Штамм Mycobacterium tuberculosis (БК-Ха ков № 841, вариант штамма Vallee типа Bovis, предназначенный дл изготовлени противотубер кулезной вакцины). Изобретение относитс к ветеринарии и пред ставл ет собой новый штамм микобактерий туберкулеза , который может быть использован дл приготовлени противотуберкулезных препаратов вакцины, других антитенов, а также сывороточных препаратов. .Дл профилактики туберкулеза людей во всем мире примен етс жива вакцина из микобактерий туберкулеза штамм БЦЖ/BCG (Ч)В ветеринарии вакцинопрофилактика ВЦЖ как в Советском Союзе, так и большинстве стран мира, в насто щее врем не осушествл етс . Одной из основных причин этого, помимо недостаточной нммуногенности БЦЖ вл етс возникновение продолжительной аллергии у вакцинированных БЦЖ животных, сохран ющейс в течение года и более, и отсутствие пригодных дл практики методов дифференциации поствакшшальных реакций на туберкулин от постинфек ционных. Одним из средств дл улучшени вакцинопрофилактики туберкулеза у людей и создани препарата дл таковой у чсивотных вл етс применение новых, более иммуногенных, вакцинных штаммов микобактерий туберкулеза. Эти штаммы должны быть безвредными и обладать достаточными дл защиты вакцинированных от развити туберкулезного процесса иммунизирующими свойствами, а также не обусловливать возникновени продолжительной аллергии. Целью изобретени вл етс получение штамма микобактерий туберкулеза, отвечающего основным требовани м, предъ вл емым к вакцинным 1итаммам: безвредный, достаточно иммуногенный , чтобы защитить вакцинированных от развити туберкулезного прюцесса. Полученный штамм микобактерий туберкулеза БК-Харьков имеет следующие морфологическую , культуральную и другие биологические характеристики. Морфологические признаки: слегка изогнутые , иногда с утолщени ми на концах, полиморфные , грамположительные палочки, длиной 2-8 мкм и толщиной 0,2-0,5 мкм, типично окращиваютс no- методу Циль-Нильсена, стойко сохран ющие эту окраску при воздействии кислот, и1елочи и спирта, не образующие спор и не имеющие капсулы . Цитоплазма бактерий имеет гомогенный вид и содержит гранулы, микрогранулы и вакуКультуральные свойства: на поверхности жидких питательных сред формируетс рыхла , морщиниста пленка светло-желтого цвета. На плотных питательных средах (Петрова, Виногра-дова ) растет в виде изолированных колоний типа R с образованием пигмента лимонно-желто го цвета. Растет в аэробных услови х. Анаэробиотические свойства не отмечаютс . Оптимальные параметры температуры 37,8-38,0С рН 4,58 ,0. Биохимические свойства: снижена активность каталазы, уреазы и дегидразы. Иммуногенные свойства: индекс защиты БК-Харьков в эксперименте на морских евинках после однократного подкожного введени им 5 мг БК-Харьков и последующего заражени 0,01 мг вирулентной культуры микобактерий туберкулеза штамм Vat lee, а также на тел тах после двукратной внутрикожной вакцинации по 2 мг/кг или трёхкратной подкожной вакцинации в дозах 0,4-0,6-0,8 мг/кг веса теленка с интервалом 5 суток и экспериментального заражени 15 мг подкожно вирулентной культурой штамма № 8, либо естественного заражени после длительного контакта с больными туберкулезом тел тами, достигает 100%. Остаточна вирулентность: незначительна , первичные туберкулезные комплексы не формируютс . Аллергизирующие свойства: аллерги слабо выражена и относительно кратковременна ( у морских свинок до двух, а у тел т до шести мес къ ). Приобретенные штаммом микобактерий туберкулеза БК-Харьков вирулентные свойства оказались стойкими, наследственно закрепленными Реверсионное пассирование в течение 10 лет не обнаружило восстановлени его исходной вирулентной характеристики.
