Abstract

DC60 This invention describes a method for generating specific and purified antibodies by inducing recombinant cells to produce antibodies specific to proteins in all types of alpha-thalassemia carriers. These antibodies are then used to create a diagnostic kit for alpha-thalassemia carriers using a colorimetric method. This method can be used to detect all types of alpha-thalassemia carriers in healthy individuals.

Claims (5)

1. Recombinant cells with the ability to produce antibodies specific to alpha-thalassemia carriers and capable of rapid division. These recombinant cells are generated by isolating and purifying the protein to be cultured against alpha-thalassemia carriers. The B cells of experimental animals are then induced to produce antibodies. These B cells are then combined with rapidly dividing cells to obtain recombinant cells with the ability to produce antibodies specific to alpha-thalassemia carriers and capable of rapid division. 2. Antibodies specific to all types of alpha-thalassemia carriers. Recombinant cells with the ability to produce antibodies specific to alpha-thalassemia carriers and capable of rapid division are cultured in the peritoneal cavity of experimental animals. The resulting recombinant cells are then purified and specificized. 3. Method for detecting all types of alpha-thalassemia carriers by staining a strip. A strip coated or dropped with the antibody is then applied. Specific to alpha-thalassemia carriers, allow the strip to dry. React it with a solution that prevents non-specific antibid reactions. Rinse the strip in buffer solution to remove excess reaction substance. React the strip with the specimen, then rinse in buffer solution to remove excess specimen. React the strip with a solution that binds to the alpha-thalassemia carrier-specific protein. Rinse off excess reaction with buffer solution. React the strip with an enzyme solution that reacts with the solution that can bind to the alpha-thalassemia carrier-specific protein. Rinse off excess reaction with buffer solution. React the strip with a color-forming solution, then rinse with water to make the color on the strip clear. 4. Ready-to-use alpha-thalassemia carrier diagnostic kit. The ready-to-use alpha-thalassemia carrier diagnostic kit consists of: 1. A strip coated or injected with purified antibid. And specific to alpha-thalassemia carriers. 2. A solution that prevents nonspecific reactions to the antibides on the strip. 3. Buffer solution for rinsing away excess residue from the reaction, along with a container. 4. A solution that reacts with the specific protein of alpha-thalassemia carriers. 5. An enzyme solution that reacts with the specific protein of alpha-thalassemia carriers. 6. A solution or powder that produces a colorimetric effect. 7. A solution or powder for controlling positive and negative results. 8. A buffer solution for diluting the specimen. 9. A dropper and container for holding the diluted specimen, ready to react with the strip. This container has a capacity of at least 96 specimens. 10. A container with the user manual for the ready-to-use alpha-thalassemia carrier diagnostic kit. 5. Pre-assembled Alpha Thalassemia Carrier Diagnostic Test Sheet: This pre-assembled alpha thalassemia carrier diagnostic test sheet consists of small membrane pieces attached to a solid strip. The membrane pieces protrude evenly from the solid strip. Each membrane piece is of the appropriate size, shape, and spacing to fit fully and properly into the specimen container, ensuring complete contact between the specimen and the membrane.
