Abstract

Die vorliegende Erfindung bezieht sich auf ein Verfahren zur hochparallelen Herstellung komplexer Molekül-Bibliotheken auf einer Substrat-Oberfläche mit hilfe regenerierbarer Matrizen-Stempel. Nach dem Stand der Technik erfordert die Herstellung von Molekül-Bibliotheken auf einem Träger einen hohen apparativen sowie Zeit- und Kostenaufwand. Durch die Verwendung regenerierbarer Matrizen-Stempel kann die Herstellung von Molekül-Bibliotheken, insbesondere von DNA und Protein-Bibliotheken, stark vereinfacht und beschleunigt werden. Der Matrizen-10 Stempel aus einem elastischen Material wie Polydimethylsiloxan trägt auf der Oberfläche ortsgebundene Matrizen-Moleküle, die als Vorlage zur Synthese von Abbild-Molekülen dienen. Die Abbild-Moleküle werden durch ein Kontaktdruck-Verfahren auf die Ziel-Oberfläche übertragen. Die Matrizen-Moleküle bleiben auf dem Stempel für weitere Synthese- und Druckzyklen, die mehrfach wiederholt werden können, zurück. Das Verfahren eignet sich auch zum Kopieren sowie zur Vervielfältigung anderweitig hergestellter Molekül-Arrays.
