Description

The subject of the invention is a method of sampling living tissue for microscopic examination, i.e., histological preparations without the use of a cover glass.

д.Gum syrup, glyceroline-gelatin, glycerol-leukemia, etc., are used in transparent media to conclude histological sections stained in the usual way, and in all cases the use of a cover glass is required. However, these substances have drawbacks: in glycerin-gelatin, preparations quickly fade, and in gumsyrope, glycerin-levulosis, fermentation, rotting, etc. can occur.

The proposed method eliminates the indicated disadvantages. In the preparation of histological preparations stained with fat, a solution of polyvinyl alcohol is used, and in other cases polyvinyl butyral is used, and in both cases no cover glasses are required. In addition, the proposed method makes it possible to manufacture large histotopographic preparations, i.e. sections through the entire organ, for example, one of the walls of the heart.

The method consists in the following: A 10–12% solution of polyvinyl alcohol is preliminarily prepared, for which the powder of polyvinyl alcohol is washed in 96 ° alcohol, then in ether and dried. After that, polyvinyl alcohol is dissolved in distilled water. Next, a cut stained for fat is placed on a glass slide and a few drops of a prepared solution of polyvinyl alcohol are applied to it, which after 6-8 hours. dries to form a durable smooth film, which encloses the cut. For the conclusion of sections stained by other methods, a 12-15% solution of polyvinyl butyral in carbol xylene or alcohol xylene is prepared. In the future, in the manufacture of the drug come the same way as in the conclusion of sections stained for fat.

A method of making histological preparations with the conclusion of sections in transparent solidifying masses, characterized in that the sections which have been cut and processed in the usual way are filled with polyvinyl butyral, and when stained for fat, with polyvinyl alcohol.
