Der Anmelder hat die Selektion in verschiedener Weise erreicht. Die Expressionskassette wurde nach Restriktionsverdau des Vektors mit Hindlll isoliert und in die Eco- RI-Restriktionsschnittsteile des Vektors pLH90001oxMel nach Auffüllreaktion mit Klenow-Polymerase ligiert. Es entstand der Vektor pLH90001oxMelGapCre. Die zweite lox-Rekombinationssequenz wurde durch Klonierung der oben beschriebenen Oligonu- kleotide in die Hindlll-Schnittstelle dieses Vektors nach Auffüllreaktion mit Klenow-Polymerase kloniert. Durch Sequenzierung wurde sichergestellt, daß die beiden lox-Rekombinationssequenzen in der gleichen Orientierung vorlagen. Es entstand der Vektor pLH90001oxMelGapCrelox. In die Sall-Restrik- tionsschnittstelle dieses Vektors wurde eine Expressionskas- sette, die das Gen für einen Einzelketten- (scFv-)Antikörper unter der Kontrolle des CaMV 35S Promotors enthält, kloniert. Das zu transferierende Fragment wurde über PCR aus einem binären Vektor mit Konstrukt Nr. 9 (Artsaenko et al.. (1998) Mole- cular Breeding 4, 313-319) mit den Primern GTC GAC AAC ATG GTG GAG CAC GAC ACT CTC G und GTC GAC TGC AGG TCA CTG GAT TTT GGT TTT AGG amplifizert, durch Restriktionsverdau mit Sall geschnitten und in den binären Vektor pLH90001oxMelGapCrelox kloniert. Es enstand der Vektor pLH9000lMGClscFv. Der Expressionsvektor pLH9000lMGClscFv wurde zur Transformation von E.coli SM10 verwendet. Transformanten wurden mit Agrobacterium tumefaciens GV 3101 gemischt und über Nacht bei 28°C inkubiert, (vgl. C.Koncz und J. Schell, Mol .Gen.Genet . (1986) 204, 383-396; C.Koncz. et al . , Proc.Natl .Acad. Sei .USA (1987) 84, 131-135) . Es wurde auf Streptomycin selektioniert, wobei das hierfür notwendige aadA-Gen in den vorstehenden Expressionsvektoren vorlag. Selektionsklone von Agrobacterium tumefaciens wurden auf abgeschnittenen und mehrfach an der Mittelrippe eingeritzten Blättern der Kartoffelpflanze cv. Desiree aufgebracht und die Blätter wurden 2 Tage bei 20°C im Dunkeln inkubiert. Danach wurden die Agrobakterien abgewaschen und den Kartoffelblättern Pflanzenwuchststoffe zugesetzt, so daß bevorzugt Sprosse regenerierten. Ferner wurden durch die Zugabe von Kanamycin in das Pflanzenmedium nicht-transformierte Zellen in den Kartoffelblättern abgetötet. Heranwachsende Sprosse wurden abgeschnitten und auf das Medium ohne Pflanzenwachstumsstoffe, aber mit Kanamycin, bewurzelt. Die weitere Kultivierung der Kartoffelpflanzen erfolgte in üblicher Weise. Alle regenerierten Linien wurden mittels Southern Hybridisier ng auf die Kopienzahl der integrierten Fremd-DNA hin untersucht. Dazu wurde mit dem Fachmann bekannten Methoden genomische DNA aus Blättern der einzelnen Linien isoliert, mit dem Restriktionsenzym Notl verdaut und auf einem Agarosegel aufgetrennt. Die Fragmente wurden auf eine positiv geladene Nylon-Membran transferiert und mit markierter DNA des Gens für den Einzelkettenantikörper als Sonde hybridisiert. Die Verfahren dazu sind dem Fachmann bekannt. Für die weiteren Analysen wurden nur solche Linien ausgewählt, die nach der anschließenden Detektion nur eine Bande aufwiesen und somit lediglich eine T-DNA-Kopie enthielten. Der Nachweis der Expression des scFv-Antikörpers wurde über den enthaltenen c-myc Tag mittels des monoklonalen Antikörpers 9E10-IgG (Cambridge Research Chemicals, Northwich, Chehire, UK) oder Protein L (Clontech, Palo Alto, CA., USA) im Western Blot bzw. ELISA erbracht. Zur Expression der Rekombinase wurden abgeschnittene Blätter von transgenen Kartoffelpflanzen aus Sterilkultur 40 Stunden anaerob in dem Anaerocult-System (Merck, Darmstadt, Deutschland) wie bei Bülow et al . , supra, beschrieben unter sterilen Bedingungen inkubiert. Danach wurden die Blätter auf Medium gelegt, dem Pflanzenwuchsstoffe zur Regeneration von Sprossen und lOO M CuS04 zugesetzt war, um die Expression des Melittins in den Pflanzenzellen zu aktivieren, in denen keine Rekombination stattgefunden hatte. Das Medium wurde alle 7 Tage erneuert, heranwachsende Sprosse wurden abgeschnitten und in kupferhal- tiges Medium ohne Pflanzenwuchsstoffe überführt. Daraus resultierte der Vektor pLH9000Hyg. Dieses Fragment wurde in den durch Restriktionsverdau mit Xhol und BamHI aufgeschnittenen Vektor pRTlOO kloniert, wodurch der Vektor pRTlOOOp entstand. Aus dem Vektor pRTlOOOp wurde die Expressionskassette durch Restriktionsverdau mit Hindlll isoliert und in die Hindlll-Re- striktionsschnittstelle des Vektors pLH9000FRTHygBar eingesetzt. In die Sall-Restriktionsschnittstelle des resultierenden Vektors pLH9000FRTHygBarOp wurde die zweite FRT-Rekombinationssequenz durch Klonierung der oben beschriebenen Oligonukleotide nach Auffüllreaktion mit Klenow-Polymerase kloniert. Durch Sequenzierung wurde sichergestellt, daß die beiden FRT-Rekombinationssequenzen in der gleichen Orientierung vorlagen. Es entstand der Vektor pLH9000FRTHygBarOpFRT. In die Apal-Restriktionsschnittstelle dieses Vektors wurde eine Expressionskassette, die das Gen für τ4-Lysozym unter der Kontrolle des CaMV 35S Promotors enthält, kloniert. Das zu transferierende Fragment wurde aus dem binären Vektor pSR8-40 (Porsch et al . (1998) Plant Molecular Biology 37, 581-585) durch Restriktionsverdau mit Hindlll isoliert und in den Vektor pLH9000FRTHygBarOpFRT kloniert. Es enstand der binäre Vektor pLH9000FHBOFlys . Der Expressionsvektor pLH9000FHBOFlys wurde zur Transformation von E.coli SM10 verwendet. Trans- formanten wurden mit Agrobacterium tumefaciens GV 3101 gemischt und über Nacht bei 28°C inkubiert, (vgl. Koncz und Schell, supra; Koncz et al . , supra) . Es wurde auf Streptomycin selektioniert, wobei das hierfür notwendige aadA-Gen in den vorstehenden Expressionsvektoren vorlag. Selektionsklone von Agrobacterium tumefaciens wurden auf abgeschnittenen und mehrfach an der Mittelrippe eingeritzten Blättern der Geranienpflanze cv. Astra aufgebracht und die Blätter wurden 2 Tage bei 20°C im Dunkeln inkubiert. Danach wurden die Agrobakterien abgewaschen und den Geranienblättern Pflanzenwuchststoffe zugesetzt, so daß bevorzugt Sprosse regenerierten. Ferner wurden durch die Zugabe von Hygromycin in das Pflanzenmedium nicht-transformierte Zellen in den Geranienblättern abgetötet. Heranwachsende Sprosse wurden abgeschnitten und auf das Medium ohne Pflanzenwachstumsstoffe, aber mit Hygromycin, bewurzelt. Die weitere Kultivierung der Geranienpflanzen erfolgte in üblicher Weise. Alle regenerierten Linien wurden mittels Southern Hybridisierung auf die Kopienzahl der integrierten Fremd-DNA hin untersucht. Dazu wurde mit dem Fachmann bekannten Methoden genomische DNA aus Blättern der einzelnen Linien isoliert, mit dem Restriktionsenzym Notl verdaut und auf einem Agarosegel aufgetrennt. The fragments were transferred to a positively charged nylon membrane and hybridized with labeled DNA of the gene for T4 lysozyme as a probe. The methods for this are known to the person skilled in the art. For the further analyzes, only those lines were selected which after the subsequent detection had only one band and therefore only contained a T-DNA copy. The expression of T4 lysozyme was demonstrated by means of a polyclonal antibody in a Western blot (cf. Düring et al. (1993) Plant Journal 3, 587-598). Protoplasts were used for marker gene elimination made from cut leaves of transgenic geranium plants from sterile culture. These were transformed with the aid of PEG with a vector which contains the gene for flp recombinase under the control of the CaMV 35S promoter. To produce this vector, the flp recombinase gene from DNA of the plasmid of the yeast Saccharomyces cerevisiae strain A364AD5 (Hartley and Donelson (1980) Nature 286, 860-864) was by PCR with the primers CTC GAG ATG CCA CAA TTT GGT ATA TT and CTC GAG TTA TAT GCG TCT ATT TAT GT amplified, the fragment cut by restriction digestion with Xhol and cloned into the Xhol restriction site of the vector pRTlOO (Töpfer et al., Supra). The resulting plasmid pRTlOOflp was used for the transient expression of the flp gene in protoplasts. The methods for protoplast isolation and transformation are known to the person skilled in the art. The protoplasts were then embedded in medium to which plant growth substances for the regeneration of shoots and 20 μM tetracycline had been added in order to activate the expression of the barnase gene in the plant cells in which no recombination had taken place. The surrounding medium was renewed every 7 days, growing shoots were cut off and transferred into tetracycline-containing medium without plant growth substances. The plants were rooted and further cultivated in the usual way. After two weeks, the transfer to tetracycline-free medium took place. The marker gene elimination was checked by means of PCR. For this purpose, genomic DNA was isolated from the leaves of the individual lines. The entire T-DNA was amplified with the border-specific primers GGC AGG ATA TAT TCA ATT GTA AAT and GTA AAC CTA AGA GAA AAG AGC GTT TA. The size of the amplified products in the agarose gel was compared with controls from the starting lines, and the marker gene-free lines were determined.

Die PCR-Überprüfung von 100 regenerierten Sprossen mittels der vorstehend beschriebenen PCR-Primer ergab, daß kein Hygromy- cinresistenz-, Barnase oder tet-Repressor-Gen mehr nachgewiesen werden konnte.
