Abstract

A synthetic nucleotide sequence that exerts regulatory control when in operative association with a transgene comprises a human elongation factor 1α promoter sequence dissociated from an intact human elongation factor 1α 'enhancer' sequence. This regulatory sequence is useful for the recombinant production of any transgene in vitro or in vivo. Various embodiments of the nucleotide sequence include a minimal fragment of EF-1α promoter, which displays minimal cell tropism and is subject to less downregulation than the native EF-1α promoter-'enhancer'. More advantageous is a chimeric promoter formed by this minimal EF-1α promoter sequence and a heterologous Intron, such as Intron A of HCMV. Recombinant molecule, e.g., plasmids, viruses, etc. containing such artificial promoters may be employed to express any number of transgenes in a wide variety of cell types.
