Abstract

The invention relates to analytical chemistry, in particular to methods for determination of doxazosin by gas-liquid chromatography, and can be used for determination of this substance in pharmaceutical analysis and at chemical-toxicological investigations. The object of the invention is in development of new method for the determination of doxazosin by gas-liquid chromatography, this through selection of optimal temperature program and balanced fixed liquid phase together with the other parameters of the process prevents destruction and provides reliable, accurate determination of doxazosin in samples of water solution with concentration 1.25-10.0 μg/ml. The method claimed is realized as follows. The determination of doxazosin is carried out with use of gas chromatograph "Agilent 19091 J-413", capillary column 30m x 320 μm with fixed liquid phase - 5%-phenyl-95%-dimethyl-polysiloxane, HP-5 0.25 mkm; movable phase – gas-carrier – helium, this is supplied to the column at rate of 45.0 ml/min. The detection of substance is carried out by flame-ionization detector, rate of hydrogen supply is 40,0 ml/min. Temperature of the column is linearly programmed: 2 min at 180 °C; temperature increase from 180 to 220 °C at rate 10 °C per minute during 4 min.; at 220 °C temperature mode is kept for 2 minutes. Temperature of evaporator is 250 °C, detector - 250 °C. Quantitative content of doxazosin is determined by method of absolute calibration by peak area.
