Description

The invention relates to the field of analytical chemistry, in particular to the polarographic determination of oxygen, and concerns calibration, i.e., determining the sensitivity of a polarographic cell using an oxygen method, allows the absolute value of the oxygen content in an aqueous solution to be calculated using measured values of cell currents and can be used in the study of photosynthesis and respiration in suspensions of microorganisms and cell organelles, as well as in all those cases where continuous measurement of concentration is necessary oxygen in aqueous solutions. Izvesti is a method for calibrating polar graphical oxygen cells using reduced nicotinamide adenine dinucleotide (NADH) as the titrant. The interaction of the added reducing agent with dissolved oxygen is carried out in the presence of a mediator - phenazine methosulphate (PMS) and the enzyme - catalase. 1 NADH is produced in the presence of catalase, the activity of which varies during storage. Changes in enzyme activity can affect the titration results. In order to simplify the process and increase the reliability of calibration, it is proposed to use an ascorbic acid solution as a titrant, and titration is carried out at a pH of the reaction medium equal to 10-12. The method is carried out in the following manner. In a sealed reaction chamber, where the concentration of oxygen dissolved in the medium is continuously measured using a polarographic cell, oxygen is titrated with an exact solution of ascorbic acid. The calibration medium is pre-adjusted to pH 10-12 by adding a few drops of 2N. KOH and saturated with oxygen at the temperature of the experiment. The saturation operation is carried out in a separate vessel by passing a stream
