Claims (1)

Журнал Лабораторное дело, 1978, № 5, с. 298-300 (прототип).With three C-GLYCINES SM electrode buffer pH 8.3. 200 µg of protein are applied to each tube with a 0.5 x 8 cm separating gel. Electrophoretic separation is carried out at a current of 5 mA per tube for 70 minutes at a temperature of 4 ° C. Proteins are stained with 1% amidoshvarts and washed from excess paint 7% acetic acid. The distribution of the protein fractions in the gel is schematically transferred to millimeter paper and the relative mobility to the bromophenol dye front is calculated. Molecular weights are determined by the Shapiro method on a 7.59) -nrm gel containing 0.1% Na dodecyl sulfate. Gels for determining molecular weights are calibrated in a range from 147,000 to 16,000. A quantitative assessment of the protein content of the electrophoregram fraction is carried out by densitometry of polyacrylamide gels on a spectrophotometer at 610 mmk. Absolute amounts of protein at den simetry are expressed by the conditional-weight method. The electrophoretic spectrum of soluble proteins of the decidual tissue consists of 13 fractions, which include a specific protein fraction with a molecular weight of 52,000. For example, DLt of pregnancy in a woman being examined take 1-2 ml of blood and get syvorochka. In the central well of the agar block, O, 20-0.25 l of antisera to specific proteins of the decidual tissue are placed. For control, 0.2 ml of phosphate buffer is placed in one of the surrounding wells: pH 6.4. Samples of the blood of the women examined are placed in other wells. The positive reaction of precipitation of evidence is yeah: the presence of the corresponding antigens in the blood and, therefore, the presence of pregnant. We examined 30 women. A positive reaction was detected in 24 women in whom pregnancy was confirmed during the observation process. In 6 women, the results were negative; their lack of pregnancy was established in subsequent examinations. The proposed method is strictly specific, since decidual tissue is formed in the body of a pregnant woman earlier. than placental tissue, therefore, the reliability and accuracy of pregnancy diagnosis at earlier periods is significantly increased. Claims of the Invention A method for diagnosing early pregnancy by carrying out an immunochemical reaction of a specific antistopus with a blood sample under investigation, characterized in that antisera against specific proteins of the decidual tissue is used as a specific antisera to increase the diagnostic specificity. .
