Abstract

L'invention porte sur des procédés de décellularisation de tissus de mammifères utilisés pour des transplantations ou la culture de tissus un vitro consistant: à soumettre simultanément les tissus à un détergent ionique, et à un détergent non ionique pendant une longue période pouvant excéder cinq jours (on utilise dans l'un des procédés le SDS comme détergent ionique, et le Triton-X 100 comme détergent non ionique); puis à les soumettre à un long rinçage pouvant aussi durer plus de cinq jours. Ce traitement de longue durée donne des tissus dont les courbes contrainte/déformation et les données DSC sont similaires à celles des tissus frais non traités. Les tissus traités, qui sont quasiment exempts de cellules et dont la structure sous-jacente est essentiellement intacte, présentent une réponse inflammatoire notablement accrue par rapport aux tissus frais, même sans fixation par le glutaraldéhyde. On a constaté une calcification in situ sensiblement réduite par rapport aux tissus fixés par le glutaraldéhyde. Les déposants pensent que le détergent ionique et le détergent non ionique agissent en synergie pour fixer les protéines au détergent ionique, et que détergent non ionique élimine du tissu le complexe détergent ionique/protéine. Lesdits procédés peuvent par exemple servir à décellulariser les valvules cardiaques porcines et les tissus pariétaux, utilisés pour les transplantations.
