Abstract
A process for the preparation of hexapeptide of formula Tyr-d-Ala-Gly-Phe-Leu-Arg by peptide synthesis in solution provides for stage-to-stage peptide elongation by connection in specified sequence of protected or unprotected amino acids or peptides followed by removal of provisional protective groups after each stage of connection, isolation and purification of intermediate peptides, repeating the process up to obtaining completely protected hexapeptide, removal of all protective groups, isolation and chromatographic purification of final peptide. In the peptide elongation process intermediate peptides of general formula Вос-AA...AA-CPG, wherein АА...АА is a chain of from two to six adjacent amino acid residues from the sequence Tyr(SPG)-d-Ala-Gly-Phe-Leu-Arg(SPG), wherein Tyr(SPG)=Туr, Tyr(Bzl), Arg(SPG)=Arg, Arg(NO), CPG=OMe, Onbz. One of protective groups Boc or CPG can be absent. Combination of protected amino acids or peptides is carried out by carbodiimide method, and addition of unprotected arginine by means of diphenyl phosphoryl azide. The purification of intermediate peptides is carried out by extraction , recrystallization or, if necessary, by chromathography. The purification of the final peptide is carried out by recrystallization and by methods of reversed-phase chromatography, ion chromatography or by combination thereof.
