Definitions

  • TSEs Transmissible Spongiform Encephalopathies
  • Scrapie Perhaps the most widely recognized transmissible spongiform encephalopathy of animals is scrapie, which is a fatal progressive neurological disorder of sheep and goats. Scrapie has been reported in many countries and has been recognized in British sheep flocks for over two centuries, having been first recorded in 1732. Scrapie can be transmitted experimentally to several species, e.g., mink, cats, deer, moose, mice and hamsters.
  • BSE Bovine Spongiform Encephalopathy
  • Bovine spongiform encephalopathy (BSE), sometimes known as "mad cow disease”, was recognized as a potential member ofthe scrapie family of diseases as soon as it was discovered in November 1986. It shows all of he pathological characteristics of a transmissible spongiform encephalopathy.
  • BSE Bovine spongiform encephalopathy
  • Creutzfeldt-Jakob Disease in man uru Creutzfeldt-Jakob Disease and its more rare variant, the Gerstmann-Straussler- Scheinker disease, are examples of spongiform encephalopathies found in man. Like the animal disorders, they are progressive and universally fatal. Kuru was, at one time, common in certain tribes in New Guinea but has not been seen elsewhere.
  • PrPSc innocuous cellular proteins
  • mice with disrupted prions (PrPc) gene are healthy and do not get infected with BSE (Weissmann 1992) is concluded that prions (PrPc) do not exhibit essential physiological functions and genetic variances can be expected.
  • Serm from bulls and sheep from all over the world is collected and screened for disrupted prion genes by using the established methods for genotyping. Using sperm for genotyping has the advantage that sperm is easily accessible, easy to store and - in case of desired screening results - can be used for breeding directly.
  • the efficacy ofthe disrupted prion gene is tested by replacing the normal prion gene in lab animals - preferable mice and hamster - with the disrupted prion gene, breeding this lab animal and testing them for susceptibility of TSE infections by application of TSE-infected tissue - preferable brain tissue from BSE or Scrapie infected animals - to the bred cattle and sheep by oral, i.p., i.v., i.m., s.c, intracranial, and/or intracerebral routes, preferable intracranial and intracerebral routes.
  • TSE resistant animals are bred by inseminating female animals with disrupted prion gene sperm.
  • the success ofthe breeding process is monitored by analyzing the genome ofthe bred animals for the disrupted prion genes using the established methods for genotyping.
  • TSE-infected tissue - preferable brain tissue from BSE or Scrapie infected animals - is applied to the bred cattle and sheep by oral, i.p., i.v., i.m., s.c, intracranial, and/or intracerebral routes, preferable intracranial and intracerebral routes.
  • the describes process for breeding of BSE and Scrapie resistant animals has the advantage that natural BSE and Scrapie resistant cattle and sheep are obtained and transgenic knockout cattle and sheep (gene modified animals) are avoided.
