、溶液状態では緩やかに構造が保たれてレ、ることを意味する(図 2A)。
[0071] MyD88 TIRドメインの構造説明および他の TIRとの比較
MyD88 TIR(157-296)の構造は、 4本の αヘリックス( a A- ct C、および α Ε)に囲ま れた、中心部に位置する 5本のストランドからなるパラレル シート( /3 A- ]S Ε)を構成 する。予測されたように、全体的な折りたたみは、以前に決定された TIRドメインの結 晶構造と本質的に同一であった (PDBコード: 1FYV, 1FYWおよび 1T3G) (図 2B)。 MyD88 TIRは、他の TIRタンパク質と比較して、 TLR2 TIRのアミノ酸配列と最も相同 性が高い(MyD88 TIRおよび TLR2 TIR間の同一性スコアは 22%で、相同性スコアは 4 8%である)。図 2Cおよび 2Dは、 MyD88 TIRおよび TLR2 TIRを重ね合わせて表したも のである(PDBコード: 1FYW)。 ]3シートコアの構造は、相互に類似している(この部 分の主鎖 N、 C aおよび C,に対する rmsd値は 0.90オングストロームである)力 レ、くつ かの領域では注目すべき非類似性が示された。 BB-ループ(MyD88 TIRの Serl94_C ys203)およびそれに続く α Βまでの残基には、大きな構造的不一致が存在する(図 2 D)。 MyD88および TLR2の両方において BB-ループは溶液に露出されている力 ル —プが向いている方向は大きく異なる。これは、 MyD88の α Βに先行する残基 (MyD8 8の Trp205-Ser209)の構造的相違によって主として引き起こされる。これらの残基は、 MyD88において伸長された βストランド様構造をとっている力 他方 TLR2において対 応する残基は αヘリックス( α Β)内に存在し、 MyD88の α Βを有意に短くしてレ、る。こ の βストランドは、 ΒΒ -ノレ一プを TLR2 TIRにおいて見られる方向とは全く異なる方向 へ向けることになる。
[0072] この伸長された βストランドは、 15N edited 3D NOSEYスぺクトノレにおけるクロスピーク のパターンによって十分支持される。この 3D NOSEYスペクトルにおいて、一連の NO Eクロスピークの強度(H a (i)から HNG+1))は、残基内強度(H α (i)から HN(i))よりも有 意に強かった。 C αおよび C βのケミカルシフト値もまた、この二次構造を支持した。
[0073] MyD88は TLR2に存在する a Dヘリックスの代わりに長いループ(DD- DEロングル一 プ)を有する。配列ァライメントにおいて、ひ Dが存在する領域に多数のギャップが見 い出されたため、 a Dの欠失は予測された。(図 1)。しかし、この長いループには、 α ヘリックスに類似した短いヘリックスコイルコンホメーシヨン (Ρ265- 1271)が存在する。
[0074] TIRドメインの表面特性の比較: MyD88 TIRおよび他の TIR
図 3は、 TIRドメインの表面静電ポテンシャルを示す。 MyD88 TIRの全体的な蛋白構 造の折りたたみは他の TLR TIRドメインに類似しているにも力かわらず、表面静電ポ テンシャルには顕著な相違が見られる。特に、 MyD88 TIRの Box3には大きな陽性ポ テンシャル領域が認められる。実際、 MyD88 (148-296) TIRドメインの計算された等 電点 (pi)は 9.25 (全長 MyD88では 5.79)なので、このドメイン自体が陽性に荷電されて いる。 MyD88は細胞の細胞質に存在する (引用文献 #21)。 yD88 TIRの大きい陽性 ポテンシャル領域は、このドメインが陰性に荷電された細胞膜を標的とするのを容易 にするであろう。それは、シグナルリガンドが IL- 1R/IL-18R/TLR等の膜受容体と結合 したときに、このドメインがそれら膜受容体と結合するのを促進するであろう。 Box3とは 対照的に、 BB-ループを含む Box2領域は、狭い陰性ポテンシャルに覆われる領域を 形成している。

MyD88 TIRの機能に関する洞察を得るため、本発明者らは IL - 18および LPSで刺激 した後に NF- κ Bレポーター遺伝子アツセィを実施した。本発明者らは、以前に刊行 されたデータに基づいて、 myc_MyD88 TIRに単一アミノ酸置換を組織的に導入し、 I L-18または LPS刺激後のルシフェラ一ゼ活性を定量した。 NF- κ B活性は、細胞中の MyD88の機能に対する突然変異の作用を反映するであろう。選択された 25個の荷電 極性アミノ酸と 3個の疎水性アミノ酸力 なるァラニン置換突然変異体を評価した (表 2)。 IL-18を用いた NF- κ Bルシフェラーゼレポ一タ一アツセィの結果を図 4Aに示す。 この図に示すように、ドミナントネガティブ遺伝子をトランスフエクトしな力 た対照細 胞では、 IL-18刺激の結果、ルシフェラーゼ活性が約 3倍に増大した。対照的に、野 生型 myc-MyD88 TIRを細胞中で過剰発現させると、 NF- κ Β活性の重大な増加は見 られず、これは当該タンパク質のドミナントネガティブ阻害作用を示している。レ、くつ かの突然変異体(例えば、 R196A, Κ238Α, R288A)は、 NF- κ Β活性の重大な増加を 示した。これは、ドミナントネガティブ阻害作用の喪失を意味し、突然変異させた残基 のタンパク質-タンパク質相互作用における重要性を示している。突然変異体のドミ ナントネガティブ阻害作用は、統計学的分析により野生型と各変異体とを比較してか ら有意と判定された。 MyD88は一般に、 IL-1R/IL-18R/TLRシグナル伝達およびさら に多種類の分子(ただし、 MyD88に結合可能な IRFファミリ一等の TIRドメイン含有受 容体を除く)にリクルートされる (引用文献 #22、 23、 24)。
[0076] ひとつの分子が複数の蛋白と相互作用可能であることおよびそのようなシグナル多 様性の土台をなす機構は、非常に興味深い。 IL-18の場合と同様に、 LPS/TLR4シグ . ナル伝達もまた MyD88によって媒介され、 NF- κ Β活性化をもたらす。それゆえ、本 発明者らのレポーターアツセィ系を TLR4シグナル伝達の研究に適用することができ る。レポ一ターアツセィの結果を図 4Βに示す。ドミナントネガティブ阻害作用は、 IL-1 8で刺激した場合も LPSで刺激した場合も、突然変異体 R196A、 D197A (BB -ループ の突然変異)、 R217A (BC -ループ)および K282A、 R288A ( α E)において共通して 消失した。さらに、 IL-18に特異的なドミナントネガティブ阻害作用の消失は、 K214A ( BC-ループ)、 238Α ( a C,)および R269A (DD-DE長ループ)に観察された。これ らのレポーターアツセィの結果および TIRタンパク質の機能に関する以前のリポートを 以下に要約する。
[0077] Boxl :保存された Boxl領域は、 IL-18および TLR4シグナル伝達の両方において 重要な役割を果たすものではなレ、。なぜなら、レポ一タ一アツセィにおいて何ら重要 な変化が検出されなかったからである。最近、 IRF-1/4/5とのある相互作用力 この 領域を含む MyD88の一部分について報告された (引用文献 #22、 23、 24)。この領域 は NF- κ Bではなぐインタ一フエロン産生のシグナル伝達系に寄与しているの力もし れない。また、 Boxl領域に隣接して、 R217Aは顕著なドミナントネガティブ阻害作用 の喪失を示し、機能的に重要である。 [0078] Box2 (BB-ループおよび Pro200):初期の研究は、 TLRの TIRドメインの BB-ループに 存在する保存されたプロリンの重要性を示した (引用文献 #12、 18)。 Liらもまた、 MyD 88の Box2における突然変異力 TIRドメインを介する同型な MyD88オリゴマー化を阻 止したと報告してレ、る (引用文献 #10)。 BB -ループの重要性は、本発明者らのレポ一 タ一アツセィで確認された。なぜなら、 R196Aおよび D197Aにおいて重大な効果が観 察された力もである(図 5A:)。また、この領域力TIRドメイン蛋白の機能に重要であるこ とを示唆する情報として、 TLR2の Arg677 (MyD88の R196に対応する)は、ハンセン病 患者において Trpに変わることが見い出された力もである (引用文献 #13)。 yD88 TIR の BB-ループ領域の構造は、 TLR1および TLR2の TIRドメインの結晶構造に見られる ものとは全く異なっていた(図 2Cおよび 2D)。 NMR実験は、 MyD88 TIRドメインの BB- ループにおけるフレキシビリティーの存在を提案した。このことが当該ドメインに機能 的多様性をもたらすのカ^しれなレ、。なぜなら、フレキシブルな構造は、相互作用の パートナーに比較的広い特異性を許すからである。実際、 MyD88 TIRは多数の異な る TIRドメインと相互作用することが知られている。 P200H突然変異の効果もまた、わ れわれの研究で試験した。爽味深レ、ことに、この突然変異は IL-18シグナル伝達に特 異的に影響するが (R丄. U.値: mean土 s.d., 1.79±0.21)、 TLR4シグナル伝達には影 響しなレ、 (R丄. U.値: mean土 s.d., 1.24±0.26)。このことは、この保存されたプロリンが 、 TIRタンパク質間の相互作用に必要ではあるが十分ではなレ、ことを意味する。 Brow nおよび Dunneは、このプロリンカ ^IR相互作用に常に必須な訳ではないと述べた( 引用文献 #17、 25)。
[0079] α B and BC loop:この領域に存在する数個のアミノ酸の重要性もまた Ronni et al. ( 引用文献 #20)によって TLR4 TIRについて提案されている。 この領域では、 MyD88の 詳細な構造は、他の TIR構造と比較して特徴的である。 α Βヘリックスは他の TIR構造 と比較して短く(構造説明の項を参照)、そしてこのへリックスに続く BC-ループはより 長レ、。本発明者らは BC-ループ領域で、 K214A、 R215Aおよび R217Aについてァラ ニン置換変異体を作製した。この領域で、 K214Aは IL- 18シグナル伝達に特異的な 効果を示した。 R217Aは、上記刺激の両方に対して重大な効果を示した。古典的に は、 Boxl、 Βοχ2および Βοχ3と呼ばれる TIRドメインの間で保存されたアミノ酸配列は、 TIRドメインの機能的に重要な部位であると言われていた。 R217は、 Boxlの F/YDAモ チーフを構成する D162の隣接アミノ酸である力 本発明者らはこの BC-ループ領域 力 MyD88 TIRの新しい相互作用部位の 1つであることを決定した。さらに、この領域 におけるこれらのアミノ酸の選択性は、 MyD88と直接相互作用する膜受容体におけ るアミノ酸残基の違いに由来する力もしれない。
[0080] a C : TLR2の BB-ループ、 a C 'および DD-ループに突然変異を数個導入すると、 ペプチドダリカンによって刺激された NF- κ B活性が失われると報告されている (引用 文献 #26)。さらに、ヒト IL-1RAPLの TIRドメインの二量体構造において、 TLR2の Cys7 13に対応する Ser488は、二量体界面の中心に位置する他の一量体の等価な Ser488 'と堅固な水素結合を形成する (引用文献 #27)。本発明者らの研究において、 a C 'に おける K238から Alaへの置換の効果は、 IL- 18シグナル伝達に特異的な効果を示し 、 LPSシグナル伝達には影響しなかった(図 5)。ただし、 K231A、 E232Aおよび D234 Aは NF- κ B活性の重大な効果を示すことが出来なかった。
[0081] a D:他の TIRドメインの a Dに対応する αヘリックスは、 MyD88 TIRには認められなか つた。このへリックスの欠如は、 TIRドメインの配列ァライメント(図 1 )によれば驚くにあ たらない。このァライメントでは、 i3 Dと J3 E間の領域の多数のギャップが見い出される 。 DD-DEロングループの Arg269を Alaに置換した場合、ドミナントネガティブ作用の喪 失が IL-18刺激に特異的に観察されたが、 LPS刺激に対しては観察されな力つた(図 5B)。 TLR2において同じ部分に相当する Argが Ginに置換された突然変異は、ブドウ 球菌感染に対する免疫不全を弓 Iき起こすことが報告されている (引用文献 #28)。
[0082] Βοχ3 ( α E helix): a Eもまた、 TIRドメインの間でアダプタ一-アダプタ一および受容 体-受容体オリゴマー化を媒介することが報告されてレ、る (引用文献 #10、 11)。 α Εに 含まれる極性残基をァラニンに置換させた場合 (Κ282Αおよび R288A)、本発明者ら の研究では IL-18刺激および LPS刺激の両方とも重大な効果をきたした(図 5)。 F285 およ I W286は TIRドメインの間で 存されており、 FWモチーフと呼ばれている。この FWモチーフにおけるァラニン置換は二量体形成にとって決定的であると以前に報告 されてレ、ることは注目に値する。しかし、 F285Aはレポーターアツセィにおいて、ドミナ ントネガティブ作用に対して何ら影響を示さなかった (R丄. U.値: mean土 s.d., IL-18: 1.04±0.10, LPS: 1.17±0.13)。そして、本発明者らは W286について全く情報を得る ことができな力つた。なぜなら、 W286A変異体タンパクは細胞中で非常に不安定だつ た力 である。本発明者らは、シクロへキシミド法を用いて、 W286Aタンパク質発現レ ベルおよび安定性を確認した(図 6)。
[0083] 考察
TLR TIRおよび MyD88 TIRの構造
TIRドメインは、典型的には 135〜160アミノ酸によって構成される。 TIRドメイン間の 配列保存は一般に 20から 30%の間であり、そして保存された残基のほとんどは、 5本の ストランドからなるパラレル i3シートによって構成される疎水性コア内に存在する。そ の長さの違い、および TIRドメイン間のアミノ酸配列におけるかなりの変化は、このドメ インの構造的および機能的多様性を示唆する。現在、 TIRドメインについて 3つの構 造 (TLRl, TLR2および IL- 1RAPL)が入手可能である力 これらはすべて受容体の ものである。本研究は、細胞アダプタ一タンパク質由来の TIRドメインの最初の構造決 定である。 MyD88 TIRは、受容体 TIRに対しかなりの類似性を示す(図 2Cおよび 2D) 。しかし、「結果」の項に記述したように、構造には相違点が認められる。 BB -ループ に認められる構造上の不一致は、顕著でかつ興味深レ、(図 2D)。これは TIRドメインの 機能的多様性を意味する。なぜなら、多数の研究力 Sこれらの領域 (Box2に対応する( 引用文献 #10、 11、 18、 19、 20))の機能的重要性を提案してレ、る力もである。他の構 造不一致領域 ( c B, BC-ノレ一プおよび DD- DE長ループ)もまた、機能的に重要であ る。さらに、 MyD88 TIRの表面における大きい陽性荷電領域の存在は興味をそそるも のであり、これは MyD88の機能と関連してレ、るの力もしれなレ、。
[0084] MyD88のマルチ相互作用部位および MyD88 TIR: IL- 18 vs LPS/TLR4 MyD88に関 して示された受容体特異的シグナル伝達
MyD88は、細胞中の IL- 1R/IL-18R/TLR媒介シグナル伝達経路に通常使用されて いる。しかし、細胞の応答はどの受容体が刺激されたかによって異なる。したがって、 MyD88のもとで細胞の運命を分ける、ある種の作用機構が存在するはずである。 IL- 1R/IL-18R/TLR媒介シグナル伝達における MyD88のシグナル伝達機構を解明する 目的で、本発明者らは培養細胞を用いて MyD88の機能分析を実施した。その結果 に基づいて、本発明者らは IL-18刺激おょぴ LPS刺激の両方にとって重要な数個の 残基を同定することができた。図 5においては、 yD88 TIR構造の表面にレポーター アツセィの結果がマッピングされている。この図には荷電残基のみをマッピングしたこ とに注意されたレ、。緑色に着色したアミノ酸残基は、 IL- 18および LPSシグナル伝達に 重要であると判断された。茶色に着色したアミノ酸残基は、活性に何ら影響を及ぼさ ない残基を示す。 3つの相互作用部位(保存された Box2、 Box3および BC -ループの 終末)は、 IL- 18および LPS/TLR4経路の両方で同定された。さらに、 IL- 18シグナル 伝達において 1つの特異的な表面が同定された。 BC-ループの一部、ひ C 'ヘリック スおよび DD- DEロングループは、 MyD88 TIRドメインの分子表面でごく近位に存在し た。 TLR2 TIRに関する別の研究もまた、 C713S、 L717E (これらは a Cに存在する) および R748E、 R748S (これらは DDループに存在する)カ NF- κ Β活性をダウンレギ ユレーシヨンすることを示した。その著者らは、このダウンレギュレーションは変異導入 によって引き起こされるこの領域での TLR2ホモ二量体化の抑制によるものであると提 案した (引用文献 #26)。本発明者らのレポーターアツセィは、これらの報告とよく一致 した。 c C '近傍に群がる 3個の塩基性残基(Lys214, Lys238,および Arg269)は、 IL -18シグナルに対して重大な効果を示した。しかし、これらの残基は LPS-TLR4シグナ ノレ伝達には関与しない。本発明者らは、 IL-18または LPSに対する特定の細胞応答 は、 MyD88 TIRドメインの異なる相互作用表面と関連しているらしレ、と推定する。 MyD 88下のシグナル多様性の理由の一部は、この表面パッチに帰する力もしれない。 本研究において、本発明者らは溶液 NMR分光法を用いて、ヒト MyD88 TIRドメイン の溶液構造を決定した。これは、細胞アダプタ一分子の TIRドメインとして初めて明ら かにされた構造である。本発明者らはまた、培養細胞系を用いて MyD88 TIRドメイン の機能アツセィを実施した。これらのデータは、このドメインの保存された Boxl、 Box2 および Box3領域は、 IL-18および LPS経路の両方にとって重要な相互作用部位であ ることを明ら力 4こするものである。さらに、 a C 'およびその近傍に群がる 3個の塩基性 残基からなる 1つの表面は、もっぱら IL-18シグナル伝達のために機能しており、 TLR 4シグナル伝達のためには機能しない。最近、 IL-18の広範な免疫調節機能 (IFN - γ の誘導および Thl分化の増強のみならず、 IgE抗体の誘導およびアレルギー性反応 との関連を含む (引用文献 #29、 30、 31、 32、 33))が報告されている。特に興味深いの は、 IL-18シグナル伝達を正確にコントロールすることである。なぜなら、それは慢性 炎症性疾患およびアレルギー性疾患の治療に寄与すると考えられるからである。本 研究により MyD88について得られた構造情報および機能情報は、受容体と MyD88と の相互作用を調節することができる、免疫不全疾患のための薬剤の開発に寄与する 。種々の TIRドメインの複合体の構造決定に関して今後さらなる研究が期待される。さ らに、 MyD88 TIRドメインと他の分子(IRF-1, IRF-4, IRF- 5および IFN- y Rl)との相 互作用が最近報告されている (引用文献 #22、 23、 24、 34)。これらの相互作用の解明 もまた将来期待されてレ、る。

(b) (a)のタンパク質のアミノ酸配列において、 214番目のアミノ酸、 238番目のアミノ酸 及び 269番目のアミノ酸以外の 1若しくは数個のアミノ酸が欠失、置換若しくは付加さ れたアミノ酸配列力 なり、かつリポ多糖刺激のシグナルは阻害する力、インタ一ロイ キン 18刺激のシグナルは阻害しないタンパク質
