Abstract

Un anticorps monoclonal de la classe IgG se lie sélectivement à des cellules T perméabilisées mais non à des cellules T non perméabilisées. L'anticorps monoclonal TIA-2 se lie à un épitope intracellulaire de la chaîne zêta du complexe récepteur des cellules T. Selon un procédé d'analyse d'une population d'anticorps monoclonaux afin de détecter un anticorps monoclonal qui se lie sélectivement à des lymphocytes perméabilisés mais non à des lymphocytes non perméabilisés, on stabilise une population de lymphocytes en mettant les cellules en contact avec un fixateur doux, on perméabilise les cellules en les mettant en contact avec un détergent glucosidique doux, on met la population d'anticorps monoclonaux en contact avec les lymphocytes perméabilisés stabilisés, on met en contact la population d'anticorps monoclonaux avec des lymphocytes non perméabilisés et on sélectionne un anticorps monoclonal qui réagit avec les lymphocytes perméabilisés mais non avec les lymphocytes non perméabilisés.
