Abstract

L'invention concerne des méthodes, des compositions et des articles d'analyse d'un échantillon à la recherche d'un polynucléotide cible. Un échantillon susceptible de contenir le polynucléotide cible est mis en contact avec un polynucléotide capteur monocaténaire complémentaire du polynucléotide cible et un agent permettant au polynucléotide capteur lui-même, lorsqu'il est présent sous forme monocaténaire, de devenir fluorescent avec l'excitation. Le polynucléotide capteur est éventuellement conjugué à un substrat, qui peut être un dispositif de détection optoélectronique, et peut être micro ou nano adressable. Un chromophore peut être utilisé pour absorber l'énergie du polynucléotide capteur excité et émettre de la lumière. Les méthodes de l'invention peuvent être utilisées sous forme multiplexe. L'invention concerne également des dispositifs de détection incorporant le polynucléotide capteur et éventuellement le chromophore. L'invention concerne enfin des kits comprenant des réactifs permettant de mettre en oeuvre lesdites méthodes.
