Abstract
This invention describes an electrochemical method for measuring the urine albumin-creatinine ratio (UACR). The amount of albumin and the UACR ratio can be used to estimate the amount of albumin and creatinine excreted in the urine by the kidneys within 24 hours. A UACR value greater than 30 mg albumin/g creatinine can be used as a preliminary indicator of kidney problems in a patient. This invention reveals a simple and user-friendly sample preparation and measurement method for measuring UACR in urine. It does not require expert knowledge while maintaining high accuracy and precision. The electrochemical method for measuring urine albumin using linear sweep voltammetry consists of a screen-printed probe with two electrodes. The electrode consists of a silver working electrode and a silver/silver chloride (Ag/AgCl) reference electrode. The chemicals used to control pH changes during UACR measurement from clinical urine samples are a 0.4 M Britton-Robinson (BR) buffer solution with a pH of 2.0. The optimal ratio of urine sample to BR buffer is 7:60 µL. The optimal electrolyte solution is artificial urine with a pH range of 4.5 - 8.0, which is the pH of human urine. The conditions used for linear sweep voltammetry electrochemical measurement are a step potential of 0.005 volts and a scan rate of 50 millivolts per second in the potential range of -0.8 to 0.8 volts.
