METHODE DE PRODUCTION ET DE CRYOCONSERVATION D'EMBRYONS BOVINS DEVELOPPES IN WRO ET PAILLETTE CONGELEE POUR LE TRANSFERT DIRECT DE CEUX-CI
L'invention se rapporte à une méthode de production et de cryoconservation d'embryons bovins développés in vitro et à une paillette congelée pour le transfert direct de ceux-ci.
Au cours de ces 10 dernières années, les techniques de production de l'embryon bovin in vitro ont atteint un niveau d'efficacité élevé si bien qu'aujourd'hui de nombreuses sociétés privées et laboratoires proposent cette technologie pour produire de façon régulière des embryons bovins. Toutefois, pour accroître l'impact de cette biotechnologie dans les élevages et l'utiliser à grande échelle dans les programmes d'amélioration génétique, il est indispensable de disposer d'une méthode fiable de congélation de ce embryons.
Chez la plupart des mammifères domestiques, les embryons développés in vivo sont congelés selon des méthodes lentes basées sur le maintien de l'équilibre osmotique entre les cellules embryonnaires et leur milieu environnant. La cryoconservation se déroule typiquement en trois étapes principales : - le pré-traitement : équilibration des embryons en une ou plusieurs étapes dans un cryoprotecteur perméable de type polyols afin de limiter la formation de glace intracellulaire et les stress osmotiques en présence de glace extracellulaire ;
- le traitement : refroidissement des embryons, stockage dans l'azote liquide (- 196 °C) et réchauffement ; - le post-traitement : étapes de dilution du cryoprotecteur perméable afin de rétablir au mieux les qualités biologiques des embryons cryoconservés.
La nature du cryoprotecteur, sa concentration compatible avec une toxicité limitée, les vitesses de refroidissement et de réchauffement qui vont régir les mouvements de l'eau à travers les membranes cellulaires sont des facteurs déterminants du succès des méthodes de cryoconservation. Ainsi, le choix d'un cryoprotecteur pour un type de cellule donné dépend non seulement des propriétés physico-chimiques qu'il manifeste en solution mais aussi de ses propriétés biologiques sur les cellules. Des études sur les modalités d'addition et de dilution du cryoprotecteur ont abouti à la simplification des protocoles de congélation de l'embryon bovin développé in vivo tout en améliorant leur efficacité. L'utilisation d'un cryoprotecteur non perméable comme le saccharose dans la solution de dilution a permis tout d'abord de limiter le nombre des
étapes de dilution du cryoprotecteur perméable. Le saccharose élève la pression osmotique du milieu extracellulaire ce qui a pour effet de limiter les mouvements de l'eau vers l'intérieur des cellules pendant que le cryoprotecteur perméable diffuse à l'extérieur des cellules au moment de la décongélation. Par la suite, diverses méthodes ont été proposées pour éviter cette étape de dilution et permettre le transfert direct de l'embryon après décongélation sans manipulation supplémentaire :
- équilibration des embryons dans une solution de glycérol 1,5 M et conditionnement en paillette en incluant la solution de dilution (0,5 à 1 M de saccharose) de part et d'autre du milieu de congélation comme décrit par J.P. Renard et coll., Ann. Méd. Vét, 126 : 23-32 (1982), et par S.P. Leibo, Theriogenology, 21 : 767- 790 (1984).
- équilibration des embryons dans une solution de glycérol 1,4 M additionnée de 0,25 M de saccharose et conditionnement en paillettes en incluant la solution de dilution (0,5 M de saccharose) comme décrit par K. Touati et coll., Ann. Méd. Vét, 134 : 249-251 (1990). La présence de saccharose pendant l'équilibration entraîne une déshydratation partielle des cellules avant le refroidissement. La conséquence est une augmentation de la concentration intracellulaire en glycérol et en solutés ce qui limite la cristallisation intracellulaire ;
- équilibration des embryons dans une solution d'éthylène glycol 1,5 M. La vitesse de pénétration de l'éthylène glycol est élevée et proche de celle de l'eau. Ainsi les embryons tolèrent une réhydratation directe dans le milieu de conservation sans passer par des étapes de dilution, comme décrit par Voelkel et Hu , Theriogenology, 37 : 687-697(1992).
Les taux de gestation après transfert direct d'un embryon bovin développé ]n vivo et congelé selon la méthode au glycérol-saccharose ou à l'éthylène glycol sont voisins de 50 % ou même supérieurs.
Les premières tentatives de cryoconservation des embryons cultivés in vitro ont souligné la difficulté d'utiliser directement les techniques de congélation appliquées à grande échelle pour l'embryon développé m vivo (Leibo et Loskutoff, Theriogenology, 39 : 81-94 (1993). De nombreux protocoles de cryoconservation ont été élaborés mais leur efficacité a été le plus souvent validée par des taux de survie in vitro après décongélation et culture in vitro. Seules quelques études rapportent des taux de gestation après transplantation d'un embryon sur un nombre significatif de receveuses.
Ces taux sont très variables et restent insuffisants pour une application dans les élevages, comme illustré par le tableau ci-après.

Ainsi donc, à la connaissance de la demanderesse, on ne dispose pas, à ce jour, de méthode de production d'embryons bovins développés in vitro et cryoconservés pouvant être utilisée par des éleveurs avec des taux de réussite (pourcentage de gestations menées à terme) satisfaisants, et susceptible d'être employée dans les schémas de sélection ou à des fins commerciales en particulier pour l'exportation.
L'invention vise à combler cette lacune en fournissant une méthode de production perfectionnée d'embryons bovins développés in vitro et cryoconservés susceptibles d'être implantés par transfert direct.
Plus précisément, l'invention concerne une méthode de production d'embryons bovins cryoconservés à partir d'ovocytes fécondés in vitro qui comprend les étapes suivantes :
(1) maturer et féconder in vitro des ovocytes bovins ; (2) cultiver in vitro lesdits ovocytes fécondés jusqu'au stade morula ou blastocyste ;
(3) sélectionner les embryons de qualité 1 ; et
(4) congeler les embryons sélectionnés, caractérisée en ce que l'étape de culture (2) est une co-culture sur un tapis de cellules Vero effectuée en l'absence de sérum, et en ce que l'étape de congélation (4) est effectuée tandis que l'embryon est conditionné dans un milieu de congélation contenant un mélange glycérol-tréhalose comme cryoprotecteur.
L'étape de culture (2) est une co-culture d'ovocytes fécondés sur un tapis de cellules Vero (lignée de cellules épithéliales de rein de singe). Cette technique de co-
culture est bien connue en soi et décrite notamment par C. Guyader-Joly et coll., 12th Congress of European Embryo Transfer Association, 136, (1996) et par L.M.C. Pegoraro et coll., Theriogenology 49 : 1579-1590 (1998).
Contrairement aux préconisations usuelles, on a trouvé que cette co-culture doit être effectuée dans un milieu exempt de sérum. Le milieu de co-culture utilisé peut être le milieu synthétique pour la culture cellulaire dénommé "B2 INRA" ou de préférence le milieu dénommé "Upgraded B2 INRA". Le milieu B2 INRA est décrit en détail par Yves
Ménézo, C.R. Acad. Se. Paris, t.282, (14 juin 1976), Série D, pages 1967-1970. Le milieu "Upgraded B2 INRA" ne diffère du milieu "B2 INRA" que par l'utilisation de constituants répondant à des critères de sécurité poussés.
On a trouvé, en effet, que la présence de sérum était néfaste à l'aptitude à la congélation/décongélation des embryons bovins résultants, les embryons développés en présence de sérum, tel que le sérum de veau, présentant après décongélation un taux de survie in vitro et un taux de gestation après transplantation inférieurs. Selon un mode de mise en oeuvre particulier préféré, le refroidissement de l'embryon sélectionné est précédée de deux étapes successives d'équilibration de cet embryon, la première étant effectuée dans un milieu de congélation additionné de glycérol et la deuxième dans un milieu de congélation additionné d'un mélange de glycérol et de tréhalose. Avantageusement, la congélation est opérée tandis que l'embryon est conditionné dans une paillette en matière plastique, dans le milieu de congélation contenant, comme cryoprotecteur, le mélange glycérol-tréhalose. La paillette peut être de tout type couramment utilisé pour la congélation d'embryons de mammifères.
L'invention concerne également une paillette congelée utile pour le transfert direct d'un embryon bovin caractérisée en ce qu'elle comprend une zone centrale dans laquelle se trouve l'embryon dans un milieu de congélation contenant, comme cryoprotecteur, un mélange de glycérol et de tréhalose et deux zones extrêmes dans lesquelles se trouve une solution de tréhalose, chaque zone extrême étant séparée de la zone centrale par une bulle d'air. Un telle paillette congelée, après décongélation, autorise un transfert direct aisé de l'embryon à la vache receveuse à l'aide d'un pistolet de transfert, sans avoir à effectuer un traitement de dilution préalable du cryoprotecteur.
De préférence, l'embryon bovin est un blastocyste.
Le milieu de congélation peut être constitué, à titre illustratif, d'une solution saline de tampon phosphate (PBS) additionnée de sérum de veau foetal (SVF) ou autre
sérum. La proportion de SVF peut aller, par exemple, de 5 % à 40 % en volume. La concentration de tréhalose peut aller, par exemple, de 0,1 M à 0,5 M. Elle est de préférence d'environ 0,2 M . La proportion de glycérol peut aller de 8 % à 1 1 % en volume, de préférence de 8 à 10 % en volume. Elle est avantageusement d'environ 9 % en volume. La concentration de tréhalose dans les zones extrêmes est, de préférence, plus élevée que dans la zone centrale. Elle peut aller de 0,2 M à 1 M. De préférence, elle est d'environ 0,5 M .
L'exemple non limitatif suivant est donné afin de bien faire comprendre la méthode de l'invention.
I) PRODUCTION DES EMBRYONS IN VITRO A) Collecte des ovocytes par ponction folliculaire
Les ovaires sont prélevés à l'abattoir immédiatement après l'éviscération des femelles. Ils sont désinfectés à l'alcool 70° et transportés au laboratoire dans du tampon phosphate (PBS) à une température de 30-35°C, de préférence d'environ 30-32°C dans un délai 3 à 6 heures, de préférence de 4 heures. Les ovocytes sont collectés par aspiration des follicules visibles à la surface de l'ovaire (2 à 8 mm) à l'aide d'une pompe péristaltique à débit constant reliée à une aiguille 18 G x 11/2 TW (1,2 x 40 mm). Le liquide folliculaire est recueilli dans un tube de 50 ml. Après sédimentation, le culot cellulaire est prélevé et dilué dans du milieu M 199 tamponné par 25 mM d'hépès (M 7528, Sigma) additionné de 10% de sérum de vache en oestrus, 5 /g/ml de gentamicine (G 1264, Sigma), 100 Ul/ml de pénicilline G (P 4875, Sigma), 50 //g/ml de streptomycine (S 9137, Sigma) et 20 /g/ml de nystatine (N 4014, Sigma).
Les ovocytes peuvent également être prélevés chez l'animal vivant, sous échographie et de façon répétée (Nibart et coll., 1995). Dans les deux cas, les complexes ovocyte - cumulus compact (COC) sont recherchés sous la loupe binoculaire. Les ovocytes immatures sont sélectionnés sur des critères morphologiques : ovocytes entourés d'au moins 3 couches de cellules du cumulus compact avec un cytoplasme non granuleux de couleur homogène. B) Maturation in vitro des ovocytes
Les COC sont matures pendant 20-24 heures à 38,5°C sous une atmosphère de 5% de CO2 dans l'air en présence ou non de cellules de la granulosa. Le milieu de maturation est composé de M199 (N°31 150014, Gibco) tamponné au bicarbonate et supplémenté de 10 % de sérum de veau foetal (SVF, Hyclone), 10 / g ml de FSH-LH (Stimufol N.D., Mérial), 1 / g/ml d'oestradiol 17b (E 2257, Sigma). Un facteur de croissance peut, facultativement, être ajouté.
C) Préparation de la semence et fécondation in vitro des ovocytes Les spermatozoïdes sont sélectionnés pour la fécondation selon la technique de migration ascendante. Après décongélation d'une paillette de semence dans un bain- marie à 37°C pendant 30 secondes, son contenu est vidé délicatement dans le fond d'un cryotube contenant 1 ml de sperm-TALP (Parrish et coll., 1988). Le sperm-TALP est une solution de lactate-tyrode modifiée (pH = 7,2) contenant 6 mg/ml d'albumine bovine sérique (BSA, A 7906, Sigma) et 1,0 mM de Na-pyruvate (P 2256, Sigma). Après 45 minutes d'incubation à 38,5°C, la fraction supérieure contenant les spermatozoïdes
les plus mobiles est prélevée puis centrifugée pendant 10 minutes à 200 g. La concentration en spermatozoïdes dans le culot cellulaire est estimée après comptage sur une cellule de Thoma.
Après la maturation in vitro, les ovocytes à cumulus expansé sont lavés deux fois dans le milieu de fécondation et placés par groupe de 20 à 25 dans des tubes à hémolyse contenant 500 μ\ de fert-TALP (Parrish et coll., 1988). Cette solution de lactate-tyrode modifiée (pH = 7,6) contient 6 mg/ml de BSA (Sigma) et 0,2 mM de Na- pyruvate. Le milieu de fécondation est également supplémenté de 10 //g/ml d'héparine (H 3393, Sigma), 20 //M de D-pénicillamine (P 4875, Sigma), 10 / M d'hypotaurine (H 1384, Sigma) et 1 //M d'adrénaline (E 4250, Sigma). Les ovocytes et les spermatozoïdes à une concentration finale de 1 x 106/ml sont incubés pour une durée de 16 à 20 heures, de préférence d'environ 18 heures à 38,5°C sous une atmosphère de 5% de
D) Préparation des tapis de cellules Vero La lignée établie de cellules Vero (Mérial) a été multipliée selon le protocole décrit par Ménézo et coll. (1990). A partir d'un cryotube contenant 2 à 3 x 106 cellules congelées, des flacons de culture de 25 cm2 (S20/C160, Falcon) sont ensemencés à raison de 1 x 106 cellules/flacon dans du milieu M199 (N°31150014, Gibco) + 10%
SVF. Après 4 jours de culture à 38,5°C sous 5% de CO2 dans l'air, les cellules sont à confluence (6 à 8 X 106 cellules par flacon de culture) et décollées avec une solution de trypsine à 0,05% (T 4174, Sigma) préparée dans du milieu de Hank's (N° 04104175,
Gibco). Après inactivation de l'enzyme par addition de SVF (V/V) et centrifugation à
1000 g pendant 10 minutes, les cellules sont ensemencées à des densités différentes en fonction de la surface de culture. L'inoculum est de 1 X 106 cellules par flacon de 25 cm2 et de 1 x 103 cellules par microgoutte de 50 μ\ recouverte d'huile minérale (M
3516, Sigma). Les cellules restantes sont congelées.
La méthode de congélation et de décongélation des cellules est celle décrite par
Ouhibi et coll. (1990) légèrement modifiée. Les cellules sont congelées à raison de 2 à 3 x 106 cellules dans du milieu M199 (N°31 150014, Gibco) supplémenté de 50% de SVF et de 10% de diméthyl sulfoxide (DMSO, D 5879, Sigma). La courbe de congélation est la suivante :
- 1 °C/min de +2°C à -10°C ; - 2°C/min de - 10°C à -30oC ; - 10°C/min de -30°C à - 150°C.
Les cellules sont alors plongées très rapidement dans l'azote liquide et stockées à - 196°C.
La décongélation est effectuée dans un bain-marie à 37°C. Son contenu est vidé dans 1 ml de SVF puis centrifugé 10 minutes à 1000 g. Le culot cellulaire est resuspendu dans du milieu Ml 99 + 10% SVF et les cellules sont ensemencées selon le protocole précédemment décrit.
E) Culture in vitro des embryons
Après la fécondation in vitro, les cellules du cumulus et les spermatozoïdes surnuméraires accolés à la zone pellucide sont enlevés mécaniquement par agitation au vortex pendant 1 minute. Les zygotes sont ensuite lavés une fois dans le fert-TALP puis deux fois dans le milieu de culture (milieu dénommé Upgraded B2 INRA, disponible auprès du Laboratoire CCD, 60 rue Pierre Charron, 75008 Paris). Les zygotes sont cultivés par groupes de 20 à 25 dans des microgouttes de 50 μ\ de milieu Upgraded B2 sans sérum (contrairement à ce qui est généralement préconisé) recouvertes d'huile (M 3516, Sigma) sur des monocouches préétablies (appelées aussi "tapis") de cellules Vero
(Ménézo et coll., 1990). La culture est réalisée dans un incubateur à 38,5°C sous 5% de
CO2 dans l'air pendant 7 jours (jour O = jour de l'insémination in vitro). Les embryons non segmentés sont enlevés du milieu de culture à J2. Les embryons jugés de qualité 1
(selon les critères de l'IETS, International Embryo Transfer Society), sont sélectionnés pour la congélation à J7.
II) CONGELATION DES BLASTOCYSTES
Les embryons de qualité 1 sont lavés dans 10 bains successifs de solution saline de tampon phosphate (PBS, IMV) selon les recommandations IETS (1990). Tous les milieux de congélation sont préparés à partir d'une base constituée de solution saline de tampon phosphate (PBS, IMV) additionné de 20% SVF, et utilisés à température ambiante. Les embryons sont équilibrés en deux étapes successives de 10 minutes dans un milieu constitué de la base ci-dessus additionnée de 5 % en volume de glycérol
(G 7757, Sigma) puis dans un milieu de congélation constitué de la base ci-dessus additionnée de 9 % en volume de glycérol + 0,2 M de tréhalose (18,835-2, Sigma). Les embryons sont conditionnés en paillettes de 250 μ\ (N°005592, IMV) par aspiration successive de : tréhalose 0,5 M (sur une longueur de 3 cm), bulles d'air, milieu de base additionné de 9 % de glycérol, et de tréhalose 0,2 M contenant l'embryon (sur une longueur de 1 ,5 cm), bulle d'air, tréhalose 0,5 M (sur une longueur de 6 cm). Chaque paillette est alors fermée par un jonc plastique puis déposée horizontalement sur un portoir métallique. Celui-ci est placé à température ambiante dans l'enceinte d'un congélateur biologique programmé (Nicoolbag MS 21, Air Liquide). L'ensemble est refroidi à une vitesse de 1,5°C/min jusqu'à -7°C. Après 3 à 4 minutes à -7°C, la cristallisation est induite manuellement par contact de la paroi des paillettes avec l'extrémité d'une tige métallique préalablement refroidie dans l'azote liquide. Trois minutes après l'induction de changement de phase, les paillettes sont refroidies à une vitesse de 0,3°C/minute jusqu'à -32°C, puis plongées directement et stockées dans l'azote liquide à -196°C
III) DECONGELATION POUR LE TRANSFERT DIRECT
La décongélation s'effectue en tenant par une extrémité la paillette 10 secondes dans l'air à température ambiante, puis 30 secondes dans un bain-marie à 20°C sans agitation. La paillette est ensuite insérée (sans agitation) dans un pistolet de transfert, son extrémité coupée et l'embryon est déposé dans la corne utérine ipsilatérale au corps jaune d'une receveuse à jour 6 à 7 de son cycle.
IV) RESULTATS D'ESSAIS Une expérimentation ayant impliqué 49 transferts directs a donné des taux de gestation qui sont de 58 % à J 35 et de 51 % (25/49) à J 90. Antérieurement à l'invention, on n'avait pas obtenu des taux de gestation excédant 35 % (voir par exemple A.M. Van Wagtendonk - de Leeuw et coll., Theriogenology, 53 : 575-597 (2000). L'invention constitue donc une avancée technique notable, résultant d'une synergie inattendue entre les moyens mis en oeuvre. Références bibliographiques :
