Abstract

A method of simultaneously measuring the Rb value and the plasma protein-binding ratio of a test compound in two blood samples having different dilution ratios which comprises the step of calculating the Rb value of the test compound in the samples and the hematocrit values of the samples in accordance with the following formula (I): Rb=(aVh+(xb+1)Vp)/(Vh+Vp)/(xb+1) formula (I) [wherein a represents the distribution ratio to hemocytes; b represents the maximum binding concentration/dissociation constant (nPt/Kd); Vh represents the hemocyte volume; Vp represents the plasma volume; and x represents the dilution ratio of plasma;] determining a and b in the above formula by the non-linear minimum square method, and then calculating using the following formula (II): protein-binding ratio=100b/(1+b) formula (II).
