Abstract

The process for production of thrombin comprises production of fraction of vitamin K-dependent blood plasma proteins with subsequent chromatographic isolation of proserosyme from this fraction, and activation of proserosyme into thrombin. At first concentrate of vitamin K-dependent blood plasma proteins is produced by modified method by sorption thereof on BaSO4 with subsequent elution of K-dependent proteins with 0.05 M tris-HCl buffer solution, pH 7.4. Proserosyme is isolated from the concentrate of vitamin K-dependent blood plasma proteins by separation on Q-Sepharose ("Sigma"), which is balanced with buffer solution and subsequent elution of proserosyme with this buffer solution with addition of 0.4 M NaCl. Immobilization of ecamulin on absorbent BrCN-Sepharose ("SIGMA-AlDRICH") is carried out, the produced absorbent ecamulin-Sepharose is standardized according to chromogenic substrate of kallikrein S2302.
