Abstract

L'invention concerne une méthodologie pour la création de capteurs et de microcircuits de structures sous la forme de feuilles, de films ou de fils, comportant un élément capteur, pour la détection directe d'interactions de liaison d'antiligand-ligand. Des nanostructures comportent des nanoparticules conductrices liées à un substrat non-conducteur, un réseau ordonné des nanoparticules ayant une densité inférieure au seuil sous lequel la nanostructure devient conductrice. Des antiligands sont fixés sur les nanoparticules du substrat et un ensemble correspondant de ligands parents sont fixés sur un ensemble supplémentaire de nanoparticules de détection, respectivement. Les nanoparticules de détection sont également conductrices. La réaction de liaison est déterminée à partir de la transition dans les propriétés électriques des structures, favorisée par la liaison des nanoparticules de détection conductrices. Les paires de ligand-antiligand peuvent être des paires d'anticorps-antigène ou des ensembles correspondants d'acides nucléiques ou d'oligonucléotides complémentaires. Une structure utile peut en outre être créée au moyen de formats de type sandwich impliquant des paires additionnelles d'interactions de liaison.
