一種蝦過敏原的用途,係用以診斷一受試者是否具有對蝦過敏的可能性,其中,該蝦過敏原為丙酮酸激酶,且具有如SEQ ID NO:1所示之胺基酸序列。本發明另關於藉由該蝦過敏原診斷受試者是否對蝦過敏的蝦過敏的診斷方法。

Description

蝦過敏原的用途及蝦過敏的診斷方法
本發明係關於一種過敏原的用途,尤其是一種診斷受試者是否對蝦過敏的蝦過敏原的用途。本發明另關於藉由該蝦過敏原診斷受試者是否對蝦過敏的蝦過敏的診斷方法。
食物過敏(food allergy)是食物安全中的一個重要的議題,其可能會造成發癢、舌頭腫脹、嘔吐、腹瀉、蕁麻疹、呼吸困難或低血壓等症狀。一般而言,只能夠藉由減少與過敏原接觸的機會,來預防食物過敏的發生,而在台灣,蝦蟹類為最主要的食物過敏原,有鑑於此,若是能夠提供一種蝦過敏原,藉此能夠用以診斷一受試者是否具有對蝦過敏的可能性,確實能夠提供該受試者飲食生活的參考,以提升國人的生活品質。
為解決上述問題,本發明的目的是提供一種蝦過敏原的用途,係用以診斷受試者是否對蝦過敏者。
本發明的另一目的是提供一種蝦過敏的診斷方法,係藉由前述蝦過敏原診斷受試者是否對蝦過敏者。
本發明的蝦過敏原的用途,係用以診斷一受試者是否具有對蝦 過敏的可能性,其中,該蝦過敏原為丙酮酸激酶,且具有如SEQ ID NO:1所示之胺基酸序列。
據此,本發明的蝦過敏原的用途,係藉由該蝦過敏原能夠與對蝦過敏的患者的免疫球蛋白E共同形成該複合物之特性,進而能夠應用於診斷該受試者是否為對蝦過敏的患者,為本發明之功效。
本發明的蝦過敏的診斷方法,包含:自一受試者取得一血清樣品;及使該血清樣品於體外接觸一蝦過敏原,該蝦過敏原為丙酮酸激酶,且具有如SEQ ID NO:1所示之胺基酸序列;其中,若該血清樣品中存在一免疫球蛋白E,且該免疫球蛋白E能夠與該蝦過敏原共同形成一複合物,則表示該受試者具有對蝦過敏的可能性。
據此,本發明的蝦過敏的診斷方法,係藉由使用如上述之蝦過敏原,進而能夠精確地診斷該受試者是否為對蝦過敏的患者,為本發明之功效。

為讓本發明之上述及其他目的、特徵及優點能更明顯易懂,下文特舉本發明之較佳實施例,並配合所附圖式,作詳細說明如下:本發明之蝦過敏原係為蝦的丙酮酸激酶(pyruvate kinase), 其具有如SEQ ID NO:1所示的胺基酸序列,藉由該蝦過敏原具有能夠與對蝦過敏的患者的免疫球蛋白E共同形成一複合物之特性,因此能夠應用於一蝦過敏的診斷方法,藉此診斷一受試者是否為對蝦過敏的患者。
詳而言之,於該蝦過敏的診斷方法中,係可以自該受試者取得一待測樣品,該待測樣品可以為一血清樣品。
接著,可以使該待測樣品於體外(ex vivo)接觸該蝦過敏原,此時,若是該蝦過敏原與該待測樣品中的免疫球蛋白E共同形成該複合物,則可以表示該受試者具有對蝦過敏的可能性。舉例而言,可以藉由二級抗體,利用ImmunoCAP或西方墨點法等方法偵測該複合物。
為證實本案之蝦過敏原確實能夠應用於該蝦過敏的診斷方法,藉此診斷該受試者是否為對蝦過敏的患者,遂製備一蝦萃取物,係取5公克之白蝦(whiteleg shrimp,Litopenaeus vannamei)混合25毫升之磷酸鹽緩衝生理鹽水緩衝液(PBS buffer,配方如第1表所示),於4℃下均質(homogenize)1分鐘,離心(7,000g,30分鐘,4℃)以取得一過濾液後,再將該過濾液離心(15,000g,15分鐘,4℃)以取得一上清液,該上清液即為該蝦萃取物。

接著,自七名已被診斷為對蝦過敏的患者(患者1~7)取得各 患者的血清樣品,及自未曾被診斷為對蝦過敏的患者取得血清樣品作為一控制組血清樣品(C)。
以十二烷基硫酸鈉聚丙烯醯胺凝膠電泳(SDS-PAGE)分離該蝦萃取物,轉染至一聚偏氟乙烯膜(PVDF membrane)之後,分別與被診斷為對蝦過敏的患者(患者1~7)的血清樣品,以及該控制組血清樣品(C)進行反應,其如第1圖所示,患者1~7的血清樣品都於63kDa的位置存在一條帶(band),而控制組血清樣品(C)則不存在此一條帶。
接著,將於63kDa的位置之條帶挖下,依序進行胰蛋白酶消化反應(trypsin digestion)及胜肽萃取反應(peptide extraction)之後,以質譜法(mass spectrometry)進行分析,結果如第2a、2b圖所例示,蛋白質序列分別為VLVVPSTGPTASIVK及IGHSYPVAIALDTK,將前述分析結果與美國國家生物技術信息中心後生動物蛋白質序列資料庫(NCBI metazoan protein sequence database)進行比對後可以確認,於63kDa的位置的蛋白質如有如SEQ ID NO:1所示之序列,即為本發明之蝦過敏原。
綜上所述,本發明的蝦過敏原的用途,係藉由該蝦過敏原能夠與對蝦過敏的患者的免疫球蛋白E共同形成該複合物之特性,進而能夠應用於診斷該受試者是否為對蝦過敏的患者,為本發明之功效。
再者,本發明的蝦過敏的診斷方法,係藉由使用如上述之蝦過敏原,進而能夠精確地診斷該受試者是否為對蝦過敏的患者,為本發明之功效。
雖然本發明已利用上述較佳實施例揭示,然其並非用以限定本發明,任何熟習此技藝者在不脫離本發明之精神和範圍之內,相對上述實施例進行各種更動與修改仍屬本發明所保護之技術範疇,因此本發明之保護範圍當視後附之申請專利範圍所界定者為準。

  1. 一種蝦過敏原的用途,係用以診斷一受試者是否具有對蝦過敏的可能性,其中,該蝦過敏原為丙酮酸激酶,且具有如SEQ ID NO:1所示之胺基酸序列。
  2. 一種蝦過敏的診斷方法,包含:自一受試者取得一血清樣品;及使該血清樣品於體外接觸一蝦過敏原,該蝦過敏原為丙酮酸激酶,且具有如SEQ ID NO:1所示之胺基酸序列;其中,若該血清樣品中存在一免疫球蛋白E,且該免疫球蛋白E能夠與該蝦過敏原共同形成一複合物,則表示該受試者具有對蝦過敏的可能性。
