Abstract

An invention relates to the process for production of polypeptide in mammalian cell culture, containing gene, coding wanted polypeptide, where the gene is expressed under conditions of cell cultivation at large scale production on the medium, containing glutamine, in which aggregate amount of amino acids per unit of volume is more than approximately , and molar ratio of aggregate amount of glutamine towards the aggregate amount of asparagines is less than approximately 2, and has characteristic of medium, selected from the group, including i) molar ratio of aggregate amount of glutamine towards the aggregate amount of amino acids is less than approximately 0.2; ii) molar ratio of aggregate amount of inorganic ions towards the aggregate amount of amino acids is approximately from 0.4 to 1; iii) pooled aggregate amount of glutamine and asparagines per unit of volume is more than approximately , and theirs combination and maintenance of culture in initial growth phase at the first set of cultivation conditions during the first time interval, sufficient for enabling the mentioned cells to reproduce till density of living cells within diapason of approximately 20%-80% from the maximal possible density of living cells, if the mentioned culture was maintained at the first set of cultivation conditions and changing of at least one of the conditions of cultivation in such a way that we could apply the second set of cultivation conditions and maintenance of the mentioned culture during the second time interval in such a way that polypeptide is accumulated in cell culture. Using of such system allows to obtain high levels of protein production and reduces accumulation of some undesirable factors, such as ammonium and/or lactate.
