- Compounds of Formula 24 where Y 9 is OH and Z is CH2 can be further transformed to compounds of Formula 24 wherein Y 9 is Cl, Br, I, O(S ⁇ 2)Me and Z is CH2 by general methods known in the art.
- Y 9 is Cl, Br, I, O(S ⁇ 2)Me or OH; PG is an acid-labile protecting group.
- Step A Preparation of 1 , 1 -dimethylethyl 4-[4-(ethoxycarbonyl)-2-thiazolyl]- 1 - piperidinecarboxylate
- a suspension of 1,1 -dimethylethyl 4-(aminothioxomethyl)-l-tetrahydropyridine- carboxylate (30 g, 123 mmol) in ethanol (180 mL) cooled to 0 °C in an ice bath, was added dropwise a solution of ethyl bromopyruvate (15.7 mL, 125 mmol) in ethanol (180 mL). The ice bath was removed, and the mixture was stirred at ambient temperature overnight.
- Triethylamine (30 mL) was added, and the mixture was concentrated under reduced pressure, diluted with ethyl acetate, washed with brine, dried over magnesium sulfate and concentrated under reduced pressure to give 31 g of a brown oil, which solidified on standing.
- a portion of this crude product (8.1 g) was heated with ether (200 mL), and the ether was then decanted. This was repeated a second time, and the combined ether solutions were evaporated under reduced pressure to give 7.6 g of the title compound as a yellow solid.
- Step B Preparation of 1 -(1 , 1 -dimethylethyl) 4-[4-(hydroxymethyl)-2-thiazolyl]- 1 - piperidinecarboxylate
- Step C Preparation of 1 -( 1 , 1 -dimethylethyl) 4-(4-chloromethyl-2-thiazolyl)- 1 - piperidinecarboxylate
- Step D Preparation of 1,1 -dimethylethyl 4-[4-[[rnethyl[(li?)-l,2,3,4-tetrahydro-l- naphthalenyl]amino]methyl]-2-thiazolyl]- 1 -piperidinecarboxylate
- the tetrahydrofuran solvent was removed from the mixture by evaporation under reduced pressure.
- the aqueous solution remaining was washed with ether and then acidified with concentrated hydrochloric acid to lower the pH to 1, resulting formation of a precipitate.
- Oxalyl chloride (3.5 mL, 40 mmol) was added drop wise to the stirred reaction mixture, which was then allowed to warm to room temperature for 3 h. The resulting mixture was concentrated under reduced pressure and placed under high vacuum to give 7.93 g of the title compound as a tan solid. This compound was of sufficient purity to use in subsequent reactions.
- Step G Preparation of l-[4-[4-[[methyl[(li?)-l,2,3,4-tetrahydro-l- naphthalenyl]amino]methyl]-2-thiazolyl]-l-piperidinyl]-2-[5-methyl-3- (trifluoromethyl)- 1 H-pyrazol- 1 -yl] ethanone
- Step B Preparation of ethyl-2-[l-[[5-methyl-3-(trifluoromethyl)-lH-pyrazol-l- yl]acetyl]-4-piperidinyl]-4-thiazolecarboxylate
- Step C Preparation of 2-[l-[[5-methyl-3-(trifluoromethyl)-lH-pyrazol-l-yl]acetyl]-4- piperidiny 1] -4-thiazolecarboxylic acid
- Step D Preparation of 2-[l-[[5-methyl-3-(trifluoromethyl)-lH-pyrazol-l-yl]acetyl]-
- reaction mixture was stirred at -10 0 C for 30 minutes more, and then allowed to warm to room temperature and stir an additional 16 h.
- the resulting homogeneous mixture was evaporated in vacuo, and the residue was placed under high vacuum for several hours to give 2.46 g of the title compound as a light yellow solid. This compound was of sufficient purity to use in subsequent reactions.
- Step F Preparation of ethyl 4-[4-[[methyl(l,2,3,4-tetrahydro-l- naphthalenyl)amino]carbonyl]-2-thiazolyl]- ⁇ -[5-methyl-3-(trifluoromethyl)- lH-pyrazol-l-yl]- ⁇ -oxo-l-piperidinepropanoate (Compound 6)
