Description

II. 具體實施方式
PRAP1最初被鑑別為於妊娠中期至晚期在子宮中特異性表現的蛋白,因此被稱為妊娠特異性子宮蛋白(pregnancy-specific uterine protein,PSUP)。之後,PRAP1被發現也有在腸道上皮中表現,且在小腸近端(proximal small intestine)大量表現,且其沿著十二指腸-迴腸軸線(duodenum-ileum axis)有一遞減的表現梯度,在盲腸或結腸中幾乎無法檢測到。PRAP1之人類同源物的過表現將在某些上皮和肝癌細胞株中引起細胞生長抑制。此外,據信PRAP1有涉及DNA損傷後的p53依賴型細胞存活。然而,PRAP1的生理功能仍尚未明瞭。
本揭示內容至少部份基於以下發現,亦即PRAP1藉由與微粒體三酸甘油酯轉移蛋白(microsomal triglyceride transfer protein,MTTP)交互作用來促進脂質吸收和apoB的組裝和分泌,而且PRAP1之缺失與突變會減少個體血漿中磷脂與TG的量。因此,引入修飾的PRAP1等位基因或多肽提供治療與高脂血症有關及/或由其引起的疾病或病症的潛在手段。
因此,本揭示內容是一種帶有弱化或摘除PRAP1活性之遺傳上修飾小鼠模型,該遺傳上修飾的小鼠模型在篩選治療高脂血症之藥物候選物的用途、藉由使用修飾的PRAP1多肽與包含修飾的PRAP1多肽的藥學組成物來治療高脂血症或高脂血症相關疾病的方法。
II-1. PRAP1 剔除(KO)小鼠

基因。修飾PRAP1多肽包含與野生型(WT)PRAP1多肽一樣的胺基酸序列,不同之處僅在於修飾PRAP1多肽的胺基酸序列有一個胺基酸修飾。具體而言,與WT PRAP1多肽相比,修飾PRAP1多肽之胺基酸修飾為麩胺酸 (E)被纈胺酸(V)所置換,即,在WT PRAP1多肽中特定位置的一個「E」殘基,在修飾PRAP1多肽的相應位置處被換成一個「V」殘基。

如表1統整之,同功型CRA a具有序列編號:1的胺基酸序列。如此,修飾PRAP1多肽具有序列編號:1的胺基酸序列,其在88殘基處具有一個由V殘基置換E殘基的置換(E88V)。在本揭示內容中,帶有E88V突變之修飾PRAP1多肽被命名為「小鼠E88V突變體PRAP1」,並具有序列編號:2的胺基酸序列。

產生基因敲入小鼠的方法為本領域中具有一般知識者所習知。根據本揭示內容的部分實施方式,是透過CRISPR/Cas技術產生E85V敲入小鼠。具體而言,包含該突變的核苷酸序列被與PRAP1-
靶向之小嚮導RNA(small guide RNA,sgRNA)及Cas9 mRNA共同注射進入小鼠胚胎(例如,原核期之單細胞胚胎),藉此產生該E85V敲入小鼠。
或者,如本文所揭露,在PRAP1多肽的遺傳突變(在同功型中之E85V或其等效物)可以在遺傳上突變的小鼠被有條件地敲入。本文所使用之「有條件敲入」意指該突變的PRAP1
基因只會表現在特定器官或組織。例如,突變的PRAP1
基因專一性地表現於遺傳上修飾小鼠的腸道(例如,在腸上皮細胞(intestinal epithelial cells,IECs)),而正常的PRAP1則表現與其他器官與組織。舉例來說,有條件之遺傳修飾的小鼠可以是可藉由建構一個可受四環素誘導的啟動子或一發育特定之啟動子以驅使PRAP1

本揭示內容的轉基因小鼠,包含PRAP1-/-
小鼠與E85V敲入小鼠,可用於各種目的。舉例來說,該轉基因小鼠可做為用於研究血脂異常(dyslipidemia)相關疾病以及/或是鑑別用於治療血脂異常及/或血脂異常相關疾病之候選藥物的疾病模型。
根據部分實施方式,該轉基因小鼠或自其所取得之細胞可以用於鑑別用於治療血脂異常及/或血脂異常有關之疾病的藥物。根據本揭示內容之任何態樣或實施方式,藉由使用轉基因小鼠來篩選用於治療血脂異常相關之疾病之藥物候選物的方法包括:
(a) 對轉基因小鼠投予一或多種候選藥物;以及
(b) 由一或多種候選藥物中選擇藥物候選物,其中該藥物候選物改善轉基因小鼠與血脂異常相關之疾病有關的表現型。
根據部分實施方式,血脂異常相關的疾病為動脈粥狀硬化症、高脂血症、高三酸甘油脂血症或高膽固醇血症(hypercholesterolemia)。在該等實施方式中,以HFD或西方飲食(Western diet)餵食轉基因小鼠。HFD是指包含60%的能量是來自脂肪的飲食,例如,TESTDIET®

亦可以是,血脂異常相關之疾病為低脂血症(hypolipidemia)、低三酸甘油酯血症(hypotriglyceridemia)或無β脂蛋白血症(abetalipoproteinemia)。如前述,相較於WT對照組小鼠,本揭示內容之轉基因小鼠展現較低的血漿TG含量與磷脂含量,因此可以作為動物模型以鑑定用以增強脂質吸收及/或增加血漿TG與磷脂含量的候選藥物,藉以減輕及/或改善血脂異常相關疾病之有關症狀。可以在投予藥物之前、投予藥物期間與投予藥物之後監測與疾病相關症狀的發展與變化。與疾病相關之一或多種症狀的減輕或改善表明該候選藥物對抗該疾病的潛在治療效果。舉例來說,藥物候選物可能會增加轉基因小鼠的脂肪組織;增加轉基因小鼠的血漿脂質、磷脂、TG、apoB48及/或apoB100的含量;及/或減少在轉基因小鼠之糞便中脂質或卡路里的含量。
候選藥物可以是任何形式的分子,例如,蛋白質、多肽、胜肽、核酸、寡核苷酸、脂質、醣類、多醣或小分子。在部分例子,候選藥劑是小分子化合物,其具有小於2,500道爾吞的分子量。
候選藥物可由多種來源獲得,包含合成或天然化合物的資料庫。例如,許多手段可用於多種有機化合物和生物分子的隨機和定向合成,包括隨機寡核苷酸和寡肽的表現。抑或是可取得或能輕易產生之細菌、真菌、植物和動物萃取物形式的天然化合物資料庫。此外,天然或合成產生的資料庫和化合物可通過常規化學、物理和生化手段輕易地修飾,並可用於產生組合資料庫。
較佳地,前述篩選將包括負對照組(例如,不含化合物)和正對照組(例如,已知對疾病有治療作用的化合物)。
II-4. 高脂血症或高脂血症相關疾病的治療方法
本揭示內容還關於一種治療高脂血症或與高脂血症相關之疾病的方法。該方法包含對個體投予一有效量的PRAP1抑制劑、修飾的PRAP1多肽或編碼修飾的PRAP1多肽之核酸。
取決於期望目的,PRAP1抑制劑可以是反義寡核苷酸、PRAP1靶向之小干擾RNA,或對PRAP1的表現及/或活性表現出抑制或壓制作用的任何其他分子。
在本揭示內容中,修飾的PRAP1多肽包含與WT PRAP1多肽相同的胺基酸序列,除了在其胺基酸序列中有一胺基酸修飾之外。具體來說,相較於WT PRAP1多肽,修飾的PRAP1多肽的胺基酸修飾是以纈胺酸(V)置換麩胺酸(E),即,在WT PRAP1多肽特定位置的「E」殘基,在修飾的PRAP1多肽的對應位置被改變成為「V」殘基。

材料及方法
產生PRAP1剔除(PRAP1-/-
)與E85V敲入小鼠
為產生PRAP1-/-
小鼠,首先從129/Svj小鼠基因體庫中分離出兩個帶有Prap1
基因體之基因座的重疊基因體片段,並用於構建靶向載體。該靶向載體是藉由PCR輔助選殖而建構的,其方式為,在同源重組後,與側位的(floxed)Neo標記(LacZ-Neo卡匣;序列編號:18)連接的LacZ報導基因則被插至PRAP1
基因座的外顯子3(第1A圖)中。將該靶向載體以電穿孔方式送入R1胚胎幹細胞,並使用適當的探針進行南方墨點法以篩選已經在Prap1
基因座同源重組的細胞殖株。將帶有突變等位基因的兩正向細胞殖株:#159及#318顯微注射到C57BL/6小鼠之胚囊中以產生嵌合小鼠。

小鼠被飼養在具有12小時光/暗週期(在上午7:00亮燈)的房間中,可以隨意獲取食物和水。除非另有說明,否則在所有研究中使用的雄性小鼠為8至12週齡。所有動物實驗均按照中央研究院實驗動物保護與使用委員會所制定的指導原則進行。
來自細菌之重組PRAP1的誘發突變、生產與純化
pGEX4T-1-PRAP1△N20是一種細菌表現載體,其主掌缺少N端20個胺基酸之訊號肽(PRAP1△N20)C端融合至麩胱甘肽S-轉移酶(GST)蛋白的PRAP1合成。另外以標準的PCR輔助誘發突變耦合選殖方法(PCR-assisted mutagenesis-coupled cloning method)採用pGEX4T-1-PRAP1△N20為模板,來建構表現多種PRAP1突變體(例如,E38V、E51、54V等)的其他細菌表現載體(也在pGEX4T-1載體中)。pGEX4T-1-PRAP1 21-40、pGEX4T-1-PRAP1 21-76與pGEX4T-1-PRAP1 21-110是額外的細菌表現載體,該些細菌表現載體主掌N端GST標籤之PRAP1突變體的合成,該突變體所涵蓋PRAP1的胺基酸殘基如其名稱所示。
為純化重組PRAP1△N20(在本實施例中稱為WT rPRAP1)與前述各種其他突變體,使用麩胱甘肽SEPHAROSE®
4B珠粒純化細菌產生的GST融合蛋白,並用凝血酶(thrombin)進行切除以移除GST部分,然後藉由添加苯甲基磺醯氟(phenylmethylsulfonyl,PMSF)達到最終濃度為1 mM之PMSF使凝血酶失去活性。除非另有說明,否則本實施例中提及的所有重組PRAP1(無論具有或不具有GST標籤的WT或突變體)均指沒有N端20個胺基酸訊號肽的蛋白質。
小腸脂質吸收之測量
僅在晚上7點至早上7點間提供食物給受測小鼠,為期三天。在第四天晚上7點,對小鼠靜脈注射對體重每公斤500毫克(mg/kg)的泰洛沙泊(以阻礙富含三酸甘油脂(TG)之脂蛋白的水解),隨後胃內推注0.5 ml含有5 µCi之[14
C]-三油酸酯與1 μCi之[3

脂質結合分析
以緩衝溶液A(50 mM、pH7.4的Tris-HCl與150 mM 之NaCl)沖洗與麩胱甘肽SEPHAROSE®
4B珠粒結合之約0.7 μg的GST或GST-PRAP1蛋白(除非另有說明,否則WT或突變體均不含訊號肽)三次,然後重新懸浮於含有所指數量之[3
H]-TG的100 μL緩衝溶液A中。在室溫(room temperature,RT)下反應4小時後,將珠粒以800 μL緩衝溶液A離心沖洗三次,然後重新懸浮於500 μL緩衝溶液E(10 mM、pH 7.4之Tris-HCl、150 mM之NaCl、1 mM之EDTA以及1%TRITONTM
X-100)內,並使用閃爍計數器(scintillation counter)分析保留下來的放射性。
密度梯度超高速離心
以apoB48、MTTP跟PRAP1-WT或E85V表現載體一起對海拉細胞株(在6公分盤中6 × 105
個細胞)共轉染。24小時後,吸出培養基,用4.5 ml富含脂質的培養基(含有1%無脂肪酸的牛血清白蛋白(bovine serum albumin,BSA)、0.8 mM油酸鈉、1 mM甘油與0.05 mg/ml膽固醇的達爾伯克氏必需基本培養基(Dulbecco's modified Eagle medium,DMEM))替換,再培養24小時。收取後,培養基被調整為含有1 mM之PMSF、0.02%之EDTA、0.02%之NaN3
與蛋白酶抑制劑,並且以1200 rpm離心5分鐘以移除細胞碎片。以密度梯度超高速離心分離條件培養基中的含apoB的脂蛋白。簡言之,被固態KBr(0.565 g) 將其密度調整為1.12 g/ml的4 ml培養基,在離心管中依順序被以密度為1.063 g/ml者3 ml、密度為1.019 g/ml者3 ml與密度為1.006 g/ml者的2 ml溶液覆蓋。在以40,000 rpm,在15°C下離心33分鐘後,收集上端的1 ml作為區分部分1。該梯度之後被以密度為1.006 g/ml者2 ml的溶液覆蓋。隨後以40,000 rpm,15°C下離心3小時又30分鐘後,收集上端的1 ml作為區分部分2。以密度為1.006 g/ml之1 ml溶液覆蓋該試管,然後,以40,000 rpm,在15°C下離心17小時。從上端收集其他脂蛋白至12個一毫升的區分部分。接著處理各區分部分的蛋白質,藉由冷的丙酮沉澱蛋白質以移除脂質。被沉澱的蛋白以包含20 mM之Tris、1 mM之EDTA、1%之SDS、pH為7.4的緩衝溶液重新懸浮,隨後進行SDS-PAGE並使用MTTP、apoB與PRAP1專一之抗體的免疫墨點分析。
血漿脂蛋白之分級分離
以快速蛋白質液相層析(fast protein liquid chromatography,FPLC)分離來自指定基因型(300 μl)(實際注射進樣量200 μl)三隻小鼠的合併血漿。在含有0.01%之EDTA與0.02%之NaN3
之磷酸鹽緩衝生理鹽水(phosphate-buffered saline,PBS)中進行洗脫,對每一區分部分收集約0.6 ml。收集區分部分12至31進行脂質和蛋白質分析。在部分狀況,藉由密度梯度超高速離心對來自所指基因型之3至5隻小鼠的合併血漿(0.6至1 ml)進行分級分離。簡言之,用固體NaCl(140.4 mg)將1 ml血漿調整到密度為1.10 g/ml,並在離心管中與3 ml密度為1.1 g/ml之NaCl混合。將由3 ml密度為1.065 g/ml之NaCl、3 ml密度為1.02 g/ml之NaCl和2 ml密度為1.006 g/ml之NaCl所組成的密度梯度依次置於血漿頂部。以40,000 rpm,15°C下離心33分鐘後,收集上端的1 ml作為區分部分1(大乳糜微粒,密度約為0.96 g/ml)。該梯度之後被以密度為1.006 g/ml之1 ml的溶液覆蓋,並再次以40,000 rpm離心3小時及30分鐘(15°C)。收集上端的1 ml作為區分部分2(小乳糜微粒)。以密度為1.006 g/ml的溶液補充該梯度,並再次離心(40,000 rpm,15°C,17小時)。收集上端1 ml作為區分部分3(VLDL大小的顆粒)。收集將其餘的梯度成為10個一毫升的區分部分。
彈式熱量測定法(Bomb calorimetry)
從單獨飼養的小鼠收集過去48小時內的糞便,在60°C下乾燥兩天,均質化並使用半微型氧氣彈式熱量計(semimicro oxygen bomb calorimeter)進行一式三份(每份0.2 μg)的分析。然後計算乾重之每克的能量含量。
糞便脂質含量之測定
從單獨飼養的,已經以HFD餵食兩週的小鼠收集過去2至4天內的糞便。
動物研究
同時對由8週齡起開始HFD的野生型C57BL/6小鼠(對每一群體,n = 10)口服投予rE85V蛋白(序列編號:4;10 mg/Kg,每天一次),或只口服投予賦形物(PBS)。一週後,測量體重增長(相對於初始體重之百分增長率)。在HFD的第5至9天,收集糞便並測定總糞便脂質含量。
統計學
使用t檢定對兩組進行比較,或者使用單因子變異數分析(one-way analysis of variance)(杜凱氏事後檢驗)以軟體比較多個實驗群體,來進行統計分析
實施例 1 PRAP1缺陷小鼠表現出小腸的長度增加
小鼠PRAP1
基因編碼149個胺基酸(amino acid,aa)的富含脯胺酸之酸性多肽,其帶有20個推定為訊號肽的胺基酸。透過使用編碼C端有HA或旗標(FLAG)標籤之PRAP1的表現載體所進行的瞬時轉染分析,則揭露出帶有兩種標籤的分子都被外泌到轉染細胞的培養基中,這顯示PRAP1是外泌的蛋白(數據未呈現)。
為了勾勒出PRAP1的生理功能,藉由傳統的基因靶向(gene-targeting)方法(第1A圖)產生PRAP1 KO小鼠(PRAP1-/-
)。第1B圖中的數據證實PRAP1-/-
小鼠無法表現PRAP1蛋白。同型合子(homozygous)突變體以預期中的孟德爾頻率產生,且牠們肉眼外型上看似正常(數據未呈現)。在正常的飲食中,PRAP1-/-
小鼠增加的體重與對照組小鼠所增加的體重非常相似(數據未呈現)。以PRAP1專一抗體對來自對照組小鼠之小腸切片的免疫組織化學分析顯示,PRAP1蛋白表現在幾乎所有上皮細胞譜系中(數據未呈現)。在剔除小鼠,PRAP1的上皮專一性表現被完全消除(數據未呈現)。藉由使用增生標記物(Ki-67)(數據未呈現)或各種細胞譜系之標記物(數據未呈現)的組織切片染色顯示,PRAP1的缺陷並未顯著影響小腸上皮的整體形態、增生或分化。以溴脫氧尿苷(bromodeoxyuridine,BrdU)對小鼠的脈衝標記(pulse-labeling)也顯示腸上皮細胞(intestinal epithelial cell,IEC)的遷移在對照組與PRAP1-/-
小鼠間沒有顯著差異(數據未呈現)。然而,有趣的是,PRAP1-/-
小鼠的小腸長度被觀察到明顯長於對照小鼠的小腸長度(第1C圖)。
實施例 2 PRAP1與MTTP交互作用並促進MTTP介導之脂質轉移及apoB48外泌
為了探索PRAP1的功能,使用缺乏N端20個aa訊號肽且含GST標籤的重組PRAP1(GST-PRAP1△N20),以及從PRAP1-/-
小鼠之IEC製備的細胞裂解物來進行沉降分析(pull-down assay)。被GST-PRAP1△N20所沉降之蛋白的質譜分析顯示MTTP是與PRAP1交互作用的首要候選物。無論使用PRAP1或MTTP專一性抗體進一步進行免疫共沉澱法(co-immunoprecipitation,co-IP)證實在來自對照組IEC中的PRAP1可以與MTTP在內源性量之下進行免疫共沉澱(第2A圖),而來自E85V敲入突變體小鼠IEC中的PRAP1則無法達成。免疫染色證實,PRAP1與MTTP部分地都位於在腸道上皮的內質網(endoplasmic reticulum,ER)處(以鈣聯結蛋白(calnexin)為標記物)(數據未呈現)。該結果促使檢視PRAP1是否能夠調節MTTP的脂質轉移活性。儘管兩種基因型的腸道細胞中MTTP量非常相似(第2D圖),但第2B與2C圖的數據指出,與來自對照小鼠的細胞相比(比較第2B和2C圖中的第一長條和第二長條),來自PRAP1-/-
小鼠的IEC展現出顯著降低的三酸甘油脂(TG)和磷脂轉移活性。有趣的是,缺乏N端訊號肽(即PRAP1△ N20,此後稱為WT rPRAP1)的重組PRAP1(rPRAP1)可以挽回在PRAP1-/-
IEC中MTTP的TG與磷脂之轉移活性,其中最大活性可以達到使用對照組IEC裂解物(第2B與2C圖)時所觀察到的非常相似之量。藉由使用這樣的分析,辨認出在位置85之麩胺酸(Glu)殘基對於PRAP1促進MTTP脂質轉移功能非常重要,如同在PRAP1△ N20中該殘基的突變為纈胺酸(Val)(E85V)顯著地減弱在活體外(in vitro
)MTTP轉移TG(第2E與2G圖)與磷脂(第2F與2H圖)兩者的活性。
然後,使用表現內源性蛋白質二硫鍵異構酶(protein disulfide isomerase,PDI)的海拉細胞株來檢視PRAP1對MTTP介導之apoB48組裝和外泌的影響。結果顯示出,PRAP1的共表現確實加強MTTP介導的apoB48進到培養基之外泌(數據未呈現)。與活體外分析結果一致,在此以細胞為基礎的分析中,PRAP1的E85V突變體對促進apoB48的外泌展現出較弱的活性(數據未呈現)。透過密度梯度超高速離心對培養基進一步分級分離揭示,在此瞬時表現系統中,當PRAP1在同樣的細胞中與apoB48共表現時(數據未呈現),apoB48被外泌到HDL(高密度脂蛋白,high-density lipoprotein)大小的區分部分。
實施例 3 PRAP1與TG結合並以三元複合物(ternary complex)與MTTP交互作用
接下來,在本實施例中檢驗PRAP1是脂質結合蛋白的假設。第3A與3B圖的數據指出GST-PRAP1△N20(標記為GST-WT)確實以可飽和的方式與3
H標記的TG結合(第3A圖),而且在胺基酸第40個殘基至其C端之間的區域似乎對於TG結合很重要(第3B圖)。值得注意的是,在全長的重組蛋白(標記為PRAP1-FL)中,訊號肽的存在並不會進一步增加PRAP1△N20的脂質結合活性(比較第3B圖中的PRAP1-FL和PRAP1△N20)。有趣的是,雖然保留類似WT之活性的PRAP1突變體(E118-119V突變體)在促進MTTP介導之脂質轉移中表現出與WT蛋白相似的脂質結合特性,但E85V突變體表現出獨特的特性,與WT或E118-119V突變體相比(第3A圖),E85V突變體與TG的結合能力顯著地增加。
使用過量無標記脂質的進一步競爭實驗揭示,磷脂(phospholipid,PL)可以顯著地與TG競爭對於PRAP1的結合(第3C圖),表明PRAP1對於這兩種脂質的結合位點彼此顯著地重疊。值得注意的是,在此分析(第3C圖),並未能觀察到二酸甘油酯(diacylgyceride,DG)、膽固醇(cholesterol)或膽固醇油酸酯(cholesteryl oleate,CE)的顯著或邊際競爭(marginal competition)。
E85V突變體以比WT對照組更強的能力去結合TG(相較於第3A圖中描繪的GST-E85V與GST-WT),但E85V突變體無法促進MTTP的脂質轉移活性(第2E至2H圖)。這些結果顯示出一個可能性,MTTP可能會將結合PRAP1的TG辨認為貨物(cargo),然後將TG轉運至像是apoB的脂質受體,而且,與E85V結合的脂質並不如與WT蛋白結合的脂質那麼地易於被MTTP接近。如果這樣的模型是正確的,一種含有MTTP和負載脂質之PRAP1的三元複合物應該要可以被檢測到,但E85V的突變將大大損害這種三元複合物的形成。為了檢驗此一假設,來自海拉細胞株受免疫沉澱之帶有旗標標籤的MTTP被拿來與重組WT(GST-WT),或與有預載、或與無預載增加劑量的3
H標記之TG的PRAP1之E85V突變體(GST-E85V)結合。此分析包括在上述各種分析中表現類似於WT蛋白的PRAP1之E118-119V突變體。第3D圖的數據指出,相較於在含有GST-E85V的反應中,在含有GST-WT或GST-E118-119V的結合反應中,顯然有較多3
H標記之TG與帶有旗標標記之MTTP共沉澱。而且,有脂質(在分析條件下為40至80 pmol的3
H-TG)時,相較於GST-E85V,顯然有較多GST-WT或GST-E118-119V與帶有旗標標記之MTTP共沉澱,但沒有脂質時卻並非如此(第3E圖)。與此結果一致,MTTP抗體可以與內源性WT免疫共沉澱,但無法與在IEC裂解物中PARP1之E85V突變體免疫共沉澱(第2A圖)。
綜上,該等結果支持PRAP1與TG交互作用的模型,在MTTP轉移反應期間,此交互作用讓TG更可被MTTP接近。
實施例 4 PRAP1在小鼠促進脂質吸收、apoB的組裝與外泌
在此實施例,檢視PRAP1是否與小鼠的脂質吸收有關。第4A與4B圖的數據表明,推注脂質的口服投予後兩小時,與對照組小鼠相比(第4A圖),在PRAP1-/-
小鼠的小腸組織中檢測到明顯更多的脂質,然而,相比於對照組小鼠(第4B圖),PRAP1-/-
小鼠在血液中可檢測到的來自[14
C]-三油酸酯之放射性則低得多。與此結果一致,在投予脂質4小時後,從PRAP1-/-
小鼠收集的血漿不如從對照組小鼠收集的血漿那麼地乳白色(第4C圖),表明PRAP1缺陷降低脂質吸收。
接下來,由於MTTP對於含有apoB之脂蛋白的生物合成是必要的,因此檢驗PRAP1是否會影響apoB的組裝以及從兩種主要生產apoB之細胞類型的外泌。短暫標記與放射標記實驗揭示,相比於對照組小鼠(第4D圖),來自PRAP1-/-
或E85V突變體小鼠之經純化所得之初級IEC中,新合成之apoB48的合成與外泌有顯著的降低,儘管PRAP1缺陷或E85V突變並沒有顯著影響apoB之mRNA的表現(數據未呈現)。在使用初級肝細胞的實驗中觀察至類似的結果,與對照組細胞相比,由PRAP1-/-
和E85V突變體之肝細胞合成和外泌的apoB48和apoB100明顯更少(第4E與4F圖)。
接下來,檢查小鼠之血漿中apoB的含量與脂質化,其中小鼠被禁食16小時,然後被靜脈注射泰洛沙泊(一種脂蛋白脂肪酶抑製劑),並被胃內推注含有因特立滋(Intralipid)與玉米油的脂質乳劑。使用FPLC對投予脂質4小時後之小鼠血漿的分級分離揭示,相較於來自對照組小鼠血漿之相同區分部分(圖5A至5C),明顯較少的apoB48和apoB100分布在PRAP1-/-
小鼠血漿之富含CM/VLDL的區分部分(圖5A至5C中區分部分14至16)。以密度梯度超高速離心從禁食16小時的小鼠中進一步分離血漿顯示,PRAP1缺陷會稍微減少apoB的脂化與外泌到肝的VLDL中(數據未呈現),表明肝臟中很低的PRAP1量仍然對MTTP介導的apoB組裝與外泌有所影響。使用來自投予脂質3小時後之小鼠血漿的額外之類似分級分離實驗表明,E85V突變體小鼠展現出與PRAP1-/-
小鼠非常類似的表現型,即,與對照血漿相比(第6A至6D圖),明顯較少的apoB48、apoB100與TG分布在PRAP1-/-
及E85V突變體小鼠血漿之富含CM/VLDL的區分部分(區分部分1至3)。
值得注意的是,在第5圖與第6圖所呈現的實驗中,在投予脂質3-4小時後小鼠的血漿中很容易檢測到PRAP1。如果以免疫墨點分析法分析更多的血漿(數據未呈現),則可以檢測到0小時此時間點(禁食4小時後)的基礎量。有趣的是,儘管通過免疫組織化學分析很容易在腸道上皮中檢測到E85V突變體蛋白(數據未呈現),但與WT蛋白相比,它較少被外泌到血流中或具有較慢的外泌動力學(數據未呈現)。
這些數據證明,PRAP1缺陷降低脂質吸收,因此,與對照組小鼠相比,在PRAP1-/-
小鼠和E85V突變體小鼠的血漿中檢測到較低量的apoB48、apoB100和TG。
實施例 5 PRAP1-/-
和E85V突變體小鼠的體重和脂肪量均增加得較少,並且在高脂飲食中可預防肝脂質變性
儘管以飼料餵食,對照組與基因剔除小鼠的體重增加非常相似(數據未呈現),但在HFD下(TESTDIET®
58Y1中脂肪的能量佔60%;表3中概述該等成分)與對照組小鼠相比,PRAP1-/-

考慮到對照組和PRAP1-/-
小鼠在HFD下的食物攝入非常相似(數據未呈現),那麼對它們的能量消耗和糞便脂質含量進行比較。兩組小鼠展現出非常相似的能量消耗,如綜合實驗室動物監測系統(Comprehensive Laboratory Animal Monitoring System,CLAMS)所示(數據未呈現)。然而,相較於對照組小鼠的糞便,PRAP1-/-
小鼠的糞便含有較多的脂質(第8A至8B圖;第8A圖:以飼料餵食小鼠;第8A圖:以HFD餵食小鼠)。彈式熱量測定法也揭示,PRAP1-/-
小鼠的糞便比對照組小鼠的糞便保留更多卡路里,這顯示在HFD下PRAP1-/-
小鼠的較瘦表現型主要是由於脂質吸收缺陷所致。
有趣的是,E85V突變體小鼠在許多方面表現出與剔除小鼠非常相似的表現型。例如,E85V突變體小鼠的小腸始終比同窩對照組的小腸長(第9A圖,以飼料餵食)。餵食管口服脂質後,與對照組小鼠相比,E85V突變體小鼠的小腸組織中積累更多的脂質(數據未呈現)。此外,在12週HFD下,與對照小鼠相比,E85V突變體小鼠的體重增加得顯著較少(第9B圖),而且展現出較小的脂肪組織(iWAT與eWAT)(圖9C)和較低的血漿TG量(第9D圖)。
最後,檢查PRAP1缺陷或E85V突變對HFD誘導之肝臟脂質堆積(肝脂質變性)的影響。對於該實驗,在對小鼠的肝臟進行處理並分析形態和脂質含量之前,先以HFD餵食12週(第10A至10C圖)或8週(第10D與10E圖)。結果顯示,與對照組相比,在PRAP1-/-
和E85V突變體小鼠肝臟中,脂滴(以油紅色O染色,第10D圖)、帶空泡的肝細胞(以H&E染色,第10A圖)、肝臟對體重的比例(第10B圖)與肝的TG含量(第10C與10E圖)均顯著降低,表明PRAP1缺陷或E85V突變減少HFD所引起的肝脂質變性。
實施例 6 重組E85V多肽(rE85V)在減少脂質吸收的作用
鑑於在脂質吸收方面,E85V突變體小鼠表現出與PRAP1剔除小鼠相似的表現型,在本實施例中檢視rE85V的投予是否會影響體重增加和以HFD餵食之小鼠的脂質排泄。
第11A和11B圖的數據表明,在HFD下,與僅餵食賦形物(PBS)的小鼠相比,口服投予rE85V(10 毫克/公斤/天)的小鼠體重增加顯著較少,且糞便中的脂質排泄趨向增加。這些結果證明,rE85V多肽的投予顯著地降低脂質吸收,因此,提供治療高脂血症或高脂血症相關疾病的潛在手段。
