Abstract

The present invention relates to methods and kits for gene shuffling, as well as to shuffled genes. In particular, the methods comprise amplification of the sequences to be shuffled using affinity tagged primers, separation of the strands of the amplified sequences using the affinity tag and cleavage of the strands. The strands may be cleaved as single stranded nucleic acids or heteroduplexes may be formed from the strands and the double stranded heteroduplexes may be cleaved. Shuffled genes may be obtained by using the cleaved fragments as both primers and templates in a nucleotide polymerisation reaction, for example PCR. The kits of the present invention comprises primers and enzymes suitable for perfoming the methods.
