Abstract

The method for determining the total number of non-sulfated glycosaminoglycans includes the preparation of solution of alcian blue, introducing of sample and solution into each cuvette, incubating and recording of colour reaction at 480 nm, creation of calibration curve. The analysis is performed in non-sterile plates, in the holes of which it is introduced per 20 mcl of test biological material. Simultaneously in appropriate calibration holes it is introduced highly purified hyaluronic acid in appropriate dilutions as standard. Then at the same time per 200 mcl of dye solution are added to all holes. It is performed the incubation for 15 minutes and with a microplate reader it is defined the optical density at a wavelength of 480-492 nm.
