Abstract

FIELD: biotechnology, biochemistry. SUBSTANCE: method of superoxide dismutase isolation from biomass producer Saccharomyces cerevisiae involves the disruption of cells by freezing-thawing, extraction with 0.04-0.06 M potassium-phosphate buffer, pH 7.7-7.9 at the ratio of wet mycelium and extractant = 1: 3, at 7-10 C for 20-24 hr. Before depleted cell mass removal suspension is acidified to pH 4.4 and removal is carried out by centrifugation. Ion-exchange chromatography of supernatant is carried out on carboxylic cation-exchange resin KMT followed by concentration of eluates by ultrafiltration. Gel-chromatography of prepared concentrate is carried out on Sephadex G-75 followed by lyophilization of the end product. Preparation can be used for treatment of joint pathology, pulmonary diseases. EFFECT: improved method of enzyme isolation. 1 tbl
