Abstract
The method for the preparation of the purified avian tuberculin (PPD) comprises the culture of the proteinogenous strain of M. avium in the synthetic culture medium, the inactivation of the bacterial mass, the filtration, the separation of the culture filtrate, the precipitation of the protein by trichloroacetic acid (final concentration of 6%), and the isolation of the final product. The culture filtrate is concentrated on the membrane retaining the proteins of 15 kDa. The product is additionally purified by ammonium sulfate pH 7.0-7.2 and sterilized in Sartorius apparatus.
